are substrates for S-glutathionylation and that this post-translational modification correlates strongly with diastolic dysfunction led to new concepts regarding how levels of oxidative stress affect the heartbeat Major sarcomere proteins for which there is evidence of S-glutathionylation include cardiac myosin binding protein C (cMyBP-C) Our hypothesis is that these S-glutathionylated proteins are significant factors in acquired and familial disorders of the heart; and We consider the molecular mechanisms for these effects in the context of recent revelations of how these proteins control cardiac dynamics in close collaboration with Ca2+ fluxes These revelations were made using powerful approaches and technologies that were focused on thin filaments Here we integrate their regulatory processes in the sarcomere as modulated mainly by neuro-humoral control of phosphorylation inasmuch evidence indicates that S-glutathionylation and protein phosphorylation promoting increased dynamics and modifying the Frank-Starling relation Earlier studies demonstrated that in addition to cTnI as a well-established biomarker for cardiac disorders serum levels of cMyBP-C are also a biomarker for cardiac disorders We describe recent studies approaching the question of whether serum levels of S-glutathionylated-cMyBP-C could be employed as an important clinical tool in patient stratification early diagnosis in at risk patients before HFpEF and as a guide in developing future therapies Our findings were confirmed and extended by other laboratories more work needs to be carried out to understand the role of S-glutathionylation more completely in early and late stages of cardiac disorders and in their usefulness as serum biomarkers in diagnosis and stratification of patients there have been significant investigations contributing to a new and dynamic evolution in understanding the structure and function of all three of the filaments of the cardiac sarcomere Detailed accounts of these advances are in recent publications to which we refer in the present paper in the context of our focus on oxidative modifications of cMyBP-C Adding to the complexity of the disposition of proteins in relaxed and active sarcomeres is evidence that these interactions are biased toward interactions with actin-Tm in the low Ca2+ inhibited state of the thin filament and more toward interactions with myosin heads at activating levels of Ca2+ As discussed below these interactions are also modified by phosphorylation and by titin induced strain with stretch stabilization of the state of distensibility of titin occurs with aggregation of distal Ig domains of parallel titin strands Variations in this mechanism produce a range of values of passive tension and can affect length dependent activation (LDA) Evidence indicates that stretch of titin signals interactions with the C-terminal domains of cMyBP-C that are transmitted to the cMyBP-C N-terminal domains which promote interactions with actin-Tm and activation of the thin filament which occur in control of contraction and relaxation dynamics and in the Frank-Starling relation are in a state of evolution in our understanding This discussion includes descriptions of accessible Cys residues expressed in each of these proteins that are targeted by S-glutathionylation Addition of the negatively charged GSH tripeptide modifies function alters signaling via protein phosphorylation The concept includes the idea that a diastolic mechanism governed by modifications in titin and troponin-tropomyosin engages ahead of and controlling the systolic state They concluded that with beta-adrenergic stimulation phosphorylation of cMyBP-C is mainly responsible for the effect to increase LDA and the Frank-Starling mechanism We also determined sites of S-glutathionylation employing mass spectrometry As with the preparations isolated from DOCA-salt hearts myofilaments incubated in GSSG had an increased Ca2+-sensitivity Sites of S-glutathionylation as determined by mass spectrometry occurred at cysteines 655 As reported previously they found the HF cells had reduced maximum tension and Ca-sensitivity By treating the myocytes with reduced GSH they were able to correct these changes They found increased S-glutathionylation of both cTnI and cMyBP-C in the HF samples compared to the donor samples Treatment with PKA failed to phosphorylate these proteins in the HF samples but worked in the donor samples there was increased titin stiffness in the HF samples that could be suppressed by treatment with PKA and GSH These data indicated a significant role for interactions of S-glutathionylation and phosphorylation in end stage human heart failure There was a correlation of cMyBP-C S-glutathionylation with increased Ca-response of tension and ATPase activity with no change in maximum or minimum values we found no evidence of actin S-glutathionylation in hearts expressing the HCM linked mutant TmE180G discussed below A main conclusion from publications from our and other laboratories is a dominant and consistent modification of sarcomere proteins by S-glutathionylation is at cMyBP-C and titin Existing evidence does indicate a need for further investigation of the potential role of actin-S-glutathionylation of integrated function of cardiac myocytes Apart from the suggestions above about use of serum levels of S-glutathionylated cMyBP-C in diagnosis and patient stratification there are several gaps in our understanding of the role of S-glutathionylation of major sarcomere proteins in cardiac physiology and pathology The unique ability of S-glutathionylation to modify Cys residues reversibly and to protect against irreversible oxidative modifications needs to be more completely understood in relation to physiological processes for example Whereas correlations with diastolic dysfunction remain a strong possibility there is evidence that this PTM may occur in a variety of cardiac disorders many of which display oxidative stress More studies on early-stage cardiac diseases are in order These studies need to integrate S-glutathionylation in the major affected proteins We think an important unexplored avenue of research is S-glutathionylation induced modulation of sarcomere length dependent activation (LDA), i.e., the Frank-Starling mechanism. Evidence of a role for titin (10, 71), cMyBP-C (56, 57), and cTnI (41, 55, 57) in altering LDA provides a rationale for this suggestion A further rationale that is unexplored is the possible modification of LDA in the oxidative stress associated with cardiac disorders demonstrating mechanical dyssynchrony Early activation of the septum associated ith late activation of the left ventricular free wall characterizes dyssynchronous heart failure (DHF) A common cause is abnormal Purkinje fiber conduction there is also evidence that DHF occurs with altered sarcomere mechano-biochemical signaling Although bi-ventricular pacing can suppress the DHF a significant number of patients do not respond to this treatment Understanding of this lack of responsiveness may involve abnormal LDA and engagement of the Frank-Starling mechanism In testing patients with non-obstructive but symptomatic HCM, Ahmed et al. (72) applied CRT (bi-ventricular pacing BiV) and tested for improvements in LV diastolic filling and exercise capacity Their approach was based on evidence of a significant incidence of dyssynchrony in HCM patients without obstructive outflow disorders The dyssynchrony manifests at rest in a maladaptive interaction between the RV and LV in diastole inducing a failure of the LV to fill properly during exercise resulting in a failure to employ the Frank-Starling mechanism there was an improvement in LV volume during exercise The authors did not investigate molecular mechanisms but did evaluate the effects of BiV on LV contractility there was no change in mechanical dyssynchrony in the study groups and no change in systolic elastance associated with BiV indicating no change in contractility with some allusions to limitation of the study the authors conclude that major mechanisms for the improvements in exercise were the effect of BiV to engage the Frank-Starling mechanism Inasmuch as length dependent sarcomere activation is the basis of the Frank-Starling mechanism this conclusion emphasizes consideration of LDA in sarcomeres with HCM-linked mutations The significance of LDA in CRT has been investigated by Niederer et al. (73) By employing a patient specific model of excitation and mechanics from the level of cellular mechanisms to heart function determined in patients before and after CRT Sensitivity of the model to various elements in cellular control mechanisms demonstrated LDA as a significant contributor to the therapeutic benefit of CRT The computations indicated a reduced effect of CRT in patients with dysfunctions in LDA A significant contribution of these findings is the idea that the Frank-Starling relation is not only important in beat-to-beat control of cardiac function but a significant mechanism in synchrony of the heartbeat Mechanical LDA and other sub-cellular mechanisms are downstream to electrical stimulation and thus dampened or amplified LDA appears to be a relatively important contributor to the efficacy of CRT This study indicates that despite the electrical effects of CRT it is critical for the heart to have robust LDA Our studies with models of HCM are relevant to these concepts. Early studies from our lab investigated the LDA of myofilaments expressing cTnT-R92Q. The data indicated a higher level of LDA in the R92Q skinned fibers compared to WT fibers (74). Other studies reported an attenuation of LDA in HCM models cTnT-R95H and cTnT-F88L (75, 76) It would be of interest to determine a role of oxidative stress and potential S-glutathionylation of sarcomere proteins in these models We think our discussions here provide a rationale for continued investigation of this powerful post-translational mechanism in sarcomere proteins The original They came together in their collective grief to pray, to reflect and to begin the healing process, as the small rural community struggles to process the tragic events of three weeks ago. “It’s been a difficult few weeks,” vigil organizer Angela Lamendola said. “Jefferson has been shaken to its core by recent events, ending in the passing of Christie Solaro.” Lamendola added that the community doesn’t think of itself as that type of place. “Jefferson is a sleepy town. My friends and I jokingly and lovingly refer to it as Mayberry,” she said. “Stuff like this doesn’t happen here. Only it does happen here. It did happen here.” Lighting a purple candle in front of a portrait of Solaro, Lamendola said that purple is the color designated for domestic violence and that she was surprised to learn how pervasive a problem it is in Jefferson Township. Since Solaro’s death, Lamendola said she has learned that the town is in the top third in Morris County in terms of domestic violence occurrences. According to county domestic violence statistics filed with the state Attorney General’s Office, Morris County had 604 total original investigations in 2018, 254 of which were pending or inactive at the beginning of the year. Seventy-nine defendants in original investigations were criminally charged through accusation. A breakdown by municipality was not available. “Now is the time for healing, now is the time for hope,” she said. “For those of you who knew Christie, as I did, I am so very sorry for your loss and the anguish you are coping with right now. God knows I’m feeling it too.” Whether it was meeting up for coffee or for their daughters’ playdates, or sending internet memes via group chat, Lamendola said that Solaro was a daily fixture in her life. “It’s a void I felt immediately and feel immensely,” she said of Solaro’s death. Solaro was a dedicated mother, Lamendola said, who fought for her children and was fiercely protective of them. “Those kids were her whole world,” she said. “There was nothing she wouldn’t have done for them.” A great friend who made everything better just by being there, Solaro was there both to make you laugh and pick you up when you were down. “Christie had a light within her that shined so bright it was a beacon of joy,” Lamendola said. “This candle I’ve lit up here represents that light. I will now use it to light my vigil candle.” “We’ll continue to pass this light, from person to person, until we are all basking in the glow of Christie’s light,” she said. Volume 10 - 2019 | https://doi.org/10.3389/fphys.2019.00478 This article is part of the Research TopicProbing the cardiac arm of the baroreflex and complementary branchesView all 12 articles Interests about the fine underpinnings of cardiovascular beat-by-beat variability have historical roots various aspects of the relationships between arterial pressure and heart period were taken as a proxy of the baroreflex in physiology and medicine stimulating the interest of investigators in several interconnected scientific fields Studies of the overall system facilitated the emergence of a simplified negative (vagal) feedback model of the baroreflex and overshadowed the simultaneous interaction with excitatory sympathetic positive-feedback mechanisms that would better suit the model of a “paired antagonistic (parasympathetic/sympathetic) innervation of the internal organs.” From the bioengineering side the simplicity of obtaining the series of subsequent RR intervals stimulated the analysis of beat-by-beat variations providing a multitude of heart rate variability (HRV) indices considered as proxies of the underlying sympatho-vagal balance and participating to the management of several important clinical conditions used in an integrated manner and controlling for age and gender biases might help shed new light on the relationship between cardiac baroreflex assessed by the frequency domain index α and the HRV indices with the varying of systolic arterial pressure (SAP) levels The focus is also on a novel unitary Autonomic Nervous System Index (ANSI) built as a synthesis of HRV considering its three most informative proxies [RR and the rest-stand difference in the normalized power of low-frequency (LF) variability component] Data from a relatively large set of healthy subjects (n = 1154) with a broad range of SAP [from normal (nNt = 778) to elevated (nHt = 232)] show that α and ANSI significantly correlate overall (r = 0.523 and that this correlation is lower in hypertensives (r = 0.444 p < 0.001) and higher in pre-hypertensives (r = 0.618 p < 0.001) than in normotensives (r = 0.5 That suggests the existence of curvilinear “umbrella” patterns that might better describe the effects of the SAP states on the relationships between baroreflex and HRV non-parametric and resampling statistical techniques we give empirical support to this study hypothesis and show that the pre-hypertensive group results at the apex/bottom in most of the studied trends now surpassing 23,000 in the Medline database witness beyond doubt the growing interest on HRV as a de facto standard Even a simple cursory look at available literature, it appears that HRV may spark interest for different reasons, i.e., biological and technical, alone or combined, risking to favor debates about semantics rather than substance (Brown, 2017): but even at maximal stimulation some level of vagal activity remains amplitude and frequency codes should both be considered in the modeling of RR V it is possible to reduce the number of significant proxies and minimize redundancy From a clinical perspective, it is crucial to recall that HRV (particularly its time domain proxies) provides sensitive markers of prognosis in several conditions, particularly in coronary artery disease, predicting mortality in post-myocardial infarction (Huikuri and Stein, 2013) It should also be considered that explicit acceptance of a mixed neural model of the autonomic (!) innervation could clear large fraction of the existing inconsistencies about HRV interpretation This aspect is beyond the aim of the present study We then assess how SAP levels affect these relationships according to the study hypothesis that non-normotensive states could induce changes in the strength and significance of this kind of relationships Results are displayed through convenient graphical tools that aim at providing valuable insights into the examined trends Data for this observational, cross-sectional study, which is part of an ongoing series of investigations, focus on the use of autonomic indices in cardiovascular prevention. They refer to a population of 1154 ambulant subjects, who visited our outpatient Exercise Medicine Clinic for reasons varying from a health check-up to cardiovascular prevention (Lucini and Pagani, 2012) for chronic conditions inclusive of hypertension (considering untreated non-smokers individuals within the 17–86 years age range) The protocol of the study followed the principles of the Declaration of Helsinki and Title 45 The project was approved by the Independent Ethics Committee of IRCCS Humanitas Clinical Institute (Rozzano All subjects provided informed consent to participate Definition of the variables (ANS proxies plus ANSI) employed in the studya Frequency and percentage distributions of the participants to the study within the three SAP groups and further subdivision in seven SAP intervals almost 84% of Nt subjects are individuals with less than or equal to 49 years of age in both female (55.98% out of 1154) and male (44.02% out of 1154) groups this percentage reduces to 64.3% within females and 62.5% within males while in the Ht group to 35.3% within females and 56.9% within males Distribution of the 1154 participants to the study by gender and classes of age within the three SAP groups For the same arguments extensively discussed in Lucini et al. (2018) and with the same methodology therein presented we accomplished the comparability among the SAP groups by statistically transforming the original ANS proxies in such a way they were free of age and gender effects we fitted a two-way full ANOVA model for each ANS proxy regarded as the dependent variable and classes of age and gender as the independent variables through their main effects and interaction the resulting ANOVA residuals (given for each proxy by the difference between observed and predicted values) were referred to as adjusted ANS proxies and accordingly used in place of the original ANS proxies in all the subsequent statistical analyses Following Lucini et al. (2002), our study hypothesis was that potential connections, observed over the total set of subjects, between the baroreflex gain (as measured by the α index) and the other ANS proxies could differ in strength, direction, or statistical significance depending on the SAP group. Focusing specifically on the linear relation (or correlation) between the α index and the other variables listed in Table 1 we were interested in assessing whether the preHt group could represent a sort of transition state from Nt to Ht group in which the correlations between the α index and the other ANS proxies plus ANSI could even strengthen This research question mostly arose from our experience in analyzing this type data where frequently we had observed non-monotonic (or curvilinear) effects of SAP groups on the correlations involving the α index there is a medium correlation of α and ANSI overall (r = 0.523 p < 0.001) as well as in the Nt group (r = 0.5 but the correlation tends to weaken in the Ht group (r = 0.444 p < 0.001) and strengthens in the preHt group (r = 0.618 the correlation of α and RR LFnu is weakly negative overall (r = -0.281 p < 0.001) and in the Nt group (r = -0.261 but it is not significantly different from zero in the Ht group (r = -0.033 All that seems then to evidence the presence of either monotonic- or curvilinear-type effects of the SAP groups on the correlations between the α index and the other variables Scatter plots of the α index against the ANS proxies and ANSI over the whole set of subjects The Pearson correlation coefficient r between the α index and each of the other ANS proxies plus ANSI is written above each panel together with the p-value of the standard procedure for testing: H0: ρ = 0 against H1: ρ ≠ 0 the regression line (with parameters estimated by the ordinary least-squares method) is depicted in red It is worth stressing that these regression lines are used only as graphical references for better visualization of the spread of the point clouds the α index is not regarded as the dependent variable of a regression model In the light of the above issues, correlations between the α index and the other variables listed in Table 1 were inspected both over the whole set of subjects and within the SAP groups by using the ANS proxies adjusted for age and gender effects and the following statistical methods (for which further methodological details are given in Figure 2): The bootstrap algorithm and the statistical methodology for non-parametric inference (2) having decided to introduce no assumption for the data distribution as a resampling technique we applied the non-parametric stratified balanced bootstrap to generate B = 5000 bootstrap replicates from the original data 5000 new datasets each of size equal to n = 1154 subjects with p = 16 variables (the adjusted ANS proxies plus ANSI and the classification variable given by the SAP group membership) (a) by balancing, over the whole set of the nB bootstrap observations generated, the same subject was randomly sampled (with repetition) for exactly B times, but he/she might not be present in each of the B bootstrap replicates or might be present twice or more in any bootstrap replicate. In such a way, simulation errors were reduced considerably in comparison with the ordinary bootstrap procedure (Davison et al., 1986); The adjusted ANS proxies and ANSI were recomputed on each of the B = 5000 bootstrap replicates obtained the WINcorr coefficient was computed on each replicate and for every comparison between the adjusted α index and the other ANS proxies plus ANSI both over the whole set of subjects and within the SAP groups we obtained 5000 values of the WINcorr coefficient (i.e. a bootstrap distribution) for each type of examination and each pairwise comparison involving the α index As a synthesis of the multitude of these bootstrap distributions we used the median rather than the mean for reducing the influence of potential anomalous values on subsequent analysis results; both on the overall set of subjects and within the SAP groups on the medians of the bootstrap WINcorr coefficients according to the following three approaches: (a) with the aim of providing plausible ranges of variation for every correlation coefficient involving the α index, non-parametric 95% bootstrap confidence intervals were computed through the BCa (i.e., “bias-corrected and accelerated” intervals, given as adjusted bootstrap percentiles) method (DiCiccio and Efron, 1996; Davison and Hinkley, 1997); (b) to test the null hypothesis of zero correlation coefficients of the α index and the other variables, a bootstrap permutation test (Hall and Wilson, 1991; Davison and Hinkley, 1997) was applied at the 0.05 nominal significance level; (c) in line with the study hypothesis above described, we applied the Hettmansperger–Norton trend test (Hettmansperger and Norton, 1987) to verify the hypothesis, at the 0.05 nominal significance level, of no effect of the SAP groups on the correlation coefficients concerning the α index against the following two sets of patterned alternatives (explicitly presented in Figure 2): (i) the SAP groups have increasing/decreasing effects on the strength of the correlations (so-called monotonic ordered alternatives (ii) the SAP groups have concave- or convex-shape effects on the strength of the correlations (so-called umbrella alternatives similar to quadratic or curvilinear contrasts) With the aim of simplifying the interpretations we will present most of the findings obtained by the bootstrap procedure and the non-parametric inference through a synoptic figure and several graphs such as the correlation plot and the ridgeline plot this last graph turned out to be a powerful tool for visualization of the SAP group effects and their trend patterns consistently with the Hettmansperger–Norton procedure Descriptive data concerning the original ANS proxies and ANSI are given in Table 4 as means and standard deviations computed over the whole set of subjects and within the three SAP groups A further inspection based on the box plots of the distributions of the (both original and adjusted) ANS proxies and ANSI within the SAP groups is reported in SM and the α index present the highest mean values in the Nt group and the smallest ones in the Ht group and SAP LFa have the highest mean values in the Ht group The preHt group has the largest mean values for RR LFnu and RR LF/HF it is worth observing that its mean values decrease from the Nt group to the Ht group thus proving its sensitivity to the different ANS states observed under the various SAP conditions Descriptive data (mean and standard deviation) of the ANS proxies and ANSI within the SAP groups and over the whole set of subjects Regarding the bootstrap analysis, Figure 3 displays panels of box plots of the within-groups bootstrap distributions of the WINcorr coefficients (with γ = 0.1) computed for every pairwise comparison involving the adjusted α index with the other adjusted ANS proxies along with ANSI (Figure 2) the umbrella pattern appears in its entire evidence in line with our study hypothesis in the first panel concerning the WINcorr coefficient between the adjusted α and HR it can be seen a convex effect of the SAP condition on the strength of the negative correlation the negative linear relationship between α and HR (the higher the HR values the smaller the α values) tends to strengthen in the preHt group in the last panel concerning the WINcorr coefficient between the adjusted α and ANSI the positive linear relationship between α and ANSI (the higher the ANSI values the higher the α values) tends to strengthen Box plots of the bootstrap distributions of the WINcorr coefficients (with γ = 0.1) of the adjusted α index and the adjusted ANS proxies along with ANSI computed within the SAP groups over the B = 5000 bootstrap replicates to higher correlation coefficients (roughly +0.2) between α and RR LFa as well as RR HFa (in the preHt group especially) and to lower coefficients (nearly -0.1) between α and RR LF/HF as well as SAP LFa although the sign of both Pearson and WINcorr coefficients does not change across the groups especially moving from the Nt group to the Ht group the strength of the linear relations with the α index reduces in the Ht group the bootstrap confidence intervals present fairly small widths in the whole set of subjects (0.096 on average) as well as in the Nt group (0.121 on average) thus suggesting that the strength of correlation in the various comparisons is appraised with high stability the confidence intervals result wider in the preHt (0.270 on average) and Ht groups (0.240 on average) thus reflecting a greater internal heterogeneity of these two groups that is bolstered by their smaller sizes than the Nt group Non-parametric 95% bootstrap confidence intervals for the medians of the WINcorr coefficients (B = 5000 bootstrap replicates γ = 0.1) between the adjusted α index and ANS proxies plus ANSI over the whole set of subjects and within the SAP groups Figure 4 displays the correlation plot of the medians of the WINcorr coefficients computed over the whole set of subjects and within the SAP groups along with the results of the bootstrap permutation procedure for testing the hypotheses of null correlation coefficients between the α index and each of the other variables Corresponding p-values are reported in the legend cells containing non-significant coefficients are marked with an X symbol the hypothesis of the absence of a linear relation involving the α index is accepted at the 0.05 level: in the Nt group All this seems to support our starting conjecture about the existence of SAP group statistical procedures combined in the perspective of a multitude of simulated replications of the study we give empirical support to the conjecture that there exist specific pressure states in which the relationship between the cardiac baroreflex and ANS proxies can strengthen or weaken We have turned this conjecture into practical terms by introducing the trend analysis of the SAP group effects on the correlations between the α index and each of the ANS proxies along with ANSI after adjustment for the potential biases by age and gender We have focused on both ordered monotonic trends increasing or decreasing effects moving from the Nt group to the Ht group concave- or convex-shape effects concerning which the preHt group is regarded as an intermediate transition pressure condition a delicate issue that we had to face concerned the fact that the results found should not strictly depend on the adopted statistical methodology we are not able yet to advance plausible explanations of such an observed phenomenon In order to avoid potential straining caused by the applied statistical methods we conducted an extensive preliminary study using alternative techniques to make the conclusions as far as possible untied from the specific analysis approach empirical support to the existence of the patterned trends related to the SAP group memberships such as the ones presented the study and the statistical analyses we addressed should be more appropriately considered as a first exploratory phase toward a broader investigation that should also take into account the role of other individual characteristics (e.g. which we guess might affect the results found here to some extent the statistical approach we adopted to meet the objectives of the study was designed in order to overcome several drawbacks inherent in the type of data under analysis (a) the different age-by-gender composition of the whole set of subjects which led us to introduce the adjusted ANS proxies (while ANSI is free of age and gender effects by construction); (b) the presence of a small subset of outlying subjects concerning certain, but not all, variables, a problem that required us to use the WINcorr coefficient as a robust measure of the linear relationship between the α index and each of the other variables listed in Table 1; (c) the necessity of providing a more general value to the statistical analyses without having the possibility of replicating the data collection on new sets of subjects a fact that we overcame by turning to the bootstrap resampling technique; (d) the improperness of the normality assumption for the bootstrap distributions of the WINcorr coefficients for which reason we preferred to apply a non-parametric approach for both the bootstrap procedure and the inference we decided to carry out the analyses between the α index and the ANS proxies according to a bivariate pairwise correlated with different strength and sign and that a multivariate approach could have taken into account these intertwined connections at best this would have required us to use methods of synthesis of the data having as disadvantages the facts of introducing additional margins of error in the analyses as well as making the reading of the findings less clear from a clinical point of view that profit from the analogous power of information hidden in the baroreflex as predictive tool This study was carried out in accordance with the recommendations of the Independent Ethics Committee of IRCCS Humanitas Clinical Institute (Rozzano Italy) with written Volume 8 - 2020 | https://doi.org/10.3389/feart.2020.574032 To understand the dynamics of magmatic systems one must first seek to characterize the time-dependent behavior of magma storage and ascent we do this through a combination of the Crystal System Approach and careful study of Fe-Mg interdiffusion in orthopyroxene This allows us to trace the pre-eruptive dynamics of magma plumbing systems We apply this novel approach on two large silicic eruptions (about 3–5 km3 DRE/eruption) that occurred in the central part of Dominica Island (Lesser Antilles Arc): the eruptions of Layou (∼51 ka) from Morne Diablotins and Roseau (∼33 ka) from Morne Trois Pitons-Micotrin two magmatic environments (MEs) are identified on the basis of orthopyroxene composition with a dominant reverse-zoning pattern from 50 to 54 to 54–59 mol% enstatite (En) three MEs are observed as represented by three populations of pyroxenes: En47-51 The normal-zoning pathway from En51-53 to En47-51 is significantly registered by crystals interpreted as convective mixing in a zoned reservoir The reverse-zoning pathway from En47-51 to En51-53 and also En53-58 is also significantly present supporting the mixing within the zoned reservoir but also suggesting mixing with a hotter magma possibly stored in another part of a sub-volcanic mush The crystal and glass compositions (melt inclusion and matrix glass) from both studied eruptions suggest heating and mixing between different magma pockets located within the mush that were the dominant process for mobilizing eruptible magma we constrain the associated pre-eruptive timescales by modeling the diffusive relaxation of Fe-Mg chemical gradients that originated within the zonation of the same orthopyroxene crystals Diffusion modeling was considered along the b-axis of 66 zoned orthopyroxene crystals for these two eruptions at a magmatic temperature of 850 ± 25°C we propose that the Layou and the Roseau magma reservoirs were rejuvenated and heated by ∼25–50°C about 10 years prior to eruption by the injection of an underplating creating the observed dominant reverse-zoning patterns of the erupted orthopyroxenes We thus have evidence that silicic mush can be re-mobilized over timescales of decades prior to eruption as previously suggested for Santorini and Taupo volcanoes How magma is stored at depth and how magma bodies assemble prior to eruption are two questions that are still widely debated. The architecture and associated dynamics of magma plumbing systems for silicic volcanoes have been in the last decade envisaged as transcrustal magma systems (Cashman et al., 2017) following the pioneering work of Bachmann and Bergantz (2003) Dominica volcanic activity thus offers the unique opportunity to investigate the magma dynamics in the plumbing system prior to repeated large silicic eruptions FIGURE 1. (2017) Red: deposits from Morne Trois Pitons–Micotrin eruptive center with dark red deposits corresponding to Roseau eruption Blue: deposits from Morne Diablotins eruptive center with dark blue deposits corresponding to Layou eruption (E) Layou pyroclastic density current deposit (F) Roseau pyroclastic density current deposit The results are discussed for their implications in terms of the more general framework of the magma plumbing system architecture and dynamics beneath Dominica we assess the rates of silicic magma rejuvenation prior to voluminous explosive volcanic eruptions The Lesser Antilles Arc results from the westward subduction of the North-South American plates beneath the Caribbean plate, at an average rate of 2 cm/yr (Wadge, 1984). The volcanic arc is 850 km long, and extends from the island of Saba in the north to Grenada in the south, with an eastward convexity. Dominica is located in the central part of the arc (Figure 1) Amphiboles in Layou are ferri-magnesio-hornblende with Mg# [Mg/(Mg + Fetot)] ranging from 0.52 to 0.60 Clinopyroxenes are augites with Mg# ranging from 0.48 to 0.71 Fe-Ti oxides are titanomagnetites (Mag71–75; Mag = magnetite mol%) and ilmenites (Ilm87; Ilm = ilmenite Phase equilibrium experiments starting from glasses of Roseau and Layou bulk compositions have been performed in an internally-heated pressure vessel, spanning a temperature range of ∼800–900°C, a pressure range of 200–400 MPa (under water saturation and under-saturation), and fixed oxygen fugacity of ≈ NNO +0.6 (NNO = Nickel- Nickel oxide buffer) (Solaro et al., 2019) By experimentally reproducing the natural melt and phenocryst rim compositions the magma storage conditions have been determined to be close to a pressure of ∼400 MPa a temperature of 850 ± 25°C with 7–8 wt% H2O dissolved in melt These parameters will be used for the diffusion modeling below Studied pumices and crystals come from the field campaigns performed previously for chronostratigraphic, melt inclusions and experimental petrological studies (Boudon et al., 2017; Balcone-Boissard et al., 2018; Solaro et al., 2019). The samples consist of white, homogeneous pumice clasts collected from the basal Plinian fallout deposits of Layou and Roseau (Figures 1C,D) Whole rock samples were gently crushed to liberate crystals and sieved 701 for Roseau) crystals were separated by hand-picking under binocular microscope and mounted in epoxy resin Orthopyroxenes were oriented during mounting to obtain b-c sections needed for diffusion modeling along the b-axis of crystals The method (Figures 2A–E) is based on the modeling as a diffusion profile of the core-to-rim traverse (Figure 2E) obtained by coupling a high analytical resolution profile by Electron Probe Microanalyses (Figure 2D) and a high spatial resolution profile by SEM (Figure 2C), following the methodology developed by Allan et al. (2013) and Couperthwaite et al. (2020) By using a library of profiles calculated for particular values of compositional contrast the appropriate curve shape to apply to a natural crystal can be selected from the library based upon compositional data and then scaled in width to solve for time The scaling factor needed is a simple expression of crystal composition the diffusivity used for the library profiles the diffusivity in the crystal and the ratio of diffusion timescales in the library and the timescale of the crystal As the scaling factor can be determined from data the timescale for the crystal can be simply solved as the only unknown The internal consistency of timescales obtained by this modeling method is tested by modeling several profiles on the same core-rim boundary of the same crystal and verifying for the acquisition of comparable timescales the pre-exponential factor (D0) and the activation energy (Ea) all have a logarithmic effect on final calculated timescales such that the final uncertainties are not Gaussian in time but asymmetric with larger error bar on the long-time side and smaller error bar on the short-time side with uncertainties approaching Gaussian behavior in log-time Here we have used the diffusion coefficient resulting from the parameterization of Ganguly and Tazzoli (1994) In summary, all modeled profiles and timescales have been here calculated for a fixed temperature of 850°C (±25°C) based upon the experimental work of Solaro et al. (2019) on these magma compositions. This yields diffusivities of the order of D ∼10–20 m2 s−1 (Ganguly and Tazzoli, 1994) without dependence of the diffusivity on oxygen fugacity The initial condition is considered to be a step-wise profile (i.e. instantaneous growth at a fixed composition followed by diffusional relaxation of the interface) which yields maximum timescales if all broadening occurs via diffusion Compositional profiles for these eruptions have been interpreted as pure diffusion profiles with no or limited recorded growth, i.e., considering profiles as due purely to diffusion. This constrains maximum timescales, because if the profiles have a growth component, the required timescale is shortened. The fitted profiles have features typical of diffusive character—smooth compositional variation of sigmoid shape (Supplementary Figure 2) Oscillations can be seen toward the rim but are minor in amplitude (±0.2 mol% or less) and could plausibly be attributed to noise growth-dominant profiles would display linear trends or curves on graphs of composition vs which would not be congruent nor consistent with a diffusion sigmoid calculated for the given initial compositional contrast The CSA technique has been successfully applied to the study of Etna, in order to define the main active magmatic environments (ME) prior eruption from crystal compositional zoning, their connections (by heating, decompression, mixing, mingling, crystal settling, etc) and the pre-eruptive processes occurring at depth (Kahl et al., 2011; Kahl et al., 2015) a ME is not necessarily a single batch of magma but may be different part of the same reservoir at depth a variety of ME were described based on olivine-melt equilibria and their occurrence with time We applied this method to orthopyroxenes (opx) that show variations in major-element composition (Enstatite content XEn) that can be related to changes in the magmatic system These changes can lead to either reverse zoning typically associated with higher temperatures typically associated with cooler and shallower environments The compositions of unzoned, single zoned and multiple zoned opx along rim-to-rim or core-to-rim traverses allow us to identify compositional plateaus (En mol% plateaus) (Supplementary Table S3) Compositional plateaus have been here defined as regions of the profiles with a minimum length of ∼15 µm (respect to 315–500 µm wide crystals) within which the En content of the crystal shows a constant value with accepted variability of ±0.5 mol% The composition of each plateau is then calculated as an average of all values defining the plateau The occurrence of plateaus can be interpreted as a result of crystal growth in different with residence times in each environment long enough to be recorded by crystal growth but also short enough that the compositional gradient has not been erased by diffusional equilibration The repetitive occurrence of plateaus indicates that compositional zoning observed in a given crystal has not been produced by pure fractionation during growth but by subsequent and fast changes during crystal growth in response to a change from one ME to another or physically transferring a crystal to a different melt assuming that crystal growth occurred sequentially from core to rim cores represent the first ME recorded by crystals while outer rims represent the last ME Analysis of compositional plateaus from core to rim allows us to track either movements of crystals and their carrying melts between different ME or modification of storage conditions by external processes (heating etc.) through the plumbing system until eruption This procedure has to be repeated on all analyzed crystals in order to define crystal populations and the frequently-travelled crystal transfer pathways This method allows a simple graphical representation that avoids much of the difficulties of combining and interpreting diverse and complicated zoning patterns between crystals of the same sample Here we applied this approach to opx to highlight magma transfers and the magnitudes of these transfers at depth between the different ME This approach is based on the recognition of the diversity of opx composition interpreted as ME changes we can identify the disrupting events at depth responsible for magma remobilization up to eruption That is also why a ME does not necessarily correspond to a physical lens of magma within the crust; this will be discussed hereafter A total of 1,452 opx were analyzed for the two eruptions (701 for Roseau, 751 for Layou), including their textural features and chemical compositions (Figure 3; Table 1; Layou crystals included both normal- and reverse-zoned examples while only reverse-zoned crystals were studied for Roseau (given the dominance of reversely-zoned crystals in this eruption). are too short for accurate discrimination by the method or that both processes are related The injection of a new batch of hot magma may be responsible for the magma convection within the intruded reservoir The complexity of the pre-eruptive magma dynamics for the Layou eruption confirms the hypothesis of a system heterogeneous in temperature, with parts of the magma stored at lower temperature (allowing quartz crystallization; Solaro et al., 2019) the CSA results suggest more direct paths and fewer magma transfers in the case of Roseau magmatic system Various studies have already used the Fe-Mg interdiffusion chronometer in opx to retrieve timescales of magma dynamics prior to eruption (Table 3) These include examples of eruptions across a large range of differentiated magma volumes Comparison of diffusion timescales obtained for different large volume silicic eruptions By studying the geochemical signature of zircon and sanidines, the timescales of magma storage and differentiation of voluminous high-silica rhyolites at Yellowstone caldera (Wyoming) have been investigated (Stelten et al., 2015) the authors argue that eruptible rhyolites are generated by extracting melt and zircons from a long-lived mush of immobile crystal-rich magma Residence times of the large-volume rhyolites (∼40–70 km3) are ≤1 kyr (conservatively <6 kyr) which suggests that large volumes of rhyolite can be generated rapidly by extracting melt from a crystal mush the Yellowstone magmatic system evidences that magma reservoirs spend most of their time in a largely crystalline uneruptible state and that eruptible magma bodies are ephemeral features The coupled investigation of crystal and melt composition lead us to identify the disrupting event at depth responsible for the magma mobilization leading to two major explosive eruptions in Dominica interpreted using the revised CSA approach provides a detailed view of the pre-eruptive architecture and dynamics of the crystal mush prior to eruption This highlights the diverse complexity of the magmatic system with several magmatic environments and short remobilization timescales several Plinian eruptions in the central part of Dominica have been described and should be investigated to detail the plumbing system dynamics in time and space The raw data supporting the conclusions of this
