Cloud is a compellingly tricky film to pin down, both stylistically and thematically.
As Yoshii moves out of Tokyo into the countryside to focus on his shady business full-time, the film continues along this path, ramping up the isolation and paranoia along the way. Scenes are quiet until they’re suddenly not. The eerie soundtrack dips in and out. The acting is performed with an intentional lack of passion. Even the most extreme moments unfold with an increasingly unsettling reserve.

Yoshii (Masaki Suda) works in a factory, but he makes most of his money from online reselling. He buys goods – therapy devices, figurines, designer handbags – cheaply, and then sells them on again at an enormous markup. He’s so good at his side hustle, soon he’s able to chuck in his day job, and buy himself and his girlfriend Akiko (Kotone Furakawa) a picturesque house in the woods, with more than enough space for all his stock.
But along the way to that success, Yoshii has made a lot of enemies, sellers and buyers alike, all of whom feel he has ripped them off. They are angry. They want revenge.
Anyone who’s seen a film by Kiyoshi Kurosawa before knows that there are few directors better at instilling a seemingly innocuous scene with almost unbearable levels of dread. Whether he’s going full pelt or not – and Kurosawa at full pelt requires a mighty strong stomach! – he has an extraordinarily precise ability to control the tone of his movies from beat to beat; his plots often contain wild twists and turns, but you never feel like he has lost control.
Cloud is split into two very distinct halves. In the first, we watch Yoshii as he goes about his days. Although he operates his business with an almost sociopathic single-mindedness, feeling no qualms about his dubious dealing, we see that he does at least have genuine feelings for Akiko – those feelings are the one thing that show he has a little bit of humanity.
On a macro level, Cloud resounds with a general sense of dismay over the depersonalisation, the societal fracturing, of the gig economy. Alliances are drawn up over the course of the movie, but they are flimsy, self-serving ones, liable to be broken at a moment’s notice if those allied see a better offer on the table. It’s a vision of everyone as their own business, staring intently at their own computer screens, waiting to see if anyone is going to buy what they’re selling.
Kurosawa has clear thoughts as to where it’s all leading, and that is nowhere good – but he sure does make the journey entertaining.

You can see it in their list of players initially called up to the training camp in preparation for Window 6 of the FIBA Basketball World Cup 2023 Asian Qualifiers at the end of January
the 24-year-old's journey is a bit different from his similar-aged peers
The 1.96M (6'5") forward was not a featured star of youth national team programs like Yudai Nishida
He hasn’t even played for a youth national team
though he was a candidate to make the final roster for the U19 World Cup in 2017
who was not a part of the youth national team programs
had made a name for himself as the NCAA Division I leader in assists back in 2018-19
Hirotaka Yoshii doesn’t have that kind of resume
He’s played in Japan’s B.League ever since 2018 and has been a key piece of the mighty Alvark Tokyo for the past two years
but it wasn’t as if he was lighting the league on fire
I never thought about playing for the national team," Yoshii confessed
suiting up as a starter for Akatsuki Japan as they tipped off against Australia in the third window of the FIBA Basketball World Cup 2023 Asian Qualifiers
Not only was it his first time starting for the national team
but it was also his first game with the national team ever
I thought it was simply a great honor to play for the team."
"Playing for Japan has been a valuable experience for me," He added
"I don’t get many minutes in my club so I am thankful for being able to play in games with the national team
I also think that it has been a good opportunity for myself and my name to be known to others."
Yoshii was not just a one-time afterthought addition to coach Tom Hovasse’s roster either
Yoshii also started three of five Asia Cup 2022 games and has been a part of the team in the Asian Qualifiers windows since
Over the 11 games in 2022 that he played for the flag
Among all of the players that have played for Japan this past year
and Yudai Nishida have played more than Yoshii
Not bad for someone who still has some doubt about his own level of play
"There are times that I think I had a good play here and there," he admits
"But I don’t think I am good at basketball yet."
Coach Tom Hovasse might not entirely agree with his player on that
While there’s still room for Yoshii to improve
he’s certainly earned his part on the court for Akatsuki Japan
His counting numbers (6.3 points and 3.5 rebounds per game in the Qualifiers) might not pop out
This matches well with his choice of jersey number ("91") which is inspired by Dennis Rodman ("When I was in high school
In his limited minutes playing for his professional club
Yoshii specializes in playing defense against foreign players
A large part of that is because of his height
which allows him to better cover these "imports" who usually have that size advantage against the general local Japanese player
His height was also a part of his basketball origin story ("I started playing basketball in third grade when the coach invited me to play because of how tall I was") and Yoshii has made good use of that ever since
When Hovasse came on board as the head of the men’s national team
Yoshii has been one of the staples of this program so far
but considering the playing time that has been invested in developing Yoshii
there seems to be a good chance that the young forward will be a part of the team that will play at the World Cup that will be co-hosted in Okinawa
"There is only a small percentage of basketball players anywhere in the world who is able to play in the World Cup for their national team," he said
"[To be a part of this program] is representing and playing with a lot of responsibilities."
Yoshii considers himself as being lengths away from being a "good player"
going up against physical teams like Australia and Iran
and just learning from Yuta Watanabe at the Asia Cup
he’s finding his role on the team and improving in that role with each passing game
Even though he might not have an extensive resume as that of the majority of his teammates
he built a strong one right now - one that could add FIBA Basketball World Cup 2023 experience real soon

the researchers identified 242 clock neurons based on morphology
“The number of clock neurons identified in this study is higher than the previously reported 150 neurons

such as Allostatin-C and SIFamine (SIFa) peptidergic neurons
Neurotransmitter analysis showed that most of the lateral clock neurons expressed the excitatory neurotransmitter acetylcholine
whereas most of the dorsal clock neurons expressed glutamate
“The inhibitory neurotransmitter GABA was absent in clock neurons
Glutamatergic (small-central projecting DN3) and cholinergic clock neurons were involved in synaptic outputs to neurons that regulate hunger
the study identified 12 neuropeptides that were upregulated in the clock network
DH44 and Proctolin were the novel neuropeptides identified in this study
This suggests that clock neurons communicate through peptidergic paracrine signaling

Please start the interview by giving us a quick introduction to your company
I would like to begin by explaining the roots of our company
There is a town under preservation in the Omi area of Shiga Prefecture
Major Japanese companies such as Itochu and Marubeni started their business in the Omi area
The first president of Toyota was also born in Omi
Our company learned a lot from the philosophy of the Omi merchants
we will have an event to spread the philosophy of the Omi merchants
I am the sixth generation of Kizaemon Tsukamoto
We have inherited the same name through the different generations
It was the third generation of Kizaemon Tsukamoto who founded our current business
My photo will be placed alongside the photos of my father and grandfather
I will not put my photo next to the different generations
We also have photos of the historic executive managers of the company dating back 150 years
This was also a custom of the Omi merchants
Itochu had similar customs a few years ago
This painting is based on a painting by Korin Ogata
Silk used to be one of the main import goods in Japan. It was the top import for around 30 years
The theme of this exhibition is Nishijin-ori and the Silk Road
the Silk Road has been the main trade route in Asia
The Silk Road extended from the west of the Eurasian continent all the way to Japan
It created a large-scale economy throughout history and
It was the way that technology for orimono passed from the West to the East
Culture and designs were also spread through the Silk Road
Chinese characters and Buddhism were spread this way
The eastern end of the Silk Road was Nara in Japan
Nara has the oldest tradition associated with the Silk Road
As the theme of this exhibition is the Silk Road
you can see the designs of each country here in our museum
Some treasures are preserved in the Shoso-in Temple in Nara
You can see the four different seasons here for example
This is the design of the orimono for this particular area
It was made using an 1800-hole jacquard weaving machine
we opened our Tango Creative Center in Kyotango City
Many of the craftsmen in the area were quite old
The purpose of the center is to train the younger generations of craftsmen
you can see a replica of the treasure in the Shoso-in Temple in Nara
You can see products from both the East and the West here
We also have Ancient Egyptian patterns on Nishijin textiles
This exhibition shows the different cities and towns that the Silk Road passed through
There are many designs from areas where Islam was the main religion
we had a visitor from Thailand who needed to learn about the Silk Road
they recognized the designs as you can find similar designs in Thailand

There are a lot of different design types in China
We have many different inspirations for designing kimonos here
Our kimono designs incorporate many different aspects of the Silk Road
Buddhism came from India to China through the Silk Road
we have kanji characters and hiragana characters
All of the writings related to Buddhism in India were translated into kanji in China
The words of Buddhism inspired many of our designs
We used the Nishijin-ori technique to make this item
You can see Buddha being brought up to heaven
The Buddha is wearing yellow clothes from India
They are all made using orimono techniques
Japanese Buddhism has many different rules from different religions
Japanese people accepted all of these differences and backgrounds
we reproduced a painting from 900 years ago with these new types of threads
We weaved famous paintings such as Monet with the same threads
We also weaved the Genji Monogatari which is an ancient Japanese story
These were all done using the Nishijin-ori technique
There is a weaving process and a dyeing process
It takes around 10 months to design one of our kimonos
you specialize in trading a wide range of fashion products with a particular focus on preserving traditional Japanese culture
Among the products you offer is the kimono
a garment that represents over 1,500 years of Japanese history
you are also dedicated to creating original in-store products through collaboration with retailers
Can you elaborate on your expertise in handling kimono products and explain how you strive to preserve tradition throughout the process
we emphasized the opening of the Tango Creative Center on July 25th
you can read the conversation that I had with the mayor of Tango
We want to collaborate with the local government so that we can revitalize the local market
The different weaving companies need to collaborate to make this happen
you can see some photos from the Kaga Yuzen Women Artists Competition
Kaga is another large area for the production of kimonos and is comparable to Kyoto
Female designers have competed in this competition for the past 42 years
We want to train young designers so that we can preserve and pass on our traditions
We are also working together with the Outrigger Hotel in Waikiki Hawaii to build a chapel
we collaborated with a coat retailer to host an event during the Gion Festival
which is one of the biggest festivals in Japan
We invited maikos (Japanese apprentice hosts
one rank below geishas) and we were able to make many sales
We do not only collaborate with retailers to develop original products
We also have these kinds of events to sell our products
and we have been able to diversify our businesses very well
Your company focuses on seven different types of businesses which shows how you have managed to diversify your business throughout your many years of history
Could you please give us an overview of your seven business divisions and the synergies that you can create among them
which one do you believe has the most growth potential both for the domestic market and the overseas market as well
The first is Tsukaki Shoji which is our original company
It runs our museum and our Tango Creative Center
The next company is Kyoto Wasou which specializes in Kaga Yuzen
Both Tsukaki Shoji and Kyoto Wasou are wholesalers
The third company is Kyoho which is a dye manufacturer for Kyoyuzen
you can see photos of the weaving process for Nishijin-ori
The next pages show the museum and the dyeing process
You can also see photos of the Narutaki Creative Center and the technique that is called suminagashi or paper marbling
It involves putting the dyeing material on the surface of the water
and the craftsmen use various tools to create various designs
you put the textile over the surface of the water
Issey Miyake and Asics also used this technology
This technology allows you to capture the moment of the design on the surface of the water
These companies loved this concept and decided to use it
we opened the Tokamachi Creative Center in Niigata Prefecture
The supply chain in Kyoto is different from the supply chain in Tokamachi
different steps are followed by different companies
These different companies are connected like a chain
There is also the issue of the aging craftsmen
and we have opened the Tokamachi Creative Center in the area
While Tsukaki Shoji was founded by the third generation
He recently met with the Minister of the Mines there
We conduct many different types of activities
The fifth company in our group is Marie Classe
They are located in Ginza and their business is wedding dress rental
Our business in Hawaii will start in February of next year
The final company is Tamura which is a foundation underwear business
They have several patents to help women have better body shapes
The first is real estate while the second is our financing business
which one do you believe has the most growth potential for the domestic market
and which one has the most growth potential for the overseas market
We are still analyzing the potential for these businesses
foundation underwear is limited to Southeast Asia due to the body sizes of the people there
Japan has a historically deep clothing culture with its kimono and yukata characterized by timeless designs that have gone through improvement and modifications over the years
Japan is famous for its cultural heritage and craftsmanship
and innovative technologies integrated into the supply chain
How would you define the uniqueness of the Japanese fashion and apparel industry
It is a difficult question as we are engaged in the kimono business
which is a little different from the apparel business
The Japanese clothing business and clothing trends have been able to gain momentum in the global market
A lot of this comes from the kimono-making process with the kimono being the traditional clothing of Japan
To what extent do you believe the kimono has influenced the Japanese clothing industry
we have an extensive network of department stores
All these traditional department stores like Takashimaya
and Mitsukoshi started their business as gofukuya
when it comes to designers who have global activities
they tend to incorporate the kimono traditions in their designs and oftentimes use the materials that are used for kimonos
The kimono is still often used for important life events
the kimono area is different from the typical apparel areas
As for your second question regarding the strengths of the Japanese fashion industry
it will be very difficult for it to continue to grow
I believe that the strength of Japan lies in its weaving technology and its technology to produce original designs
we have the tradition of incorporating international design elements. These are Japan’s strengths in the global market.
You just mentioned that Japan’s strengths lie in its craftsmanship and its technology
This is something that is under threat due to Japan’s aging population
The first is a shrinking labor force which makes it difficult to pass on the accumulated know-how and manufacturing experience
The second challenge is that the shrinking population will lead to a shrinking domestic market
which is forcing Japanese companies to look abroad to find new opportunities
How is your company navigating the challenges of a shrinking labor force and shrinking domestic market
and to what extent are you looking overseas to continue the company’s corporate growth
I think that those same trends can also be found in countries like Italy
To overcome the challenge of the shrinking labor force
we have been engaged in activities such as the opening of the Tango Creative Center which is focused on the training of the young generations
We are collaborating with the local government and local associations of the industry
it is very difficult to tackle the overall trend of the aging craftsmen
The first involves training the young generation as soon as possible
The other strategy is to ask the seasoned craftsmen to expedite the process of weaving so that we can stock the orimono
There are some materials such as threads that will be difficult to produce in the future because the craftsmen who make those threads are also aging
We are therefore asking these seasoned craftsmen to accelerate the production of these threads so that we can stock the important threads
It is also important for us to look toward the overseas market
We are now considering supplying the obi that we have weaved to overseas designers
so we have not been good at collaborating with non-Japanese people
That is what produced Japan’s closed society
What types of artists are you looking to collaborate with
Do you have any specific countries in mind that would be appealing to your obi business
so we are trying to collaborate with the designers there to attract wealthy potential customers in the country
we have a lot of similarities with Chinese dresses
We are collaborating with a famous Chinese designer and are supplying the textiles for kimonos to them
They carry out the dyeing process to make Chinese dresses that are sold at very high prices
We have already spoken at length about your kimono business
We are also very interested in understanding why your company decided to venture into the luxury jewelry business where you have been able to purchase demantoid garnet from Madagascar and bring it to Japan
where you cut it to meet the needs of many customers not only in Japan but also around the globe
What was the motivation behind expanding into the jewelry business
could you please introduce the different brands that you have
One of our motives was that we had the know-how needed for the jewelry business and the fur coat business
as the know-how is very similar to the kimono business
Some of the products that we make are worth JPY 1-2 million
We also have channels for distribution in department stores and kimono retailers
we were able to distribute our fur coats and jewelry
Our diamonds are our best-selling products
especially rare diamonds such as pink diamonds and red diamonds
These diamonds are worth between JPY 10 million and JPY 100 million
The other aspect of our jewelry business is to develop new color stones
We have sold many different colored stones
We noticed the need to find new colored stones
That was why we decided to look into Madagascar
Across the ocean from Madagascar lies Tanzania
Madagascar is an isolated island with unique gemstones
We dispatched our employees to the mines there and discovered different gemstones that we brought back to Japan
it takes time for customers to learn about new color stones
we were able to spread the information regarding these new stones and begin to distribute them
what ambitions or goals would you like to achieve in the years to come
and how would you like your company to be seen in the eyes of the global market
We believe in the philosophy of the Omi merchants
which is known as Sanpo-Yoshi or “three-way satisfaction.” This philosophy means being good for the buyers
My dream is to spread this business philosophy to the world
Kanji is one thing Japan has in common with those countries
We also have similarities that derive from Confucianism and Buddhism
That is why the philosophy of Sanpo-Yoshi has been widely accepted in East Asia
It is a business style that teaches what society should be like
I have made a lot of friends not only in Japan but also in China
This company represents my business and my life
I want to communicate Japanese traditions to the global market
I would like to link these assets with the global market
I will do my best for the company so that my sons and my employees can inherit these assets

Their value is deemed by what the market for them is making them Schrodinger’s boxes
an infinite number of unseen goods that matter only because of the demand of unseen buyers
It is never entirely apparent whether Yoshii wishes for wealth in any extreme sense
He clearly wishes for more than the exhaustion of the daily grind of the average worker with nothing to show for it
Yoshii has some assemblage of aspirations beyond being a factory worker
but he doesn’t yearn for material wealth
His growing addiction to reselling goods seems instead to be his attempt at gaming life
Yoshii’s downfall is not particularly unique or special
Nor is there anything extraordinary about Yoshii’s desires or ambitions
which is very much in keeping with Kurosawa’s characters
Alongside an electronic hum that haunts much of his filmography
even while descending into a protagonist's personal hell
Kurossawa’s latest techno-thriller Cloud is arguably two movies in one

released in 2024 was a massive hit taking home a record-breaking 19 Primetime Emmys
The critics and audiences were all in on the samurai drama set in 1600s Japan based on James Clavell’s novel
but Jerry London was not feeling the most modern adaptation
“Mine was based on the love story of Shogun between Blackthorne and Mariko
and this new one is based on Japanese history
who was the Shogun,” he told the outlet
“It’s very technical and very difficult for an American audience to get their grips into it.”
In the FX show, Blackthorne is played by Cosmo Jarvis, Mariko by Anna Sawai, and Toranaga by Hiroyuki Sanada
According to London everyone he spoke to about the FX series couldn’t understand it and said the new filmmakers “didn’t care about the American audience.”
What’s it all about?’ I watched the whole thing
It’s very difficult to stick with,” he said
London was happy the new series was not too similar to his because he “didn’t want my show to be copied.”

although the 2024 drama was originally conceived as a limited series with just one season
the popularity and acclaim for the series have made a second season more than just a wish
there are several reasons to believe that Shogun Season 2 will actually happen sometime in the near future
and Tadanobu Asano all taking home Golden Globes recently
the details of Shogun Season 2 are still a bit murky
Here's everything we know about the return of the samurai-heavy epic that is easily one of the best TV series in recent memory.
As of right now, there is no concrete release date for Shogun Season 2. The reason for this is because Shogun's new season has not started filming. After the 2025 Golden Globes, co-showrunner Rachel Kondo told Variety that the writing process is "ongoing," while her partner
Justin Marks said the writers' room was about "six weeks" from mapping out Season 2
it's unlikely we'll know about the release date
But work is happening on the show's story right now
What Is Shogun's New Season About?The ambiguous ending of 'Shogun' Season 1
The final episode of Shogun Season 1 was Episode 10 "A Dream of a Dream," and it did not end on a cliffhanger
one could argue that because the final episode showed Blackthorne (Cosmo Jarvis) rebuilding his ship
it follows that Season 2 would have to let us know whether he was successful or not
deal with the fallout of the deaths of Lady Mariko (Anna Sawai) and Kashigi Yabushige (Tadanobu Asano)
There's also a scene of an elderly man on his deathbed
who may or may not be Blackthorne at some point in the future.
Speaking to Slate in 2024 about the ending of Season 1
husband and wife co-showrunners Justin Marks and Rachel Kondo said: "There’s a little ambiguity that we’d like to stay there...we initially frame the story as if it were the recollection of an old man looking back on his life with regret in some way
only to find that what we were really seeing was the dream of a young man looking forward with regret to the life that he could possibly have."
Clearly, Season 2 would have to delve into these narrative complexities, and either make things more clear, or perhaps, even more layered. The show is now moving beyond the events of the original novel
there will be even more creative narrative freedom than ever before

Tuberous sclerosis complex (TSC) is an autosomal dominant neurogenetic disorder affecting the brain and other vital organs
TSC is caused by a loss-of-function mutation in the TSC1 or TSC2 gene
These gene products form a protein complex and normally suppress mammalian target of rapamycin (mTOR) activity
mTOR inhibitors have been used to treat subependymal glioma (SEGA) that is a brain tumor characteristic of TSC
neuropathology of TSC also involves dysregulated cortical circuit formation including neuronal migration
It is currently unknown to what extent mTOR signaling inhibitors correct an alteration in neuronal morphology that have already formed prior to the treatment
we address the efficacy of rapamycin treatment on neuronal migration and dendrite formation
we suppressed Tsc1 expression in a fraction of neuronal progenitor cells during the fetal period
we found that more Tsc1-suppressed cells remained within the periventricular zone
and rapamycin treatment facilitated neuronal migration
Tsc1-suppressed pyramidal neurons showed more complex branching of basal dendrites and a higher spine density at postnatal day (P) 28
Aberrant arborization was normalized by rapamycin administration every other day between P1 and P13 but not P15 and P27
abnormal spine maturation improved by rapamycin treatment between P15 and P27 but not P1 and P13
Our results indicate that there are multiple critical windows for correcting different aspects of structural abnormalities in TSC
and the responses depend on the stage of neuronal circuit formation
These data warrant a search for an additional therapeutic target to treat neurological symptoms of TSC
These microscopic structural changes and functional alterations underlie neurological disabilities in TSC
It is likely that the response to rapamycin is maximal while the abnormal morphology is formed
it remains unclear whether each of these cellular processes has a sensitive period to respond to the mTOR inhibitor treatment
we address the efficacy of rapamycin treatment on neuronal migration
we transferred a DNA construct encoding Cre recombinase tagged with green fluorescent protein (Cre-GFP) into E 15.5 neuronal progenitor cells in a Tsc1fl/fl mouse fetal brain and suppressed the gene expression in a group of cells that are born around the same time
we found that more Tsc1-suppressed cells remained within the periventricular zone and that rapamycin treatment facilitated neuronal migration
the lamination pattern of Tsc1-suppressed neurons was widened and scattered more than WT cells
Tsc1-suppressed pyramidal neurons showed more complex branching of basal dendrites
which was normalized by rapamycin administrations between postnatal day (P) 1 and P13 but not between P15 and P27
abnormal spine maturation improved with rapamycin between P15 and P27 but not between P1 and P13
These results suggest that there is a critical time window during neuronal circuit formation to correct abnormal neuronal morphology in TSC
Timed pregnant mothers were anesthetized with 2–3% isoflurane and oxygen
the uterus was externalized and the lateral ventricle of E15.5 embryos was injected with 1–2 μg Cre-GFP alone or in combination with 0.1–0.2 μg DiO-YFP
Using an ECM 830 apparatus (Harvard Apparatus
Mouse embryos were electroporated at embryonic day 15 (E15)
and acute coronal brain slices (240 μm) were prepared at E17 and E18
Occipital slices were transferred onto slice culture inserts (Millicell) in cell culture dishes (35 × 10 mm; Corning) with Neurobasal medium (Invitrogen) containing the following: B27 (1%)
Slices were used for imaging (1–2 h after slicing) or for pharmacological treatments (incubated at 37°C in 5% CO2
A subset of slices was incubated with the medium containing rapamycin (100 μM)
Cre-GFP- and mCherry-positive cells were imaged on an inverted Nikon microscope (TE 2000-S) with a 20 × objective lens [numerical aperture (NA) 0.45]
mCherry was added to ensure fluorescent signal detection during serial imaging
slices were kept in an acrylic chamber at 37°C in 5% CO2
We captured time-lapse images with a Cool SNAP EZ camera (Roper Scientific) using NIS-Elements software (Nikon)
The neuronal migration velocity was measured using a plugin for ImageJ (Mouse Tracker, programmed by P. Malatesta, IST Genova) that allows tracking the cell position over time. Using the coordinates obtained with ImageJ, the velocity was calculated with Excel (Microsoft) (de Anda et al., 2010)
Averaged pixel intensity was calculated by dividing total pixel intensities with the pixel area of cytosol
averaged intensity of Cre-GFP signal was measured
A cell was considered Cre-GFP positive when its averaged Cre-GFP intensity in the nucleus was above 100
Immunolabels of TSC1 and phosphorylated S6 were compared between neurons with and without Cre-GFP
neurons expressing Cre-GFP (indicated with a dotted circle) shows suppression of Tsc1 protein as compared with neurons without Cre-GFP

Because layer 2/3 cortical neurons project to the contralateral hemisphere
mRFP+ axons were followed to measure pS6 levels in cells of the non-electroporated region
ROIs were generated using an elliptical selection tool
and average intensi- ties for each ROI were determined
The ROI did not include the nucleus that had low pS6 staining
For both cell size and pS6 staining measurement
3 sections per mouse and 38–111 cells per section were analyzed
Neurons were blindly traced and analyzed based on spherical shells concentric with the somal centroid and spaced at a 5μm interval
The number of intersections and nodes were counted from the soma to the distal end of dendritic branches
basal dendritic branches within the interval between 15–40 μm and number of spines were counted to calculate the spine density (number of spines per μm)
To examine the migration of Tsc1-suppressed neuronal progenitor cells
we combined a Cre-lox recombination system and in utero electroporation
we used a Tsc1fl/fl mouse which has exon 17 and 18 flanked with Lox sequences and electroporated a DNA construct encoding Cre-GFP into wild-type (WT) or mutant fetuses at embryonic day 15.5 (E 15.5)
We performed Immunohistochemistry in both WT and mutant brains at P28 and confirmed that Cre-GFP expressing neurons in the mutant showed suppression of the TSC1 protein (Figures 1A,B) and increased phosphorylation of ribosomal protein S6 (p-S6) (Figures 1C,D)
indicating an enhanced mTOR signaling as a result of suppressed TSC1 function
Tsc1-suppressed neurons show a scattered distribution and some cells are mislocalized outside of layer 2/3
the distribution of Cre-GFP (+) neurons is scattered
Arrowheads indicate mislocalized cells and some of them are positive for the layer 2/3 marker Brn2
the distribution of neurons that were electroporated with Cre-GFP at E15.5 is consistent with layer 2/3

These results indicate that Tsc1-suppressed post-mitotic neurons remain in the ventricular zone/intermediate zone longer than WT neurons and that rapamycin normalizes the departure timing
once neurons leave the ventricular zone/intermediate zone

we find a scattered distribution of Tsc1-suppressed neurons
Dyslamination results from the delayed departure of the mutant progenitors from the ventricular zone and is correctable with rapamycin treatment during neurogenesis
postnatal administration of rapamycin did not correct malpositioning of Tsc1-suppressed neurons
Our results indicate that mTOR inhibitor can correct the cortical lamination defect if it is given during corticogenesis
While the result suggests cortical tubers and dyslamination are potentially treatable with a mTOR inhibitor
the treatment is currently not feasible in a human embryo due to a concern for teratogenicity
Neurons in different brain regions and ages have different local connectivity
which is also likely to contribute to the interpretation
abnormally elaborate dendritic arborization occur in response to up-regulation of mTOR signaling pathway
Recent evidence suggests that each dendritic arbor process distinct information. For example, in a pyramidal neuron of the mouse visual cortex, each dendritic branch is tuned to a different orientation, and they are summated in the soma (Hausser, 2000; Jia et al., 2010; Grienberger et al., 2015)
Further study is needed to address whether dendritic integration and sensory processing are dysregulated in Tsc1-suppressed neurons
Our results indicate that there is a critical period for rapamycin response to normalize exuberant dendritic branching of Tsc-1-suppressed neurons that correspond to the first 2 weeks of postnatal life
mTOR pathway is responsible for dendritic arborization in an early postnatal period before spine maturation occurs
We also find that the rapamycin effect on aberrant spine formation is optimal between P15 and P27 when activity-dependent synapse formation is at its peak
While rapamycin treatment is effective in correcting dendritic spine formation
our results suggest that there may be a limit in compensating for the structural changes preceding mTOR inhibition
further studies are needed to understand whether there is an additional therapeutic target to further improve neuronal connectivity

A district court on Thursday referred to the family court a case involving a 16-year-old boy charged with murdering his parents in the city of Sagamihara

"The incident wouldn't have happened if he had not been placed in an inappropriate environment by his parents," the presiding judge said
Yoshii said that the boy "regrets" what he did
and that he needs to receive special education
Public prosecutors had sought a prison sentence of 10 to 15 years for the boy
The judge found the boy guilty of killing his 52-year-old father and 50-year-old mother at their home near Tokyo on Feb
by stabbing them multiple times in their necks with a knife
He was in his first year of high school at the time
The boy had admitted to murdering his father while claiming that he killed his mother because she had asked him to
The defense argued that this amounted to a case of assisted suicide
Yoshii noted that the mother had gone out to the balcony to seek help
which contradicted the defense's claim
The defense pointed out that the boy was a victim of severe maltreatment and had become desperate after enduring many years of physical and mental abuse by his parents
waited for his mother to return home to kill her due to his strong homicidal intention

Kumiko Kakehashi's So Sad To Fall In Battle: An Account of War Based on General Tadamichi Kuribayashi's Letters from Iwo Jima is a masterpiece. I find this book challenging
as it reveals the depth of my own ignorance
This is the story of Lieutenant General Tadamichi Kuribayashi. Five months before Japan's surrender on August 15, Kuribayashi led a fierce battle against overwhelming American forces on the isolated island of Iwo Jima
Kakehashi's book reveals that Japan's Imperial General Headquarters (IGHQ) altered his famed death poem
"Unable to complete this heavy task for our country
so sad we fall" was the original first line in the poem
the IGHQ changed "so sad we fall" to "so bitterly we fall."
Kakehashi presents 41 letters that Lieutenant General Kuribayashi sent to his family from Iwo Jima
I also read Kuribayashi's letters and telegrams from Iwo Jima in 1995
This was a decade before the publication of Kakehashi's book
I was then conducting interviews for the series "Voices Without a Voice: The 50 Years of War Dead's Families."
Despite the chaos during and after the war
the family managed to preserve the letters from Iwo Jima carefully
I focused not on their content but on how the family read them
"Have you ever seriously considered developing your willpower
establishing a family as a man will be challenging
Lieutenant General Kuribayashi had addressed this letter to Taro
He drew red lines next to "developing willpower" and "challenging future," hoping Taro would remember his father's teachings
There was also a letter addressed to Yoshii
Yoshii had personally obscured these sections
which included lines such as "My remains will likely never return..."
The post-war life of the Kuribayashi family
once venerated as the "House of the War God," underwent a dramatic change
Both Yoshii and Taro spoke little about their post-war hardships during the interviews
the silent voices of Kuribayashi's wife and children can be heard clearly through his letters

I also had the honor of interviewing Takako decades ago. Back then, she was the head of a kindergarten in Kawaguchi City, Saitama Prefecture
"Relatives of the soldiers who died on Iwo Jima came to our house on several occasions asking for money," she revealed
Takako related this story in an interview at the kindergarten
I remember seeing posters of her eldest son
In July, I met with politician Yoshitaka Shindo (66) in his ministerial office, which overlooks the National Diet Building
Shindo is Takako's son and Lieutenant General Kuribayashi's grandson.
Some 30 years after my interview with his mother
he has become a member of the National Diet
Yoshitaka Shindo is shouldering a significant responsibility
always telling me to reflect on how I should conduct myself," he said
"Our relatives often say that she's the one who most inherited Tadamichi Kuribayashi's personality."
Shindo remembers her as "elegant and never angry
I can't recall ever seeing her laugh genuinely
"We can never truly heal the sorrow of war," Shindo opined
"It is our responsibility to ensure that our country never experiences such devastation again."
I recently visited the Kuribayashi family for the first time in about 30 years
Yoshii passed away at the age of 99 in 2003
Kuribayashi's eldest son Taro passed away the year after that
to review the letters from Iwo Jima once more
She kept them in a plastic file along with paraffin paper to protect against moisture
also a granddaughter of Lieutenant General Kuribayashi
faces the challenge of preserving these historically valuable documents
who published her new book War Museum: Connecting the Circuits of Memory
"As long as the family wishes to keep the letters
there comes a point when these personal items become 'historical testimony.'" She worries where the family should entrust the Iwo Jima letters at that juncture
"There is no museum in Japan that provides a complete overview of the war," she explained
We are approaching the 80th anniversary of the end of the war next year
I could hear the "silent voices" from Iwo Jima.

objective mode as a result of the coursework I was required to do," Yoshii said
After a coaching stint with the SoftBank Hawks and a second with the Fighters
he oversaw fire-balling young right-hander Roki Sasaki's careful initiation into pro baseball and helped guide him on the path to becoming Japan's youngest perfect-game pitcher in 2022
Yoshii said the Marines did not let Sasaki pitch in any games in 2020
because of how slowly he recovered from the arm inflammation that comes with pitching
Sasaki's bone density was widely reported to be low
a sign that his bones had not fully developed
something Yoshii had learned was relatively common
"Baseball needs to be very careful with players turning pro out of high school because adding too much muscle on those with immature bones can lead to injuries."
without depending solely on practical experience
Through talking about different things with different players
the scope of my understanding grew broader still."
His experience and education made Yoshii an attractive candidate for a managing job he didn't particularly want
suddenly quit the Marines after the 2022 season
"It (managing) is a lot of trouble," Yoshii said
But as to whether he has learned anything from sitting in the big chair since last season
"I'll let you know after we win a championship."

Rika Yoshii grew up in Point Loma wanting to become a rock star
she found her passion at Cal State San Marcos
where she has served as the chair of the Computer Science and Information Systems Department and is now the interim faculty director of the new Master of Science in cybersecurity program
“San Diego is a cybersecurity hub,” said Yoshii
“We have many military installations and we have many defense industry partners all in the area
So it is really important to have a good cybersecurity program.”
But this isn’t just any cybersecurity program
It is the only professional science master’s program in cybersecurity in the state and one of just a handful in the nation
“We are creating the future leaders who can be chief information security officers
in computer science from UC Irvine and worked as a project leader at Rockwell and a researcher for the Northrop Research and Technology Center before joining the computer science department at CSUSM in 1993 to pursue her love of teaching

The character of Yoshii Toranaga in Shōgun is inspired by the life of warlord and later shōgun
From more than a century of near-constant civil war
laid the foundations in the late-16th century
who fought alongside both Nobunaga and Hideyoshi
He went from child hostage to feared warlord to military dictator, and in the process established a shōgunate that lasted 265 years
Born with the name Matsudaira Takechiyo in 1543
the future Tokugawa Ieyasu was the son of a daimyo (large landowner)
Although the Matsudaira family claimed ancestry to the prodigious Minamoto clan
which ruled Japan from the 12th to 14th centuries and counted the first shōgun among its number
they were a minor power based in Mikawa province
The Sengoku (‘warring states’) period had been raging for decades by now
It was a time of relentless fighting and civil strife between the daimyos that kept Japan divided
The ever-shifting loyalties of Ieyasu's father meant that the boy was just four or five when he was offered as a hostage to secure an alliance with a strong neighbouring clan
While being transported from his home at Okazaki Castle to the Imagawa base at Sunpu
who threatened to kill him if the Matsudaira did not abandon the proposed alliance
The boy was spared and lived with the Oda for several years
Takechiyo received military training and education in matters of governance
He also became clan leader after his father’s death in 1549
and father 11 sons and five daughters – and personally led forces on campaign against the Oda
circumstances had changed: the Imagawa clan was in disarray
while an impressive warrior named Oda Nobunaga had taken over the Oda and launched a mission to unify the country
by force.Ieyasu saw an opportunity to return to his family’s domain and assert his own control
Emerging as a new daimyo to be reckoned with
Takechiyo would take a new name too: Tokugawa Ieyasu
Ieyasu rose in prominence and expanded his territory thanks to the alliance with Oda Nobunaga
who had become the most powerful man in Japan
Nobunaga died after being betrayed by one of his vassals
Into this vacuum stepped Nobunaga’s brilliant general Toyotomi Hideyoshi
As part of the ongoing attempt to unify the country
the two joined forces to defeat the dangerous Hojo clan
Ieyasu was offered Hojo provinces to the east
as long as he gave up his existing territory
Despite the risks of starting anew in unknown lands
Ieyasu moved his administration to a small fishing village called Edo
where he immediately embarked on a massive construction project to transform the area into a political centre to rival Kyoto
and a cultural hub for artisans and businessmen
became the foundations of the modern-day Imperial Palace
Tucked away in his eastern corner of Japan
he avoided getting caught up in Hideyoshi’s ill-fated military invasions of Korea
Tokugawa was among the most powerful daimyo
and appointed one of the ‘council of five elders’ tasked with ruling on behalf Japan of Hideyoshi’s young son
he would seize the chance to take power for himself
Those loyal to Hideyoshi’s wishes for his son Hideyori to rule formed a faction against Ieyasu
but his so-called Eastern Army defeated the Western Army of Ishida Mitsunari at the decisive battle of Sekigahara in 1600
Ieyasu shook up the political landscape by moving them to different parts of the country
confiscating land from enemies and giving it to allies
he split the daimyos into three classes – shinpan (those related to the Tokugawa clan)
fudai (his loyalists) and tozama (those who swore fealty after Sekigahara) – which each had varying degrees of restrictions within his new system
He was awarded the title of shōgun by the emperor of Japan in 1603
marking him as the pre-eminent warlord in the land
this meant little as he continued to rule anyway
yet the move established his shōgunate as a hereditary title in the hopes of preventing future claims from other daimyos
Having let Hideyori remain at Osaka Castle for years
Tokugawa ultimately defeated this last rival in 1615
Tokugawa initially welcomed trade with European merchants and allowed missionaries to operate
he came to see foreigners as a threat to his dictatorial order
so he forbade Christianity and curtailed traders
This laid the groundwork for Japan’s isolationist policy
William Adams began working in a shipyard at the age of 12
and was involved in the defeat of the Spanish Armada in 1588 before joining a fleet of Dutch merchant ships bound for the East Indies
his ship anchored off Japan and he was brought before Ieyasu in the months before his victory at the battle of Sekigahara
The Englishman impressed Ieyasu so much that he became a trusted advisor
and the shogun lavished him with land and a title
where he helped build ships and set up a factory for the East India Company
Just as Ieyasu was the inspiration for Toronaga in Shōgun, John Blackthorne is loosely based on William Adams
Tokugawa died of an illness at the age of 73
A magnificent mausoleum and shrine would be erected in Nikko
the Tokugawa Shōgunate ruled Japan: a time of peace
but achieved through rigid and often ruthless control
It ended only with the Meiji Restoration in 1868
which re-established direct imperial rule once again

I became project leader for the SENDAI UMINO-MORI AQUARIUM in July 2014
My job was to manage the schedule and make sure we hit all our milestones
There were seven companies involved in the project
some from Sendai and some from the Tokyo region
they all had their own views on what sort of aquarium we should make
My role—in conjunction with Yokohama Hakkeijima
the firm responsible for the marine exhibits—was to interpret and realize all the different parties’ aspirations
but one thing was never in doubt: we all wanted to create something very special
there was a lot of discussion about whether to include an exhibit of the local marine life of northeast Japan
people there felt understandably ambivalent toward the sea
we were keen to create an aquarium that had a distinct
and we wanted to make people aware of that
We ended up installing a giant aquarium with a northeast Japan theme
It accounts for about one third of the total exhibition space and has proven very popular
I spent several years working on the Hiroshima Carp baseball team which is based in the Hiroshima Zoom-Zoom Stadium
I brought that skill set up to the aquarium in Sendai
No one had sold such a large sponsorship program for an aquarium before
but I devised a variety of packages to pitch to potential sponsors
visitor numbers fluctuate according to the team’s performance whereas the flow of people to aquariums is pretty constant
we managed to raise in sponsorship for the first year
The most important thing is to make sure that’s sustainable and to get our partners to renew next year
necessarily has an impact on the local community
One aspect of my job was to get up on stage at town hall meetings
explain our plans and reassure residents that we would do our best effort to prevent traffic jams
There was a lot of anticipation locally before the opening
I used to spend about half the week up in Sendai and when I was on the train or in restaurants
I’d often hear people talking about it
It attracted 500,000 visitors in its first two months of operation
We are now expecting to exceed our original target for the first year
I was lucky to be put in charge of this project and given so much latitude
I’m relatively young—in my mid-thirties—so it was a precious opportunity
I took my own kids to the aquarium in August 2015 and they had a great time
I think it was one of their best memories of the summer holidays
Now that SENDAI UMINO-MORI AQUARIUM is up and running
in the sense that we both own and operate the facility
so before we embark on another similar project
we need to verify that it works as a business and is delivering all-important value for our shareholders
we may explore working on another facility somewhere else

"Em Va Trinh," a biopic on Trinh Cong Son (1939-2001)
has a character playing Yoshii and reveals a lot of details about her relationship with Son
Yoshii wanted an apology through a press release within seven days
She also wants the apology included in the film every time it is screened and the producer to make a pledge not to do something similar again
The production company merely said it has received the notice from Yoshii and made no further comment
claimed her client remained silent when the film was released for fear of affecting its revenue

and examiners at the University of Paris VII graded it as excellent
Now Yoshii frequently visits Vietnam and burns incense at Son’s grave

A 23-year-old man was found guilty Wednesday of killing a protected deer in a popular park in the western Japan prefecture of Nara earlier this year
The Nara District Court sentenced Hayato Yoshii to 10 months in prison
for attacking the deer with an ax around 2 a.m

"The defendant did not value the animal's life
His act was brutal and malicious," Judge Risa Ishikawa said in handing down the ruling to Yoshii
noting his motive for the attack was that he became furious after one of the deer suddenly rammed his car but that cannot justify his action
He was accused of violating the law that protects cultural properties after killing the female deer
Police were notified by a passerby about the attack and identified the suspect using surveillance camera footage of the area
the court also found Yoshii guilty of drunken driving in Matsusaka
and possessing a Japanese sword without authorization
The sword was found when his home in the city was searched by investigators on March 2
