The transcription factor CCAAT-enhancer binding factor alpha (C/ebpα) is a master controller of myeloid differentiation that is expressed as long (p42) and short (p30) isoform
Mutations within the CEBPA gene selectively deleting p42 are frequent in human acute myeloid leukemia
Here we investigated the individual genomics and transcriptomics of p42 and p30
Both proteins bound to identical sites across the genome
they induced a highly similar transcriptional response with the exception of a few isoform specific genes
Amongst those we identified early growth response 1 (Egr1) and tribbles1 (Trib1) as key targets selectively induced by p42 that are also underrepresented in CEBPA-mutated AML
Egr1 executed a program of myeloid differentiation and growth arrest
Trib1 established a negative feedback loop through activation of Erk1/2 kinase thus placing differentiation under control of signaling
differentiation elicited either by removal of an oncogenic input or by G-CSF did not peruse C/ebpα as mediator but rather directly affected the cell cycle core by upregulation of p21/p27 inhibitors
This points to functions downstream of C/ebpα as intersection point where transforming and differentiation stimuli converge and this finding offers a new perspective for therapeutic intervention
we investigated C/ebpα isotype-specific genomics and transcriptomics in primary hematopoietic precursor cells and its relation to HoxA9 mediated transformation
Our findings suggest a particular role for p42 in establishing a signaling-dependent differentiation program
The oncogenic activity of HoxA9 perturbs this program downstream of C/ebpα itself
Our results emphasize the importance of a loss of-function for leukemogenesis and give potential perspectives how to bypass this defect for therapeutic purposes
Transduction was done with CD117 (Kit) selected cells enriched with magnetic beads (Miltenyi
Germany) essentially as recommended by the manufacturer
cells were cultivated in methylcellulose (M3534
Germany) for two rounds under antibiotics selection
then explanted and maintained in RPMI1640 (Thermo-Scientifc
and 50 ng/ml recombinant murine SCF (Miltenyi
All other chemicals were provided either by Sigma (Taufkirchen
Antibodies were purchased either from Thermo Scientific (Darmstadt
Germany) or from Cell Signaling Technologies (Leiden
0.5% N-lauroylsarcosine 1 mM PMSF and 1% HALT complete protease inhibitor cocktail) (Pierce
Precipitation for all samples was performed with protein G coupled paramagnetic beads (Cell Signaling Technologies)
Antibodies used for ChIP: anti-HA rabbit monoclonal
Cell Signaling Technologies (#3724) 5 µl per 5 x 10e6 cells; anti-C/ebpα rabbit monoclonal
Cell Signaling Technologies (#8178) 5 µl per 5 x 10e6 cells
ChIP sequencing libraries were prepared using NEBNext® Ultra™ II DNA Library Prep Kit reagents (NEB
MA) according to the procedure recommended by the manufacturer
Size selection was done after final PCR amplification with Illumina index primers for 14 cycles
Nascent RNA was converted into Illumina compatible libraries with NEBNext® Single Cell/Low Input RNA Library Prep reagents according to the standard protocol
Sequencing was done at the in house core facility yielding 100 bp single- or paired-end reads
Transcripts were quantified by Homer analyzeRNA routines and further analyzed with standard spreadsheet tools
Where appropriate two-tailed T-test statistics were applied

A short upstream reading frame (uORF) controls use of ATG start codons within the main coding sequence leading to expression of long (p42) and short (p30) C/ebpα isoforms
p30 retains only one of three transactivation domains present in p42
Replacement of the uORF by an optimized Kozak site allows isoform specific expression as shown in the western blot
B Experimental set up of inducible C/ebpα expression in HSPCs
Hematopoietic precursors isolated from animals with a knockout of neutrophilic proteases (Elane
Ctsg triple ko) were immortalized either by HoxA9 or MLLENL and subsequently transduced in the presence of the degrader dTAG13 with p42-FKBP or p30-FKBP constructs
Expression of the respective proteins was tested in the presence of dTAG13 and 24 h after release from degradation by western blot
HoxA9 x p42-FKBP cells were cultivated for the indicated time in medium without dTAG13 and morphological aspect
as well as expression of Myc and the differentiation sentinel gene Ngp (neutrophilic granule protein) were followed by May-Grünwald-Giemsa staining
Micrographs were taken on a Zeiss Axioskop with a Nikon camera at 63x magnification

Plotted are average expression values in the Egr1 off and on-states for those genes that showed a log2-fold difference between these conditions of at least 1.0
For representation expression values were averaged between duplicates
E Gene set enrichment analysis of the Egr1-controlled expression pattern
Please note that the Egr1-on state induces genes that are down-regulated by transformation with HoxA9/Meis1 (anti-correlation)
Trib1 was either added to cells pretransformed with HoxA9 or it was used for direct transduction of primary HSPCs
As Trib1 protein is highly unstable and cannot be detected by western blot
qPCR was used to confirm successful Trib1 RNA expression as depicted in the bar diagram
The lower panel shows a representative result (one out of three) of a replating assay demonstrating the transforming potential of Trib1 in comparison to HoxA9
Western blot experiments with extracts of HoxA9 (control)
and precursors directly transformed with Trib1 (Trib1) cells
The bar diagram depicts Myc RNA expression in the same cells (average and standard deviation of a technical triplicate)
Immunoblots were developed with antibodies as indicated
C Trib1 increases resistance against differentiation stimuli
Cells co-expressing Trib1 and HoxA9 (or HoxA9-FKBP) as well as controls were subject to induced differentiation either by cultivation in G-CSF or by degrading HoxA9-FKBP through addition of dTAG13 for 48 h
RNA was isolated at the indicated timepoints and expression of the differentiation sentinel gene Ngp was determined by qPCR
A Morphological as well as genomic response of HoxA9/HoxA9-FKBP transformed cells to induced differentiation
Cytospins were stained with May-Grünwald-Giemsa and photographed with a Zeiss Axioskop and a Nikon camera
Shown are 63x magnifications and the scale bar corresponds to 10 µM
Gene expression was determined for Myc and Ngp by qPCR
The figure shows a representative example of a duplicate experiment
B Western blot following endogenous protein expression in a time course during differentiation
Blots were developed with antibodies as indicated
C Intersection of Hox-mediated transformation with differentiation pathways at the level of the core cell cycle control machinery
CDK6 is a direct downstream target of HoxA9 and also MLL fusion proteins and CyclinD1 (CCND1) transcription is strongly induced by activated Myc
The schematic overview shows only part of the known interactions
these studies compared steady state wild-type cells that contained both
p42 and p30 to Cebpa mutants with p30 only
Here we manipulate each isoform directly and individually
although with the caveat that this always occurs on top of wt-expression
these findings point to cell cycle regulators as unexpected major players that coordinate differentiation control by C/ebpα and transforming inputs through Hox-proteins to determine cellular fate
Raw NGS reads were submitted to the European Nucleotide Archive under accession number PRJEB62028

A story is unfolding in Savannah involving several coincidences in history - long-term and short - uncovered by a disbelieving committee doing research at Savannah's Congregation Mickve Israel
They're calling it the Slany-Savannah connection
• The museum committee of Congregation Mickve Israel is working to showcase its 280-year past
Co-chairs Phoebe Kerness and Herbert Victor are spearheading the research and put together much of the Slany information
• A Torah collected during the Nazi occupation of Czechoslovakia is now on permanent loan and in weekly use at the Savannah synagogue
The base of the Torah is inscribed with its provenance
"This Scroll came from Slany and was written in 1890." It came to Savannah in 1968
She discovered via the internet that a B-17 bomber from the Eighth Air Force was shot down over the village of Slany
At the airport in Slany is a monument and a plaque dedicated "In memory of the crew of the American bomber shot down at this spot on 2 March 1945" - two months before the end of World War II
at Savannah Army Air Base-Hunter Field and generally credited with winning World War II in Europe.)
• Former Savannahian Jeffrey Young moved 20 years ago to the Czech Republic
but became intrigued with the growing connection of Savannah and Slany
His father is Savannah attorney Arnold Young.) He took pictures and sent them home
Most startling were the photos of the monument
that had been on a mission to bomb oil fields in Bohlen
He also translated the memorial's inscriptions
Vivian Rogers-Price is research center director of the National Museum of the Mighty Eighth Air Force
she directed a Mickve Israel museum subcommittee to original records about the mission
He was in the tail section of the plane and fell to the ground before it crashed
(Rogers-Price noted that the D-Day invasion would not have happened without the support of the Eighth Air Force
until this day the European people are grateful to the Mighty Eighth.)
• Mickve Israel Rabbi Robert Haas had planned a vacation this summer to the Czech Republic before the latest Slany-Savannah connection was uncovered
said a prayer at the monument and toured the synagogue building
the original home of Mickve Israel's Holocaust Torah
It still retains on its façade a large marble tablet of the Ten Commandments and a Star of David
the Torah is the centerpiece of its history
on parchment (or deerskin in earlier times) it contains the first five books of the Bible
week by week throughout the year - Genesis
It's the Old Testament and forms the basis of Western religion
At Mickve Israel it is read aloud in Hebrew
The Slany Torah is one of 1564 Czech Memorial Torahs confiscated and saved in Prague during the Nazi occupation
They have been distributed throughout the world as "permanent memorials to the martyrs from whose synagogues they come." A condition for custodianship of a Czech Torah is that it must be maintained in perfect condition
used regularly in religious observance and for education and it must be returned if a synagogue is re-established in that town
They were allowed to return in the early 19th century
In 1890 the Jewish population in Slany was 290
the 81 remaining Jews were deported to Theresienstadt (Terezin) concentration camp
In all the atrocities that the Nazis perpetrated during World War II on Jews and others
they were determined to preserve some of the trappings of the people they were destroying and created a stockpile of Jewish ceremonial objects
the Torahs were found stacked on wire shelving
During the 1960s an effort was made by Jewish organizations to find homes for the sacred scrolls where they would be used and cherished and would bring meaning to the lives of the victims who had perished in death camps in Europe
Savannah's Congregation Mickve Israel was one of hundreds of Jewish groups who accepted the responsibility to acquire a "holocaust" Torah on permanent loan
one was brought over with the original Jewish settlers
They are among the historic treasures of the congregation
taken out once a year at the anniversary of the synagogue's founding and are believed to be the oldest documented Torahs in the United States
now and probably forever will be known as the Slany Torah
and the modern Hebrew script is more legible
a Torah must be in perfect condition - no tears in the parchment
It was on condition that the Czech Torah be properly maintained that it was sent to Mickve Israel
the congregation will plan an appropriate welcome celebration

Export diversification has been among the most cited policy recommendations for African countries to spur structural transformation and increase resilience
export diversification that benefits structural change is not an automated process and requires an analytical approach and complex decision-making
Applying an adjusted economic complexity and product space methodology on trade data of 54 African countries and their trading partners
this paper assesses export diversification opportunities that are feasible to realize
associated with structural change and of high demand in the world and on the African continent
Increasing complementarities of African exports and imports are crucial to yield higher benefits from the African Continental Free Trade Area (AfCFTA)
The paper’s focus on intra-African diversification opportunities allows for a continental mapping of current exports with export diversification opportunities and the identification of niche areas of individual countries
The paper finds that almost all countries have some potential for product diversification into light manufacturing (machinery and mechanical appliances; electrical machinery) and processed base metal products (articles of iron and steel)
The paper’s findings can guide policymakers and development partners in identifying industrialization strategies and productive capacity needs

The Amorous Adventures of Bold Sir Hans Capon is the latest expansion for Warhorse’s hardcore medieval RPG
Some spoilers for the opening of the Amorous Adventures of Bold Sir Hans Capon follow
One of the standout characters in Kingdom Come is undoubtedly Hans Capon
But over time he warms to you and reveals himself to be a charismatic
thoroughly likeable character—something Warhorse was clearly aware of
wine-loving heir to the Rattay throne the focus of its latest paid-for DLC expansion
The Amorous Adventures of Bold Sir Hans Capon
can be accessed after completing the quests Next to Godliness and Robber Baron
Capon will tell you about a woman in Rattay he’s fallen for
and asks for your help in wooing her—without her knowing
and he wants me to help him win her heart secretly
The first stage of which is recovering a family heirloom
I travel to the tavern in Ledetchko where Capon lost the necklace
but the local dice champion tells me he already lost the heirloom to a fancy-looking chap who was last seen at a river crossing near the town of Sasau
I travel to Sasau and comb the river for clues until I find the nobleman in question—murdered and
The savage nature of the killing leads me to believe bandits were responsible
I'm wearing a raggedy shirt and battered armour
As I’m investigating the crime scene
a well-spoken man calling himself Sir Anselm of Domky emerges from the woods and tells me the nobleman was killed by bandits
I instinctively reach for my sword and get ready for a bit of brigand slaying
and infiltrate the camp to find out who the killer is—and maybe find that necklace I’m looking for in the process
Making sure I know enough about Alphonse to deal with any questioning in the bandit camp
Apparently he’s a keen dice player (no surprises there) from Slany and once worked for Prokop of Luxembourg
hoping they’ll save my skin later on
But there’s one problem: I look like shit
I’m wearing a raggedy shirt and a ramshackle collection of battered armour
I visit my house in Rattay and access my stash
where I have heaps of clothes and other junk I’ve pilfered and purchased over the course of the game
I find a fashionable cap with a feather in it
battle-scarred Henry looks like a prancing toff
I also stop in at the bathhouse to clean myself up and get a fresh new haircut and beard trim
I head to the bandit camp and announce myself to the guard
Keep up to date with the most important stories and the best deals
Immediately I’m glad I quizzed Anselm about Alphonse
saying I’m actually a native of Slany
but tells me that the bandit leader—presumably Alphonse’s killer—is busy
and that I should play some dice to pass the time
As I explore the camp and eavesdrop on the foul-mouthed bandits
I learn that the champion of a dice tournament wins a precious necklace: the very same one that once belonged to Capon before he foolishly lost it
I throw 1000 groschen into the pot and enter the tournament
but I absolutely stink at Kingdom Come’s dice minigame
and he tells me it’s a guy called Pechuna
I corner him and congratulate him on his victory
and consider buying the necklace off him or pickpocketing it while he sleeps
Instead I ask him how he’ll be celebrating
and he tells me he’s walking through the woods to Sasau to drink
I follow Pechuna through the trees until we’re well away from the camp
then I pull out my sword and run it through him
so the ‘fight’ is over in seconds
which will help the cocky lord begin his quest to win the heart of that lovely lady in Rattay
A nice final detail: as I look through Pechuna’s possessions I notice that he’s carrying loaded dice
Suddenly I don’t feel too bad about stabbing him in the back and leaving him to rot in the forest
We’ll have a full review up on the site soon
And next time you decide to try and swindle someone in a dice game
make sure they don’t have a sword stuffed down their silky pantaloons

Czech Republic (AP) — Some orders are impossible for manufacturers to turn down
A Czech maker of hospital beds received one this month
Hours before the Czech government announced that the armed forces would be building a field hospital for 500 COVID-19 patients at Prague’s exhibition ground
Prime Minister Andrej Babis approached the Linet company with a request to deliver the hospital beds for the facility
“It’s a very special situation for us,” Linet executive director Tomas Kolar told The Associated Press on Monday
Hectic negotiations between the company and the government are expected to result in the signing of a deal shortly for a total of 1,620 hospital beds
Kolar said such an order would normally take 12-18 months to fulfil
“it would be one week between the prime minister’s call and the first delivery.”
The local health system has been under pressure and hospitals are filling up with patients
As the number of people currently with the virus surpassed 100,000 in the Czech Republic
over 3,500 of them need hospitalization and almost 600 of those are in intensive care
according to government figures released on Monday
The number of new cases has tripled in the past two weeks
from a seven-day rolling daily average of 23.81 per 100,000 people on Oct
The Health Ministry predicted the system could reach its capacity around the end of October
such an order would not be a problem for a firm that is expected to make 120,000 beds this year
10,000 more than in 2019 and is considered the top European producer of hospital beds
But with the virus putting pressure on health systems across the globe
Kolar said the only thing to do to meet the government’s request was to delay some other contracts
“We have been negotiating that with our clients,” he said
we are able to meet the Czech demand,” he said
the United States and other European countries
It expects revenues to reach some 325 million euros ($383 million) this year
Czech hospitals have been working recently to increase the number of beds available for COVID-19 patients to some 10,000
The field hospital in Prague is meant to be used as a last resort

He’s a jokester known for his sense of humour and vitality—and did you know that Carl Booth once danced with Princess Elizabeth?
Booth is a 99-year-old resident at Evanston Summit Retirement Residence and he will be turning a spry 100 in March, prompting those at the facility to ask the public to send birthday cards to help celebrate this centennial milestone.
Victoria Slany works at the facility and says during the pandemic, they did the same thing for another resident when he turned 100. He received 651 cards so they wanted to do something similar for Booth, who is a beloved resident.
“Carl is one of our very funny residents who has been with us for about two years now. Let me tell you, he's quite the jokester and he is loaded with personality. He’s a really great guy, quite incredible,” Slany says.
A veteran, Booth joined the Army in 1943, taking his basic training in Edmonton and his advanced training in Calgary before serving at Aldershot in England, just outside of London.
That’s where he not only met the future queen of England, but he also had the confidence to ask her to dance.
“He claims that one night there was a dance at the base and Princess Elizabeth was in attendance. He was pestered by his buddies to go ask the Princess for a dance,” Slany says.
“Carl who is not shy about that sort of thing, went up to her and asked her to dance. She accepted and that's how Carl got to dance with the future monarch.”
Booth was also stationed in North Africa and Italy and is currently a member of the Army Navy & Airforce Club in Edmonton. He's also a massive Edmonton Oilers fan.
The greetings have already started flowing in for Booth, and Slany is touched by all of the sweet words and attention the man is starting to receive. He’s even received a birthday card from as far as Amsterdam already.
“It’s so incredibly heartwarming and really cool to see all of the people that want to make it happen,” Slany says.
The goal is to receive at least 100 cards, which will be hung around the residence. On his actual March 7 birthday, there will be a full day of celebrations and surprises.
“It’s so incredibly special to be able to see the reactions and to be able to share that moment with them. To be able to create a memory for them, it’s so special.”
Slany says this special celebration wouldn’t be possible without the help of gracious citizens from all over the world.
“I just want to say thank you so much for helping to make Carl’s 100th birthday so memorable,” Slany adds.
Birthday cards can be dropped off or mailed by Feb. 15 to Evanston Summit:

Germany is realigning its development policy partnerships around migration
Hubertus Heil is delighted. Heil is Germany’s Federal Minister of Labour. He and Svenja Schulze, the Federal Minister for Economic Cooperation and Development
children welcomed the two ministers with singing and dancing
“It really makes your heart swell,” says Heil
Karola Slany from Berlin set up this primary school 27 years ago
Many of her protégés have since gone on to secondary education and even university
but no-one has yet made it as far as Germany
she says: “Germany is a long way off for our children.”
The Federal Government agreed the key points in late November 2022 and the proposed amendments to the Skilled Immigration Act will be presented to the Bundestag in the near future

The intention is to ensure as far as possible that the movement of skilled workers to Germany does not worsen the situation in their countries of origin
Heil and Schulze used their visit to Accra to send a positive message about migration
“This is about the economies of both countries being able to benefit,” Heil says
“That’s why it’s good to work together and create a win-win situation,” Schulze adds
While in Accra, Schulze and Heil are setting a new strategic direction for the Ghanaian-German Migration Advice Centre
The centre is located right next door to Ghana’s Ministry of Employment and will help those interested in moving to Germany and the EU find ways of getting there
Ghanaians who want to emigrate will also be able to find out how to start by first trying a move to a different country in Africa
The Centre will also continue its work supporting Ghanaians returning from Germany
helping them to regain a foothold in their homeland
Germany has promised support which includes help with setting up a business
“If this gives the impression that we want to take bright and skilled professionals away from this country
He notes that despite Ghana having a population of just under 34 million
but there is a surplus of well educated people who cannot find a job
Ghana’s population is predicted to grow by a further 7 million over the next 10 years
“That’s why it’s important that we ensure in more ways than one that migration is fair.”
Compared to some other countries in West Africa
Many young people who attended Karola Slany’s school as children now work in Ghana as teachers
Ghana still suffers from inflation running at over 50% and the economy is struggling
On the issue of illegal movements of refugees
Heil explains that “What we’re trying to do is prevent people from taking lethal journeys across the Sahara.” Under the Skilled Immigration Act
people wanting to emigrate to Germany will be chosen according to their professional experience or links to Germany
More centres like that in Accra are planned for Morocco
