New Bern woman to compete in senior pageant for friendship, not prizesby Valentina Wilson
NEW BERN, N.C. — Margaret Cobb Fairchild, a New Bern resident, is set to participate in the "Miss North Carolina Senior America" pageant next weekend in Pinehurst, northwest of Fayetteville. This will be her second time competing in the pageant, which is exclusively for women aged 60 and above. Last year, Fairchild made her pageant debut and was voted Miss Congeniality.
Fairchild, originally from Missouri and a retired flight attendant, expressed that her motivation for participating is not the prizes but the opportunity to forge new friendships.
"I wanted to be able to meet more friends, make new friends again," Fairchild said. "That was the number one thing when I was asked about joining. I said - I would love to make new friends - and I've got some more new ones, and the ladies are genuine."
In addition to her pageant pursuits, Fairchild enjoys sewing and playing the piano.
NEW BERN, N.C. — Hundreds of protesters gathered at the intersection of Pollock and Craven Streets in downtown New Bern on Saturday, joining nationwide demonstrations marking the 250th anniversary of the Battles of Lexington and Concord. The rallies, dubbed the "Hands Off" protests, are aimed at opposing what participants see as government overreach by the Trump administration.
Penny Saulmier, who organized the New Bern protest, said, "It seems to me like we were having a king who’s ruling instead of governing, and I just thought that was outlandish." Initially expecting around 45 attendees, Saulmier reported that over 300 people had joined the protest by noon. "I just can’t express how overjoyed I am at the turnout," she said.
Protesters chanted slogans such as "Hands off our constitution," "Hands off Free Speech," and "Hands off equal rights," expressing their concerns over various issues. Diane Lemieux, one of the demonstrators, said, "This is reality—this is the truth about how American people feel." Another protester, Jennie Adams, added, "There’s a lot of people who have the same needs, same irritations, same objectives."
Lemieux highlighted specific concerns, stating, "Federal positions are getting cut, we’re-- services to the VA are getting cut. Threats to cut Medicaid and Medicare being cut. I’m especially concerned about the save act." Adams expressed her motivation for participating, saying, "There’s just been too many things that are not good right now, and this is the only way I can participate to be able to feel better about it and release some energy."
The protest in New Bern was part of a larger movement across the United States, with similar events taking place in cities like Washington, D.C., as citizens voiced their opposition to perceived governmental overreach.

The installation will remain on view through June 1st as the Kunsthalle moves toward a full reopening with an ambitious program featuring exhibitions by artists including Melvin Edwards and Tuli Mekondjo
This new commission arrives at a pivotal moment for Kunsthalle Bern. As the institution undergoes renovations to improve safety
it has embraced the opportunity to rethink its mission
Under the leadership of director iLiana Fokianaki
the institution has been engaged in a yearlong process of what it has termed ‘fermentation’
referring to a radical rethinking of its operational
and administrative practices by drawing from the ecological principles of permaculture
so too does the institution’s vision: toward a more inclusive
model that considers its social and ecological responsibilities in the context of the environment and global cultural shifts — ‘A ‘work in progress’ where our new vision for an institution of the future emerges: permeable and with access for all,’ as the team notes
carries particular resonance in this project

The new SPRQ chemistry for the Revio system is making high-quality long-read sequencing more efficient than ever—delivering more data from less input without compromising accuracy or cost efficiency
But what does that actually look like in a real lab
As the head of University of Bern’s sequencing facility
Pamela Nicholson and her team are known for taking on some of the trickiest genomic projects
and pushing sequencing technology to its limits
the results were even better than they expected
and what it could mean for the future of sequencing
Pamela Nicholson: I’m the head of the sequencing facility here at the University of Bern
We serve all the researchers at the university who want to do sequencing from extractions all the way to basic data processing
We also serve one of the biggest hospitals in Switzerland
epigenomics and we have a particular focus on microbiomics
People come to us because they’ve got difficult projects they want to do
and they know we’re strong on long reads
I really focus on finding the technology that will bring them the most for their research questions
If it’s going to provide them with better results to meet their research question
HiFi sequencing has really made a lot more projects feasible and addresses all the major considerations for a sequencing project
Accuracy and length are not in question with Revio
We struggled with the sequencing yield and throughput of earlier PacBio instruments and Revio really has proved itself to be a game changer for our facility with regards to throughput
In many projects we routinely use all the barcodes that PacBio sells and this just shows the leap forward in sequencing output
It often comes down to budget for researchers and now
a PacBio full-length16S rRNA-Seq project is cheaper than an Illumina V3-V4 16S rRNA-Seq at our facility
It’s impressive to follow the relentless innovation
there are questions like “How can we ensure consistency in our longitudinal projects
What will it mean for people who are between projects
We started with SPRQ late last December with some really diverse sample types
we had a project working with naked mole rats
which is genetically a very curious animal
and a project for the Bovine Pangenome Consortium with some really difficult older samples
and typically would have expected between 70–80 Gb
The quality scores were also up with the new chemistry as well
We had a similar experience with the Bovine Pangenome Consortium –– these were blood samples
shipped all over the world for the consortium
we have processed many of these tricky samples
and some didn’t get quite enough reads
we took the opportunity to pool some of these old libraries together and run them with SPRQ
Previously such libraries returned 60–80 Gb and here with SPRQ we were nearly up to 100 Gb
We are very satisfied with the results and critically
we’ve consistently been seeing results like this since we have switched over to SPRQ chemistry
PacBio offers fully kitted solutions and protocols so that we don’t have to buy all the important aspects from three different vendors
It becomes expensive when you start counting the other vendors that you have to buy things from for every protocol
Before it was only feasible for certain researchers and certain projects
but now it can really cover everything extensively and this is because the protocols and sequencers are constantly evolving and getting better
We do high-quality gold-standard reference genomes with HiFi
and Kinnex full-length RNA sequencing led genome annotations
It is exciting for microbiomics where we clearly see that full-length 16S sequencing with Kinnex is extremely powerful and gives a new dimension to phylogenetic resolution that you will not get from a V3-V4 16S rRNA-Seq project
University of Bern data yield from recent Kinnex single-cell and full-length libraries
There’s possibly a bit of a latency to see adoption with academic researchers or in the clinic
it can really switch a light on in a project
we have had a few long-read RNA sequencing projects that have uncovered critical gene fusions
short-read sequencing was not giving enough information and only with PacBio full-length Kinnex kits were we able to detect transcripts spanning two genes and sequence full-length fusion transcripts
The amount of alternative splicing has probably been underestimated and undervalued for years

recently pled guilty to two counts of Possession of Methamphetamine
Swindell received an active sentence of 128-166 months in the North Carolina Department of Adult Corrections
Swindell pled guilty in Craven County Criminal Superior Court
District Attorney Scott Thomas and Craven County Sheriff Chip Hughes made the announcement on Tuesday
"the charges stemmed from two separate incidents involving Swindell
The first incident occurred when Swindell was a suspect in a larceny and breaking or entering at a job site in the Gracie Farms Mobile Home Park in New Bern
Swindell was on probation at the time and probation officers went to Swindell’s home to conduct a search of Swindell

one bag containing 0.11 grams of oxycodone
and one bag containing 20.25 grams of Methamphetamine
Later Craven County Sheriff’s Office Investigators received information about Swindell selling narcotics and began investigating Swindell
