Tell us about your participation in the Walla Walla Valley
I was born 39 years ago at Saint Mary's Hospital in Walla Walla to Mexican immigrant parents who
After spending twelve years away—pursuing my education and exploring the world—I returned to the valley two years ago
now accompanied by my two children and partner
tending to the roots that have sustained me while putting down new roots in the place that shaped me
I currently serve as the Executive Director of the Walla Walla Immigrant Rights Coalition
an organization founded in 2016 that formally established its first board of directors and executive director just last year
My work and my family are at the heart of my life
my participation in the Walla Walla Valley
each deeply intertwined and inseparable.
My commitment to racial justice is woven into the fabric of my daily life and participation in the Wally Walla Valley as well
I strive to ensure that immigrants in our valley are not only recognized for their inherent value
but also have access to the resources and opportunities that empower them to thrive
equitable community that will foster a brighter future for everyone who calls the Valley their home
Outside of work, I serve on the board for the Walla Walla Community Council and love to explore new businesses in the valley
I coached our son’s soccer team and enjoy supporting and spending time with my community of friends and family.
How does what you do fit into or address larger issues in our region
As the Executive Director of the Walla Walla Immigrant Rights Coalition
I see my role and our organization as pivotal in addressing the intersection of immigration
and community integration within a region deeply shaped by agriculture and the agriculture economy
relies on immigrant labor to sustain its agricultural economy
yet immigrant communities often face barriers to full participation in the social
Our organization seeks to help bridge these gaps
ensuring immigrants not only have access to vital resources and legal protections but also that their contributions to the region’s prosperity are recognized and valued
The challenges faced by immigrants in Walla Walla—whether related to legal rights
or healthcare—are emblematic of larger national issues surrounding immigration reform
By advocating for policies that protect immigrant workers
ensuring they have access to education and healthcare
our work addresses these systemic inequalities on a local level
while contributing to broader efforts for justice and equity within the region and beyond.
What is something you’ve learned about this area that surprised you or that you weren’t expecting
There isn’t enough room to adequately answer this question–and that is why I love the question
I read this question as an invitation to reflect on the very nature of diversity itself–what is it
the diversity of Walla Walla is perhaps most visible in the tapestry of people who call this place home—from long-established families whose roots go back generations
to the more recent waves of immigrants drawn here by seasonal work in the vineyards and fields
bring with them not only their labor but also their languages
enriching the cultural fabric of the valley in ways that continue to evolve
Walla Walla is a microcosm of the broader American experience—a place where generations of settlers
sometimes uneasily but often with a shared sense of place.
What do you think this region will be like in five or 10 years
I believe Walla Walla will undergo significant changes
particularly in terms of its economy and demographics
driven by the increasing influx of high-wealth individuals
I’ve witnessed firsthand how concentrated wealth in mountain towns can dramatically shift the dynamics of a community
often leading to rising living costs and widening economic disparities
While Walla Walla is different in many ways—its diverse economic institutions
and manageable population size give it a unique stability—there is a growing trend toward attracting more affluent residents
particularly with the arrival of wealthier individuals and families
Walla Walla will need to find innovative solutions to ensure that the community remains accessible to people of all income levels
The challenge will be balancing the benefits of increased wealth and investment with the need to create affordable housing options and maintain a healthy
vibrant community where families can thrive without being displaced by rising costs
it will likely see greater diversification
with more opportunities in industries like tech
This shift could attract even more people from outside the region
leading to a more cosmopolitan mix of residents while still preserving the small-town charm that makes Walla Walla unique
The next decade will likely be a time of growing pains
but also an opportunity for the community to define its identity in a way that ensures its economic and cultural vitality is accessible to all
If you could be involved in an aspect of Walla Walla life that you are not currently involved with
If I had the time and opportunity to be more involved in an aspect of Walla Walla life that I’m not currently engaged with
I would love to return to working with youth
While my current commitments—raising two young children and serving as Executive Director of the Walla Walla Immigrant Rights Coalition—are incredibly fulfilling
I look forward to being more active in my children’s activities as they grow and discovering where there might be gaps in available youth programming
I have a background in teaching and coaching dance
and I would love to eventually bring those skills back into the community
I see tremendous value in extracurricular activities
I hope to be able to contribute to Walla Walla’s youth programs
or helping to shape new opportunities that are accessible to all kids
especially those in underserved communities
Is there anything else Whitman students should know about our community that you haven’t mentioned yet?
One thing I’d like Whitman students to know about our community is how deeply interconnected Walla Walla’s people are—across different backgrounds
it’s a place where diverse experiences and histories meet
particularly when it comes to its immigrant communities
but it’s also a place of transformation and opportunity
with a growing focus on inclusivity and community-building
The changes happening here—such as changes in business
and the changes in our national administration—bring both challenges and exciting opportunities
especially in terms of creating a more equitable
accessible town for people of all backgrounds
There’s also a real sense of belonging here
and residents work together to support one another
there’s a shared understanding that the health of this community depends on how well we care for and uplift each other
Whitman students who engage with Walla Walla have the chance to make a meaningful impact—whether it’s through academic work
or simply connecting with people who call this place home like me.

For lovers of vinyl and streetwear, there’s a new drop spinning into focus. Japanese hip-hop icon and globally respected DJ, Takayoshi Murota—better known as DJ Muro—has teamed up with Japanese beer brand Kirin Ichiban and Australian label Deus Records to launch a limited-edition capsule celebrating the golden age of crate digging
Fresh off his three-stop tour through Sydney and Melbourne
the artist has debuted a specially crafted ‘record hunting jacket’ and matching tee exclusively for the Australian market
This collaboration isn’t just merch—it’s a wearable tribute to the artistry of analog sound
and the unshakable legacy of first pressings
More Than Merch: A Nod to the Diggin’ Lifestyle
DJ Muro has earned his global reputation not only through his skills behind the decks but also as a tastemaker with an encyclopedic knowledge of records
“The King of Diggin’,” isn’t just a moniker—it’s a lifestyle
The launch of the record hunting jacket is a tangible extension of that ethos
created for those who share a reverence for the hunt
Crafted in Tokyo and designed in partnership with Deus Ex Machina
the jacket reimagines functionality for vinyl heads who know the value of a good find
Its rear-facing pocket is built to hold up to 10 full-sized LPs—a feature made with serious diggers in mind
The utilitarian build combines traditional Japanese craftsmanship with subtle but intentional design upgrades
The collaboration draws on the energy of Muro’s live performances, particularly his recent sets across Australia
Each show during the tour added momentum to the release
as the DJ sported the jacket on stage—turning the piece into a live
A closer look reveals its thoughtful construction: multiple storage compartments resized to cradle records
a cable-secure lapel outfitted with a Kirin Ichiban press stud
and a visual flourish in the form of an embroidered illustration adorning the sleeve and back pocket
The entire piece serves as an homage to first pressings—not just the records themselves
but the idea that there’s something sacred about the initial moment music meets medium
I think it’s the most special.” That sentiment drives the capsule’s ethos: preserving the emotional resonance of the first encounter—whether with a rare record or a limited-edition piece of wearable art
Australian-Only Pressing with Global Vibes
Adding to the rarity of this drop is its geographic exclusivity
This ‘first pressed’ edition was created specifically for Australia
featuring a unique colorway and exclusive embellishments that won’t appear in global versions
The Kirin Ichiban accents—like the branded snap closures and distinctive illustrations—tie the piece directly to the collaborative spirit behind the collection
The partnership also includes a black unisex cotton tee
created in the signature graphic style of Deus
with vintage Kirin-inspired branding that tips its cap to vinyl lovers everywhere
and made to accompany the jacket without competing for attention
a collectors’ edition DJ Muro vinyl is also included—a fitting final detail for a drop that places so much weight on tactile authenticity
Stevie James of Deus Ex Machina summed it up best: “The Record Hunting Jacket combines Japanese values of function and craftsmanship with crate-digger culture.” That fusion is exactly what makes this collaboration stand out
This drop creates a space where underground music culture
it speaks directly to a community that values stories
this release offers a moment of connection—a tribute to analog love in a digital world
The capsule is available exclusively through Kirin Ichiban’s website and Instagram
and quantities are expected to move quickly
With the jacket’s previous iterations already selling out twice
this Australian version brings both scarcity and cultural depth to the table
For anyone chasing the high of the first press—or just looking to add a uniquely crafted piece to their rotation—this collaboration delivers
It’s not just a nod to vinyl culture
stitched and pressed into something you can wear

The watershed legislation of the Biden years continues to loom large as the Biden administration winds down
the bills loom historically large and account for some $3.8 trillion in outlays aimed at modernizing the U.S
What comes after them in the next administration
And yet, beyond the big bills’ scale, something else matters intensely about this legislation: the inclusion within it of nearly $80 billion in explicitly “place-based” industrial policy that is now being tested in the nation’s regions
Authorized by the 117th Congress but in many cases under appropriated, the nearly two dozen individual place-based economic development programs embedded in the big policy packages stand out as a critical experiment for a nation that has been struggling with stark regional divides and uneven economic progress
The “place-based” experiment matters because “place-based” policies—which channel investments or other interventions into particular locations—reflect a growing recognition that the nation’s economy is itself place-based—shaped by a tilework of distinct local economies—and requires place-focused problem-solving
As such, the new place-based economic investments have put into action compelling new programs such as the Economic Development Administration’s Regional Technology and Innovation Hubs and the National Science Foundation’s Regional Innovation Engines that take a fresh stab at engaging more directly
and efficiently with the roots of the nation’s economic issues
Such programs purport to support “bottom-up” local problem-solving that gets at the “micro” underpinnings of broader “macro” challenges
whether it be building up the nation’s semiconductor sector in Phoenix to compete with China or improving worker training for an advanced mobility cluster in Tulsa
the presidential election may bring a major decision point
The presidential candidates each point to the China competition and the struggles of places “left behind,” but they would likely train very different views on place-based initiatives
murmurings on Capitol Hill continue to suggest that there could be bipartisan investment in place-based programs like the Tech Hubs in the next administration
The investment would likely be oriented toward technology scale-up on topics like AI that are vital to the China competition
the election will likely greatly affect the scope of the next phase of place-based policy in the U.S
While a Harris administration would likely seek to extend the experiment
Trump might well let the experiment peter out
the stakes are high as the nation continues to test new “place-based” programs to counter the regional inequality that continues to divide the nation
the nation has on the table sizable new tools for addressing the stubborn economic problems that wrack the scores of local communities following deindustrialization
MEMPHIS, Tenn. (10/9/24) – Freshman Carla Muro Castellvi advanced to the singles main draw on Wednesday
as the Arkansas State tennis team picked up five singles wins on the first day of the ITA Central Regional Championships
Fellow freshman Lily Cvanciger later prevailed in a marathon three-setter that lasted nearly four hours
6-1 outcome to Kansas State's Veronika Kulhava
Sarah Millard took the opening set against Iowa's Nikita Vishwase
but ultimately fell in three by a score of 2-6
Millard then dropped a consolation match to Saniya Singh of Tulsa
The doubles main draw begins Thursday, with Fernandez and Thurmer opening up against Nebraska's Lucy Loy and Ana Carmen Zamburek at 9 a.m. At 10 a.m., Cvanciger and Anna Grigoreva will face off against another Nebraska pair in Reya Coe and Emma Rizzetto
while Millard and Muro Castellvi will take on Nicolly Ferreira and Aryn Walker of Saint Louis
in the main draw against Alice Amendola of host Memphis

Victor Muro Velásquez, a respected Mexican lighting engineer known for his work on Alfonso Cuarón's Oscar-winning film "Roma" (2018)
was found dismembered in two separate locations
shocking the film industry and the general public alike
Authorities have apprehended a suspect believed to be connected to the crime
which they have stated is linked to personal issues rather than organized crime
Muro's death has left a significant void in the Mexican film industry
He had an extensive career that spanned several decades
contributing to substantial productions like "The Crime of Father Amaro" (2002) by Carlos Carrera and "The Sons of the Jaguar" (2019) by Arturo Sánchez del Villar
His work on" Roma," which won several Academy Awards
elevated his industry status and highlighted his cinematography expertise
"Muro Velásquez" worked on other titles such as "Cero y van 4" (2004)
and "Estoy todo lo iguana que se puede" (2022)

Muro's dismembered remains were discovered on October 7
Authorities reported that one of the suspects was an acquaintance of the victim
confirmed that a suspect has been detained
and investigations are ongoing to clarify the motives behind the brutal crime
A connection to organized crime has been ruled out
Governor García's statements regarding Muro's death have sparked outrage on social media
with many users accusing him of not taking Muro's profession into consideration in light of the heinous nature of the crime
The remarks have been interpreted as insensitive

authorities were led to Poeta Jesús Díaz Street
near the intersection with 5 de Febrero Avenue
where traces of blood were found in a room
detectives discovered a human torso and head
Reports indicate that Muro Velásquez had rented this place along with three other individuals with whom he was reportedly planning to make a short film
Three suspects were initially brought in for questioning
with evidence reportedly suggesting his involvement in the case
Muro had recently arrived in Xalapa to teach a workshop on cinematographic lighting at a private school—an endeavor he often engaged in to share his knowledge and experience with aspiring filmmakers
the production company that had worked with Muro Velásquez
expressed its condolences and lamented his untimely death in a public statement
His colleagues and friends will remember him not only for his professional excellence but also for his human qualities and the mentorship he provided on set

Fentanyl was found on the nightstand of the victim’s bedroom
The Boone County Sheriff's Office wrote in a Friday night social media post that Victoria Chapman was the victim in the case

Court documents say law enforcement examined the Chapman's phone and allegedly found text messages between her and Muro that appeared to set up a drug deal
Location data described in the heavily redacted probable cause statement says Chapman went to a store before heading home
The phone did not leave the residence after she returned
Muro and Chapman were allegedly seen on camera at the store earlier
Muro allegedly sent a text message to another person claiming he sold Chapman a “gram of fenny,” the statement says
Muro was later found with drugs during a traffic stop on June 25

Galleria Continua Habana is pleased to present Muro de sonido (Wall of sound)
a solo exhibition by Cuban artist José Yaque
This pictorial and spatial immersion engages in dialogue with energy
Yaque has developed a deep interest in the study of minerals and tectonic formations of the Earth
exploring the origin of color from mineral pigments
he has evoked the geological processes that shape the natural world and has captured the energy of nature from its entrails
This fascination with geology has translated into a methodology where paint flows freely
allowing chance and gravity to play a crucial role in the construction of the image
he obtains an organic relationship between the different acting elements
and the resulting painting becomes a reflection of the processes of nature
his lyrical abstraction has expanded his horizons and the colors have emerged from the sediments to the Earth’s surface
The artist is enthusiastic about the idea of painting as nature paints rather than seeking realism in representation
the center of the gallery is occupied by a river of pigments that seems to be born from the distillation of colors generated from the entrance
It is a pictorial spring that sprouts and expands freely
appropriating the space and generating chromatic combinations almost independently
the water obtain protagonism : not only for the tones that can be perceptible but for the flow that is generated in the form of painting and that intervenes the main room of the gallery more than simply occupying it
The exhibition becomes a great landscape where each work pays tribute to a new step in the pictorial inquiries within the artist’s career
José Yaque experiments here with a new way of looking at the landscape
the landscape is not a figurative representation
but is rather constructed as an allusive landscape
forces and vibrations of different places visited by the artist and which served as inspiration for his paintings
he works with the idea that nature is not only a theme in painting
but that painting itself can be conducted as a natural process
The artist takes references from native areas
such as the Salinas de Brito (Brito salt flats) in Ciénaga de Zapata (Zapata swamp)
from which he extracts the stimulus that the different tonalities provoke and of which he only leaves a slight hint in the pieces’ titles
Also inspired by the concept of the “wall of sound” generated in the field of music in the sixties of the last century
the artist seeks to translate the intensity
resonance and immersion of sound into painting
The large installation and the paintings exhibited for this show function as visual chords
where the superimposition of layers and the fluidity of the material generate a sensation of almost audible vibration
This approach allows the viewer to “hear” the painting through the vibration of color and composition
expanding preset boundaries and amplifying the senses
The viewer not only has the opportunity to observe the work
experience it and immerse him / herself in it
as if he/ she were within the pictorial landscape itself
Muro de sonido (Wall of sound) invites viewers to experience painting not only as a visual representation
a flow of matter and sensations that envelops them
The exhibition seeks to expand the limits of perception and provoke in the public a reflection on the relationship between art
José Eduardo Yaque Llorente was born in Manzanillo
He studied at “ Carlos Enríquez ” Professional Academy of Fine Arts in Manzanillo (2005) and the Higher Institute of Art (ISA) in Havana (2011)
In 2010 he participated in the first Biennial of Contemporary Art in Portugal and exhibited at Wasps Artists’ Studios

I spent the day with the renowned chef and restaurateur sourcing rainbow trout in Montebello and watching her assemble a perfect crudo at her restaurant in the Village
One of the most complete and enjoyable restaurant experiences anywhere
April 9, 2025e-Paper
Brigade Road and its adjoining lanes have always been a favoured haunt for old Bangloreans — the bookshops
jamming sessions and a chance to hobnob with kindred spirits
While most of it is now changing with familiar landmarks being replaced by newer ones
its quintessential soul still remains with a newer generation walking down its once-green paths
One place to capture that vibe of old is Muro
It is kind of hard to remember what used to stand there but then you stop wracking your brains as this beautifully lush restaurant beckons
The greenery and earth tones are a throwback to the city of yore and the fact that they have live music every Wednesday evening is a huge plus
Muro translates to wall in many languages and “is a symbol of a culinary challenge for those brave enough to scale it.” With places offering the “pan-Asian” experience popping up all over the city
Muro rests its case in the capable hands of chefs Somporn Chaisuntorn and Sanil Mohan
the duo serve up a little bit of Thailand with every bite
We begin with a pomelo salad with bird’s eye chilli and cashew nut
which was a refreshing change from the tried and tested
No matter what you order — be it a smoothie
it seems almost a shame to deconstruct what was so beautifully put together
The staff recommends the crispy pork cheung-fun and we are glad we took them up on it
The soft casing belies the crunch underneath as well as the delicate flavour coriander root and scallions add to each bite
Muro has a wonderful choice of vegetables in their menu — from chive flowers and banana blossoms to Chinese greens
We try the vegetables in pumpkin sauce next — lotus stem
yam bean and shimeji mushrooms come together in a mildly spiced
creamy dish that is quite unlike other small plates we have had anywhere else
The other recommendation we got was to try the turnip cake
puttu and sushi rice was forming at the back of my mind
but I am so glad my imagination disappointed
carrot and mushroom are steamed and then stir fried
served as cute bite-sized cubes and garnished with garlic; there was nothing turnip-y about them
I would say it was the biggest delight of the afternoon
we settled for the lamb massaman curry and jasmine rice
making it a comfortable choice for those who are not too keen on experimenting
we strayed from our usual choices of chocolate or cheesecake and tried the coconut caramel coffee flan
“expecting the unexpected” pretty much summed up our time at Muro
and the caramel ice cream with creamy coconut and coffee crumble was nothing less
There is no way to do justice to their spread in one sitting with noodles
baos and more beckoning you on a gastronomic trip

explained some of the ways her health system is using technology to make nurses’ jobs easier — including software that flags patients who are at risk of adverse outcomes and a tool that helps frontline nurse managers better engage with their staff
Before she became the CIO of Bay Area-based El Camino Health
She knows firsthand just how stressful and draining the nursing profession can be — which is part of the reason why she believes hospitals should be finding more ways to leverage technology to make these workers’ days easier
“For our labor force, we want to remove responsibilities that can be automated — because then we can focus the limited resources that we have on the most important work,” Muro said this month during an interview at the Reuters Digital Health conference in San Diego
have begun to automate the entry of vital signs into the EHR
She also said that El Camino is starting to explore virtual nursing and its potential to speed up processes like admission and discharge
one of the things that concerned her most deeply was that she couldn’t spend very much time with any individual patient when she had a high patient load
most nurses have a pretty high patient load
El Camino rolled out a tool in its EHR that its workers refer to as the deterioration index.
“It takes data from the patient record — the vital signs
all of those parameters — and predicts which patients are trending downward and who could have a bad event
It then signals a team to come and assess the patient when they appear to be on a downward spiral,” Muro explained
“That has prevented code blues — and it’s prevented the nurses who are caring for those patients from having to see their patient have a bad outcome
She noted that this tool fits in well with the greater philosophy at El Camino — which is that it’s always better to be predictive than reactive
Muro highlighted Laudio’s technology as another tool that has been helpful in El Camino’s journey to make nurses happier
The startup’s technology assists frontline nurse managers by integrating their core workflows into a single platform and automating their repetitive work
and it also has features to help managers better engage with their staff
Laudio’s platform sends nurse managers reminders about team members’ birthdays and work anniversaries so that they can recognize them
It also gives managers reminders to schedule check-ins with new hires
as well as alerts them when a nurse has been working long hours and might be at risk of burnout
“We’re excited to use more of that technology to enable the manager to know which nurse or staff member they need to spend more time with
they can intervene before the person decides to leave
or decides to leave the field totally,” Muro declared
She also pointed out something important El Camino does that not all health systems do — involve their nurses in the IT department
For the nurse informaticists in El Camino’s IT department
the main goal is to make sure that nurses’ wants and needs are accounted for when the hospital develops
These nurse informaticists always present plans for new tech programs to their nursing councils and gather feedback
which is used to inform the program’s design and rollout

Intracellular organelle communication enables the maintenance of tissue homeostasis and health through synchronized adaptive processes triggered by environmental cues
Mitochondrial-Endoplasmic Reticulum (ER) communication sustains cellular fitness by adjusting protein synthesis and degradation
and metabolite and protein trafficking through organelle membranes
Mitochondrial-ER communication is bidirectional and requires that the ER-components of the Integrated Stress Response signal to mitochondria upon activation and
mitochondria signal to the ER under conditions of metabolite and protein overload to maintain proper functionality and ensure cellular survival
Declines in the mitochondrial-ER communication occur upon ageing and correlate with the onset of a myriad of heterogeneous age-related diseases such as obesity
the exploration of the molecular mechanisms of mitochondrial-ER signaling and regulation will provide insights into the most fundamental cellular adaptive processes with important therapeutical opportunities
we will discuss the pathways and mechanisms of mitochondrial-ER communication at the mitochondrial-ER interface and their implications in health and disease
the maintenance of mitochondrial-ER communication ensures the execution of appropriate cellular responses upon external stimuli and the maintenance of organismal health
MERCs provide dual signaling routes that synchronize nuclear-transduced signals with rapid and local adaptive responses that regulate mitochondrial function
The landscape of protein-protein contacts that occur at the MERCs is at the intersection of several signaling pathways that connect mitochondrial-ER communication and transduce signals to other organelles
we will discuss the mechanisms of mitochondrial-ER communication that allow cellular adaptation and survival
We will focus on the subset of protein interactions and signaling pathways upon external stimuli that act as messengers between the ER and mitochondria and shape the nature of the adaptive responses at the ER-OMM crossroad
Mitochondrial-ER communication regulates protein homeostasis
OMM protein biogenesis is regulated by the insertases MTCH1/2 for α-helical proteins and the tandem TOM40-SAM50 for β-barrel proteins
Nuclear encoded mitochondrial proteins routed towards the intermembrane space
IMM and mitochondrial matrix are imported through the TOM40 pore at the OMM
which is facilitated by OMM receptors TOM70 and TOM20 through the binding of mitochondrial targeting sequences
PERK activation regulates TOM70 activity and dependent mitochondrial protein import through OGT
Quality control mechanisms in mammals at the OMM is dependent on the activity of the OMM AAA-containing ATPase ATAD1
which extracts clogged proteins and presents them to p97/VCP with subsequent proteosome degradation
Import and assembly of nuclear-encoded mitochondrial proteins is coordinated with mitochondrial-encoded proteins
The mitoribosome co-translationally inserts nascent peptides into the IMM through a direct interaction with the OXA1L insertase
Nuclear-encoded proteins that have been imported into the intermembrane space are further translocated into the IMM through the TIM complexes
nascent peptides derived from both nuclear and mitochondrial genomes are assembled into ETC complexes
mitochondrial and cytosolic translation rates are coordinated during mitochondrial biogenesis
and the ER membrane serves as a buffer of mitochondrial targeted peptides through the GET- and ER-SURF pathways during this process
the translation rates and ER-association capacity of cytosolic ribosomes may be dictated by mitochondrial translation
These processes may impact the UPR through the proximity of IRE1α to the Sec translocon
the PERK ISR attenuates cytosolic translation through direct phosphorylation of eIF2α
and potentially their presence at the ER-membrane
These pathways play important roles in normal physiological processes including thermal adaptation
Defects herein are associated with obesity
whether it be mTOR or ISR regulation has been implicated in pro-survival mechanisms in a variety of diseases including cancer where targeting mTOR and PERK have been proposed as new generation therapeutic strategies in different contexts
The role of the mitochondria and concomitant MERCs in the regulation of global translation processes is an area of current interest in the field and representative examples of mitochondrial and cytosolic translation regulation will be presented in the coming sections
nuclear-encoded mitochondrial subunits are imported
and assembled with mitochondrial subunits in a tightly controlled receptor- and chaperone mediated process; to be further detailed in a subsequent section of this review
cytosolic and mitochondrial translation systems are coordinated to ensure proper stoichiometry under dynamic and changing nutrient environments; tuning respiratory processes in cooperation with ISR and UPR (ex
further illustrating the benefit of localized protein translation and mitochondrial import in close proximity to the ER
The importance of LRPPRC in maintaining mitochondrial translation and coordinating proteostasis between the mitochondria
and ER highlights a complex organization across compartments that may require cross-talk between mitochondrial translation factors
These studies provide further insights into potential factors involved in the maintenance of mito-nuclear balance and potentially their underlying translation systems that when dysregulated can result in pathology
and mitochondrial translation system are in close proximity to the ER where protein translation
and degradation can be dynamically monitored by the UPR
The involvement of TRAP1 in numerous aspects of mito-nuclear balance offers an additional example of chaperone-mediated translational control that supports protein homeostasis at MAMs
It can be reasoned that the dynamic expression of TRAP1 and related chaperones can respond to challenging microenvironments supporting protein homeostasis in the context of mitochondrial biogenesis in normal physiology and disease
we will discuss their roles in context of physiology and disease
The incorporation of proteins to the outer mitochondrial membrane is possible through three main pathways: i) spontaneous insertion (Doan et al., 2020), ii) chaperone-driven (Drwesh et al., 2022) and/or iii) insertase-driven (Becker et al., 2011; Diederichs et al., 2020; Guna et al., 2022)
The biochemical nature of the different proteins defines the pathways utilized
Two distinct protein conformations populate the OMM
β-barrel proteins such as TOM40 or SAM50 and α-helical components including TOM70 or TOM20
While the involvement of mitochondrial protein import by mammalian MTCH1/2 and the development of pathologies needs further study
it is clear that the understanding of the mechanisms of OMM insertion may provide new therapeutical opportunities for metabolic and age-related diseases
These studies highlight the relevance of mitochondrial protein import in the maintenance of mitochondrial homeostasis which otherwise degenerates in pathologies
the transduction of stress signals from the ER to mitochondria can dictate the dynamics of mitochondrial protein import and provide new mechanisms to understand the regulation of these machineries in a pathophysiological context
the overload of unfolded proteins needs specific processing to avoid the accumulation of undesirable protein conformations that can negatively impact cellular functions
30% of recently translated proteins need to be removed due to improper folding
This phenomenon can also happen at the MAMs and hamper the adaptive capability of the cells
The ER is one of the main stores of non-imported mitochondrial precursors to avoid the saturation of the import machineries
and the ERAD systems are quality control mechanism that surveil the MAMs space to facilitate the transduction of stress signals from the ER to mitochondria
we will focus on the protein quality control mechanisms at the MAMs and their involvement in pathophysiological processes
A limiting aspect in understanding protein clearance from mitochondria resides in the absence of some mammalian orthologs that need to be discovered explored
Lipid and calcium trafficking through the mitochondrial-ER contacts
Lipid trafficking is possible through subsets of protein-protein interactions between the ER and OMM
Phosphatidic acid (PA) is generated by three different pathways in the ER
PERK can also generate PA which is supplied to mitochondria to synthesize cardiolipin (CDL)
Mitochondrial DHODH activity can also crosstalk to the ER to generate ether lipids that can modulate mitochondrial IMM compositions
Phosphatidylserine transport is allowed through multiple pathways including ORP5/8-SAM50-MICOS
NME3 can be attracted by clusters of PA molecules and participate in mitochondrial membrane dynamics
VPS13D provides a structural hydrophobic tunnel which is thought to transport lipid species across the ER and mitochondria
Calcium trafficking allows mitochondrial function and the release from the ER to mitochondrial is controlled by SERCA2 and IP3R channels
Calcium can be transported through the VAPB-PTPIP51 and the IP3R-VDAC-MCU pathways
Non-canonical roles of IRE1α suggest a control over the latter
TOM70 has been found to also participate in the signaling of calcium to mitochondria
multiple protein-protein contacts participate in the delivery and transport of PS species between the ER and mitochondria
albeit their mechanisms and/or targets remain still unsolved
lipids are signaling molecules that constitute a hydrophobic solvent that defines cellular components and separates chemical reactions and need to be synchronized with the protein machineries
The capacity to selectively recruit non-mitochondrial-ER proteins under specific cellular conditions illustrates how the surrounding protein environment define the function and dynamics of MERCs and cellular adaptive responses
mitochondria represent the predominant source of cellular ROS and their proximity to ER membranes may provide a direct target of ROS in cancers that lead to oncogenic Ca2+ dysfunction and ER-stress
These representative examples provide clear evidence of the importance of Ca2+ in cancer and how the ER and mitochondria respond and communicate to its dysfunction
Mitochondrial-ER communication is at the intersection of fundamental biological processes and associates with the development of several metabolic diseases described before including
Initial approaches targeting the ISR were focused on the treatment of pathological processes where protein accumulation and aggregation occur
The discovery of other roles for the three main branches of the ISR (ATF6
IRE1α and PERK) controlling multiple aspects of mitochondrial function and physiology have highlighted the importance of targeting mitochondrial-ER communication as emerging therapies
As such the beneficial or deleterious effects of UPR responses hinge on the degree of activation
repurposing of FDA approved tetracyclines for mitochondrial disease therapy may be promising
the therapeutical applications of mitochondrial-ER activation require fine tuning and need further investigation in order to provide efficient therapies for metabolic disorders
The mechanisms that allow mitochondrial-ER communication provide an effective evolutionary adaptive cellular fitness
we have covered the regulatory mechanisms that regulate protein synthesis
trafficking and degradation focusing at the ER-OMM interface
Despite the current knowledge of some of these processes in mammals
their connection with pathological disorders or even adaptive processes is still elusive
some of the essential regulatory mechanisms that have been studied in non-mammalian systems do not have orthologs
yet they participate in quality control processes such as protein insertion or unclogging that are fundamental
studying the influence of ISR component activation in shaping the ER membrane-OMM landscape will provide insights into how stress responses crosstalk to mitochondria to promote cell survival and/or protect against diseases
A common signature upon defective activation of the ISR branches is the association with the development of age-related diseases such as obesity
This agrees with the current models where mitochondrial-ER communication vanishes over time and impinges on the importance of studying how the reversal of these interferences can protect from the development of these pathologies
Along with the protein-protein interactions
metabolic fluxes are central for the buildup of cellular structures such as mitochondria
which undergo profound cristae remodeling and biogenesis during stress responses such as fasting in liver or cold stimulation in brown adipocytes
calcium and lipid trafficking results essential for the function and remodeling of mitochondria respectively
it is highly unclear how lipid species are transported from the ER to mitochondria
whether the known pathways can reverse the flow of lipids and which forces drive such transport
VPS13D has been postulated as major transporter of lipids due to the hydrophobicity of its central tunnel
many other protein contacts have been shown to participate in the delivery of phospholipids to mitochondria even though the species are not well known
Calcium flux between the mitochondria and ER has been established through the protein-protein interactions and activation of ER and mitochondrial Ca2+ channels
in cases where there are not direct mitochondrial ER contacts (ex
mechanisms by which ER-resident Ca2+ is directed specifically to the mitochondria to promote respiration requires further investigation
the role of Ca2+ in the development of cancer has been appreciated
but pharmacological targeting has not yet been effectively explored in the context of clinical oncogenesis
Future work on the study of ER and mitochondrial Ca 2+ and lipid compositions across pathophysiological conditions in cells and in vivo is necessary to precisely understand lipidome remodeling in the context of cellular and tissue adaptation and the molecular basis of disease
