The evidence base supporting treatment with BNZ comes primarily from retrospective observational studies that have documented a reduced rate of progression to cardiomyopathy and lower mortality in BNZ-treated patients (Cardoso et al., 2018; Hasslocher-Moreno et al., 2021) We present the three-year follow-up results of a prospective cohort of BNZ-treated chronic Chagas disease patients We aimed to characterize the parasite clearance rate using a high-sensitivity quantitative qPCR assay and describe the trajectory of anti-T cruzi antibody levels throughout early treatment and long-term follow-up relative to parasite persistence by PCR as a marker of therapeutic response and most were followed for three years following treatment with longitudinal clinical assessments and characterization of persistence and levels of T *All follow-up PCRs are depicting those drawn after BZN treatment We collected medical data and blood samples at multiple intervals 30 days prior to the BNZ treatment (T-30d) Another round of data collection occurred on the day before treatment initiation (T0d) We then gathered follow-up data at 60 days (T60d) and thirty-six months (T36M) post-treatment initiation At the enrollment interview and follow-up visits 12mL of blood anticoagulated with ethylenediaminetetraacetic acid (EDTA) was collected and immediately mixed with an equal volume of 6M guanidine/HCl-0.2M EDTA solution for PCR Serum separator tubes (SST) were also collected at each visit and centrifuged at 3000G for 15 min and serum aliquots were frozen at -20°C Aliquots of guanidine-lysed blood and serum were shipped to the US Central Laboratory (VRI; previously BSRI) for qPCR and serology Data from each patient were uploaded into REDCap (Research Electronic Data) (Harris et al., 2009; Harris et al., 2019) and results were then de-identified from specific patient-sensitive information We assessed changes in parasite loads measured through qPCR and serology levels by comparing the differences between the levels at multiple time points (T0d Our defined outcomes were as follows: “Negative” indicated a negative PCR result at T6M and T36M; “Positive” indicated a positive result on the T36M visit regardless of the earlier results; and “Inconclusive” referred to patterns that were negative on the T36M visit but had at least one PCR positive result at T6M or T12M visits we classified them as “Inconclusive” we conducted a multinomial logistic regression with the Negative group as the reference considering the age at study initiation and sex We presented these outcomes graphically using a serial boxplot to show the trend across visits regarding the antibody levels between these groups To predict the likelihood of experiencing a “Non-Positive PCR outcome,” which includes the Negative and Inconclusive groups we conducted a logistic regression with “Positive” as the reference category The models were adjusted for the same variables as in the multinomial analysis We presented the associations between predictor variables and the outcomes using odds ratios (OR) with 95% confidence intervals (CI 95%) we employed paired Wilcoxon rank-sum tests and Chi-squared tests with Bonferroni adjustment post-hoc for comparison across visits and the T0d visit We used the Kruskal-Wallis test to analyze the differences in antibody levels across visit time points and outcomes for T cruzi-infected patients treated with benznidazole Continuous variables were reported as medians and interquartile ranges or mean and standard deviations based on their distribution while categorical variables were reported as frequencies and percentages Our conclusions are based on a two-tailed hypothesis test with an alpha error probability of 0.05. The Mount Vernon home where Robert Espada grew up was intended to be his inheritance The house that Marlon Young bought last year in Mount Vernon was supposed to be a ticket out of Queens for him Only one of these stories will have a happy ending That’s because the address in each narrative is 440 Garden Ave. a modest two-story home across from Memorial Field that is at the center of a court fight over who really owns it Both sides have claims against the City of Mount Vernon allege that the city improperly foreclosed on his late grandmother’s home in 2012 The Youngs claim their deed is the only valid one and that Comptroller Deborah Reynolds was wrong last year to reject it "Mount Vernon has disappeared with people's inheritance," Cynthia Espada said last week who would die only five months later. Nellie had not left a will and Robert didn't go to surrogate court to transfer ownership claims he continued paying taxes on the house But the city began foreclosure proceedings in 2011,a when nearly $14,000 was owed in back taxes Espada claims he never received notification that the city was threatening foreclosure A March 2012 court order cleared the way for the city to take ownership of the house as a creditor, could not take the property of a dead person without petitioning Surrogate Court to appoint an administrator to represent Nellie Espada's interests It is unclear whether city officials knew Nellie Espada had died although the Espadas insist her death certificate was filed with the city The house was included in a 2013 auction of city-owned properties did not bid but told the finance department he was interested in the property.  the city sold the property to Moreira for $51,000 giving him a deed signed by then-Mayor Richard Thomas Espada claims he didn't even know that the city had owned it – finding out only when his stuff was removed by Moreira while he was in Georgia from February to April that year "I'd been in that house my entire life and my grandparents told me I was going to pass it on to my daughter after me." Moreira was adamant that Espada was aware he was losing the house saying he had conversations with him several times before buying the property. He said Espada never made any effort to pay the back taxes to recover the property from the city Moreira said he gave Espada more than 60 days to vacate the house at which point Moreira removed whatever belongings Espada left behind The Espadas claim Moreira was required to seek court action to remove him suggests he was worried the city marshal would have discovered there was something wrong with the title transfer The Youngs move inThe sale to Moreira came just five weeks before the city was going to auction off more than a dozen foreclosed properties The list approved by the City Council had 440 Garden Ave. removed from it because the sale both bodies that year approved the sale to Moreira although the sale was not completed until three years later who in the early 2000s was a night distribution manager for The Journal News a Mount Vernon native who he married last year took it upon herself to research what Mount Vernon had done She said her husband never should have paid any taxes on the property once his grandmother died And convinced that the city couldn’t take the property of a dead woman with no will the Espadas  went to Westchester Surrogate Court in May 2019 detailing what she alleged was the city’s fraud and giving him 30 days to vacate the house he had spent $100,000 to $150,000 to renovate the house in anticipation of selling it He said he was not concerned about Espada's letter and believed the city had gotten the house properly and so had he Moreira sold the house to Young and his father The following month Robert Espada was named the administrator of his grandmother's estate and obtained a deed showing he transferred the property to himself and his wife the Youngs’ deed has not been recorded by Westchester County — but Robert Espada’s deed has the city comptroller, would not sign off on the title transfer to Young She agreed with the Espadas that her predecessor had improperly obtained the property in 2012.  Marlon Young said he was shocked to learn someone else had a claim to the house “I did everything I was supposed to,” he said Walker said she did not remember details of the 440 Garden Avenue sale She said property owners would be sent several notifications that their homes were subject to foreclosure and that it was the law department's responsibility to make sure the right people were notified.  Corporation Counsel Brian Johnson had no comment as the matter was being reviewed and was the subject of pending litigation.   the Espadas began eviction proceedings in Mount Vernon City Court to force the Youngs out of the house though that was quickly dismissed when they didn't pay a $400 filing fee or apply to have fees waived The Youngs then sued the Espadas and Reynolds in state Supreme Court Among the court exhibits was a letter from the title insurance company detailing Reynolds' refusal to approve the Youngs' deed That mirrored a criticism of the comptroller levied this spring by the head of the New York State Land Title Association who said members were no longer providing title insurance in Mount Vernon because Reynolds was taking too long to process the paperwork Reynolds responded that paperwork was often submitted with incomplete or erroneous information Cynthia Espada knows all about the attacks on Reynolds since she took office in 2018 — including that she doesn’t pay bills and keeps other officials in the dark about city finances But Espada insists Reynolds is the only official willing to look into what she considers the city's fraudulent takeover of her husband's home The Espadas have answered the Youngs' lawsuit with their own counterclaims against the city Joseph Altman, to seek a default judgement ordering Reynolds to accept the Youngs' deed and blocking any effort to remove the Youngs from the house.  Altman argued that the Espadas have no legal claim to the property and that the transfer of ownership from the city to the Moreiras to the Youngs was valid And even if the city erred in taking ownership the Espadas allowed too much time to pass before making any claim He said there was no reason for the title search company to go beyond the chain of title "The documentary evidence firmly establishes that the (Youngs) are the proper legal owners of the property," Altman wrote While the two families are in a legal battle the Espadas also think of the Youngs as victims in the case “The Youngs are now in the unenviable position of losing a real property for which they allegedly paid good money,” the Espadas’ lawyer wrote in court papers last month seeking a court judgement validating his clients’ claim to
