Metrics details
the silkworm possesses a restricted feeding capacity
To investigate the impact of varying sucrose concentrations on silkworm growth and development
transcriptomic analyses were conducted on the silkworm midgut
A total of 909 differentially expressed genes (DEGs) were identified
with down-regulated genes predominantly associated with starch and sucrose metabolism
and amino sugar and nucleotide sugar metabolism
These alterations suggest that excessive sucrose consumption may suppress the carbohydrate metabolism pathway in silkworm
This study offers a comprehensive analysis of transcriptomic studies on high sucrose intake in silkworm
providing insights into the effects of varying sucrose concentrations on these organisms
silkworm enters a voracious feeding period
with a sharp increase in mulberry leaf consumption
begins to synthesize mass silk proteins in the silk gland
The digestion and absorption of nutrients in silkworms are inseparable from the digestive tract
accounting for approximately 78% of the total length of the digestive tract
Cylindrical cells are the main components of the midgut epithelium
especially well-developed in the posterior part of the midgut
capable of secreting digestive fluid and absorbing nutrients
the majority of the enzymatic digestion and the absorption of nutrients largely take place in the midgut
carbohydrate metabolism is a critical aspect of insect physiology
By gaining a deeper understanding of the mechanism underlying glucose conversion and metabolic regulation in insect
we can develop more effective strategies for silkworm rearing
potentially leading to increased silk production and more sustainable agricultural practices
we investigated the effects of adding sucrose with four different concentrations (6%
from the 5th instar molted silkworm (I5D1) to wandering (spinning the cocoon)
the diet supplemented with sucrose had positive effects on the development of silkworm
the growth situation of the 12% treatment group was relatively the better
we conducted a transcriptomic comprehensive analysis of gene expression of midgut in I5D3 silkworm under condition of sucrose 12% treatment
The results revealed various alterations in carbohydrate metabolism and glycolytic pathways induced by changes in sucrose consumption
The findings indicate that the appropriate sucrose concentration and the optimal feeding time will positively impact the development of silkworms
This research not only can provide valuable understanding regarding the impact of sucrose on the midgut of silkworm but also holds significant guiding implications for enhancing silkworm breeding efficiency
The silkworm 871 strain (monovoltine race) was provided by the Sericulture Research Institute of Chinese Academy of Agricultural Sciences
The rearing temperature should be maintained between 25 and 26 °C
and the humidity should be controlled between 70 and 85%
Ensure appropriate lighting and ventilation
Control the stocking density to avoid overcrowding
and uncontaminated mulberry leaves as the feed
In the preliminary stage of the formal feeding trial
we conducted a pre-experiment to select the appropriate concentration levels
as evaluation criteria and found that the silkworms grew slowly and appeared mass death in the 30% concentration groups
we decided to refine the concentration gradients to 6%
18% and 24% to allow for a more reasonable assessment of the experimental results
Select well-developed and similar-sized female silkworm at the 1st of the 5th instar (molted silkworm
Establish a Control Group (or C-Group) and four treatment groups with sucrose supplementation (or S-Group)
each with three replicates consisting of 150 silkworm per replicate
The C-Group was fed mulberry leaves washed with deionized water
while the S-Groups were fed mulberry leaves soaked in 6%
we firstly placed the calculated amount of sucrose into a certain volume of sterile water and heated it until fully dissolved
additional sterile water was added to reach the desired volume
All leaves were selected to ensure a similar state
minimizing significant differences among them
the petioles and stems of the mulberry leaves were removed
During the mulberry leaf immersion process
each leaf is individually soaked in a sucrose solution for 15 min to ensure that the mulberry leaves are fully exposed to the sucrose solution
we lay out the mulberry leaves taken from the solution flatly on a sterilized wire mesh for natural drying until that the mulberry leaves were moistened with sucrose solution without any dripping
Then the leaves were placed in the silkworm box for consumption by the silkworms
the density of silkworms was also controlled to ensure that each silkworm had access to a sufficient amount of leaf
the growth situation of the 12% treatment group was relatively the better among all treat groups
we conducted RNA-Seq of midgut in I5D3 female silkworm under condition of sucrose 12% treatment
The midguts of 10 female larvae per sample were collected from the sucrose 12% group and the control group
The total RNA of the sample was extracted by EASYspin Plus rapid tissue/cell RNA extraction kit of Beijing Aide-Lai Biotechnology Co.
and integrity number were assessed using the Nanodrop 2000 Bioanalyzer
Utilizing the fragmented mRNA as a template and random oligo-nucleotides as primers
the first cDNA strand was synthesized in an M-MuLV reverse transcriptase system
and the second cDNA strand was synthesized in a DNA polymerase I system using dNTPs as substrates
The purified double-stranded cDNA underwent end-repair
After the library construction was complete
initial quantification was performed with Qubit2.0 Fluorometer and the library was diluted to 1.5ng/ul
the insert size of the library was tested using Agilent 2100 bioanalyzer
different libraries were pooled according to the requirements of effective concentration and target data volume for Illumina sequencing (NovaseqX Plus)
The raw data have been submitted to the NCBI public database (NCBI.nlm.NIh.gov) with the accession number PRJNA1186106
Genes were represented as DEGs when the adjusted p-value < 0.05 and |log2 (fold-change)| ≥ 1
a functional enrichment analysis was conducted to pinpoint the GO terms and metabolic pathways that are significantly overrepresented within the set of DEGs
To determine the significance of these enrichments
with a p-value < 0.05 being considered significant compared to the entire transcriptome background
To identify genes that may not show significant differences in overall expression level but have significant biological significance
we conducted Gene Set Enrichment Analysis (GSEA)
and normalized enrichment score |NES| > 1 were selected for further analysis
The quantitative real-time PCR (qRT-PCR) was conducted to confirm the DEGs in the cDNA libraries, which was constructed from midgut of 5th instar larvae. BmGAPDH was selected for housekeeping gene. All the primers (Supplementary Table S1) were designed using primer 5.0 software (Premier Biosoft International
CA) and DNAMAN (Lynnon BioSoft) according to the rigorous principle
and the data was expressed as the mean ± standard error (SE) (n = 3)
the growth situation of the 12% treatment group was relatively the better among all treatment groups

Climbing returned to Indonesia for the second IFSC Lead World Cup of the year as the stunning Peninsula Island welcomed 55 men and 51 women for qualification
it was a first World Cup start of the year for Japan’s Mori Ai who once again showed her Lead strength by topping both routes for a joint first-place in the standings with Great Britain’s Erin McNeice
McNeice already has two World Cup medals from her two events in Keqiao and Wujiang
and the Brit is hoping that form continues
It does certainly look that way as she was the only other climber to top the two qualification routes with Mori
South Korea’s Seo Chaehyun and Italy’s Laura Rogora both progress to Sunday’s semi-final
Sitting in fifth was Slovenia’s Rosa Rekar who is taking some learnings from her previous event but also trying to battle the elements: “I feel a bit better than Wujiang
but it is different because it is so hot and humid here
We did a training session here in Bali before to try and get used to the conditions
“It’s a really nice place here with the water so close to the wall
but the most important thing is to focus on the competition and then enjoy everything when we are finished.”
Talking about her qualification round Rekar said: “I really enjoyed the qualification
The first route I think I could have done a bit better
Rekar is joined in the semi-final by teammates Mia Krampl
Lukan was one of five climbers in joint 23rd place meaning there will be a 27-climber semi-final
The others joining Lukan in the 23rd spot were Czechia’s Tereza Siruckova
Austria’s Flora Oblasser and Ukraine’s Anastasiia Kobets
France’s Hélène Janicot progressed into the semi-final in tenth position in her 75th Lead World Cup – a number that gives her ninth position on the all-time appearance list
Full women's Lead results can be found here
The only two climbers to top a route in the men’s competition were Japanese teammates Anraku Sorato and Yoshida Satone
With both reaching hold 32+ for their other attempt means the pair share first position
After climbing Yoshida said: “I’m happy I could send the second route
and I was struggling with the temperature.”
Yoshida is liking his time in the new World Cup venue: “It’s very nice here
It’s the first time we’ve had an event in like a resort so I’m excited for the competition and it is definitely a place you can relax
Yoshida has his sights set firmly on the road ahead in Bali: “I’m ready for the semi-final and final
it’s all part of the road to being a World Cup champion.”
Team Japan will have all six climbers in the semi-final with the likes of Wujiang silver medallist Suzuki Neo also making it through
but they aren’t the only team with multiple climbers – there will be lots to cheer for the home crowd also
Indonesia will have three representatives in the semi-final as Putra Tri Ramadani was eighth
Raviandi Ramadhan was 12th and Muhammad Rizky Syahrafli Simatupang was in the 24th and final position
Another country with three to progress was Switzerland who had Jonas Utelli
Sascha Lehmann and Nuno Grünenfelder all inside the top ten while Spain’s Alberto Ginés López continued his good start to the season moving through in third
Twentieth on the all-time men’s appearance list
Germany’s Sebastian Halenke progressed in 23rd position in his 61st Lead World Cup
Full men's Lead results can be found here

Mori Hosseini has been the UF Board of Trustees chairman since 2018
When nine top donors and administrators recounted the full story of former UF President Ben Sasse’s resignation
one name came up for each of them — Morteza “Mori” Hosseini
has served as the UF Board of Trustees’ chairman since 2018.
The board has influence over the employment of Sasse’s former staff members. It’s unclear how many of the staff Sasse hired have remained employed at the university, including previous aides from his time as a United States Senator. Several remain in the UF directory, but they are missing main web pages or profiles on the President’s Cabinet website
Hosseini’s position comes with considerable power
including the ability to override certain presidential decisions
Hosseini also influenced major initiatives across the university
In a Board of Trustees meeting on July 23
Hosseini said he asked UF’s Chief Financial Officer Taylor Jantz to allocate $25 million to support faculty “who work so hard to serve our students and do great research.”
Hosseini did not elaborate on how the $25 million would be distributed
Sasse's office employed more than 30 staff members
a stark contrast to interim president Kent Fuchs
who had fewer than 10 during his presidency
James Wegmann and Dan Dillon are among those still listed on the UF President cabinet website and are believed to still be employed
is Sasse’s former Senate communications director and serves as UF’s vice president of communications
Wegmann replaced UF spokesman Steve Orlando, who made $270,000 a year in the position. Additionally, Orlando had nearly 30 years of experience in UF media relations before he was demoted to Wegmann’s deputy
Orlando has not responded to The Alligator’s inquiries
It still remains unclear the status of:
The university has not responded to The Alligator’s public records request regarding the status of employment for Shirley
Several reports from The Washington Post and other news outlets outline Hosseini’s notable contributions to Gov
DeSantis had taken several undisclosed trips on private jets belonging to Hosseini.
In 2019, Hosseini donated a golf simulator worth $27,500 to DeSantis’ mansion and provided him with private flights as recently as February
DeSantis reappointed Hosseini as chair of the Board of Trustees.
Hosseini has also served as a trustee at Embry-Riddle Aeronautical University since 1999

In 2022, Hosseini’s companies gave at least $361,000 to political groups aiding DeSantis’ reelection campaign, with a report by The Orlando Sentinel noting
“Hosseini has donated nearly $3.5 million over nearly two decades” to conservative committees
For DeSantis’ 2024 presidential campaign, Hosseini was amongst several donors who donated more than $1 million to DeSantis’ political action committee, The Alligator previously reported
UF interim president Kent Fuchs said he is unsure of Hosseini’s political affiliation
but maintains that the chairman of UF’s Board of Trustees is expected to work closely with the Senate President
regardless of where they stand politically
“Does he have strong personal relationships
“He uses that to work on behalf of the university
Hosseini is not compensated for his work at the University of Florida but dedicates extensive time and energy to the institution due to his passion for it
Although Fuchs has had disagreements with Hosseini and previous chairs
he added that they have always found common ground for the sake of the university
a leading Democratic fundraiser who graduated from UF’s law school
referred to Hosseini as a “money guy for the [government]” in an email to The Alligator
Morgan alleged he most recently gave the university $1 million. In a series of social media posts on X
Morgan said he knew many of the UF Board of Trustee members — especially James W
when asked by The Alligator if he had any comments on the UF Board of Trustees
Morgan wrote in an email response that he didn’t know enough about them
“they are political appointees who do what the governor wants
toothless tigers who donated to his campaign.”
Others who took to X to share their frustrations
such as Republican Representative Matt Gaetz and the DeSantis administration
Hosseini has not responded to The Alligator’s inquiries about Sasse’s resignation or the donors and officials who testified to the extent of his involvement
On Aug. 15, Jimmy Patronis, Florida’s Chief Financial Officer and State Fire Marshal, took to X to announce that his agency would be reaching out to the Florida Board of Governors to offer auditing support and said that the Board should
“investigate this issue to ensure tuition and tax dollars are being properly used.”
DeSantis’ office has not responded to inquiries regarding the audit’s current status
Sasse has expressed an interest in becoming president emeritus and will be teaching college classes at the Hamilton Center
a GOP-mandated civic program with a focus on the education of classic Western literature
Western philosophies and the foundation of American politics.
As part of his severance package, Sasse will also receive a $1 million base salary until 2028
the realization of Sasse’s plans and the future of his employees are all dependent on the approval of the UF Board of Trustees — and especially on Mori Hosseini

the newest exhibition at Boston’s Museum of Fine Arts invites visitors to contemplate mortality through the eyes of young curators
“Curated by Teens: Death as a Constant Companion” was developed by five Boston-area high school students — Robert Cronin (Boston Collegiate Charter School)
Alayssa Diaz (Cristo Rey Boston High School)
and Nicole Wu (Braintree High School) — in collaboration with the MFA’s Center for Netherlandish Art
exploring artistic representations of death across cultures and centuries
The teen curators behind “Death as a Constant Companion” made thoughtful choices in selecting artworks that invite viewers to engage in a broad dialogue about mortality
The exhibition successfully explores different facets of death
from its impacts on humans and animals to the role of religion in coping with loss
while the artworks themselves are compelling
the organization of the gallery could have been more intentional to create a smoother narrative flow
As visitors enter gallery 243A and turn to the right
the exhibition begins with a striking display of “Vanitas Still Life” by Cornelis Norbertus Gijsbrechts
reminding viewers of life’s inevitable end
an extinguished candle signifies the swift passage of time and the brevity of life
An hourglass further emphasizes this notion
illustrating the relentless flow of time and the fleeting nature of existence
viewers come across “Still Life with Dead Partridge and Kingfisher” by Simon Pietersz
The painting presents a partridge hanging by its feet
positioned just above a lifeless kingfisher
“Still Life with Dead Heron and Swan” by Elias Vonck continues the theme with another striking portrayal of lifeless birds
“The Butcher Shop,” by David Teniers the Younger
a graphic scene of a pig being slaughtered
introduces the idea of human intervention in the cycle of life and death
These initial works immediately confront viewers with death in a stark
the exhibition transitions to a more philosophical exploration of mortality
A painting featuring a man and a sphinx head introduces a nearby text on the wall discussing humanity’s reluctance to directly talk about death: “Most humans don’t openly discuss death
it feels uncomfortable.” This section establishes the exhibition’s intent — to create a space that encourages reflection on death as a transformative experience rather than something to be feared
The gallery then presents a sequence of eight diverse artworks that depict humanity’s relationship with death
and Monstrous Demon” by Philips Wouwerman portrays a knight in battle
reinforcing the belief that while the body may perish
“Young Man with a Skull” by Lucas Leyden presents a contemplative young man gazing at a skull
George Reverdy’s “Allegorical Subject with Death Holding a Globe” takes a more direct approach
personifying Death as a skeletal figure clutching a globe
an ominous reminder of death’s universal power
This section culminates in “Still Life with Carp” by Pierre Niehen
unlike the previous works focusing on human mortality
is depicted as a religious motif rather than a source of sustenance
its placement at the tail end of a series dominated by human death feels slightly disjointed
Wall text at this point addresses the connection between animals and death
noting that depictions of dead animals in art are often killed at the hand of humans
two additional paintings — one featuring a dead partridge and another with two quails and a kingfisher — bring the exhibition to a close
While the exhibition succeeds in prompting reflection on death
the placement of artworks could have been more cohesive
the three paintings of animal death at the beginning might have been more effective if positioned after the text discussing animals and death
the sequence of seven human-focused works would have felt more unified without the sudden interruption of “Still Life with Carp,” which would have been better placed alongside the other animal death pieces
Although “Death as a Constant Companion” effectively sparks conversation about mortality
the exhibition does not seek to romanticize death
The teen curators bring a fresh perspective to this age-old subject
using art to explore their understanding of life’s impermanence
Curated by Teens: Death as a Constant Companion is on view in the William A
Coolidge Gallery at the MFA at Boston through Nov

When renovating a historic home, it helps when the owners have an appreciation and respect for design. And luckily that was the case with this Portland, Oregon
house designed by renowned architect Saul Zaik
who was known for his contributions to Pacific Northwest architecture
“Our clients are a young couple with a deep appreciation for Pacific Northwest mid-century and contemporary minimalist design,” says Project Architect Robert Arlt of SHED Architecture
“They came to know and admire Saul Zaik’s work through open houses and the local design community
The property came up for sale just a month after their wedding.”
The home, which is called The Mori House, was designed for Zaik’s friend and next-door neighbor, interior design Howard Hermanson. It features an elevated deck, which connects to a detached teahouse pavilion. Zaik was known for his modernist residential designs
integrating homes into their natural surroundings and prioritizing natural materials and light
“We inherited a unique structure within a beautiful forested site and the primary driver for the project engaged the principle of addition through subtraction,” says Arlt
“The house had accumulated many layers of finishes
and a tight spiral stair connected the walkout basement and loft under the large skylight which inhibited circulation
Given the task of reconfiguring and providing safer circulation
the question we kept coming back to during the design investigation was
‘How do we do that so the large central skylight can organize and impact every space in the home?’”
the clients knew they wanted to reconfigure the main floor’s living
and flow while maintaining clear separation from private areas and renovate the two main level bedrooms into a large primary suite,” says Arlt
the existing spiral staircase needed to be reconfigured to optimize safe vertical circulation for their growing family and further define the division between public and private spaces.”
The final goal was to update the home’s design to reflect the homeowners’ personal style
which is described as “‘East meets Pacific Northwest’ modernist minimalism
and natural-light-accentuating materials.”
When planning the renovation
the SHED team took the mindset of “What would Saul do?” and used Zaik’s other PNW mid-century remodels as inspiration
The team removed the existing loft and spiral staircase
unveiling a nail-laminated wood roof that extends to a central skylight
There’s now an entry closet and powder room in the space where the spiral staircase used to be
The SHED team incorporated a new horizontal window above the cooktop
which offers views of the mature rhododendrons outside
The kitchen opens into a dining nook that includes a custom fir bench in front of floor-to-ceiling glass
A new glass door provides direct access to the deck and teahouse pavilion
The newly renovated living room features a relocated fireplace
giving more views of the forest (and Mount St
The former fireplace area has a built-in bar and tech closet
“The vaulted living space and skylight are made possible through a unique hip-roof structure,” Arlt explains
and solid 2×6 nail-laminated framing free-spans between to form the roofline and exposed wood ceiling
The finished side of the nail-laminated 2×6 was exposed in the primary bedroom and the whole surface was blasted with walnut shells to restore and brighten the surface.”
The primary suite features fir slats above the door
“This suite transforms an existing bedroom and bathroom into a spacious primary bathroom with an adjacent walk-in closet,” Arlt says
“The primary bathroom includes a Japanese soaking tub
custom medicine cabinets with integrated LED lighting
and transom windows to borrow light from the skylight.”
On the southwest side of the house is a new switchback stair that leads to the lower level hangout space that overlooks the forest
The lower level also includes three private
The renovated home feels like you’re amongst the trees—true to Zaik’s natural surroundings ethos
“We have heard they enjoy the quality and detail and coming home to a feeling of lightness,” Arlt says of the homeowners

BUT DIFFERENTClose at the top of the women's Lead competition
but there will be no repeat of Wujiang shared-gold drama in Indonesia
Looking at the finals start lists for the last act of the IFSC World Cup Bali 2025
the two names at the top of the list seem very familiar
Great Britain’s Erin McNeice and South Korea’s Seo Chaehyun shared the top step of the podium
and in Bali they are again at the top of the standings
Both McNeice and Seo topped the semi-final route to progress to another Lead final
but this time its advantage McNeice who had the better qualification round
It means the British climber is in first and the South Korean in second
so no chance of what surely would have been an historic moment with a double-double gold
After her climb Seo said: “I’m super happy to reach the top
After falling on the first route in qualis I was quite nervous today because it looked a bit hard and a super pumpy route
Mori fell just short of the top on that dyno move Seo was worried about but still did enough for a final place in third
Also coming up just shy of the dyno was Laura Rogora
The Italian had done more than enough though on hold 48+ for a final spot sitting fourth
Despite only just making the semi-final in a four-way tie for 23rd position
young Flora Oblasser of Austria put it out of her mind and climbed to hold 38 early on in the round
the Austrian then had the long and agonising task of sitting and watching
her climb was enough for fifth position and another final appearance
the final eight was confirmed with Slovenian duo Rosa Rekar and Mia Krampl joined by Australia’s Oce Mackenzie
Finishing in 21st position was Japan’s Kuzuu Mashiro who
made the semi-final for the first time which meant a personal best score
don’t be surprised to see her in a lot more
Japan’s Yoshida Satone topped the qualification standings and kept up that form also topping the semi-final
His 36+ was the high point on the men’s route
and the Japanese climber is looking good for a Bali podium place
Germany’s Yannick Flohé was next best on hold 35+ and is feeling a lot better in Bali than he did last week in Wujiang: “I think I did what I could even though I slipped in the end
I’m happy I’m safe into the final and I didn’t have to suffer and wait for people to fall like last time
Talking about the semi-final route Flohé said: “We all thought it was relatively easy until the head wall
it looked quite juggy but turned out to be quite cruxy with some rest
“There was a few uncomfortable moves to deal with and the pockets section below the head wall was way harder to deal with than we expected as there were some little jibs inside which made the pockets way worse.”
Fresh off the Lead podium in Wujiang are Spain’s Alberto Ginés López and Japan’s Neo Suzuki
and both stay in with another medal opportunity progressing in third and fourth position
Japan once again has multiple finalists with Omata Shion moving into the final in sixth
with South Korea’s Lee Dohyun splitting the Japanese teammates in fifth
the home crowd of Indonesia love supporting their climbers
and they will have something to make some noise about in the men’s final when Muhammad Rizky Syahrafli Simatupang steps out onto the stage
The Indo climber made it into the final in seventh position
one place above France’s Max Bertone who again makes a Lead final
Positions three to seven all hit hold 33+ with Bertone making hold 31+
which gave him a final place thanks to countback to qualification with Italy’s Filip Schenk just missing out on the same hold
Another climber to miss out was the only 2025 World Cup golden winner so far in Boulder or Lead
Olympic champion Toby Roberts missed finals in China
South Korea’s Noh Hyunseung finished in 12th
Indonesia’s Putra Tri Ramadani finished in 17th and France’s Victor Guillermin was 19th
Metrics details
mori is a model species of lepidopteran insects
The genome assembly has been successively updated since the whole genome sequences were first determined in 2004
chromosome-scale genome assemblies of not only standard strains but also practical strains have been reported
We successfully constructed a chromosome-scale female genome assembly of p50ma
a standard strain developed by Kyushu University
This assembly is now certified as a ‘reference’ in the NCBI datasets
here we report the achievement of gene model construction based on the transcriptome information
The assembly harbours 16,295 protein-coding genes
we performed ATAC-seq in early embryos and comprehensively identified open chromatin regions
small RNA-seq (sRNA-seq) targeting PIWI-interacting RNA (piRNA) was performed in testes
and early embryos to identify piRNA clusters comprehensively
These data will increase the usability of p50ma as a standard strain and facilitate NBRP users to exploit this strain
assembly by short reads would be easily confused
a new standard strain that Kyushu University and the National BioResource Project (NBRP) silkworm recently developed
we present the annotation information of the p50ma female genome assembly to facilitate the use of this strain
we performed embryonic ATAC-seq to identify open chromatin regions
Since we considered that any studies which are conducted in the context of ignoring organisms
piCs in the p50ma genome were identified in a tissue-comprehensive manner
These datasets will increase the usability of this strain as a platform for piRNA research
B. mori strain, p50ma was provided from NBRP silkworm (https://shigen.nig.ac.jp/silkwormbase/)
Kyushu University developed p50ma strains by hybridization of p50 strain and T-series strain: F1 individuals were repeatedly sib-crossed for at least twenty generations
Larvae were fed on fresh leaves of Morus alba under a long-day condition (16 h light/8 h dark) at 25 °C
The outermost to the innermost circle shows I

Total RNA was extracted using TRIzol reagent (Invitrogen) according to the manufacturer’s protocol
Embryos were sampled 24 hours after oviposition
The two aliquots of testis and ovary-derived RNA samples were subjected to RNA-seq and sRNA-seq
The three aliquots embryo-derived RNA samples were subjected to sRNA-seq
The sRNA-seq library was prepared using TruSeq small RNA kit (illumina) according to the manufacturer’s protocol with a slight modification
a region of 147–158 nucleotides was extracted in the purification step of the cDNA construct using BluePippin (Sage Science
The constructed library was sequenced on the illumina Hiseq2500 platform (illumina
RNA-seq library was prepared using NEBNext Poly(A) mRNA Magnetic Isolation Module (New England BioLabs) and NEBNext® Ultra™ ll Directional RNA Library Prep Kit (New England BioLabs) according to the manufacturer’s protocol
The constructed library was sequenced on the illumina Novaseq6000 platform (illumina
the library was constructed using Sequel Iso-seq Express Template Prep (Pacific Bioscience
USA) according to the manufacturer’s protocol
The constructed library was sequenced on the PacBio Sequel platform (Pacific Bioscience

a talent of hololive production’s very first English generation hololive English -Myth-
this solo concert marks the start of hololive production’s continued growth at bringing its amazing performance events overseas
We hope that this report conveys the excitement and energy of the concert
Though hololive production has hosted group concerts in the United States such as hololive English 2nd Concert -Breaking Dimensions- on August 24th and 25th
Mori Calliope 2nd Concert “Grimoire” is the first concert in hololive history to star a single performing talent overseas and outside of Japan
made her debut within hololive English back on September 12th
As an early pioneer for English-speaking VTubers
this year marks her fifth year together with the agency
and within an impressive two years of activity
signed with major record label Universal Music Japan
a total of 22 of her original songs were performed over the course of the Los Angeles solo concert
With a sold out venue and lines of fans present long before the doors were set to open
the concert once more reaffirmed hololive production’s growing popularity in its efforts to expand the sensation of VTubing across the world
the concert welcomed them with an introductory video into Mori Calliope’s world as a grim reaper— who rather than aspiring to climb higher and higher to reach the skies as a typical idol might
instead journeys further and further into the deepness of the abyss
This introduction was followed by ongoing dark and edgy aesthetics which laced each following song performances
Clad in a cap and fashionable street-style outfit
Calliope opened with her first song of the night
The energizing performance featured pink burning lights as well as bursts of smoke pillars
Calliope was joined by four skeleton back up dancers
and supporting her through “未来島 ~Future Island~”
a song released in 2023 made to commemorate the release of popular manga series ONE PIECE’s 106th volume
The friendly skeletons continued to dance with Calliope through “Yona Yona Journey”
which shifted the atmosphere of the stage to that of a futuristic yet somehow simultaneously nostalgic dance club
Next up came a snazzy and high-energy performance of “NEZUMI Scheme” with an animated backdrop that featured bold animations and cartoons
During “FLASH BANG” which followed shortly after
Calliope introduced us to Japanese graffiti aesthetics and a groovy beat
“Make ‘Em Afraid” further amped the excitement of the audience with its playfully hypnotic melody line
With the crowd clapping along to help carry the tempo
Calliope embodied her grim reaper image and empowered the audience with her song
With a shift in mood first induced by visual noise and glitch art
Calliope appeared on stage with a bass guitar shaped like a large scythe
A stand mic of bones gave the performance an extra heavy metal touch
and with a genre change to hip-hop rap rock
some fans even began to throw their heads and hair along
a collaboration song with METAL GEAR SOLID
a large snake and a code matrix were featured in the backdrop while Calliope drove through the earworm chorus again and again
which enraptured the audience in a haunting melody
Large skeleton hands enveloped Calliope after
and revealed a new costume shown on the concert’s official artwork— a black outfit with lace and other gothic elements as well as horn-like headpiece
she continued with the lively song “DONMAI”
“Last Days” which showcased Calliope’s singing ability and higher voice register near a cathedral gate and beneath a large
allowed Calliope to similarly maintain a slower and relaxed energy
the stage presented an underwater scene with tropical fish while Calliope sang “through the blue”
the crowd waved their pen lights along to the acoustic strums of a guitar
and Calliope strutted about the stage and inspired confidence through her dancing
pulling the energy up from the previously relaxed three pieces
heavier guitars returned and the audience sang along to the soulful and powerful melody
skeleton backup dancers returned to join Calliope for “Go-Getters” the ending theme song of the anime “Suicide Squad: ISEKAI”
and Calliope once again emerged on stage in a casual concert T-shirt and cap
“Scuffed Up Age” showcased urban and savvy style first
while “glass slipper” embodied the loneliness that comes with the nature of being a grim reaper after
Calliope plunged the crowd into her dark world with the song “Black Sheep”
as the final song and epilogue to the show
Calliope dedicated the spooky “Left for Dead Lullaby” to her fans
which conveyed the concert’s preluding image of falling deeper and deeper into the abyss
Calliope’s evolution over the past five years is clear with this concert
and she displayed the prowess and skills she gained as an artist in this mega milestone for not just herself
but also paved a way for hololive production itself in its continued expansion
It’s amazing that we were able to share this with you
and we can’t wait to see where this journey takes us all

Fans waved the electronic instruments popular in the anime community
and cheered on a digital avatar while its creator performed from behind the stage
The creator conceals her true identity to fans
Mori Calliope has 2.5 million subscribers on her main YouTube channel
where she posts music videos in both English and Japanese
and streams herself playing games like Minecraft
Some of her livestreams in the last three months were almost eight hours long
On Wednesday, I went to Mori Calliope's second solo concert. Tickets ranged from $95 for general admission floor tickets to $400 for four balcony table seats, according to the show's official website
I was invited to check out the event for free with a press pass
If you're still wondering what a VTuber is
Although small in terms of the overall creator economy
the genre has a surprisingly dedicated fan base — one unlike I've seen before
most of the fans at Mori Calliope's show appeared to be in their late teens and 20s
It was the type of engaged fandom I've really only seen YouTubers conjure with younger audiences before
The crowd seemed like one you'd find at an anime convention
The show was also more highly produced than other YouTuber events I'd seen
The lasers and lighting matched the color of the VTuber's virtual stage
making it appear like she was physically there
Mori Calliope is one of dozens of VTubers taking over YouTube
Hololive manages Mori Calliope's business ventures and YouTube channel
similar to the way talent agencies do for other creators and stars
The company's market capitalization is about $160 billion yen
It recently opened an office in Los Angeles as part of a US expansion
Cover CEO Motoaki Tanigo told Business Insider
Hololive's English-speaking audiences are still a fraction of those in Japan
Hololive's VTubers had 59 million subscribers in Japan and 32 million subscribers overseas as of December 31
with about two-thirds of those in English-speaking regions
according to the company's February financial presentation
Tanigo said music like Mori Calliope's is one of the top ways its talent reaches new audiences
"The popularity of these events proves that North American audiences have a tremendous appetite for VTuber content," Tanigo said in a January interview conducted through translators
"Our goal is to elevate VTubers alongside popular Japanese exports like manga
One concertgoer told me fans lined up as early as 3 p.m
When I arrived about an hour before the start time
a line to get into the venue wrapped around the block
Fans held plushies of Mori Calliope and other characters from her videos
They dressed in merch and other apparel that matched her character's logo and signature pink
I also saw several fans ahead of me dressed in cosplay
people stood in line for Mori Calliope merchandise
Hololive also sold the event merch online and at a Los Angeles pop-up earlier in the month
Part of the show was livestreamed for free on YouTube for Mori Calliope's global audience
the VTuber directed those viewers to a link where they could buy tickets to watch the rest of the performance online
The tickets ranged from $43 to $51 on platforms like SPWN and Streaming+
I overheard groups of fans outside the venue exchanging information including social media handles and Discord groups
They cheered as a pink car wrapped in images of Mori Calliope drove swiftly past them
2025 — An earlier version of this story misstated the number of Hololive's VTuber subscribers from a financial presentation
There were 59 million subscribers in Japan as of December 31

The third-generation property scion opened up to Forbes Asia about her approach behind the rebalancing act from the firm’s headquarters in Tokyo World Gate
a 38-story complex in the bustling Toranomon district that combines green spaces
residences and a hotel from Marriott’s luxury brand
It builds on the legacy of her late grandfather
a force majeure whose large-scale urban redevelopments shaped the capital city's towering skyline in the last half of the 20th century
The fiscal year is the 12 months through March
Date has implemented an ambitious spending program to unlock more value for the Mori group company that her father
built into one of the country’s biggest property players
Mori Trust is targeting ¥70 billion ($457 million) in operating profit on ¥330 billion in operating revenue
hotels and property sales each contributing ¥100 billion to the top line
“It’s not enough to just keep doing the same thing,” Date says
you can take risks to gain that something extra
that is hotels.” Japan’s luxury hotel market is projected to see annual compound growth of over 4% over the next eight years
according to India-based consultancy Imarc
as the government looks to position itself as a premium destination for those with cash to spend wanting a unique travel experience
As part of a ¥1.2 trillion investment across its group businesses
Mori Trust will add 2,000 hotel rooms to the existing 5,300
It has some two dozen hotels under construction or in planning stages across Japan
in addition to the 35 hotels already operating—most managed in partnership with some of the world’s leading hotel chains
group income was flat at almost ¥263 billion from a year earlier
But its hotel revenue jumped by half to over ¥66 billion—outpacing earnings growth at the group’s other businesses
providing evidence that Date made the right choice
while property sales (which mainly consist of condominium units) fell by a quarter
The last two segments are already near ¥100 billion of operating revenue—with property sales in recent years far above that
but hotels still have the most room to expand
Mori Trust’s hotel “business base is solid
as indicated by the efforts to attract foreign-affiliated luxury hotel brands to Japan ahead of [its] peers,” Tokyo-based Japan Credit Rating Agency analysts Mikiya Kubota and Takeshi Rikawa wrote in an August ratings note
While other major Japanese developers’ hotel units account for a minor percentage of revenue
such as Mitsui Fudosan (9%) and Tokyu Fudosan Holdings (5%)
hotels accounted for about a fourth of Mori Trust’s operating revenue of ¥263 billion in fiscal 2024
with office leasing making up 35% and property sales
Interior construction work for offices and commercial spaces accounts for the rest
Under its latest strategic initiative released in November
Mori Trust will keep investing in traditional projects along with those going into hotels
with plans to add 379,000 square meters of new office
commercial and retail space—on top of a current 1.1 million square meters—and 800 high-end condo units
Date declined to give an investment breakdown
including its overseas investment—targeted at 20% in a preceding strategic plan—but indicates that offices will account for largest portion of the outlay
“It depends on the opportunities,” she says
“Offices are a stable business [because of long-term leases]
but there’s the potential that it will grow rapidly
condo sales are a business where investments can be recouped quickly,” she adds
I thought about making it possible to respond quickly to different economic changes.”
Given its geographic and asset spread—Mori Trust has 64 office buildings
condos and commercial properties in addition to its hotels and resorts
mainly across Japan and the U.S.—there’s no one-size-fits-all approach to navigating global turbulence
Mori Trust emerged from the pandemic relatively unscathed
with operating revenue up almost 13% between fiscal 2019 and fiscal 2023
But it now faces an aftermath of fluctuating exchange rates
tight labor markets and soaring construction costs
Operating income fell 18% to nearly ¥54 billion in the fiscal year through March 2024 year-on-year on a drop in real estate sales after booking a large office sale the previous year
The majority of assets are owned by Mori Trust
with some also held by two listed subsidiaries
Tokyo-based Mori Trust Reit consists of 20 office
develops and sells mid-tier to luxury condos
which had over ¥80 billion in revenue in the year through March
is focused in the region of Japan’s second-most populous metropolis
and accounts for the majority of Mori Trust’s property sales unit’s operating revenue
Shares of the larger Mori Trust Reit (market cap: ¥217 billion)
which haven’t yet recovered to pre-pandemic levels
which had ¥3.8 billion in revenue last fiscal year
Date says a shift in focus to hospitality not only spreads risk
but also shows investors and lenders that it has a stable earnings base
“Raising funds during good times is a given but being able to during the bad is a must for a company,” she notes
That includes expanding its property portfolio beyond Japan to ensure steady cash flow
She spent $700 million in 2023 for Mori Trust’s first investment in New York City
in partnership with New York-based SL Green Realty
which followed a $531 million purchase to buy an office tower in Washington
buys included acquiring an interest in a life science lab and office in Boston last year
two office buildings in Virginia (2022) and an office complex in Silicon Valley (2019)
the company announced its investment in a housing development in Bangkok with Thailand’s Major Development
saying it expects the 44-unit gated community to garner ¥9 billion in sales
Mori Trust entered the hotel space in 1976 through its domestic Laforet Hotels & Resorts
a corporate membership club that provides priority access to client companies
Its first upscale tie-up with an international brand was in 2005
the 291-room Conrad Tokyo—owned by Mori Trust and operated by Hilton—was among the international high-end luxury hotel brands that could compete with Japan’s iconic brands such as Hotel Okura and Imperial Hotel
Five years later came the 292-room Westin Sendai in northern Japan
which Mori Trust owns and operates as a franchisee—its preferred approach
where it shoulders the investment and operating risk but has all the upside potential and management freedom
(Westin is a Marriot marquee.) “We thought
‘How can we make it viable to invest in new hotel development?”’ she says
and that we could manage an overseas franchise with the Westin.”
From there Mori Trust joined forces with Marriott International
Today about half of the hotels it wholly or partially owns are operated through Marriott brands
including three from its The Luxury Collection
a Sheraton on Okinawa and two Edition properties in Tokyo
And while Mori Trust owns five hotels nationwide under the Laforet brand
another eight were rebranded as Marriott and Courtyard by Marriot hotels
Mori Trust has also expanded its partnership with Hilton to two more hotels under the U.S
and inked an agreement to open a hotel with IHG Hotels and Resorts in Nagasaki
a Marriott Luxury Collection hotel in Kyoto
The current outlook for the Japanese hotel property market is positive and overseas investors have “very high interest,” James Yukio Abe
investment sales chief at consultancy JLL Hotels & Hospitality in Tokyo
JLL forecasts overseas investment into Asia-Pacific hotels will rebound to $11.6 billion
An essential component of Date’s vision is to attract more premium guests outside of Japanese cities
Last year Mori Trust completed the 43-room Shisui hotel
the former prefecture governor's residence in the ancient capital of Nara
which falls under its in-house brand Sui and is operated by Marriott’s The Luxury Collection
Other dual-branded properties are the 39-room Suiran in Kyoto and the 58-room Iraph Sui on Okinawa
where Mori Trust owns mainly Grade A office space by floor area
international property consultancy CBRE notes
there’s a lag between a decision to reduce space or move out
“A 2% or 3% vacancy rate is extraordinarily low
but they’re at the cusp of rising again because Japanese firms are becoming more active in [office] searches,” Date says
there’ll be a dearth of space and prices will quickly jump.”
One lingering Covid concern is that firms may downsize their office space as more staff work from home
Those that reduced their offices probably realize that they don’t have enough space,” she says
and we’re now heading toward a recovery.” By late next year
Mori Trust plans to open its latest Tokyo development
a 13,100-square-meter site including a yet-to-be-named hotel
(The firm hasn’t disclosed investment costs.)
Based on improving office and hotel earnings and cash flow as new properties open
and a “conservative financial management policy” balancing cash and investment
Japan Credit Rating Agency upgraded Mori Trust Holdings’ credit a notch to AA from AA-
Meanwhile Date has worked to keep the group’s debt in check: At 0.78
Mori Trust’ debt-to-equity ratio is about half the gearing of its peers such as Tokyu Fudosan and Mori Building
an issue for Mori Trust will be maintaining good financial standing while making large-scale investments
An independent Mori Trust came into existence after the 1993 death of Date’s grandfather Taikichiro
an economics professor who pivoted to property development at age 55
and whose Mori Building helped transform Tokyo's residential streets into a modern urban hub
Following a disagreement over corporate strategy
parted ways starting in 1999—with Minoru taking over the family’s Mori Building and Akira turning Mori Trust
Today Akira and his family share a net worth of $4.4 billion
Minoru’s widow who inherited part of his estate including a share of Mori Building
Date joined Mori Trust at the age of 27 in 1998
after a stint at the now-defunct Long-Term Credit Bank Research Institute
where she earned a graduate degree in urban planning
After her two older brothers left the company
she was appointed president in 2016—a rare corporate move in a country where the eldest son usually takes the top job at family-run firms
who likely has decades to go before retirement (Akira is 88 now)
says that there is no plan to have a family member succeed her nor are there other family members
“My father used to tell us that ‘it’s not a given that your generation will run the company,’” she says
“He always said that ‘the most qualified person at the time will be chosen.’”
to increase luxury hotel offerings in Japan dovetails with government efforts to bring high-end tourists to Japan
The country aims to double visitor arrivals to 60 million by the end of the decade from a projected 35 million this year
who sits on several tourism advisory panels across government and business organizations
acknowledged the problems facing the hospitality sector
especially given the labor shortage and need to improve working conditions and wages
Other issues exist like transportation bottlenecks
including domestic flights that have yet to return to pre-pandemic levels and a lack of capacity to move people from airports and train stations to their final destinations
she says the current goal of 60 million visitors is too conservative and the government looks to fall short of the targeted ¥15 trillion ($98 billion) in inbound spending by 2030
That’s partly because fees for activities and cultural attractions
“The government needs to step back and reassess its target
what kind of visitors its strategy wants to attract and how the country is going to host them,” Date says
Steps include relaxing rules to allow more ride-sharing and increasing earmarked hotel and visitor taxes to help pay for tourist infrastructure

Today’s Gospel begins shortly after Jesus left the Temple where
he had condemned the Scribes’ desire for honors and flattery and wealth and had praised the widow’s embrace of poverty and her absolute trust in God’s providence
Those messages are clear to us if we listen
They’re a call to seek first the Kingdom of God
and trust that all you need will be added unto you
Don’t start with your focus on worldly desires and cares
On the heels of that lesson on keeping our eternal priorities straight
it seems like an abrupt shift when Jesus today speaks in apocalyptic terms of the days to come when the sun will be darkened and the stars will fall from the sky
It seems the Gospel message has made a hard pivot from “How to do right” to “The end of the world.” But this is not a departure – it’s an exclamation point
Last week’s Gospel closed out chapter 12 of Mark
and today’s Gospel picks up at verse 24 of chapter 13
Jesus set the stage for what we hear today
one of his disciples turns around and looks back and exclaims
look how awesome this temple built by men with stones is!” While that is a natural sort of expression of wonderment at an impressive building
has done something awesome for – and without — God
That assertion is self-reliance writ large
the very thing Jesus has been warning us of
Stephen tells the Sanhedrin before his martyrdom
“Yet the Most High does not dwell in houses made by human hands
As the prophet says: ‘The heavens are my throne
Jesus underscores this with his reply to the awestruck disciple: “Do you see these great buildings
There will not be one stone left upon another that will not be thrown down.” And it’s from here that Jesus explains in no uncertain terms that things are going to get very
There will be false prophets looking to lead us astray
The faithful will be handed over to worldly authorities
All those wars and natural disasters and the divisions in society and even within our families sound quite familiar to our ears today
but we can be sure that they sounded familiar to the ears of every generation that preceded us since Jesus spoke of them
None of this is new or unique to our present circumstances
Which gets us to the point of what Jesus tells us today
The end is the salvation of the elect by the Son of Man
the salvation of those who have kept the faith and endeavored to spread it come what may
After Jesus has told us of the certainty of what is going to happen up to and after the great tribulation to come
he pointedly makes it clear that there’s no certainty of when it’s going to happen
but only the Father.” It is the same for each of us
we recognize that our own personal end will come
Memento mori – remember that we will all see death
allowing us to guard our intellect and giving us unceasing prayer
We might be reluctant to accept the inevitability of our own death and respond with denial
or we might accept it and respond with a fatalistic “Why bother
but in the final verses of chapter 13 that follow today’s Gospel
Jesus tells us our response is not denial or deflated passivity
You do not know when the time will come” and “What I say to you
Abbot Hesychios described four ways we can be watchful
heaven and hell – is a sure way to keep focused on doing God’s will
The second type of watchfulness is to closely scrutinize the thoughts and images that we allow to enter or persist in our minds
we have to be aware that social media and the unrelenting news cycle come with messages that will lead us astray or drag us into anger or despair if we allow ourselves to dwell on them
The third way to be watchful can be the hardest sometimes
and that is free our hearts from distractions and enter into silence and stillness in prayer
as all the cares and worries and to-do lists in my mind parade on by to the exclusion of quiet contemplation
We might think that stillness and prayer is more like passive meditation than active watchfulness
but it really does take effort to be still
When we see those distractions creeping in but don’t allow ourselves to be carried off
when we make the conscious effort to turn back and focus on our conversation with God
The fourth type of watchfulness is simply the habit of continually and humbly calling on our Lord for help
until the end of the age” to reassure us because he knows our weakness
and in his great love wants only to help us
This acknowledgment of our dependence on God and his mercy is no better reinforced than in the Jesus Prayer: “Jesus Christ
and gives us the path to that fullness through the sacramental life of the Church
We have access to the cleansing and healing grace of Confession and the life-giving real presence of Jesus in the Eucharist
As we approach the close of the liturgical year and the start of a new one with Advent
let us think about ways we can be more watchful in our daily lives
In what situations would remembering my own mortality be particularly helpful in keeping me on track
In what areas of my life do I need to keep a closer guard on the thoughts I allow in
What can I do – or what can I give up — to carve out some true quiet to sit in the presence of God
can I make my first step be to simply call on God for his help with genuine humility and with the absolute confidence that he will provide it
In the acclamation before the Gospel for this Sunday
“Be vigilant at all times and pray that you have the strength to stand before the Son of Man.” May we embrace the grace God gives us to keep those words ever in mind and keep our hearts watchful each day

and the overall effect is not dissimilar to my experiences at Niki Nakayama’s two Michelin-starred n/naka
which happens to be the best restaurant in Los Angeles
Mori is operating at or near the highest level of L.A
She takes pride in sourcing beautiful gold-lacquered ceramics—serving vessels being a key element of both high-end sushi and kaiseki—and across both my meals
Mori tends to lean closer to slightly firm in texture and close to room temperature
arrived perfectly grilled over binchotan charcoal
Live hairy crab is presented with the necessary fanfare (I even saw a little bubble of seawater burst from one’s mouth!)
and so is the official paperwork for a bluefin tuna sale from Tokyo’s Tsukiji Market
I’ve seen all of these performative status-signaling aspects of omakase before and more
but Mori also serves a handful of more unique cuts
including a piece of thinly sliced pen shell clam wrapped in nori that’s absolutely spectacular
is also a welcome change of pace from the sweeter
more cake-like Tokyo-style pieces more commonly found around town.
But is there a good reason to choose Mori Nozomi over the others? Absolutely. Unlike most other omakase experiences, Mori serves delicate hand-shaped wagashi (a category of Japanese sweets, of which mochi is just one kind) and ceremonial-grade matcha at the end of every meal. As first reported by Eater
she integrates elements of traditional Japanese tea ceremony into the high-end omakase experience
The team’s level of thoughtfulness is unparalleled; on my second visit in mid-June
Mori swapped out my grilled tilefish course for barracuda
since she had noticed I had already tried this course on my first visit two months earlier
For those who appreciate artisanal tea and anyone avoiding alcohol
there’s a $55 five-tea pairing made with premium varieties imported from Japan
Brewed directly in front of you and served out of glassware
the tea pairing option strikes me as more of a non-alcoholic pairing than traditional tea ceremony
Despite the chef’s relatively young age and her having less than a decade of formal training (Mori has also worked at West Hollywood’s Sushi Ginza Onodera and Moto Azabu in Marina del Rey)
I would say Mori Nozomi already holds its own against other standard-bearing edomae-style sushi spots like Morihiro and Sushi Kaneyoshi
there is simply nothing else like this in Los Angeles
but feel free to come casually dressed—this is the Westside
and rich people often dress like slobs. The food: 15 to 17 unique courses including appetizers
plus seasonal fruit and Mori’s subtly sweet handmade wagashi. The drink: A high-end sake bottle list
plus a more limited selection of sake by the glass, hot green tea
as well a $55 tea pairing. Time Out tip: Have a particular date in mind
Set your alarm—Tock reservations for Mori Nozomi are released at midnight on a 30-day rolling basis.

A goth piece of jewelry reminds a columnist to seize each day
I recently received an intriguing piece of jewelry in the mail. It’s a prototype of a design I hope to sell someday
But I loved it so much that I wanted one for myself
featuring a coffin-cut black onyx gem in the center flanked by two leafy vines
The interplay of life and death in a single piece fascinates me while also sending shivers down my spine
These experiences made Jared cherish life more
Some are tough because of his chronic conditions
but he wakes up each day with a renewed sense of purpose and a willingness to embrace life again
Alliah Czarielle’s new ring reminds her to seize the day
A related concept is carpe diem, which was popularized by the movie “Dead Poets Society” and, before then, Saul Bellow’s novel “Seize the Day.” It reminds us to make the most of our time
This phrase has a special relevance to Jared
who often jokes about “seizing” the day because of his seizure disorder
Jared’s high level of self-awareness and acceptance of his condition show maturity
He confronts his challenges without letting them define him
and I will make the best of what skills and talents I have.”
For many, including myself, the thought of death can be scary and difficult to process. Yet I hope to challenge this feeling by studying and reframing my perspective. Mexican culture offers a unique view with Día de los Muertos” (Day of the Dead)
a holiday honoring loved ones who have passed
They create colorful altars with the deceased’s favorite things
This tradition teaches us to see the light in death
His dedication to a good quality of life remains constant
as does his drive to make the best of each day
memento vivere helps us appreciate every moment and find joy even in adversity
it’s a reminder that life’s fragility makes it all the more precious
By valuing each day and continuing to do the things we love
we can lead fulfilling lives and hopefully inspire others to do the same

Memento Mori in Hollywood will leave a lasting impression
the store has a collection of taxidermied animals
statues and figures made from animal – and sometimes human – bone
In keeping with the store’s name – which translates into “remember death” – all items follow a theme of death and the afterlife
Cofounders Bradley and Deirdre Hartman said they wished to bring some positivity to counteract the negative stigma toward death when they planned the store
They sell their own artwork as well as that consigned and from throughout the world
Jewelry is produced by local artists and all products made with animal remains are ethically sourced meaning that nothing is killed for the express purpose of making the item
The couple said the shop generates about $300,000 in revenue annually
largely driven by 90% of the sales being in the shop
They noted that nearly their entire customer base is women and that bestsellers include taxidermized ducklings and chicks dressed to look like witches
As Memento Mori approaches its 10th anniversary in November
the Hartmans plan to take the shop on the road as a vendor at various gift fairs and themed events
The self-taught taxidermists also plan to start hosting classes to teach the art form
The pair also recalled perhaps their most serendipitous transaction
One person offloaded a Voodoo doll described as haunted at the store
insisting they take it and asking no money for it
the Hartmans received a customer who wanted to buy something haunted

Now is the season for pumpkin launching and carving and lattes
Ghouls run rampant and phantoms fly freely on a certain night every year.
Young trick-or-treaters like to don many different costumes
but not many costumes hit as close to home as a plain skeleton outfit
The skeleton has been with us since there have been men
or rather assists us from our first moments to our last.
Can you imagine picking up an actual human skull
Perhaps a man’s skeleton seems naked compared to the fleshed body of a full man
The skeleton is so lacking that it simply seems beautiless
Chesterton argues that Man should quit being afraid of his bones
In his essay “A Defense of Skeletons,” he claims that the remnants of a man’s body should not be a horrific sight
“One would think it would be most unwise in a man to be afraid of a skeleton
since Nature has set curious and quite insuperable obstacles to his running away from it.”
Chesterton relates the strangeness of man being disturbed at his own structure to that of his being disturbed at the sight of leafless trees
“There was evidently a general feeling that I had caught the trees in a kind of disgraceful deshabille
the disgrace man feels while looking at a skeleton is comparable to the shame he feels at looking at a naked human body
Keeping with the metaphor of leafless trees
“Surely the idea that its leaves are the chief grace of a tree is a vulgar one.”
How can something as transient as a tree’s leaves be its primary beauty
If it is the case that leafed trees are more beautiful than the leafless variety then should it not be said that Man’s bones are more ugly than Man’s fleshed body
I am much inclined to agree with Chesterton that “the highest and most valuable quality in Nature is not her beauty
but her generous and defiant ugliness.”
the true value the leafless wintry twigs possess lies not in the fact that they can be beautiful at another time
but rather that they are so undeniably and inescapably ugly now.
Man’s left-behind bones do not possess value for what once clothed them
but rather for their blatant unattractiveness
as solemn and as happy as a child…This is the deepest
the most wholesome and religious sense of the value of Nature–the value which comes from her immense babyishness.”
can be reduced to the “shapes that a baby scrawls upon a slate–simple
a million years older and stronger than the whole disease that is called Art,” says Chesterton.
There is no longer the idea of work and play
of joy and sadness or of accomplishment and failure
“Memento Mori” is the term given to this realization
remember that you will be a skeleton.
So when you see a skeleton this All Hallows Eve
feel free to think about the grotesqueness
the ugliness and the simplicity conveyed by those bones
that “the bones of [your] skull beneath it are laughing for ever.”

Memento Mori
a macabre and mystical plant-based coffee shop
a Latin phrase that translates to “remember you must die,” informs the dark and eclectic vibe of the place
The cafe comes from husband and wife team, Correna and Justin Hernandez. The pair have been appearing across Southern California with their pop-up, Wicked Garden Cafe, since 2019. In 2021, they set up a permanent location in the Cartlandia pod to serve their Crypt Keeper caramel latte and Corpse cold brew
The duo paused operations at Wicked Garden on October 10 to focus on their dream — a permanent cafe
Memento Mori serves a full line-up of oat milk-based coffee drinks like the Werewolf mint mocha and the Wolfsbane with lavender and honey
The theme continues into the food menu with the Cemetarium Salad served with vegan barbecue smoked “chicken” and the various toasts offered as the Bread of the Dead
a blended Goth-a-Chino with flavors including mint cookie mocha
The theming for the cafe was inspired by Correna’s love for anything dark and gothic
She’s been collecting decor for the cafe for years in the hopes she would be able to open something permanent one day

Cemeteries are not usually very entertaining
I have been to my fair share of graveside services for folks I knew and loved over the years
and I readily acknowledge the solemnity of these places of remembrance
When we lived in Massachusetts in the 1970s
and teenaged me visited churchyards in Boston
to gather rubbings of epitaphs engraved on stone
There were prominent people and families galore in my collection
There were also the heart-tugging memorials to deceased babies and children (SO many too many of those)
my sister Maureen is buried at Arlington Cemetery in Atlanta
Mo was devoted to Saint Therese (the “Little Flower” of Jesus)
whose feast day (October 1) was the same day as Maureen’s death—and Therese was Mo’s age
Hence the inscription: “Our Beautiful Little Flower” marking the spot where my sister was laid to rest
is mute testament to the large number of people who have left this earth over the years
are the days when families habitually brought flowers to Granny and Gramps at Meadowview Cemetery on Sunday afternoons
We 21st century denizens HATE to be reminded that we will die at some point
I would much rather be cremated/donated to science than placed in a casket and buried six feet under
there is definitely an urge to have some sort of permanent memorial for posterity—you know
a sacred space where my children can feel sufficiently guilty over NOT visiting
And this way I can program a changeable series of snappy sayings too
grabbing their phones to play Candy Crush—but first they pop over to momsrestingplace.com
curious to see what witty/sentimental inscription is posted today
Kicking around a few ideas for online epitaphs:
