Extracellular vesicles (EVs) play a crucial role as potent signal transducers among cells
with the potential to operate cross-species and cross-kingdom communication
Nanoalgosomes are a subtype of EVs recently identified and isolated from microalgae
Microalgae represent a natural bioresource with the capacity to produce several secondary metabolites with a broad range of biological activities and commercial applications
The present study highlights the upstream and downstream processes required for the scalable production of nanoalgosomes from cultures of the marine microalgae Tetraselmis chuii
and conditions were assessed to improve EVs isolation by tangential flow filtration (TFF)
The optimization of the overall bioprocess was enhanced by quality control checks operated through robust biophysical and biochemical characterizations
we showed the possibility of recycling by TFF microalgae cells post-EVs isolation for multiple EV production cycles
The present results highlight the potential of nanoalgosome production as a scalable
cost-effective bioprocess suitable for diverse scientific and industrial exploitations
we demonstrate the possibility to recycle microalgal biomass after EVs harvesting to renew the cell culture and continue EVs production in a cyclic bioprocess (renewable processing)
This capability to go through several production/isolation cycles further increases the interest of microalgae as a sustainable and renewable biosources of EVs

USA) was used to isolate microalgae-derived EVs
Microalgae cultures (1.6 L) were clarified by sequential micro- and ultra-filtration using TFF hollow fiber filters (MiniKros Sampler) with cut-off of 650 nm (S04-E65U-07-N
Three different settings were evaluated: feed flow 750 ml/min and permeate flow 60 ml/min
feed flow 450 ml/min and permeate flow 6 ml/min
and feed flow 450 ml/min and permeate flow 6 ml/min followed by a wash of the TFF cartridges with 100 ml of culture medium
During all filtration processes transmembrane pressure (TMP) was kept constant at 0.02 bar
The small and large EVs recovered from the retentate of the 500-kDa and 200 nm cut-off TFF filter modules
were concentrated until a final volume of almost 150 ml
using a smaller 500-kDa cut-off TFF filter module (C02-E500-10-N
MicroKros) with a feed flow 75 ml/min and a permeate flow 2 ml/min
samples were further concentrated and diafiltrated seven times with PBS
reaching a final volume of approximately 5 ml
the retentate obtained from the 650 nm cut-off TFF cartridge (100 ml) was diluted in modified f/2 medium to reach the initial batch volume (1.6 L) and used to start renewed cultures in bottles via a 25% v/v inoculum
microalgae were again processed by TFF to isolate EVs
necessary for the recycle of the microalgae cell culture
the 650 nm TFF membrane filter was washed with 1 L of sterile water before starting the clarification process
for the first TFF step the instrument and the bioreactors are connected in a closed system to maintain sterility
The EVs protein content was quantified using the colorimetric BCA protein assay (Thermo Fisher Scientific
The protein concentration was measured at 562 nm
according to the manufacturer’s instructions

The Western blot analysis was executed using sodium dodecyl-sulfate (SDS) polyacrylamide gel electrophoresis (PAGE); 10 μg of cell lysate and 5 μg EV samples (in PBS) were incubated at 100°C for 5 min with 5× loading buffer (0.25 M Tris-Cl pH 6.8
0.25% bromophenol blue) and loaded on 10% SDS polyacrylamide gel for electrophoresis
Polyvinylidene fluoride (PVDF) membranes are used to blot proteins
The membranes were blocked with BSA-TBS-T solution [3% powdered with bovine serum albumin in TBST (50 mM Tris HCl pH 8.0

In the present work, we isolated EVs from the marine chlorophyte microalgae T. chuii (Figure 1A). This species was selected from a set of several microalgal strains as one of the best candidates for EVs production (Adamo et al., 2021; Picciotto et al., 2021)
We established a permanent platform for microalgae cultivation at pilot scale
Different cultivation methods are available for microalgae, in brief (Henley, 2019):
• batch—a given volume of culture medium is inoculated with cell culture at low density and then processed during exponential growth to obtain a maximum yield;
• fed batch—a supplement of culture medium and nutrients periodically feeds the cell culture without removing the biomass before harvesting;
• semicontinuous—a fixed volume of cell culture is harvested at given time intervals and replaced with fresh culture medium;
• continuous—the harvested culture is drained out of and the fresh medium is fluxed in the bioreactor continuously to maintain a constant biomass concentration
The harvested cultures are then pooled and processed at the same time
The harvesting time depends upon the life cycle of T. chuii as well as the culture conditions, such as the amount of the inoculated culture. Specifically, after 4 weeks the cell density increases to reach a maximum, as measured by periodic optical density measurements and cell counting. Along with the cultures fated to EV production, we maintained a refreshed stock culture, as described in the general flow chart of Figure 1C
In our previous work, we have used both differential ultracentrifugation (dUC) and tangential flow filtration (TFF) to isolate EVs from microalgae, confirming the above described expectations for TFF performance (Adamo et al., 2021). Thus, we implemented the following procedure based on sequential TFF filtration steps (Figure 2)
and feed for the three filters used in sequence: (i) 650 nm
(iii) 20 nm (namely 500 kDa)
The harvested cell culture is fluxed through a fiber with a cut-off of 650 nm to remove the biomass
The clarified permeate from the previous step is fluxed through a fiber with a cut-off of 200 nm; this step allows to isolate EVs smaller than 200 nm in the permeate by removing larger objects
including large EVs or cellular debris which escaped the first clarification
The isolated permeate from the previous step is fluxed through a fiber with a cut-off of 500 kDa
which corresponds to approximately 20 nm; this step allows to remove small particles
maintaining small EVs in the retentate; at the same time
the retentate volume is reduced down to the fiber volume (150 ml in the current setting)
The ultrafiltered retentate from the previous step is fluxed through a fiber with the same cut-off (500 kDa) and a lower volume and filter surface; this step allows to further concentrate the sample by reducing the volume down to the volume of the filter module and the tubing (5 ml in the current setting)
a culture with high protein content may result in quick membrane fouling affecting all TFF steps and preventing an efficient recovery
In order to optimize the TFF protocol, the specific parameters controlling the process must be adjusted individually for each type of culture medium (Moleirinho et al., 2019)
Two important parameters to maximize EV yield are the inlet and outlet flow rates: more specifically
We evaluated three different conditions for each filtration step:
(A) Fin = 750 ml min−1
Fout = 60 ml min−1;
(B) Fin = 450 ml min−1
Fout = 6 ml min−1;
(C) Fin = 450 ml min−1
Fout = 6 ml min−1 and filter module wash
likely due to the prevention of any membrane fouling
An additional washing step (condition C) may also improve particle recovery

After the optimization of upstream and downstream processing, we were able to produce EVs at a lab scale, with a maximum yield of 2 mg EVs for every 5 L of cell culture and approximately every 5 g of dry biomass. Also, our production platform showed high reproducibility and quality over different production cycles, as discussed in the present work and in our previous studies (Adamo et al., 2021; Picciotto et al., 2021)
we explore the possibility to link upstream and downstream processes by implementing a cyclic bioprocess for nanoalgosomes isolation
Since microalgae cells are concentrated during the first TFF filtration step
the corresponding retentate could be used to seed a new culture via appropriate dilution with fresh medium (modified f/2 medium) and subjected to further sequential filtration after 4 weeks
This requires that microalgal cells are not damaged during the first TFF step
when the cell culture is depleted from the released vesicles

protein mass concentration determined by BCA assay
This quantity is a standard parameter in the EV field; its measure can be done by using a specific kit and a spectrophotometer or a colorimeter; thus it is quite cheap and easily accessible in every laboratory
it is a widely used procedure and thus it is very useful to compare measurements from different studies and different samples
Other colorimetric methods measuring protein mass concentration
an accurate measurement of protein concentration typically requires a concentrated sample
so it is often taken after a concentration step
which introduces the possibility of sample loss and a biased measurement
particle number concentration measured by NTA
The number of particles is the ideal quantification for each sample and it is becoming another standard parameter with the increasing availability of NTA instruments
or also Resistive Pulse Sensing techniques
which allow to track and count each particle in a sample
There are two main drawbacks: the first is the intrinsic limit of detection of NTA instruments which are less sensitive to objects with a size below 100 nm (or with a very large size)
giving a constitutive bias to the measure of number concentration; the second is the intrinsic incomplete sampling of the particle population; indeed
both a short experimental duration and a different setting of the acquisition parameters may lead to large differences in the particle count
which only a highly trained operator can reliably and partly suppress
one may note that the variability in the data does not allow to infer one quantity by simply measuring the other one
we recommend to measure both quantities to complete any batch characterization

In any case, the correlation shown in Figure 7B works as an a posteriori calibration of the particle number with respect to the “hard number” of Rayleigh ratio
which can be quickly measured to assess EV concentration and make a reliable and accurate batch to batch comparison

The role of EVs in cell communication is attracting increasing interest from several clinical and biological fields. This emerging relevance is also supported by the application of EVs for clinical diagnosis and liquid biopsy (Ayers et al., 2019; Trino et al., 2021)
their potential exploitation as efficient drug delivery systems boosted the interest in their biotechnological exploitation

it is required to adopt new strategies for their massive production at high purity level or
Here, we addressed the production of nanoalgosomes, EVs derived from microalgae recently identified and characterized in our recent work (Adamo et al., 2021; Picciotto et al., 2021)
Both upstream and downstream processing steps have been optimized to maximize EV yields
we showed that it is possible to operate microalgal production cycles using TFF-derived culture inocula to facilitate a cyclical production of nanoalgosomes
The optimization of EV production was achieved by implementing quality control checks
which included the use of several biophysical and biochemical methods for EV characterization
As highlighted in the present work and accounted in previous studies (Paganini et al., 2019; Adamo et al., 2021), nanoalgosomes have different competitive advantages with respect to EVs derived from other sources (Figure 8): 1) Sustainability
They are obtained by a sustainable “green” biosource: they can be seen as more appealing for an exploitation as drug carriers than EVs from human or animal sources
which have inherent safety and ethical issues
The optimized TFF based bioprocess is suitable for a large scale production: for any large-scale exploitation
a cell suspension has a definitive advantage with respect to other green sources
which require more time-consuming and expensive treatments
3) Renewability—the potential recycling of TFF-concentrated microalgal cells to facilitate the scaled production

Post-traumatic stress disorder (PTSD) and anxiety disorders are among the most prevalent psychiatric conditions worldwide sharing many clinical manifestations and
neural mechanisms as suggested by neuroimaging research
While the so-called fear circuitry and traditional limbic structures of the brain
have been extensively studied in sufferers of these disorders
the cerebellum has been relatively underexplored
The aim of this paper was to present a mini-review of functional (task-activity or resting-state connectivity) and structural (gray matter volume) results on the cerebellum as reported in magnetic resonance imaging studies of patients with PTSD or anxiety disorders (49 selected studies in 1,494 patients)
regarding the direction of effects and anatomical localization
cerebellar structures like the vermis seem to be highly involved
the neurofunctional and structural alterations reported for the cerebellum in excessive anxiety and trauma are complex
although mixed results are found across disorders
While it can be hypothesized that individual differences in cerebellar activation underlie reactivity to stressors (Moreno-Rius, 2019) and the risk for developing anxiety and stress-related disorders (Caulfield and Servatius, 2013)
a clear understanding of how the cerebellum contributes to excessive anxiety and stress is lacking
The aim of this mini-review was to describe the main findings
of human neuroimaging studies using structural (sMRI) or functional (fMRI) magnetic resonance imaging
in adult patients suffering from PTSD or anxiety (SAD
with adult participants (>18 years of age) that had received a clinical diagnosis
in either patient-control or pre-post-treatment comparisons were included
Exclusion criteria were research articles that used: (I) another neuroimaging modality than sMRI or fMRI; (II) pediatric or adolescent populations
or healthy individuals only; (III) neuropsychiatric disorders different than the targeted PTSD/anxiety disorders; and (IV) meta-analyses

Over 500 children from FBCD Catarroja and Atlético Sedaví CF begin training at the Club's facilities in Paterna
Valencia CF opens the Ciutat Esportiva de Paterna to the clubs of the #EscolesVCF programme affected by the flash floods
whose facilities have been seriously damaged
so that children and youngsters can resume their sporting activities
Around 500 players from FBCD Catarroja and Atlético Sedaví CF have started training this week at the Valencia CF Ciutat Esportiva facilities that the Club has offered to the 4 most affected VCF Schools so that they can make use of them over the coming months
The entrance was already an emotional experience for these young footballers who have been affected by the flash floods
For some of them it was their first time at the Ciutat Esportiva del Valencia CF
they received the warmth of the VCF Academy players and coaches who made a corridor as a sign of support and solidarity with all of them
Just by seeing the children's faces and their smiles
you realise how they have experienced it and how much it helps them
Seeing the children on a football pitch is what we have been wanting for a long time

50-year-old Michael Paterna is charged with DUI manslaughter and vehicular homicide for a crash on County Road 3280 and Magnolia Road from July 2.
Paterna was operating a 2005 Yamaha motorcycle and traveling east on County Road 3280 with a female passenger
throwing both him and the passenger into a fence
The passenger did not survive and was pronounced dead on scene
Paterna suffered serious injuries and was taken to Fort Walton Beach Medical
A search warrant was obtained for Paterna's blood samples
which came back with results that tested positive for alcohol
Florida Highway Patrol Corporal James Durham was able to reconstruct the crash and discovered Paterna was traveling between 66 and 70 miles per hour in a 35 mile per hour zone
Paterna was arrested and booked into Walton County Jail
He was released Thursday afternoon on $50,000 bond

On Saturday July 28, Salvatore (“Tori”) and Susanne Paterna celebrated their 50th wedding anniversary at Boston’s Seaport Hotel. Tori, now retired, formerly owned and operated John Solomon’s Inc. on Somerville Avenue in Somerville. Until retirement, Tori and Susanne lived on Bailey Road, in Somerville’s Ten Hills neighborhood. They recently moved to New Hampshire and keep a winter home in Florida. Happy 50th wedding anniversary Tori and Sue!

Families finally recover the remains of 21 of the 3,400 Republicans executed after the Spanish civil war
The families who gathered in a Spanish cemetery beneath Saturday’s burning sun had waited 82 years
They weren’t going to let the 30C heat stop them doing what they had come to do
Pit 111 of Paterna cemetery in the Valencia region finally gave up the remains of 21 of the 3,400 Republicans who were summarily executed by Franco’s forces after the end of the Spanish civil war
and whose bodies were tossed into an enormous and infamous mass grave
were returned to the families of the murdered in a solemn ceremony
which has worked to reunite the bodies of the dead with their descendants
said a last goodbye to his wife and children

we’d already been through a year of misery
We didn’t have to look for his remains for years because my mother always knew which grave he was in.”
For those who gathered in Paterna on Saturday, the event was as overdue as it was welcome. Eighty-three years after it ended, the civil war and the subsequent dictatorship continue to cause personal pain and political skirmishes
Spain’s socialist-led government is hoping to build on landmark legislation from 2007 with a “democratic memory” bill intended to “settle Spanish democracy’s debt to its past”
The plans have been criticised by the opposition conservative People’s party
which says the proposed legislation will serve only to “dig up grudges” and churn up the earth that was carefully tamped down with the post-Franco amnesty law and the so-called pact of forgetting

countries with a growing interest in football
have enjoyed a unique experience in VCF's Training Stage
The players who traveled to Valencia had participated in the VCF Soccer Camp last summer in Albania and Montenegro and with this Training Stage they have had the opportunity throughout a training session with the coaching staff of the VCF Academy
The experience allowed them to get to know first-hand based on the methodology of one of the best academies in Europe in training players for the elite
according to the latest reports from the CIES Football Observatory
they were able to play some games with teams from the VCF Academy and the VCF Escoles
as well as enjoying with their families and coaches a complete experience on the Mestalla Tour
a Valencia CF home game in Mestalla and a VCF Femenino match in the Antonio Puchades
The presence of these players at the Ciutat Esportiva de Paterna is one more step in their approach to Valencia CF after having participated this summer in the VCF Soccer Camp held in Tivat (Montenegro) and in Tirana (Albania)

photographer and graphic designer Daniel Paterna is dishing up more than mouthwatering recipes in his new cookbook
“Feast of the Seven Fishes: A Brooklyn Italian’s Recipes Celebrating Food & Family.” Through lovingly rendered photographs
readers also get a taste of the neighborhood’s little Italian bakeries
alongside a highly personal family memoir.
The Bensonhurst backdrop gives rich context to recipes for dishes like arancini
a traditional Italian cheesecake made with ricotta instead of cream cheese
“I think anyone that has it is kind of over the moon,” said Paterna
Paterna began to think about the food and the culture of Bensonhurst as something to be preserved after a few years away in Boston in the ‘90s
he wanted to document his family’s Italian traditions like the Feast of the Seven Fishes
a meal with seven different seafood dishes customarily served on Christmas Eve.
I started to photograph my mom cooking,” Paterna said
this is a very unique thing that we do every year.”
Paterna also started visiting Bensonhurst’s “negozi,” or little Italian shops
many of which have been run by the same families for generations

“I was committed to shoot everything on film,” Paterna said
“I felt that the work I was presenting and the concepts
the emotions I was evoking were all taking place in the film era.”
Paterna’s grandfather left Naples and settled in Red Hook around 1915
right at the tail end of a massive wave of immigration that brought roughly two million Italian immigrants to the city
Like many of the grandchildren of those immigrants
known for its Italian shops and restaurants and for its annual street fair honoring Santa Rosalia
When Paterna was growing up, the neighborhood was also known for its mob presence, and for outbursts of racial violence and bigotry
One of the goals for the book that Paterna had from the outset was to portray Bensonhurst’s “B-Side,” the side of the middle class
Italian-American neighborhood that defies stereotypes.
Paterna says he encountered those stereotypes often
People would ask if he knew anybody in the Mafia
Coworkers found his Brooklyn accent intimidating
and one even suggested speech therapy.
“I always felt like we grew up in a very dignified house,” said Paterna
“My mother abhorred ‘The Sopranos.’ There’s no identification
And I think that’s because it was all too real for us… you step out in the street
you could run numbers or be part of an organized crime outfit very easily
sang in the choir at church and was a seamstress and a bank teller
She spoke Italian and would help new immigrants to set up bank accounts
establish themselves in the neighborhood.
Her reputation in the community helped Paterna as he was visiting shops in Bensonhurst to photograph and interview the sometimes-reluctant owners
It also came in handy after he finished the book
he mailed an advance copy to Michael Lomonaco
the former Food Network host and chef at Windows on the World.
he had worked at a bank with Paterna’s mother.
‘Had I not seen the word ‘Paterna’ on that FedEx
I would have never opened it,’” Paterna said
“I really feel like my mom had a direct hand.”
Lomonaco read the book and wrote a glowing preface
“This deeply personal book is meant to move you
to help you understand the story of one family
and to see that so many other families lived much like this.”
Paterna says the book is a way to preserve not just his own family’s traditions
but the traditions of a changing neighborhood.
“Me photographing these stores and the area was sort of like a way to hold onto what’s left,” Paterna said
“I think that’s what people are reacting to when they see the book.”

A mini VCF World Cup will be held at the Ciutat Esportiva in Paterna
The Valencia CF Academy is already preparing the second edition of the VCF Academy World Cup which will take place from 29 June to 2 July 2023 with a representation of teams from America
Valencia and the Ciutat Esporta de Paterna will host hundreds of young players under 12 years of age from all over the world to enjoy football and participate in this second edition of the VCF Academy World Cup for a week
aiming to repeat and improve the great experience of 2022
"The idea is to consolidate the VCF Academy World Cup as an international reference and make it an event where every season the different VCF Soccer Academies that we have around the world
as well as other clubs and academies linked to the VCF Academy Programs come together," explained those in charge of the VCF Academy Programs
"Last season's experience was incredible because of the atmosphere that was generated
the training camp in the buildup to the tournament
the cultural exchange between players and coaches
the entertainment provided by family members during the days of competition and
above all the footballing performance displayed by every team
The teams participating in this II VCF Academy World Cup are already booking their places to take part in the tournament and in some cases also to carry out the full training camp at the Ciutat Esportiva de Paterna with training sessions

- Two men charged with illegally cutting and removing trees from a Riviera neighborhood in Santa Barbara were in court Monday for their arraignment.
James Allen Carr and Enriquez Calles Vasquez both pleaded not guilty to two felony counts of vandalism and four misdemeanor counts.
The incident happened back in December of 2020 on Paterna Road
Prosecutors allege Carr hired Vasquez and a crew to chop down four mature Eugenia trees on or near Carr's newly purchased property
It's estimated the trees were more than 50 years old; three were owned by the city
Seven months later neighbors are still up in arms
who live right across the street from Carr
"It's really hurt our privacy," Malki told NewsChannel 3
anybody at that house can look directly into our master bedroom
Malki was there the day the trees were chopped down and taken away
He said he asked the workers to stop so he and other neighbors could talk to Carr about the situation first
Park rangers also responded, telling the crew a permit was needed.
Neighbors told NewsChannel 3 Carr put the home on the market within two months after the tree cutting incident
Many believe the Elk Grove man intended to "flip" the property all along. Eventually
One large Eugenia tree still stands out front to the left of the house
"He left this one which is equivalant to the size of the other ones because it blocks his view to the telephone pole," said Malki
Malki said he and Carr have never spoken.
