Time series data are recorded in various sectors
the continuity of these data is often interrupted
Several algorithms are used to impute the missing data
and the performance of these methods is widely varied
the effective imputation depends on the nature of missing and available data
We conducted extensive studies using different types of time series data
specifically heart rate data and power consumption data
We generated the missing data for different time spans and imputed using different algorithms with binned data of different sizes
The performance was evaluated using the root mean square error (RMSE) metric
We observed a reduction in RMSE when using binned data compared to the entire dataset
particularly in the case of the expectation–maximization (EM) algorithm
We found that RMSE was reduced when using binned data for 1-
with greater reduction observed for 15-min missing data
We also observed the effect of data fluctuation
We conclude that the usefulness of binned data depends precisely on the span of missing data
and quantity of the missing and available data
binned data can impute a wide variety of data
including biological heart rate data derived from the Internet of Things (IoT) device smartwatch and non-biological data such as household power consumption data
we extend our studies on the utilization of binned data for missing data of different time spans of univariate time series data collected from different sources
We primarily used heart rate data derived from smartwatches worn by the healthy volunteers
We synthetically made the missing data for 1- and 5-min time spans and then imputed the data using different algorithms
The performance of imputation was quantified by using root mean square error (RMSE) as a metric
we report on the circumstances that determine that binned data will provide better imputation compared to the imputation using the entire dataset
sampling rate of the data are crucial factors
We also report the role of data fluctuation using missing data from different time periods of day depending on whether the volunteers remain physically active or inactive
We further extended our studies to non-physiological data
We investigated the effectiveness of binned data in another time series data of power consumption
heart rate data which are biological data and power consumption data
and the second was publicly available data
Heart rate data was collected as a part of the clinical trial NCT05106725
This is an IRB-approved non-interventional study
and informed consent was collected from all subjects recruited in the trial
The data were extracted from the wearable IoT device Fitbit Inspire 2 worn by four healthy male volunteers
The volunteers wore their respective devices over the whole day
we selected 30 random days for each volunteer having no missing data
The heart rate data were available at 5-s intervals and converted to 1-min intervals after preprocessing
The household electric power consumption database was downloaded from the UC Irvine Machine Learning (Hebrail and Berard, 2012)
It is an energy consumption dataset for a house
containing over 2 million measurements recorded at an interval of 1 min from December 2006 to November 2010
we used 1 month's (March 2007) data to evaluate the efficiency of using binned data for imputation
We synthetically generated missing data by deleting data from days having complete data
we imputed the missing data and compared the imputed data with the original data
We used data for two different time frames of a day
which we refer to as the “active period” and “inactive period.” The “inactive period” refers to 3–4 a.m
when our volunteers were asleep and hence inactive
The “active period” refers to 3–4 p.m
when our volunteers were active in their different daily activities
A similar time span was chosen for power consumption data too
during the “inactive period,” the power usage was stable compared to the “active period.”
The data from different days were binned into different sizes around the missing data period
we used bins with bin sizes starting from 5 min
In the case of power consumption data—while imputing 15-min missing data—we used bin sizes starting from 15 min
RMSE calculates the disparity between the actual and imputed values
providing a quantitative measure of the accuracy of our models in handling missing data
during the “active period” of the volunteer

As lifestyle follows some daily repetitive activities
heart rate pattern also carries that profile
for imputing missing data of a particular period of any day
data from other days around the same time period are adequate
the time span should be sufficient to carry the signature of the regularly repetitive work
Reduction of time span also implies reduction of data points
the sampling rate of the data is also crucial
This is the reason that the binned data are more effective for imputation of missing data of a higher time span of 15 min compared to the 5 min and even lower 1-min missing data
We conclude that the time span of missing data and sampling rate play a competitive role

The use of binned data implies the use of data from a specific span. In other words, it uses censored time span data (Støvring and Kristiansen, 2011)
It is advantageous as it uses less amount of data
hence making faster imputation of real-time data possible
As it considers data from a specified span
the presence of noise and outliers beyond that span does not affect the imputation process
The binning method is also applicable to a wide variety of imputation algorithms
the major challenge of using binned data for imputing missing data occurs when data of missing periods deviate from the usual characteristics of the available data of similar time periods and the span of other days
The activity of a person is reflected in his or her heart rate data
If a person deviates from their routine activity for a particular day and if that data are missing
the use of binned data may not be useful for imputation
The technical challenge of using binned data includes generating binned data of optimal bin size
Since time series data often carry personalized features
the choice of bin size also needs to be personalized
The so-called “active” and “inactive” periods may vary for different data sources
We observed that using binned data provides better-imputed data for different types of time series data
The advantage of the use of binned data includes the requirement of a lesser amount of data along with faster computation
It can be integrated into big data analytics to fill the missing part
It will help in the faster functioning of automated devices such as IoT devices
Our extensive study shows that the usefulness of binned data is determined by the innate properties of the data
which include the span of the missing period
and the fluctuation within the available data
the optimal bin size could not be defined because of the lesser effectiveness of binned data when the sampling rate is comparable to the missing data time span
the current study will be helpful for time series data management across wide domains

We further compare our results with other records from the West/Central Mediterranean and West African region to discuss the similarities/differences between the identified paleoclimate periods from the penultimate glacial period to the Holocene and suggest the atmospheric-oceanic circulation mechanisms that may have caused the different humidity conditions.

The newly developed SnSiZer method was applied to identify the statistically significant precipitation changes as well as the relative magnitude of the humidity oscillations
we identify six main WMHPs: WMHP-6 (180–155 kyr BP)
WMHP-2 (27–18.5 kyr BP) and WMHP-1 (15.5–5 kyr BP)
with the identified WMHPs and their comparison with West/Central Mediterranean and West African records
both West/Central Mediterranean and West African climates are in phase
with roughly coeval humid periods in both regions
west Mediterranean and west African records are out of phase during low seasonality
This could be explained by the westerlies and the influence of the local Mediterranean rainfall flux affecting the Mediterranean region

The two 15-year-old students from San Jose National High School returned home on Saturday night after seeing the missing persons reports filed by their families.
Jobert Padul and Trisha Jane Teodones were reported missing on September 28 when both teens failed to come home after their classes. Their parents filed police reports and alerted local media outlets when they were unable to contact them.
In an interview with Palawan News, Arlene Teodones, the mother of Trisha, said that they returned on September 30 to the Padul residence in Brgy. San Manuel, where they were reunited with their parents and questioned by the police about their whereabouts.
The teens had stayed in cottages near BM Beach but decided to return on Saturday after turning on their phones and seeing the messages and posts searching for both of them. They reportedly took a long commute from BM Beach and got lost in Brgy. San Jose before circling back to San Manuel, as they did not have experience in taking public transportation by themselves.
Arlene was relieved upon learning of the teens’ safety, noting that the two were unharmed and feared the backlash from their noted absence.
Arlene said that she was more disappointed by the vulgar comments found under the teens’ missing person’s posts on social media outlets.
“Grabe po kasi yung mga comments nila, masakit sa dibdib yung mga nababasa ko. Sana hindi naman ganoon kasi mga minor naman ito, wala naman sa isip nila yung mga ganoong bagay,” Arlene said.
She said that her, her husband, and Jobert’s parents were mostly worried about the two facing harassment from their peers once they return to school. The parties had agreed that for the benefit of their well-being, they will temporarily have modular classes instead of going to school.
Arlene expressed her gratitude to the authorities who helped assist in bringing the two of them home.

Neoantigen vaccines are based on epitopes of antigenic parts of mutant proteins expressed in cancer cells
These highly immunogenic antigens may trigger the immune system to combat cancer cells
Improvements in sequencing technology and computational tools have resulted in several clinical trials of neoantigen vaccines on cancer patients
we have looked into the design of the vaccines which are undergoing several clinical trials
and challenges associated with the design of neoantigens
We searched different databases to track the ongoing clinical trials and their reported outcomes
the vaccines boost the immune system to combat the cancer cells while maintaining a reasonable margin of safety
Detection of neoantigens has led to the development of several databases
Adjuvants also play a catalytic role in improving the efficacy of the vaccine
we can conclude that the efficacy of vaccines can make it a potential treatment across different types of cancers
and even frameshift mutations in genes that may or may not be oncogenes or tumor suppressor genes
they are more often used as neoantigen candidates
Below we have discussed the reported outcomes in the neoantigen vaccine research

Each of these pipelines has its selection process and results in the lists of neopeptides
these lists very often differ from each other
we will discuss the generalized approach required for vaccine design
The personalized neopeptide vaccine design requires information on the mutations in proteins which are translated from the mutation sites of the DNA in cancer cells
This information is extracted by comparing the DNA sequences of normal and tumor cells
Either whole genome sequencing (WGS) or whole exome sequencing (WES) data of DNA of both tumor and normal cells is required
only exonic parts are only translated to peptides
WES is sufficient to detect somatic mutations
WES is more economical considering both clinical and computational costs
the mutated proteins should be expressed in the tumor cells
to ensure that mRNA sequencing of tumor cell mRNAs is also performed
This post-processing includes the removal of duplicate reads
which originate from the same fragment of the DNA
Indel realignment is also recommended by realigning reads near detected indels to remove alignment artifacts
the base quality score recalibration (BQSR) is performed using GATK
the systematic errors made by the sequencer while calling the bases are estimated and base quality scores are calibrated accordingly
These recalibrated BAM files are further used for identifying different genetic variants

Since a vcf file may contain hundreds of mutations
the associated information is used for prioritizing the possible neopeptides
The mRNA expression strength and variant allele frequency are also considered for the final selection of peptides
and helper peptides like tetanus are also used in the formulation
The administration process includes subcutaneous
and intravenous injections mostly in limbs
The targeted somatic mutations should ideally be present in all cancer cells
These can be founder mutations that initiated cancer and thus possibly may be present in all lineage cells that form the bulk of cancer tissue
There may exist multiple subclonal mutations; consequently it may be better to target the dominant clone which may be present in the bulk of the cancer tissue
Selection of clonal and subclonal mutations can be achieved by establishing cancer cell content in the tissue used for sequencing and comparing mutant allele frequency with wild type/normal allele frequency
the variant allele frequency should be preferably up to 50%
for the mutation to be considered for the vaccine target
the maximum allele frequency will approach 1.0
should be compatible with the physiological environment
the entire process should be cost-effective in terms of time
We noted that there are many clinical trials currently ongoing and can be accessed via the NIH ClinicalTrials website (https://www.clinicaltrials.gov/ct2/home). The trials which involve the administration of vaccines on human subjects are listed and tabulated in Table 1
We observed that trials mostly involve multiple types of cancers
and are also dedicated to specific sites like pancreatic cancer and breast cancer
We provide a brief review of different cancer types covered by neoepitope/neoantigen clinical trials

The response of the vaccines also depends on the adjuvant drugs
The ongoing trial NCT04161755 uses the drug atezolizumab along with mFOLFIRINOX in the context of pancreatic cancer patients undergoing neoantigen vaccine therapy
Neoantigen vaccine therapy is being tried in breast cancer patients; specifically
triple negative breast cancers (TNBC) where genetic instability is associated with a high mutational burden
up to 25 neopeptides are going to be administered by the autologous dendritic cells over a 16 week span
Another clinical trial NCT04879888 also uses peptide pulsed autologous dendritic cells at six doses on nine TNBC patients
The clinical trial NCT03199040 is designed to evaluate the response of neoantigen vaccines in the presence and absence of the drug durvalumab in triple negative breast cancer patients
is an ongoing phase 1 trial of a polyepitope DNA vaccine against triple negative breast cancer
The immunogenicity and safety of the vaccine are being evaluated in the trial
The anti-PDL1 antibody drug pembrolizumab is being assessed in the neoantigen vaccine trial NCT03568058 in NSCLC
head and neck squamous cell carcinoma (HNSCC)
classical Hodgkin lymphoma (cHL) and other solid tumors
This study will observe the immune response when pembrolizumab is administered six weeks before vaccination
Genocea Biosciences is also conducting a clinical trial (NCT03633110) on 24 participants having different cancers
This trial also uses the drug pembrolizumab along with nivolumab to evaluate the efficacy of vaccine therapy
NCT3300843 was also initiated for pan-cancer study using peptide loaded dendritic cell vaccines but was terminated due to low accrual
Individualized VB10.NEO vaccine and bempegaldesleukin (NKTR-214) are being used in the clinical trial NCT03548467 for patients at the metastatic stage
It plans for 14 vaccinations for each of the 65 patients and bempegaldesleukin (NKTR-214) will be given after at least four doses of vaccinations
The primary goal is to measure the safety and adverse effects of the vaccine
Secondary outcome measurement includes measuring immunogenicity by T cell activity to each neoepitope
Similar to the trial NCT04161755, clinical trial NCT03359239 aims to determine the effects of atezolizumab in combination with a personalized cancer vaccine, PGV001 (84) for locally advanced or metastatic urothelial cancer patients
A clinical trial is also evaluating the immune response of a shared antigen vaccine PROSTVAC and tumor specific antigens generated DNA vaccine with nivolumab (anti-PD-1)
and ipilimumab (anti-CTLA-4) for checkpoint blockade (NCT03532217)
The ongoing open label phase 1a/1b clinical trial (NCT03970382) is focusing to evaluate the efficacy
and safety of NeoTCR-P1 T cells in subjects with metastatic hormone-sensitive prostate cancer
NCT03040791 is another trial involving pancreatic cancer patients which also utilizes nivolumab to explore DNA repair defects (DRD)
mainly in the Homologous Recombination (HR) pathway
The effect of nivolumab is also being investigated with or without ipilimumab in female patients suffering epithelial ovarian
or fallopian tube cancer in clinical trial NCT02498600
The outcomes will be measured as per response evaluation criteria in solid tumors
and incidence of adverse events in advanced stages of the disease
Clinical trial NCT04455503 also treats advanced melanoma patients but with two types of EVX-02 vaccines with nivolumab in two cohorts
Depending on the study results the third cohort will receive either one of the two types of EVX-02 vaccine
This study will measure safety and tolerability by measuring vital signs like heart rate
Neoepitope-specific T cells will be monitored by ELISPOT
Other pharmacodynamic responses of EVX-02 will be assessed by MHC I multimer analyses detecting neoepitope-recognizing CD8+ T cells and by flow cytometry to detect vaccine induced intracellular cytokine response
Relapse free survival period will be measured as secondary outcomes
A trial (NCT03422094) based on neoantigen vaccine therapy on glioblastoma patients was initiated but later focus was changed to cell therapy
Clinical trial NCT02510950 targeting glioblastoma patients did not proceed due to financial limitations
We observed variations in the vaccine formulation and administration strategies followed by different trials
The number of chosen peptides varied from 5 - 20 depending on the mutational burden
These peptides are often applied with adjuvants
Poly-ICLC is used as an adjuvant in multiple trials
Poly-ICLC stimulates the release of cytokines and the production of interferon-gamma
The administration process and doses also vary
and subcutaneous injections at limb organs are used for administration
The dose typically remains around 100μg per peptide
The treatment typically continues for several months
Table 1 lists different formulation and administration strategies observed in the trials
we observe that the clinical trials employing neoantigens are showing promising results in terms of immunogenicity and safety
on-time delivery of these personalized vaccines to patients remains a challenge
These databases help to find neopeptides whenever a mutation is detected
the proper selection of adjuvant drugs at appropriate doses and times plays a crucial role in the success of neoantigen immunotherapy
We observe that the neoantigen vaccines appear safe with limited side effects
the survival period is still not promising
This may be a result of the majority of the trials currently ongoing are conducted on patients who have already reached the metastatic stage or the late stage of the disease
An early intervention with neoantigen vaccines may provide a longer survival period for the patients
It needs to be validated by clinical trials in the future
It makes neoantigens specific to the tumor stage and more trials are needed for exploring patients of different stages
As mentioned in the introductions section and based on the NIH clinical trial website
we noted several clinical trials that are about to initiated
The results from those studies will provide a better landscape on the therapeutic efficacy of neoantigen immunotherapy
we conclude neoantigen vaccines are capable of exhibiting tumor-specific immunogenicity in different types of solid tumors
They leverage CD4+ and CD8+ effector T cells across cancer types
there is an enormous requirement for improvements in several aspects like the optimized design of neoantigens to ensure the efficacy of the vaccine
Conducting ex vivo studies on the effect of peptides on tumor cells collected from patients will be helpful for a well-defined vaccine design
Further studies are required to evaluate the possibility of the existence of patient subtypes based on the responses to neopeptides
it will make the vaccine production process more economical both in terms of money and time
Researchers and clinicians should explore the possibility of applying vaccines to patients at the earlier stages of the disease which may provide a longer survival period

the prosecutors junked the motion for reconsideration against the resolution that dismissed the murder complaint against cops tagged in the killing
The motion, dated May 2, 2023, was filed by Rosenda Lemita, mother of Ana Mariz “Chai” Lemita-Evangelista, and Ariel Evangelista’s mother-in-law. Lemita’s motion came after the prosecutors junked the complaint against the cops in March this year due to insufficiency of evidence

Complainant only alleges that ‘Spouses Evangelista were targeted pursuant to a virtual kill list under COPLAN ASVAL,” the resolution read
adding that no evidence was submitted to support the claim
