The policy response to the COVID-19 shocks to education needs to be comprehensive and well-planned to ensure that education systems are “built back better than before”
Learning from successful experiences in education is key to help develop this response
Many lessons on how to successfully fight learning poverty and improve foundational learning can be learned from Ceará - a relatively poor state in Northeastern Brazil with 9 million inhabitants - and Sobral - a 200-thousand-inhabitants municipality where 20 years ago 8 out of 10 kids were illiterate
These lessons are even more important now that Brazil and the world will have to recover from the effects of the pandemic on learning outcomes and
have an opportunity to deeply reform their education systems to accelerate learning.
As part of an effort to learn more about globally successful cases
a team of education specialists from the World Bank visited Ceará and Sobral in early February
We were a group formed by people of eight nationalities with the expectation of discovering a great secret to spread to the world
vice-governor of Ceará who led the main educational reforms in the state as secretary of education and from Ivo Gomes
who also led several of the main education reforms in that municipality
we heard the same answer: There is no secret
really! It boils down to a strong focus on the basics
teaching children how to read by the right age
and making everyone and the system to coherently work towards that goal
Let us here tease out the main ingredients of their formula
the facts: Learning improved much faster in Sobral
had the biggest increase in education quality (as measured by Brazil’s Basic Education Development Index
Almost all of the 184 Ceará municipalities initially had very low levels of quality in teaching and very limited resources
investing about a third of the per student education spending of wealthier Brazilian states
Ceará improved the quality of education so much faster than the rest of Brazil
Sobral went from a bottom-ranked municipality to become first for both primary and lower secondary education and to reach levels of education quality comparable with those of world-class education systems as measured by PISA
Today Ceará has the lowest rates of learning poverty in Brazil and features 10 of the 20 top-ranked municipalities
and Sobral has some of the best primary schools in Brazil
the outstanding educational results of Ceará and Sobral students are well above what would be expected given the socioeconomic context in which they live and learn
what are the key ingredients of the formula for the success in education of Ceará and Sobral
what municipal schools in Ceará have accomplished is firmly grounded in insights from the Science of Learning
which emphasize three key principles: first
opportunities and incentives to get better; and third
that sustained performance improvement requires constant and focused feedback
and then Ceará have embraced these principles in the day-to-day practices and interactions of students
principals and even district officials in classrooms and schools
these principles manifest in having a clear diagnostic of learning and system performance
focusing the efforts on top priorities for improvement
and then choosing the right set of strategies and implementing them in a coordinated and persistent fashion with all key education stakeholders.
the formula for education success of Ceará has five main ingredients
which are supported by international evidence on what works in education
The first three ingredients involve sound financing and pedagogical policies
for municipalities to achieve better education outcomes; (ii) technical support to the municipal school networks struggling to improve learning goals
with a strong focus on enhancing teacher effectiveness and achieving universal literacy at the right age; and (iii) the use of a robust and reliable monitoring and assessment system
which continuously assesses key results of education
These three ingredients emphasize instructional coherence in terms of the alignment of the curriculum (what students are expected to learn)
teachers’ pedagogical approaches (how students are taught)
and learning assessment (how the system checks whether students are learning)
They also show consistent implementation over time building on earlier preparatory reforms.
The fourth ingredient is a high degree of autonomy for municipalities to design and implement their education policies
this leads to the creation of a culture where feedback is essential to improve performance throughout the system
What stroke us the most in visiting Ceará and Sobral is that when asked about the potential drawbacks of the use of assessments and incentives to drive performance
and administrators generally reframed the conversation: they see feedback and incentives as critical to achieve learning for all rather than as instruments of accountability
ingredient that makes it all stick together and enables action is a sustained political leadership and commitment to education quality
Recipe for success in education in the state of Ceará
No other municipality exemplifies this non-miraculous recipe better than Sobral
What has led Sobral to become the best education system in Brazil is the coherent mix of: (i) Effective and continuous use of student assessment
through written and oral evaluations of all children focused on reading fluency
and using learning outcomes extensively to guide education policy and in-classroom pedagogical practices; (ii) Focused and well-aligned curriculum
with a clear learning sequence and prioritization of foundational skills
as well as its full alignment with national
textbooks and teacher training; (iii) Prepared and motivated teachers
with focused and practical professional development
Recipe for success in learning in the city of Sobral
All of the successful education policy reforms in Ceará and Sobral involved a crucial first step: political leadership and a commitment to put learning first or as a high-level official put it “to take politics outside of schools”
Strong political leadership is needed to confront the resistance from stakeholders whose interests are not always well-aligned with student learning
Again in Sobral sustained and strong political leadership has placed education as a top priority
as made explicit by a sharp overarching goal: ensure that all students are fully literate by grade 2.
Countries that are firmly committed to achieving learning for all can well start with establishing similar time-bound
ambitious targets to eliminate learning poverty
and adapt the key ingredients of the success formula of Ceará and Sobral to their own country contexts

Both Ceará and Sobral had low levels of literacy 20 years ago and went on to improve the quality of their education systems much faster than the rest of the country
achieving levels of education quality comparable with the most developed countries
“The State of Ceara in Brazil is a Role Model for Reducing Learning Poverty” presents the key education reforms implemented in the state that led to substantial improvements in quality in primary and lower secondary education
Despite having the 5th lowest GDP per capita among the 26 Brazilian states
Ten of its municipalities are now among the nation’s top 20
“Achieving World-Class Education in Adverse Socioeconomic Conditions: The Case of Sobral in Brazil” analyzes the main aspects of the reforms carried out by Sobral that led to rapid improvements on education quality
Sobral did not even rank in the top thousand among Brazil’s municipalities
it ranked first for both primary and lower secondary education in the national index of education quality
The improvements in education in both the state of Ceará and the municipality of Sobral are the result of a comprehensive package of education reforms
In a time when the reformulation of the educational system is necessary due to the impact of the Covid-19 pandemic
the education models in Ceará and Sobral can provide an example to all countries that wish to build back better than before their educational systems by aiming to eradicate learning poverty and build human capital

Cancer is a leading cause of mortality responsible for an estimated 10 million deaths worldwide in 2020
and its incidence has been rapidly growing over the last decades
as well as high systemic toxicity and chemoresistance associated with conventional anticancer therapies reflect these high levels of incidence and mortality
efforts have been made to search for novel anticancer drugs with fewer side effects and greater therapeutic effectiveness
Nature continues to be the main source of biologically active lead compounds
and diterpenoids are considered one of the most important families since many have been reported to possess anticancer properties
Oridonin is an ent-kaurane tetracyclic diterpenoid isolated from Rabdosia rubescens and has been a target of extensive research over the last few years
It displays a broad range of biological effects including neuroprotective
and anticancer activity against a variety of tumor cells
Several structural modifications on the oridonin and biological evaluation of its derivatives have been performed
creating a library of compounds with improved pharmacological activities
This mini-review aims to highlight the recent advances in oridonin derivatives as potential anticancer drugs
while succinctly exploring their proposed mechanisms of action
future research perspectives in this field are also disclosed
There is no curative treatment option available for cancer, and current anticancer therapies, especially chemotherapy, have limited therapeutic potential associated with adverse side effects, chemoresistance, and high systemic toxicity to the patient (Kashyap et al., 2021)
studies have been performed to search for more efficient and selective anticancer drugs with greater therapeutic properties and better safety profiles
there has been a growing attention toward the development of natural anticancer agents
they continue to hold great potential in the search for novel lead compounds in drug discovery
Herein we seek to briefly overview the biological activities of oridonin and highlight the emerging therapeutic potential of recent oridonin derivatives in anticancer therapy
while also exploring their proposed mechanisms of action
we provide a discussion of future research perspectives for the development of these derivatives in the clinic
Oridonin (C20H28O6, compound 1 listed in Table 1) is an ent-kaurane tetracyclic diterpenoid isolated from the traditional Chinese medicinal herb Rabdosia rubescens. It was first reported in 1967 (Fujita et al., 1967), and the relationship between its anticancer activity and its structure was demonstrated a few years later (Fujita et al., 1976)
Recent approaches of oridonin optimizations (modifications of hydroxyl groups)
and 357 research articles published as of September 2022 while searching “oridonin AND cancer”
This indicates that a major focus is being given to its anticancer activity
Nevertheless, oridonin’s anticancer mechanisms of action are not yet fully understood. The suggested main ones include the suppression of the cell cycle progression, induction of apoptosis, and autophagy, by modulation of signaling pathways, for instance, regulation of intracellular reactive oxygen species (ROS), Bax/Bcl-2, p53/p21, NF-κB, MAPK, PI3K, and fatty acid synthase pathways (Ding et al., 2016)

Moreover, oridonin induced apoptosis in OCM-1 and MUM2B uveal melanoma cells (Gu et al., 2015), and SW480 and SW620 colorectal cancer cells (Kwan et al., 2013), by suppressing fatty acid synthase. In HepG2 cells, oridonin induced G2/M phase arrest and apoptosis via MAPK and p53 pathways (Wang et al., 2010)
Furthermore, oridonin also induced autophagy by inhibiting glucose metabolism in colorectal cancer cells (Yao et al., 2017), and recent studies suggest that oridonin not only can suppress cell migration and invasion, (Li et al., 2016c), but also revert drug resistance (Kadioglu et al., 2018)
Oridonin is recognized as a logical hit compound for anticancer therapy research. It has an appropriate molecular weight (364.4 g/mol) and plenty of functional groups that provide numerous synthetic routes to create different libraries of derivatives. Oridonin also meets the criteria of Lipinski’s rule (Lipinski et al., 2001) and it is relatively commercially available (Ding et al., 2016)
Evidence shows that structural modifications on oridonin usually encompass four typical optimizations, as represented in Figure 2: 1) modifications of hydroxyl groups; 2) modifications of A-ring; 3) modifications of D-ring (α, β-unsaturated ketone); and 4) transformations of the skeletal structure (Zhang et al., 2020; Guan et al., 2021)
Although this research has been active for a few years
and substantial progress has been achieved in the identification of novel derivatives
herein we have selected the most representative work
Typical optimization sites and molecular structure of oridonin
In the same year, Hou et al. synthesized novel C14-1,2,3-triazole oridonin derivatives via copper-catalyzed alkyne-azide cycloaddition (CuAAC). Compound 6 (listed in Table 1) proved to be the most potent (IC50 = 3.1 μM) against the PC-3 cancer cell line. Preliminary mechanistic studies reported that this compound caused G2/M phase arrest and induced apoptosis in a dose-dependent manner in the same cell line (Hou et al., 2019)
The same research group also synthesized oridonin-coupled nitrogen mustard derivatives, and all of them showed better anticancer activity than oridonin against a variety of cell lines. Compound 9 (listed in Table 1) proved to be the most potent, exhibiting an IC50 value of 0.50 μM against the BEL-7402 cell line. This compound also induced apoptosis of BEL-7402 cells and caused G1 phase arrest (Xu et al., 2014)
In 2020, Li and collaborators synthesized oridonin derivatives with H2S-releasing groups. Compound 10 (listed in Table 1) showed the most potent anticancer activity against the K562 cell line, with an IC50 value of 0.95 μM. Further studies revealed that compound 10 caused S phase arrest in K562 cells and G1 phase arrest in HepG2 cells (Li et al., 2020)
The reported structural modifications generally improve the solubility of the derivatives by introducing aqueous solubility-enhancing moieties via esterification of the 1-O and 14-O-hydroxyl groups. Such derivatives usually act as prodrugs since ester bonds suffer from poor in vivo metabolic stability (Ding et al., 2013a)
Recent approaches of oridonin optimizations (modifications of A-ring)
In 2013, Ding et al. (2013a) developed novel derivatives by introducing a thiazole ring at C1 and C2 of oridonin’s A-ring. Most of the nitrogen-enriched derivatives exhibited higher potency and aqueous solubility. In the form of its HCl salt, compound 13 (listed in Table 2) exhibited approximately 62-fold improvement in aqueous solubility when compared with oridonin (1.29 mg/mL)
compound 13 showed an IC50 value of 0.20 μM
approximately 147-fold more potent than oridonin
and mediated apoptosis of MDA-MB-231 cells
α, β-unsaturated ketones (enones) are well-known Michael acceptors. For this reason, the enone system is considered an important pharmacophore of natural products, and oridonin’s D-ring enone appears to be critical for its anticancer activity (Ding et al., 2013b)
few modifications have been performed on that part of the molecule that have successfully produced derivatives with improved activity
Nonetheless, Shen et al. (2018) demonstrated that α, β-unsaturated ketones can be targets for structural modifications to achieve promising derivatives. Shen’s group synthesized oridonin derivatives with substituted benzene moieties at the C17 position, and compound 16 (listed in Table 3) proved to be the most potent with an IC50 value of 1.05 μM against the HCT-116 cell line
compound 16 induced apoptosis and caused G2 phase arrest in HCT-116 cells
Recent approaches of oridonin optimizations (modifications of D-ring - α
Li et al. (2013a) synthesized ent-6,7-seco-oridonin derivatives by the conversion of oridonin to spirolactone-type diterpenoids. All the synthesized compounds exhibited better anticancer activity than oridonin, in vitro. Compound 17 (listed in Table 4) exhibited IC50 values of 0.39 μM against the K562 cell line and 1.39 μM against the BEL-7402 cell line
similar values to that of the positive control Taxol (IC50 values of 0.41 μM and 1.89 μM
Further mechanistic studies of compound 17 revealed that it induced apoptosis in BEL-7402 cells and caused G2/M phase arrest
Recent approaches of oridonin optimizations (transformations of the skeletal structure)
The global cancer burden is expected to be 28.4 million cases in 2040, corresponding to a 47% rise from 2020 (Sung et al., 2021)
Chemotherapy continues to be the main therapeutic option for cancer treatment
and oridonin has recently emerged as a promising hit compound due to its anticancer activity
and the exact mechanisms of action remain to be further elucidated
these findings lead us to believe that oridonin and its derivatives have tremendous potential yet to be discovered in a variety of cancers
Although no oridonin-based drugs have been approved for clinical use by the U.S. Food and Drug Administration (FDA) or by the European Medicines Agency (EMA) (Liu et al., 2021b)
compound HAO472 has already advanced into a phase I clinical trial in China
and we anticipate that new oridonin derivatives may emerge as anticancer drug candidates and enter additional clinical trials soon
It is imperative for oridonin and its derivatives to be the subjects of more robust pre-clinical studies
to ensure the safety and potency of the compounds before developing them as anticancer drugs
Further investigations are required regarding the single or combined use of oridonin and its derivatives in anticancer therapy
while also exploring their role as anti-chemoresistance agents
for they have the potential to be viable therapeutic options

Newly qualified teachers now undergo a preparatory internship and all teachers
receive on-the-job training for a day each month
Every term there is a bonus system for teachers whose students perform well in assessments
Student welfare is a high priority; pupils receive two free school meals a day and there are plans to hire a mental health professional in each school
All this has led in part to the success of former students such as Chelton Santos, 22, and his sister Maria, 20. The siblings live with their parents and grandmother in a poor and dangerous area. The family have survived on one minimum wage (now $258 a month) for many years

I’ve seen my mother suffer with health problems since I was a child and now I want to help the poorest in society.”
is hopeful about her children’s future: “I never believed that one day my son would be studying to be a lawyer at university
We did everything we could to keep [the children] in school
I wanted to give them a nicer place to live … it’s dangerous here
they’re going to move from here to a better life.”
Sobral’s success is now being replicated across the country. Clodoveu Arruda, a former mayor of the city, who oversaw a large part of the transformation in the education department, has teamed up with the Lemann Foundation to spread best practice
So far they have worked in 25 municipalities across five states
Not everyone in Sobral is content with what has been achieved
however – they are hungry for further success

NORTH KINGSTOWN – You could easily drive past Alpha & Omega Deaf Church without noticing it
The congregants meet in a nondescript red brick building that used to be a physical therapist’s office
The only hint of its presence is a modest sign with its name and address: 7291 Post Road
But the church’s quiet demeanor on the outside belies its lively interior. On a recent Sunday, the fellowship room was abuzz with energy. Hands performing American Sign Language gestured warm greetings and made quick jokes
Perhaps no presence loomed larger than Pastor Seth Sobral
broad-shouldered and with an inviting smile
he exuded a magnetic force that drew people to him
Sobral brings this charisma to his sermons
While preaching about Samson – a Bible character known for his supernatural strength – he joked about his physique
While speaking during a service at other churches would be considered rude
congregants sign back at him in the middle of his sermons
The whole affair does not bother Sobral – he seems to harness the feedback back into his sermon
“I should not be afraid,” Sobral signed during his sermon on Samson
but that did not stop him from hearing God’s calling
It started with a cartoon Bible when he was a kid and with his mother’s instruction
He went to a hearing church but found it unrelatable
“It's hard to really relate to the hearing world
even though I'm looking for God's word,” Sobral said through an interpreter
but Sobral said attendance dips during the summer
While some churches offer ASL interpretation for services
Alpha & Omega Deaf Church is the only deaf-owned
Sobral estimated the nearest one would be in New York City or St
Rhode Island’s deaf community is small. Only 6% of people 18 or older in the state have a hearing disability, according to the federal Centers for Disease Control and Prevention
making the church an oasis for deaf people who are looking for a community of faith to join
Some commute to Sunday services and mid-week Bible studies from Connecticut and Massachusetts
The services are conducted entirely in ASL
Sobral uses visual aids to help congregants follow along

“We just want to try to connect to something greater or connect with something deeper and worship beyond our day-to-day life,” Sobral said
The church attracts a wide range of people from the deaf community
while others lost their hearing through an illness

“It was a really tough challenge because I didn’t speak English
so I had to learn English first and then ASL,” Medina said

She said her favorite part of Alpha & Omega Church is the fellowship that she and her family have found
finding the church itself was a spiritual experience
who has been a congregant for about 10 years
“God showed me the way and said go in this direction,” Buchanan said
someone pointed her to Alpha & Omega Deaf Church
Buchanan had previously attended a hearing church
“There’s a lot of spoken language in the hearing world
But now I can understand clearly,” Buchanan said
who lost his hearing due to spinal meningitis when he was 14 months old
also attended a hearing church without an interpreter
It was hard to socialize or learn anything in that environment
“I really have fallen in love with this church and learning more and more about God’s word,” Geisser said
Sobral’s desire is that people understand that
a deaf church is not so different from a hearing church
Alpha & Omega Deaf Church may be the only place where they can step into a fully immersive ASL community
It is not unusual for several to arrive early or stay behind for hours after the service just to spend time chatting with others in ASL
Their time spent together in reverence of something much larger than self becomes all the richer for the rarity of their unity
for the way they hunger for conversation and are nourished by each other's presence
for the way they linger because they can relate so easily in the church they have made their joyful and liberating haven
a baking-hot north-eastern state that has one of Brazil’s highest rates of poverty
When local officials ordered tests in 2001 they found that 40% of Sobral’s eight-year-olds could not read at all
One-third of primary pupils had been held back for at least a year
He remembers a head teacher who signed documents with a thumbprint
because she lacked the confidence even to scribble her own name

The final result of the observations made at Principe demonstrated an average deviation of 1.6 arcseconds
while the data gathered at Sobral had a deviation of 1.9 arcseconds
which was almost twice the estimated number in the gravitational theory devised by English physicist Isaac Newton (1643-1727)
Newton’s universal theory of gravitation became a specific example of Einstein’s general relativity,” explains Veiga
“We can view the photographic plates produced in Sobral and Principe as a decisive moment in the history of 20th century science
when ideas proposed by the theory of relativity had not yet been embraced,” says physicist and historian of science José Luiz Goldfarb of the Center Simão Mathias at the Pontifical Catholic University of São Paulo (PUC-SP)
“Challenging the ideas derived from Newton related to classical mechanics met with strong opposition
given that proposals from Einstein and other scientists completely changed our understanding of the world.” Goldfarb believes that conserving the photographic plates keeps alive the memory of the transformations that occurred in science early in the 20th century and still affect how we see the world today

Having studied Communication Design in Porto
the Glasgow-based practitioner originally hated drawing classes where the expected output was old-fashioned realism
These classes were “an absolute nightmare” to João as the drawing he experienced were not creative or stimulating
more “something with very specific rules that [he] was constantly doing wrong”
with the guidance of an enthusiastic illustration tutor Rui Vitorino Santos
João was introduced to “totally different drawing practices” that opened his mind to “new and fascinating worlds” beyond the realm of realism
The cartoonist started doodling in sketchbooks that were meant for ‘real’ drawings but João “didn’t dare to call those doodles ‘drawings’”
he embraced his own style and took pleasure in the unique way the drawings were created
João learning to draw in his own way and developed a practice that evolved from an idea of drawing that to some people is the “wrong” way
“I’m interested in the accidental nature of drawing — the things that happen when the hand is not trained," says João
Like the subtle expression seen through a wobbly line rather than something that is ruler-straight
João is more interested in a practice that evolves as a result of limitation
The medium of comics consequently suits the cartoonist as it contrasts the “clumsy and awkward” way of drawing with a rigid layout
João depicts his characters as “fragile beings”
“I think I have a soft spot for people who are seen as losers or weirdos because they’ve been mistreated by the system.” His characters are “people who are kind and just want to love and be love but have a hard time navigating the world they’re stuck in.” Such characters continually crop up in João’s work
firstly through the natural setting of comics which have now become “an obsession”
João can further explore the deeper narrative behind the characters
Drawing and writing come together to enhance the overall tone of the story and what the subjects are going through
João can delve into his writing “without worrying too much about whether it is fiction or a poem
this provides the viewer with emotive and often humorous storylines that are concisely delivered as well as visually stimulating

May 14 (Xinhua) -- Portuguese singer Salvador Sobral won the 62nd edition of the annual Eurovision Song Contest here on Saturday night with an emotional and romantic song Amar Pelos Dois (Love For Both) written by his sister
During the final held at the International Exhibition Center in Kiev
winning the highest possible score from the judges and the second highest score from the public vote
Bulgaria's Kristian Kostov took the second place with 615 points
followed by Moldova's participant with 374 points
Portugal had not won the Eurovision Song Contest
A total of 26 singers participated in this year's Grand Final of the Eurovision
The race was viewed by more than 204 million TV audience in European countries and other countries across the globe
"The Eurovision is the most famous song contest in the world. Each time millions of people from all over Europe, and recently from Australia as well, freeze in front of their TV screens rooting for their countries," said Ukrainian President Petro Poroshenko ahead of the contest

And all the while Salvador Sobral knew exactly what he was talking about, as he was involved in a long-term relationship with girlfriend — the French actress Jenna Thiam.
Well today we were pleased to confirm that his girlfriend is now his wife, as Salvador and Jenna tied the knot on December 29th. That was just one day after he turned 29. Talk about a gift that will keep on giving!
