Metrics details the silkworm possesses a restricted feeding capacity To investigate the impact of varying sucrose concentrations on silkworm growth and development transcriptomic analyses were conducted on the silkworm midgut A total of 909 differentially expressed genes (DEGs) were identified with down-regulated genes predominantly associated with starch and sucrose metabolism and amino sugar and nucleotide sugar metabolism These alterations suggest that excessive sucrose consumption may suppress the carbohydrate metabolism pathway in silkworm This study offers a comprehensive analysis of transcriptomic studies on high sucrose intake in silkworm providing insights into the effects of varying sucrose concentrations on these organisms silkworm enters a voracious feeding period with a sharp increase in mulberry leaf consumption begins to synthesize mass silk proteins in the silk gland The digestion and absorption of nutrients in silkworms are inseparable from the digestive tract accounting for approximately 78% of the total length of the digestive tract Cylindrical cells are the main components of the midgut epithelium especially well-developed in the posterior part of the midgut capable of secreting digestive fluid and absorbing nutrients the majority of the enzymatic digestion and the absorption of nutrients largely take place in the midgut carbohydrate metabolism is a critical aspect of insect physiology By gaining a deeper understanding of the mechanism underlying glucose conversion and metabolic regulation in insect we can develop more effective strategies for silkworm rearing potentially leading to increased silk production and more sustainable agricultural practices we investigated the effects of adding sucrose with four different concentrations (6% from the 5th instar molted silkworm (I5D1) to wandering (spinning the cocoon) the diet supplemented with sucrose had positive effects on the development of silkworm the growth situation of the 12% treatment group was relatively the better we conducted a transcriptomic comprehensive analysis of gene expression of midgut in I5D3 silkworm under condition of sucrose 12% treatment The results revealed various alterations in carbohydrate metabolism and glycolytic pathways induced by changes in sucrose consumption The findings indicate that the appropriate sucrose concentration and the optimal feeding time will positively impact the development of silkworms This research not only can provide valuable understanding regarding the impact of sucrose on the midgut of silkworm but also holds significant guiding implications for enhancing silkworm breeding efficiency The silkworm 871 strain (monovoltine race) was provided by the Sericulture Research Institute of Chinese Academy of Agricultural Sciences The rearing temperature should be maintained between 25 and 26 °C and the humidity should be controlled between 70 and 85% Ensure appropriate lighting and ventilation Control the stocking density to avoid overcrowding and uncontaminated mulberry leaves as the feed In the preliminary stage of the formal feeding trial we conducted a pre-experiment to select the appropriate concentration levels as evaluation criteria and found that the silkworms grew slowly and appeared mass death in the 30% concentration groups we decided to refine the concentration gradients to 6% 18% and 24% to allow for a more reasonable assessment of the experimental results Select well-developed and similar-sized female silkworm at the 1st of the 5th instar (molted silkworm Establish a Control Group (or C-Group) and four treatment groups with sucrose supplementation (or S-Group) each with three replicates consisting of 150 silkworm per replicate The C-Group was fed mulberry leaves washed with deionized water while the S-Groups were fed mulberry leaves soaked in 6% we firstly placed the calculated amount of sucrose into a certain volume of sterile water and heated it until fully dissolved additional sterile water was added to reach the desired volume All leaves were selected to ensure a similar state minimizing significant differences among them the petioles and stems of the mulberry leaves were removed During the mulberry leaf immersion process each leaf is individually soaked in a sucrose solution for 15 min to ensure that the mulberry leaves are fully exposed to the sucrose solution we lay out the mulberry leaves taken from the solution flatly on a sterilized wire mesh for natural drying until that the mulberry leaves were moistened with sucrose solution without any dripping Then the leaves were placed in the silkworm box for consumption by the silkworms the density of silkworms was also controlled to ensure that each silkworm had access to a sufficient amount of leaf the growth situation of the 12% treatment group was relatively the better among all treat groups we conducted RNA-Seq of midgut in I5D3 female silkworm under condition of sucrose 12% treatment The midguts of 10 female larvae per sample were collected from the sucrose 12% group and the control group The total RNA of the sample was extracted by EASYspin Plus rapid tissue/cell RNA extraction kit of Beijing Aide-Lai Biotechnology Co. and integrity number were assessed using the Nanodrop 2000 Bioanalyzer Utilizing the fragmented mRNA as a template and random oligo-nucleotides as primers the first cDNA strand was synthesized in an M-MuLV reverse transcriptase system and the second cDNA strand was synthesized in a DNA polymerase I system using dNTPs as substrates The purified double-stranded cDNA underwent end-repair After the library construction was complete initial quantification was performed with Qubit2.0 Fluorometer and the library was diluted to 1.5ng/ul the insert size of the library was tested using Agilent 2100 bioanalyzer different libraries were pooled according to the requirements of effective concentration and target data volume for Illumina sequencing (NovaseqX Plus) The raw data have been submitted to the NCBI public database (NCBI.nlm.NIh.gov) with the accession number PRJNA1186106 Genes were represented as DEGs when the adjusted p-value < 0.05 and |log2 (fold-change)| ≥ 1 a functional enrichment analysis was conducted to pinpoint the GO terms and metabolic pathways that are significantly overrepresented within the set of DEGs To determine the significance of these enrichments with a p-value < 0.05 being considered significant compared to the entire transcriptome background To identify genes that may not show significant differences in overall expression level but have significant biological significance we conducted Gene Set Enrichment Analysis (GSEA) and normalized enrichment score |NES| > 1 were selected for further analysis The quantitative real-time PCR (qRT-PCR) was conducted to confirm the DEGs in the cDNA libraries, which was constructed from midgut of 5th instar larvae. BmGAPDH was selected for housekeeping gene. It’s unclear how many of the staff Sasse hired have remained employed at the university, including previous aides from his time as a United States Senator. Several remain in the UF directory, but they are missing main web pages or profiles on the President’s Cabinet website Hosseini’s position comes with considerable power including the ability to override certain presidential decisions Hosseini also influenced major initiatives across the university In a Board of Trustees meeting on July 23 Hosseini said he asked UF’s Chief Financial Officer Taylor Jantz to allocate $25 million to support faculty “who work so hard to serve our students and do great research.” Hosseini did not elaborate on how the $25 million would be distributed Sasse's office employed more than 30 staff members a stark contrast to interim president Kent Fuchs who had fewer than 10 during his presidency James Wegmann and Dan Dillon are among those still listed on the UF President cabinet website and are believed to still be employed is Sasse’s former Senate communications director and serves as UF’s vice president of communications Wegmann replaced UF spokesman Steve Orlando, who made $270,000 a year in the position. And luckily that was the case with this Portland, Oregon house designed by renowned architect Saul Zaik who was known for his contributions to Pacific Northwest architecture “Our clients are a young couple with a deep appreciation for Pacific Northwest mid-century and contemporary minimalist design,” says Project Architect Robert Arlt of SHED Architecture “They came to know and admire Saul Zaik’s work through open houses and the local design community The property came up for sale just a month after their wedding.” The home, which is called The Mori House, was designed for Zaik’s friend and next-door neighbor, interior design Howard Hermanson. It features an elevated deck, which connects to a detached teahouse pavilion. Zaik was known for his modernist residential designs integrating homes into their natural surroundings and prioritizing natural materials and light “We inherited a unique structure within a beautiful forested site and the primary driver for the project engaged the principle of addition through subtraction,” says Arlt “The house had accumulated many layers of finishes and a tight spiral stair connected the walkout basement and loft under the large skylight which inhibited circulation Given the task of reconfiguring and providing safer circulation the question we kept coming back to during the design investigation was ‘How do we do that so the large central skylight can organize and impact every space in the home?’” the clients knew they wanted to reconfigure the main floor’s living and flow while maintaining clear separation from private areas and renovate the two main level bedrooms into a large primary suite,” says Arlt the existing spiral staircase needed to be reconfigured to optimize safe vertical circulation for their growing family and further define the division between public and private spaces.” The final goal was to update the home’s design to reflect the homeowners’ personal style which is described as “‘East meets Pacific Northwest’ modernist minimalism and natural-light-accentuating materials.” When planning the renovation the SHED team took the mindset of “What would Saul do?” and used Zaik’s other PNW mid-century remodels as inspiration The team removed the existing loft and spiral staircase unveiling a nail-laminated wood roof that extends to a central skylight There’s now an entry closet and powder room in the space where the spiral staircase used to be The SHED team incorporated a new horizontal window above the cooktop which offers views of the mature rhododendrons outside The kitchen opens into a dining nook that includes a custom fir bench in front of floor-to-ceiling glass A new glass door provides direct access to the deck and teahouse pavilion The newly renovated living room features a relocated fireplace giving more views of the forest (and Mount St The former fireplace area has a built-in bar and tech closet “The vaulted living space and skylight are made possible through a unique hip-roof structure,” Arlt explains and solid 2×6 nail-laminated framing free-spans between to form the roofline and exposed wood ceiling The finished side of the nail-laminated 2×6 was exposed in the primary bedroom and the whole surface was blasted with walnut shells to restore and brighten the surface.” The primary suite features fir slats above the door “This suite transforms an existing bedroom and bathroom into a spacious primary bathroom with an adjacent walk-in closet,” Arlt says “The primary bathroom includes a Japanese soaking tub custom medicine cabinets with integrated LED lighting and transom windows to borrow light from the skylight.” On the southwest side of the house is a new switchback stair that leads to the lower level hangout space that overlooks the forest The lower level also includes three private The renovated home feels like you’re amongst the trees—true to Zaik’s natural surroundings ethos “We have heard they enjoy the quality and detail and coming home to a feeling of lightness,” Arlt says of the homeowners and so much more delivered straight to your door every quarter Copyright ©2025 S Media International Corporation. stillness and prayer is more like passive meditation than active watchfulness but it really does take effort to be still When we see those distractions creeping in but don’t allow ourselves to be carried off when we make the conscious effort to turn back and focus on our conversation with God The fourth type of watchfulness is simply the habit of continually and humbly calling on our Lord for Absolutely. Unlike most other omakase experiences, Mori serves delicate hand-shaped wagashi (a category of Japanese sweets, of which mochi is just one kind) and ceremonial-grade matcha at the end of every meal.
