'I’ll do him a favor and score it for him,'” laughs Moore
Although the news of the possible Harmon adaption provided a spark
Moore had always been fascinated by Vonnegut’s sophomore effort
which focuses on a Martian invasion of Earth and the journey of the story’s protagonist
And every once in a while I’ll pick one out and thumb through it again — 'cause I’ve read them so many times,” says Moore
“But I grabbed [‘Sirens’] and just loved the name Malachi and wrote a surf-sounding song based on the name
I just kept going with the Vonnegut theme until I had four songs.”
Moore called his guitarist pal Whittemore to flesh out the material and eventually they enlisted drummer Banks and keyboard player Fusco to help round out the project
Taking their name from the robot messenger in "Sirens," Salo Pallini formed just as the COVID-19 pandemic hit
This group of usually busy gigging musicians suddenly found themselves with a lot of time on their hands
all that sort of thing — that was a part of it,” says Whittemore
“But Landon and I had done a fair number of things together over the years
it was just exciting to work on something together of our own
Because usually we’re all serving as sidemen supporting someone else’s music
It was fun to write our own stuff and do something fresh with it.”
Moore had initially penned a quartet of instrumental songs tied to “Sirens.” Offering a musical translation of Vonnegut’s work involved some good
“The year it was written — he started it in ‘58 and it came out in ‘59 — we took that into account, and also how quirky a guy he was,” says Moore. “I started out with this idea of composing late–‘50s-style elevator music. Recently there was a Vonnegut documentary that came out called ‘Unstuck in Time’ and his daughters were interviewed in it and they complained about all the awful elevator music he listened to while writing
the music has a lounge kind of feel,” adds Moore
“And once the other guys got more involved
we expanded into a slightly more involved sound that John calls ‘progressive Latin space country.’”
After writing and rehearsing for six months during COVID
the band recorded a set of demos at Whittemore’s home rehearsal space
The group was almost ready to release those tracks when — in another literary twist — an opportunity came to rerecord the songs at a new Memphis studio that had been set up in the former home of Civil War historian and novelist Shelby Foote
Moore began sending the resulting 11-song LP
“Sirens of Titan: A Preemptive Scoring" out to Vonnegut-connected festivals and organizations
Salo Pallini quickly landed a booking playing in Indianapolis as part of Vonnegut’s 100th birthday celebrations this past November
The gig — which marked Salo Pallini’s live debut — went over big with the assembled Kurt-heads
“Just to be playing these songs for such a dedicated audience of Vonnegut fans
I thought I knew a lot about his books and work
but there are folks out there who are super deep into his stuff and everything associated with it.”
Salo Pallini is set to play another event this summer in Bloomington, Indiana, called Granfalloon: A Kurt Vonnegut Convergence. The group may also appear at Big Ears
the annual experimental music festival held in Knoxville this year or next
In the meantime, the group will make its local debut, playing a release show on Jan. 20 at Crosstown Arts’ Green Room, where they will have copies of the album available for sale. The record can also be purchased at local retailers and online at salopallini.bandcamp.com/releases
Although there’s been no recent news on the Dan Harmon led-adaptation of “The Sirens of Titan,” through some mutual contacts
Moore has gotten a copy of Salo Pallini’s album to the writer/director
“I don’t know if Harmon will ever make the project,” chuckles Moore
“but we’ve got the soundtrack all ready for him just in case.”

My first time back in an airplane since the pandemic struck was unlike anything I've experienced before in a lifetime of flying.

My flight down was non-stop on Delta and the way back was on American connecting in Charlotte.
After writing about the effect of this pandemic on the airlines
I was curious to see just how exactly the flights would go and was surprised by how differently the two airlines have adjusted their practices in response to calls for social distancing in the skies
Delta was very proactive in ensuring peace of mind during the flight
especially for someone like me who hadn't flown since the pandemic
I would've felt differently about flying had my first flight back been on American where there were very little social distancing efforts
Even though the practice will incur losses for Delta
the airline is extending its policy of blocking middle seats until the end of September
instead of giving up on service altogether like American
Delta still provided some essentials like hand sanitizer and water.
I saw firsthand how social distancing on airplanes is a myth
some airlines are still making the effort and that will undoubtedly go a long way for some passengers as it did for me on my first flight back.

I've always enjoyed the occasional train trip.
Traveling by rail long has been overlooked by the public
as airlines have been able to provide faster
But Amtrak's new health and safety protocols should have travelers giving it a second chance for regional travel
Travelers can even see how full their trains are by checking Amtrak's website so they know exactly what to expect when stepping aboard.
And that's even while keeping its fares pretty low on its most popular routes
The 3-hour hop from New York to Washington still starts at $29 on the Northeast Regional while the slightly longer journey from Boston to New York is often the same price
with all the nostalgia of inter-city train travel included.
I took Amtrak to and from New York's Pennsylvania Station
I started on the Northeast Regional and returned on the high-speed Acela to get adequately acquainted with the service.

Both of my train trips were highly enjoyable
and showed how Amtrak is a hidden gem for regional travel during the pandemic

is that traveling by train is often a more stress-free experience than flying when everything is going right
I didn't have to arrive two hours before my train
I arrived just a few minutes before my train was to depart and didn't have to wait in line at all
reserving a seat ahead of time also allowed me to sit as far away from others as I could without having to worry about finding the perfect seat upon stepping aboard.
Amtrak is a great option for regional travel thanks to its new health and safety protocols
And I'll certainly be looking forward to my next train trip.

the answer is clear: flying beats rail every time
the clear choice is not always as obvious and each traveler has a personal preference on which is better
Flights are shorter than train trips but require getting to an airport
all of which increase the overall travel time
Train travel offers the ability to arrive at the station minutes before departure with no security checks required

I decided to answer the question once and for all and timed myself as I traveled between the two cities
I flew Spirit Airlines for the first half of my journey and took Amtrak's high-speed Acela for the second half to see which one was faster
Full disclosure: I'm an aviation enthusiast
but I do also enjoy a good train trip.
Here's which one was the better option for the 200-mile trip between New York and Boston.
Train travel failed to beat airplane travel
even with the added extras of getting to the airport
especially as the Acela was achieving speeds of nearly 150 miles per hour in some stretches.
But I was able to do things on the train I was not able to do on the plane journey
While I undoubtedly enjoyed the plane journey
I was constantly on the move going from one train to another

The engagement of the SARS-CoV-2 spike protein with ACE2 is a critical step for viral entry to human cells, and, therefore, blocking this interaction is a major determinant of the efficacy of monoclonal antibody therapeutics and vaccine elicited serum antibodies. The emergence of SARS-CoV-2 variants has necessitated the development of adaptable assays that can be applied to assess the effectiveness of antibody-based therapeutics.
Through the testing of a range of recombinant spike proteins, we have developed a cell-based, ACE2/spike protein interaction assay that characterises monoclonal anti-spike protein antibodies and neutralising antibodies in donor serum. The assay uses high-content imaging to quantify cell-bound spike protein fluorescence.
Using spike proteins from the original “Wuhan” SARS-CoV-2 strain and the Delta and Omicron variants, we identified differential blocking activity of three monoclonal antibodies directed against the spike receptor-binding domain. Importantly, biological activity in the spike interaction assay translated to efficacy in a SARS-CoV-2 infection assay.
The spike protein interaction assay can be used to monitor anti-spike antibodies against the major known SARS-CoV-2 variants and is readily adaptable for quantification of the impact of antibodies against new and emerging SARS-CoV-2 variants.

We therefore sought to develop a reductionist cell-based model
that quantifies the inhibition of SARS-CoV-2 spike protein interactions with mammalian cells and is suitable for assisting in therapeutic antibody development and immune monitoring
See Table 1 for a list of recombinant spike proteins used in this study. Hoechst-33342 was obtained from ThermoFisher Scientific (H21492), His tag® antibody [HIS.H8] was obtained from Abcam (ab18184), Rho1D4 antibody was obtained from Cube Biotech (40020), and goat anti-Mouse IgG H&L (Alexa Fluor® 488) was obtained from ThermoFisher Scientific (A-11001). See Table 2 for a list of antibodies against spike proteins
were obtained from Invivogen (a549 or a549-hace2
Cells were cultured in F-12K Ham Nutrient Mixture media (ThermoFisher Scientific; 21127022) and supplemented with 10% fetal bovine serum (Sigma-Aldrich; F9665) and 1% penicillin-streptomycin (ThermoFisher Scientific; 15140122) in a tissue culture incubator at 37°C (5% CO2) using standard cell culture practices
Cells were seeded in 96-well plates overnight prior to incubation with recombinant spike proteins
Recombinant spike proteins (4.0 μg/mL
unless otherwise indicated) were pre-incubated with anti-His detection antibody (8.0 μg/mL)
and blocking antibodies or plasma were pre-incubated for 1 hour prior to the addition to cells for 1 hour at 37°C
This was followed by preparation for confocal microscopy
Media was removed from cells and wells washed with PBS (ThermoFisher Scientific; 14190144)
Cells were then fixed with 4% paraformaldehyde for 10 minutes before being washed twice in PBS
once in permeabilisation buffer (ThermoFisher Scientific; 00–8333–56)
prior to blocking (1% goat serum in permeabilisation buffer)
Cells were then incubated with Alexa Fluor 488-conjugated anti-mouse IgG (1:500) and Hoechst (1:1,000) prepared in permeabilisation buffer before being washed three times with permeabilisation buffer
Confocal microscopy was carried out using a Yokogawa CQ1 spinning-disc microscope using a × 40 objective and appropriate excitation/emission settings for Hoechst and Alexa Fluor 488
Z-stack images were acquired and displayed as maximum intensity projections
Image analysis was carried out using Yokogawa image analysis software
and IgM antibodies in plasma samples were measured by protein ELISA using the IgG/A/M Sars-COV-2 ELISA kit
following the manufacturer’s instructions (Binding Site; MK654)
SARS-CoV-2-England 2 (Wuhan strain) at 106 IU/mL (GSAID Accession ID EPI_ISL_407073) was kindly gifted to us by Christine Bruce
Antibodies were pre-incubated with virus for 1 hour prior to being added to the Vero cells
cells were fixed with ice-cold methanol (5 minutes)
and stained with rabbit anti-SARS-CoV-2 spike protein
detected by Alexa Fluor 555-conjugated goat anti-rabbit IgG secondary antibody (Invitrogen
Cell nuclei were stained with Hoechst 33342 (ThermoFisher Scientific)
Cells were washed with PBS and imaged and analysed using a ThermoFisher Scientific CellInsight CX5 High-Content Screening (HCS) platform
Infected cells and cell viability were detected by measuring perinuclear fluorescence above a set threshold that was determined by positive (untreated) and negative (uninfected) controls
Automated quantification algorithms were developed with assistance from Dr Henri Huppert
Data are presented as mean + SEM or mean ± SD from at least three independent experimental setups unless otherwise indicated
Curve-fitting data analysis was conducted using KaleidaGraph (version 3.5)
All statistical analysis was carried out using GraphPad Prism software (version 9.2)
Data were tested for normality using the Shapiro–Wilk test and normally distributed data were analysed using one-way ANOVA and post hoc Tukey tests (unless specified)
with corrected p-values where multiple comparisons were carried out
A Kruskal–Wallis test was used for data that were not considered to be normally distributed
All samples were obtained with informed consent and with approval from the appropriate research ethics committee (REC Reference 20/WA/0216)

To expand the potential utility of the spike interaction assay, we first determined the total IgG/A/M against SARS-CoV-2 spike protein of five donor plasma samples by ELISA (Figure 6A). A value of ≥ 1.0 was considered positive in this assay, and the plasma samples containing levels of anti-spike IgG/A/M >3 exhibited concentration-dependent spike-blocking activity, whereas those with levels <3 did not (Figure 6B)

To further assess the relationship between spike interaction blockade, levels of anti-spike antibodies, and viral neutralisation, we screened the activity of donor plasma in the spike-binding assay, the anti-spike IgG/A/M ELISA, and an in vitro viral infection assay (Figure 6C)
and analysed the correlation between the three assays
Although there was a correlation between all three assays
the correlation was strongest (R2 >0.75) between the spike-binding assays and the viral infection assay
This emphasises the fact that the spike interaction blockade assay provides information about the relevant functional activity of anti-spike antibodies
these data suggest that the spike-binding assay is suitable for both monoclonal-antibody characterisation and monitoring neutralising antibody activity in plasma
shows consistent relative potencies across independent experiments

we have developed a high-content imaging assay to quantify SARS-CoV-2 spike protein interaction with ACE2-expressing target cells
ACE2 expression in A549 cells was sufficient to detect various recombinant spike proteins from the Wuhan strain and from the variants of concern: Alpha
We demonstrated that some commercially available monoclonal antibodies were able to neutralise recombinant spike labelling in a manner comparable to replicating virus neutralisation
these results indicated that our platform was in accordance with previous data
although they can identify blocking antibodies and have benefits of convenience
do not utilise ACE2 expressed in its native cellular environment and do not enable quantification at the single-cell level
As with all studies defining pharmacological potency
when ligands (in the present case antibodies) display similar potency (e.g.
anti-RBD14 and anti-RBD7 antibodies against the Delta variant)
establishing a consistent rank order of potency is challenging
when there are clear potency differences in blocking activity
the rank order of potency for the antibodies was consistent across independent experiments
further emphasising the utility of the assay
As this assay system uses a “His tag” for detection
which is frequently used for the purification
His-tagged recombinant viral variant spike proteins are often rapidly available and accessible
enabling the assay to be quickly adapted to test the activity of therapeutics as new strains of SARS-CoV-2 emerge
Across viral variants, although antibody affinity for spike protein may remain approximately constant, the affinity of spike protein for ACE2 may increase significantly and be associated with reduced antibody neutralisation (21)
Measuring functional antibody blockade of the spike protein/ACE2 interaction is therefore likely to become increasingly important
spike protein interaction assay would complement existing biochemical and pseudovirus assays in the development of therapeutics and may be adapted for other cell types to investigate the potentially ACE2-independent binding of spike protein

Whether or not that's a bad thing is up to each passenger

It's ultimately meaningless if a flight fills up but for less crowded flights
like the one I took from Orlando to Washington
it did ensure passengers weren't needlessly assigned middle seats before all of the aisle and windows were filled.

which is a luxury in flying during the pandemic.

bad limoncello will cause "allappa," which she described as "thick tongue" — "it's not a pleasurable feeling afterwards," she said.
From the verb "allappare," a better translation might be that it makes your mouth pucker due to the sourness
it's not overbalanced and leaves your tongue fresh and clean," she said
you smell lemon," Luca said — and he means real lemon
He added that many brands will use a strong alcohol percentage to cover up the fact that there are little or no natural ingredients in their product
the first thing you smell should not be the alcohol
"You can really feel the perfumes of the Amalfi coast in a good limoncello," Micaela said

The Finnish flag carrier has been faithfully flying the route for over half a century
celebrating its 50-year anniversary in 2019
while operating the only current nonstop link between the US and Finland with connections into Europe
I noticed an unbeatable flight deal to Copenhagen from New York for Martin Luther King Jr
Day weekend that involved two legs on Finnair with a return on British Airways for only $280 in economy.
The catch: the ticket was a basic economy fare.
Finnair joined the growing list of airlines adopting a basic economy fare for transatlantic flights in 2018 when it introduced an economy "light" fare
The fare has allowed full-service carriers such as Finnair to fight back against the low-cost carriers that entered the transatlantic market in recent years
selling deconstructed tickets where passengers could select what add-ons they wanted.
according to the American Airlines website where I booked the ticket
it seemed I was being intimidated to pay more for the standard economy fare
which had a difference of a few hundred dollars.
As it would only be a quick weekend trip with no reason to check bags
I decided to book it and flew my first transatlantic flight in basic economy from New York to Helsinki on Finnair
While the Finnair in-flight experience was normal for a transatlantic hop to Europe
I wasn't treated as a sub-par passenger who was trying to skirt the system to get a good deal and instead
When I checked-in for the flight on the app
I had free reign to choose any available seat while other airlines charge for seat changes at check-in for basic economy
I was also offered the opportunity to check my bag for free at the airport counter
as well as having the seat next to me blocked off so I could have it to myself.
This shocked me as my other basic economy experiences have paled in comparison to Finnair and I was glad to be treated as a passenger rather than punished for being a basic economy flyer
Though it wasn't initially my airline of choice for heading to Europe
I certainly would take it again — if the price was right.

I flew seven flights on the largest airlines in the US
I'm adequately reacquainted with flying after having been grounded since February
The experiences have been unlike anything I've seen before in a lifetime of flying
with each airline having its own way of handling the pandemic
No two airlines have been exactly alike on any of my journeys
and seemingly ever-changing policies are creating confusion for passengers
has different definitions depending on the airline
Some airlines have chosen to block middle seats and limit capacity in an effort to achieve social distancing
while others have given up entirely or give only the appearance of social distancing
Here's what you can expect on each airline
Blocking middle seats or allowing free flight changes
American began filling its flights to capacity and not blocking any middle seats
there is an option for a free flight change if an alternative flight is available
and passengers flying in basic economy may be automatically assigned a middle seat
even if other aisle or window seats are available
Only check-in or gate agents typically have the power to change seat assignments if a passenger isn't happy with a seat location
American has not said which factors determine whether the option to change flights is offered
The airline has been operating a reduced flying schedule
so alternative flights have not always been available for passengers
but an airline representative told Business Insider that more flights being flown starting July 7 should give passengers more options
American operates a normal boarding process
and passengers still board in their assigned groups
This results in economy passengers in the back of the plane walking through an entire aircraft of people before arriving at their seat
Signage at the gate informs passengers that masks are required and that the airline has adopted new cleaning standards but does not go into detail
American is limiting the in-flight service depending on the duration of the flight
Flights under 2,200 miles will no longer have a snack or drink service
with nonalcoholic canned or bottled beverages being served on request in economy
Flights greater than 2,200 miles will see a beverage service but no snack service in economy

Passengers who still do not wish to travel on a crowded flight even with the capacity restriction will have the option to request a free rebooking to a later flight

Gate agents are asking passengers to scan their own boarding passes when they board to reduce interactions between staff and passengers

There is some signage at the gate asking passengers to socially distance and informing them of the new boarding procedure but no visuals or anything pertaining to the airline's new cleaning procedure
Southwest is suspending the in-flight service on flights under 250 miles
Passengers on flights over that threshold will receive a cup of ice water and a snack bag served by flight attendants
as nearly every aspect of a flight has been revised to become more passenger-friendly during this pandemic while not compromising too much on service
From placards and informational signage in the gate area to blocking middle seats and maintaining an in-flight service
Delta is leading the way in multiple aspects
Southwest Airlines comes in a close second
with the low-cost airline earning its reputation for good customer service even more so during this crisis
The only downsides were the boarding process
the lack of informational signage at the gate area that I found on most other airlines
and a lack of consistency in staff following the new procedures
United Airlines is the second-runner up mainly because I found its policies to be more empty gestures than actually helpful
The airline is offering free flight changes despite having few backup options and restricting the advance selection of middle seats rather than blocking them but are still allowing flights to fill up
United did have some positives in that it revised its boarding procedure and offered sanitary wipes upon boarding
but I did find a lack of consistency in informational signage at different airports
as the in-flight service was also suspended
American Airlines was the least passenger-friendly airline I found on my travels with a complete lack of social-distancing policies and abandonment of in-flight service on most of its domestic flights
It's largely business as usual when flying on American
with the airline happy to assign middle seats to basic-economy passengers when entire empty rows are available and keep the standard boarding procedure
that all aircraft I flew on from all airlines were clean and I was never worried I was getting on a dirty aircraft

Regardless of whether you are in favour or against
the announcement of the formation of the ESL came as a shock to everyone
But what was more shocking was the confident attitude of the 12 clubs
which seemed to have not considered the impact that this decision would have had on their own fans/customers
The backlash and reaction were unprecedent - fans
media and even politicians bluntly showcased their disapproval and disdain towards the ESL
There was a call for a boycott of the ESL and the clubs faced multiple accusations - from acting for ‘pure greed’ to not respecting football traditions and the feelings of their own fans

These clubs have not only lost the trust of their fans but also their peers (other clubs)
and in doing so have significantly reduced the negotiating power that they once had
Although apologies have been issued by some clubs to do some level of damage control and regain the sympathy of the fans; this is unlikely to be enough
but others believe that the clubs have shown their intent and that an apology will not ‘do the trick’
How will these clubs regain their fan/customers trust
It will be interesting to see in the next few weeks and months if there will further resignations – following the one of Ed Woodward
This could bring in new top-management and it could work as a ‘breath of fresh air’ that could wipe away that ‘greedy intention’ perceived by most fans
with some calling for the owners of these clubs to now leave and further demonstrations are predicted
especially when fans are allowed back into stadiums
There are also reports that suggest giving fans a stake in their clubs
which could help clubs understand footballing traditions better
Whatever the outcomes at these clubs over the next few months
this is certainly a prime example of how easy it is to do reputational damage in the blink of an eye

The Centers for Disease Control and Prevention has reignited the debate over whether airlines should be blocking middle seats just as airlines thought it was settled

"Multiple scientific studies confirm that the layers of protection significantly reduce risk
and research continues to demonstrate that the risk of transmission onboard aircraft is very low," a spokesperson for Airlines for America told Insider
indicating no changes would be recommended to airlines
The trade organization also pointed to Department of Defense and Harvard studies, as well as data from the International Air Transport Association
finding low transmission rates among passengers since the start of the pandemic
"Since the start of 2020, there have been 44 confirmed or possible cases of COVID-19 associated with a flight," IATA found
some 1.2 billion passengers have traveled."
Delta Air Lines is the only major US airline still blocking middle seats but will stop doing so May 1
told Insider: "We do not currently have plans to change any of our existing seating policies."

I definitely would not have paid the $159 upgrade price for what was basically a larger seat unless I was looking to earn extra miles.

as we settle into what will undoubtedly be a protracted recovery for aviation
airlines are examining their lounges differently and are slowly bringing back or modifying the luxuries flyers once enjoyed.
