“We haven’t really been profitable since COVID, so we’ve been examining how to transition for the last few years. We can’t change the size of our building, our location, or the number of employees, so we have to change course altogether,” says Honn. That new course includes three separate but related ventures: Nikkei, a Creole izakaya opening soon in the Broadside in Mid-City; Eatwell Kitchen
a newly opened fast-casual restaurant in Metairie; and Tempero’s Market Kitchen
a pop-up and events catering business that will revive several Carmo favorites
Honn says the menu at Nikkei is a mix of traditional Japanese dishes like chashu pork
a version of shrimp tempura found in Japan but also as popular street food in Liberdade
a street dish of Japanese-Brazilian origin
“You find pastel carts all over Brazil,” says Honn
“But I’m not sure how well-known it is that Japanese immigrants created it.”
One common thread throughout the menu is “localizing” the dishes through the Honns’ use of Gulf seafood and local produce. After all, it’s what they’re known for. In a report from Eater and Civil Eats

The Holy Father spoke them just over seven months ago during the longest journey of his pontificate to Oceania and the Far East
Cardinal da Silva recalled this moment—and the Pope’s deep affection for East Timor—during an interview with local journalists at Dili Airport on Tuesday
as he prepared to embark on a 12,500 km journey to Rome to attend the funeral of the late Holy Father
The Salesian Cardinal was already well aware of the Pope’s love for his country
When he was appointed cardinal in May 2022
he remarked: “I’m convinced that Pope Francis didn’t offer this to me
but to the Church and the people of East Timor.” And when he traveled to Rome on May 7
he stated during the ceremony at his titular church
Albert the Great in northern Rome: “Making me a cardinal was a gift from the Pope to the youth and people of Timor.”
That appointment made him not only one of the youngest members of the College of Cardinals at the time (a distinction he still holds at age 57)

we asked him how he’d started at Olympiacos on the back of last season’s hugely successful campaign
Stam exclusively told Nottingham Forest News: “Olympiacos made a great effort to keep him this year and finally succeeded with the help of Nottingham Forest
The player missed the entire pre-season and only on August 31st he played for the first time in the red and white again
“If we take stock of the first two months of Carmo back at Olympiacos
I think we have to agree that we expected better things from the centre-back
but he’s certainly not been as good as he was at the end of last season
He needs to figure out how to become a safe bet for the team’s defence again.”
Although his second spell with Olympiacos hasn’t quite started with red-hot form
there are clear strengths when it comes to his defensive game
Stam continued: “He is a 6ft 5in left-footer
has a good long pass on him and is dominant in the aerial challenge.”
we asked Stam if there was a Forest player who he felt Carmo could be deemed similar to and he delivered an intriguing response
He told us: “I’m no expert on Forest but he reminds me of Nikola Milenkovic
We face him in the European final against Fiorentina
I see Carmo as a big-league player and if he rediscovers last season’s form he can play for Nottingham Forest
Stoppers of his size who aren’t slow are not easy to find.”

The Reds might lose Murillo next summer and with Carmo being left-sided

Tg+/− rats displayed a working memory impairment in the Y-maze and higher anxiety levels in the Open Field (OF) at 6 and 12 months of age

Multiple behavioral tasks should ideally be used to characterize the phenotypes of potential animal models of AD since in the clinical setting, a wide range of neuropsychological tests are employed to determine the profile of the cognitive impairment (Gainotti et al., 2014; Webster et al., 2014)
by using a longitudinal assessment procedure
we sought to determine the levels and accumulation of soluble Aβ and iAβ peptides and the development of impairments in the exploratory activity
emotional behavior and working and spatial reference memory in Tg+/− rats
we will test if this model recapitulates some of the biochemical
emotional and cognitive alterations seen in pre-symptomatic AD
Our results could be relevant to define in this animal model the critical window for therapeutic interventions that may be applied to early stages of AD
The study was carried out in strict accordance with the guidelines of the OLAW-NIH (Office Laboratory Animal Welfare)
The protocol was approved by the local Animal Care Committee of Fundación Instituto Leloir (FIL) Assurance# A5168-01
Hemizygous transgenic McGill-R-Thy1-APP (Tg+/−) rats, harboring the human APP751 transgene with the Swedish and Indiana mutation under the control of the murine Thy1.2 promoter (Leon et al., 2010)
were provided to Fundación Instituto Leloir (FIL) by The Royal Institution for the Advancement of Learning/McGill University
An in-house breeding colony was established at FIL by cross-breeding Tg+/− males with wild-type Wistar females rats
Each litter was culled to 10 pups (up to 8 males per litter were left)
dams were removed from the litters and pups were genotyped as described below
When the results of the genotyping were available (between 22 and 23 post-natal days)
rats were pseudorandomly assigned to experimental groups
groups were made up of pups from 3 to 4 different litters
Animals were maintained on a 12-h light/12-h dark cycle
group-housed 2–4 per cage and had ad libitum access to water and a standard rodent diet
Tissue samples were obtained by ear punches from the litters to determine the genotype of each rat
Genotyping was performed using a validated protocol
genomic DNA was prepared from ear biopsy tissue using proteinase K digestion
followed by phenol/chloroform/isoamyl alcohol extraction (Life Technologies)
DNA quantification was determined in a Nanodrop 2000 spectrofotometer
Samples with a ratio of absorbance at 260/280 and 260/230 nm above 1.80 were used for PCR to recognize whether or not the rat carried the human APP (hAPP) gene
The following sets of primers were used: APP-F 5′-AGGACTGACCACTCGACCAG-3′; APP-R 5′-CGGGGGTCTAGTTCTGGAT-3′; CyclophilinG-F 5′- TACAACAGTAGAACAAGGGAGCGAAG-3′ and CyclophilinG-R 5′-ATCCCTCCTTCTTCTCCTCCTATCTTT-3′
Cycling conditions were: 95°C (15 min) and 40 cycles of 94°C (30 s)
72°C (1 min) followed by one step at 72°C (2 min) and kept at 4°C until processed
PCR products were resolved by 2% agarose mini-gel electrophoresis in TBE buffer and visualized with ethidium bromide (10 mg/ml) staining under UV light
Cyclophilin G amplicon (248 bp) was used as a housekeeping gene to verify DNA integrity and PCR conditions
The hAPP amplicon (387 bp) indicates the presence of the human transgene
To verify the hemizygocity of the transgenic rats
quantitative PCR (qPCR) was performed by using the same set of hAPP primers as described above and the following set of GAPDH (used as housekeeping gene) primers: GAPDH-F: 5′-GGGGAAGGACGCTGTACGGG-3′ and GAPDH-R: 5′-AAGGGGAGC AACAGCTGGGGT-3′
A total of 1 ng of genomic DNA was used per well (5 μl of a 0.2 ng/μl dilution)
SYBR-Green qPCR was performed by using the KAPA SYBR® FAST Universal 2X qPCR Master Mix (Kapa Biosystems)
Reactions were run in a Stratagene Mx3005P cycler and results analyzed by the MxPro software in a Comparative Quantitation mode
In all of the runs a genomic DNA sample of a hemizygous animal was included and used as an internal calibrator
the relative quantity (dRn) value should be between 0.8 and 1.3; if the unknown sample is homozygous the result is close to a value of 2 (between 1.8 and 2.3)
Only male rats were used to avoid any potential effects of changes in the female estrus cycle on behavioral performance
Three groups (n = 3/group) of naïve Tg+/− male rats of 3, 6, and 12 months of age, and one group (n = 3) of naïve WT male rats of 12 months of age, were anesthetized with ketamine (50 mg/kg) and xylacine (10 mg/kg), perfused transaortically with 0.9% sodium chloride with 200 U/l heparine and brains were quickly removed as described previously (Leal et al., 2006)
Brains were divided into left and right hemispheres
Hippocampus was isolated from left hemisphere and retained for biochemical studies
The right hemisphere was post-fixed in 4% paraformaldehyde in 0.1 M phosphate buffer (PB)
and finally transferred to a solution of 30% sucrose in 0.1 M PB for 5 days
or until sectioned for immunohystochemistry into 40 μm coronal sections with a freezing sledge microtome (SM 2000R
None of the animals assigned to immunohistochemical and biochemical experiments were submitted to behavioral testing
Immunoreactivity was detected by Western blot using an enhanced chemiluminescence detection system (ECL
Thermo Scientific) and membranes analyzed and quantitated with a STORM 840 Phosphor Imager (GE Healthcare)
Group values were obtained simultaneously and normalized with respect to APP immunoreactivity

All the experimenters were blinded to the genotype of the animals
The EPM test assesses anxiety-like behaviors (Pellow et al., 1985)
The apparatus consisted of a black melamine central square platform (11 × 11 cm) from which four black melamine arms radiate (50 × 11 cm) separated from each other by 90°
Two of the arms (“closed arms”) have a wall (40 cm in height) all around its perimeter but not in the entrance and the other two arms (“open arms”) do not have any wall but with raised edges (0.25 cm) around its perimeter
The maze was elevated one meter from the floor by five legs
Light intensity in the open arms was 80–90 lux
Each rat was placed onto the central platform facing an open arm and allowed to freely explore the maze for 5 min
An arm entry was counted when rat introduced its four paws into an arm
The following dependent variables were recorded: total distance moved

The OF test was used to assess spontaneous exploratory activity and anxiety-related behaviors (Walsh and Cummins, 1976)
The apparatus consisted of a black melamine square (60 × 60 cm) surrounded by walls of 40 cm in height
A central area was arbitrarily defined as a square of 30 × 30 cm and it was drawn over the image of the OF in the video-tracking system
A rat was considered to be into the central area when its four paws were on it
Light intensity in the center of the OF was 70–80 lux
Each rat was placed in the center of the maze and its behavior was analyzed for 5 min

The NORT evaluates recognition memory of one previously explored object (familiar) compared with one novel object (non-familiar) (Ennaceur and Delacour, 1988; Ennaceur, 2010)

Rats were tested during two trials (sample and retention trials) separated by an interval of 1 h during which they were returned to their cages
each rat was faced with two identical objects (called familiar objects) placed in a symmetrical position and the time exploring freely each object was recorded
one of the two familiar objects was replaced by a novel object and the time exploring each was recorded
Sets composed of three copies of the same object were used to prevent odor cues and all combinations and location of objects were used to prevent bias due to preference for a particular object or location
Exploration time was computed when the snout pointed to the object at a distance ≤2 cm
Discrimination index (d1) and discrimination ratio (d2) scores were calculated using the following formulas: d1 = tn − tf and d2=tn−tftn+tf
where tn = the amount of time rats explored the novel object and tf = the amount of time rats explored the familiar object
An arm entry was scored when rat placed the four paws within that arm
The following dependent variables were registered: total number of arm entries
number of triads (sequence of three consecutive visits to different arms)
An alternation was defined as an entry into three different arms on consecutive choices
The percentage of alternation was calculated as the ratio of actual to maximum number of alternations
The maximum number of possible alternations was defined as the total number of arm entries minus 2
A low percentage of alternation is indicative of an impaired spatial working memory because the rat cannot remember which arm it has just visited
and thus shows decreased spontaneous alternation
The apparatus used was previously described in detail by Galeano et al. (2011)
it consisted of a circular galvanized steel pool (180 cm in diameter and 60 cm in height)
filled with water at 22 ± 1°C to a depth of 36–40 cm
and imaginarily divided into four quadrants: North (N)
A cylindrical platform (10 cm in diameter and made of transparent acrylic plastic) was placed 2 cm above (visible escape platform) or beneath the water surface (hidden escape platform) in the center of one of the quadrants
To enhance the visibility of the platform during the cued learning training
a “flag” was attached to the platform
The pool was located in the center of an experimental room with multiple extra-maze visual geometric cues hanging on the wall
Indirect illumination was provided by four spiral compact fluorescent lamp in each corner facing the walls
The following dependent variables were recorded: latency to the platform
the number of crossings over an annulus zone of 20 cm in diameter (defined as a zone surrounding the location where the platform was located during the spatial learning training)
and time spent in the outer ring of the pool (25 cm wide)
During cued learning the platform protruded 2 cm above water surface and a “flag” was attached to it (visible escape platform)
The maze was surrounded by black curtains to minimize the availability of extra-maze cues
For each of the four trials conducted on each day
the platform was moved to a different quadrant and a different start location was used
If a rat had not located the platform before 120 s elapsed
it was gently guided to the platform location and was allowed to remain there for 15 s
Inter-trial interval duration was 20–30 min
Spatial learning and reference memory. We used procedures described previously with some modifications (Vorhees and Williams, 2006; Galeano et al., 2011)
Black curtains were removed and the spatial learning phase of the task was performed over five consecutive days with four trials per day
rats were released into the pool from one of the four starting positions (one per each quadrant) and the order of the sequence was changed pseudorandomly between days
Animals were able to escape from the water using the hidden escape platform that was kept in the same location throughout the five spatial learning sessions
A trial was finished when the animal found the escape platform or when 120 s had elapsed
Rats remained on the platform for 15 s with an inter-trial interval of 20–30 min
Twenty four hour after the last trial of the learning phase
spatial reference memory was assessed with a probe trial in which the escape platform was removed from the pool and rats were released from a new starting position not used during the learning phase
the number of annulus crossings and the time spent in the outer ring of the pool (thigmotactic swimming) was recorded
After each trial rats were dried and when the session finished they were returned to their colony room
For stereological estimations the contours of ventral and dorsal hippocampus showing immunoreactivity for iAβ was drawn in each section with Image J software (NIH, USA) tool to determine the affected area. The Cavalieri's principle (Gundersen and Jensen, 1987) was used to calculate the affected volume by using the following formula: V = ∑AxNxE
where V = volume (mm3); ∑A = sum of area counted (mm2); N = number of sections analyzed; E = thickness of each section (mm)
All data are shown as the mean ± s.e.m
Data were analyzed by Two-Way ANOVA or by Two-Way mixed ANOVA tests followed by Tukey's HSD post-hoc tests for multiple comparisons

Our data strongly suggest a subtle phenotype in Tg+/− rats with an “age-independent” accumulation for SDS-soluble monomeric Aβ40 and iAβ but an “age-dependent” accumulation for SDS-resistant Aβ oligomers

As it was mentioned in the “Materials and Methods” Section
the groups of animals that underwent the behavioral assessment were not the same used to study Aβ accumulation
This effect could be attributable to the levels of illumination used (80–90 lux in the open arms)
To rule out the possibility of a floor effect that could be masking an anxiety phenotype in Tg+/− rats in the EPM
future studies are needed to investigate the behavior of these animals in this test under different experimental conditions
In summary, even if the emergency (Holmes, 2001) or the light/dark box (Pinton et al., 2011) tests were not assessed in this study
our report is the first analysis of the anxiety disturbances in a transgenic rat model of early AD that does not develop Aβ plaques
It is of interest that the anxiety pattern described here for the Tg+/− rats appear to closely mimic neuropsychiatric symptoms described in some pre-clinical AD patients
we did not find in Tg+/− rats any impairment in the learning phase of the MWM test
These discrepancies can be explained both by the number of animals used and by the gender of the animals utilized (males and females in the first report while males only in our study)
Although both tests are hippocampal-dependent
the NORL is a more complex task that includes the spatial component that is absent in the classic NORT test
and may explain the discrepancy between results
While in the former test one unfamiliar object had to be discriminated among a total of five objects during the novel object recognition phase of the task
we employed a more classical approach with two dissimilar objects (one unfamiliar and one familiar) that had to be discriminated during the retention trial
Discrepancies in results reveal that when the complexity of the task is increased
recognition memory may be also impaired in this transgenic rat model
These observations are in agreements with our results
since Tg+/− rats lack parenchymal Aβ plaques but thus accumulate soluble oligomers intraneuronally
we investigated emotional and cognitive symptoms in Tg+/− rats
The most relevant result is that transgenic rats showed
from adulthood (6-month-old) to middle age (12-month-old)
increased anxiety levels and working memory impairment
transgenic rats showed an impaired spatial reference memory in comparison to WT rats
This deficit was also observed at 6 and 12 months of age
recognition memory and spatial learning were spared
Given that Tg+/− rats capture many behavioral features of early stages of AD
this new model may represent an important tool for the neuroscience community to enable future studies in basic and translational AD research

The Catholic faithful of Timor-Leste have been waiting for Pope Francis’ visit for a long time
Joy permeates the streets of this small nation
and then occupied by the Indonesian military until 1999
When the Pope’s visit to Timor-Leste was announced
the entire population welcomed the news with enthusiasm
joy and a desire to welcome the Holy Father
Pope Francis will be the second Pontiff to visit the country

The visit to Timor-Leste is part of the papal trip in Asia and the Pacific

The two institutions have been working well together from the start
The solemn Eucharistic celebration that will take place on 10 September will be held in a strategic location
which will gather Timorese faithful and a number of pilgrims from Indonesia and Australia
Some 700,000 people are expected to participate
the Pope will also meet with young people at the “Centro de Convenções”; more than 4,000 young people will likely attend
seminarians and catechists in the cathedral of the Immaculate Conception
He will then meet with children with disabilities from the “Irmãs Alma” school
The Pope’s visit to Timor-Leste will be an important event for building the faith and affirming the cultural and religious identity of the Timorese people
The central theme of the visit is: May your faith be your culture. Faith
was one of the important pillars in the fight for freedom
which allowed us not to lose hope in the long moments of suffering
The liturgical commission has circulated a short prayer for parishes
religious communities and religious formation houses to say after daily and Sunday Mass
The intentions of this prayer are for Pope Francis’ visit and health
the objective of the Timorese Bishops’ Conference is to help the Christian faith live and grow with more ardour in the heart of the Timorese people
The bishops have set up some meetings to inform and form the faithful from the three Dioceses of Timor-Leste: Dili
The theme of formation and catechesis is tied to the Pope’s primacy and to Pope Francis as a person
The goal is precisely to help the faithful deepen their understanding of the primacy of the Pope and of Pope Francis’ teachings
but also to urge them to harmonize faith and local culture
International speakers from Indonesia and the Philippines intervened
as did national speakers who presented their own ideas
The theme of the first academic seminar was: Asian culture and the Christian faith. There were three speakers: Verbite Fr
professor at the Catholic University of Timor-Leste; and
a Franciscan missionary and professor at the “Dom Jaime Garcia Goulart” Institute of Philosophy and Theology in Dili
The theme of the second academic seminar was: The Christian faith in a multicultural context
professor at the national university of Timor-Leste
a national retreat will be led by Fr Antonius Eddy Kristiyanto
The Church in Timor-Leste hopes that Pope Francis’ presence will bring peace and hope to the Christian faithful and transform them interiorly
The local Church also hopes that with the Pontiff’s presence among us
the Catholic faithful will understand their faith
celebrate it and bear witness to it in their daily lives
The Christian faith should become culture in the heart of the country’s Catholic Christians
as Pope Francis said in his address to Timorese bishops on the occasion of their ad limina visit at the Vatican
the Pontiff also prayed that the Church in Timor-Leste may evangelize culture and inculturate the Gospel in the local context
so that the faith may translate into the daily life of Timorese Catholics

'In this moment of sorrow, we join together with your children Fabricio, Isabela and Talita Xavier, also doctors and collaborators of Nair Alves de Souza Hospital, in extending our solidarity to the entire family, friends, colleagues and everyone who had the privilege of knowing him.
'Out of respect and recognition for his achievements, we are declaring three days of mourning.
No further details were revealed about the woman whose child he had been preparing to deliver and what happened to her as the tragedy unfolded.
Colleague Adilma Souza said online it had been an honour to have worked with Francisco, saying he loved his work and always treated his medical team to chocolate cake and Coca Cola after surgery.
Another said: 'Rest in peace. Francisco always worked out of his love for people.'
Banda Amorim wrote in a tribute: 'A great human being who brought sensitive, empathy and care to medicine.'

has visited South Korea to take part in a Congress on migrant workers from Timor-Leste
after arriving in South Korea on the morning of 14 September
visited the Timor-Leste Embassy in Seoul and was then welcomed at the Salesian Provincial House
Accompanying the Cardinal was Ambassador Gregorio de Sousa
There are about 6,000 migrant workers from Timor-Leste in South Korea
mostly young people in their 20s who work on fishing boats in fishing villages or are employed in rural areas of the country doing simple but very hard jobs
The Cardinal shared a warm and fraternal moment with the Salesians
once he arrived at the Salesian Provincial House
Having already visited South Korea three times - in 2000 when he was Formation Delegate for the Timor-Leste-Indonesia Vice-Province; in 2017 when he was Vice-Provincial and in 2021 when he was ordained Archbishop of Dili - Cardinal do Carmo da Silva has a great interest and knowledge of this country
the number of young migrant workers in South Korea has increased dramatically in recent years
and he has made great efforts to provide them with pastoral assistance
His visit to this country is therefore intended to meet these migrant workers
with 65% of its 1.3 million inhabitants under the age of 30
More than 6,000 of these young people are migrant workers in South Korea
hundreds of young people leave Timor-Leste for foreign countries such as Australia and South Korea
which means that all this is scarce in Timor-Leste."

they are not only people who fill the labour shortage in South Korea
but renew the vitality and witness of faith of the local Church
The devastation of rural areas has led to the emptying of local churches
and it is hoped that migrant workers from Timor-Leste who work in these areas will be able to fill the void for some time and integrate well enough to visibly witness their deep faith in Korean society

which will also host World Youth Day in 2027
"The young people want to meet the Pope," he said
"We bishops were very surprised to see how enthusiastic the young people were about the Holy Father's presence alone." The Archbishop of Dili prayed that the next WYD would be a place of "reconciliation"
that it would heal the many wounds in Korea and throughout East Asia
"Hundreds of thousands of young people will come from all over the world" he recalled "and if only we could have a party of reconciliation with them..
and I hope that Seoul 2027 will be the centre
because there will never be another opportunity like this
with young people coming together and playing a leading role in advancing reconciliation
They will themselves become messengers of reconciliation and will spread it throughout the world," the Cardinal said

“Psychotherapy teaches you to look beyond what people say on the surface,” she explains

as she guides her mentees through the nuances of balancing user experience with business goals
Requejo’s journey into design was not straightforward
she pursued a degree in chemistry but soon realised it wasn’t the right fit
“Chemistry required too much imagination for me,” she laughs
where I could see the tangible impact of my work.” This led her to industrial design
where she found fulfilment in transforming ideas into reality
from initial concepts to finished products
Requejo has navigated the intersection of design
she balances user needs with market demands and business objectives
“There’s always a tension between what the business needs and what the users need,” she explains
It’s not enough for a product to look good; it must make sense from a business perspective and
Requejo achieves this through close collaboration with designers
“By bringing different perspectives together
we can create solutions that are not only functional but also innovative and impactful.”
Requejo is especially passionate about supporting startups in their early stages
and aligning solutions with real-world needs
“These early stages are critical,” she emphasises
“Helping teams approach user research without bias is foundational to their success.” She adds
Requejo highlights resilience as essential to entrepreneurship
“It takes enormous strength to bring an idea to life,” she says
even when the journey becomes difficult.” She often reminds her mentees that setbacks are part of the process and that success rarely comes quickly
Requejo advises her mentees to remain adaptable
emphasising that initial ideas are just starting points
the more your approach may need to evolve.”
Sustainability also plays a central role in Requejo’s work and mentorship approach
“Designers have a responsibility to create solutions that are not only effective but also environmentally responsible,” she explains
“It’s about thinking beyond the product’s lifecycle and considering its impact on the planet.” She encourages budding entrepreneurs to consider the broader implications of their innovations
“It’s not just about immediate goals,” she says
“Entrepreneurs must consider how their solutions contribute to a more sustainable world
I firmly believe that sustainability should be at the heart of innovation.”
Requejo is excited about the potential for design to play a pivotal role in building a more sustainable and empathetic world
“Design has the power to shape the future,” she says
