Individuals with bipolar disorder (BD) exhibit deleterious decision making
Such aberrant decision making can be quantified using the Iowa Gambling Task (IGT)
which requires choosing between advantageous and disadvantageous options based on different reward/punishment schedules
The mechanisms underlying this behavioral deficit are unknown
but may include the reduced dopamine transporter (DAT) functioning reported in BD patients
we tested whether reduced DAT functioning would recreate patterns of deficient decision making of BD patients
We assessed the IGT performance of 16 BD subjects (7 female) and 17 healthy control (HC) subjects (12 female)
We recorded standard IGT performance measures and novel post-reward and post-punishment decision-making strategies
We characterized a novel single-session mouse IGT using C57BL/6J mice (n=44)
The BD and HC IGT performances were compared with the effects of chronic (genetic knockdown (KD; n=31) and wild-type (n=28) mice) and acute (C57BL/6J mice (n=89) treated with the DAT inhibitor GBR12909) reductions of DAT functioning in mice performing this novel IGT
BD patients exhibited impaired decision making compared with HC subjects
Both the good-performing DAT KD and GBR12909-treated mice exhibited poor decision making in the mouse IGT
The deficit of each population was driven by high-reward sensitivity
The single-session mouse IGT measures dynamic risk-based decision making similar to humans
Chronic and acute reductions of DAT functioning in mice impaired decision-making consistent with poor IGT performance of BD patients
Hyperdopaminergia caused by reduced DAT may impact poor decision making in BD patients
which should be confirmed in future studies
these findings support differing mechanisms underlying poor decision making in these disorders
Assessment of mechanisms that may contribute to impaired IGT performance in BD could therefore be conducted in mice by using a single-session IGT that enables direct comparisons with human IGT findings
the relationship between reduced DAT functioning and its contribution to impaired risk-based decision-making in BD patients may be important
To determine whether reduced DAT functioning could impair decision making under risk
we compared single-session IGT performance of patients with BD and mice with chronic (DAT KD) and acute (GBR12909) reductions of DAT functioning
A single-session IGT was used for consistency with the human IGT and to measure dynamic changes in decision making after rewards or punishments
We predicted that: (1) decision making would be impaired in BD patients; (2) subpopulations of mice would be identifiable in the mouse IGT based on risk learning; and (3) both chronic and acute reductions of DAT functioning would impair IGT performance similar to deficits observed in BD
Participants were excluded for: (1) current alcohol or substance dependence; (2) a history of neurological conditions
or seizures; (3) treatment with electroconvulsive therapy; (4) stroke or myocardial infarction; and (5) a positive result for cocaine
or phencyclidine on a urine toxicology Rapid Drug screen (Pharmatic Inc.
All subjects provided written informed consent to the current protocol approved by the UCSD Institutional Review Board known as the Human Research Protections Program

All mice had ad libitum access to water and were food-restricted at 85% of their free-feeding weight during the periods of testing (during the dark phase of the day–night cycle between 0800 hours and 1800 hours)
All procedures were approved by the UCSD Institutional Animal Care and Use Committee
The UCSD animal facility meets all federal and state requirements for animal care
GBR12909 was injected intraperitoneally with a volume of 10 ml/kg
Free-base drug weight was used in drug calculations
After stabilization of responding on a simple fixed-ratio schedule (Supplementary Materials and Methods)
single-session performance of C57BL/6J mice (n=44) was assessed in the IGT
DAT KD (n=31) and WT littermates (n=28) were trained to holepoke
the IGT performance of these mice were assessed and compared
Based on previous observations (Rivalan et al, 2009)
we examined the performance of individual mice and identified three different subpopulations
The total trials of each group were split into three trial periods based on their total trials completed
These subpopulations were quantified by subtracting % advantageous choices of trial period 1 from % advantageous choices of trial period 3
and poor decision makers were stratified as (1) >0.5
and (3) <0.5 standard deviations from the mean
This stratification was made for each genotype or drug treatment group separately
The likelihood of a subject repeatedly choosing a card/stimulus following a reward from the advantageous options (safe-stay) and disadvantageous options (risky-stay) was compared with their likelihood of selecting a different choice following punishment from the advantageous (safe-shift) and disadvantageous options (risky-shift; Supplementary Methods)
Human and mouse choices were analyzed over three equal blocks of trials
Human IGT net score was analyzed using analysis of variance with group (BD
HC) as a between-subject factor and trial block as a within-subject factor
Animal choices were analyzed using analyses of variance with trial period as a within-subject factor
Group was determined by quantification of learning as described above
Subjects with ⩽10 completed trials per trial period were excluded from analysis
The % advantageous choice preference was compared with chance (50%) using a one-sample t-test
Tukey post hoc analyses of statistically significant main or interaction effects were performed where applicable and Cohen’s d effect sizes were calculated

these data support the use of the single-session rodent IGT to examine risk-based decision making and highlight the importance of inter-individual differences in risk preference
the findings of reduced DAT functioning in mice are consistent with BD patients making less repeated picks from low-reward decks
both DAT model animals resemble patients with BD in both behavior and putative etiology
DAT levels in BD patients in the manic phase have yet to be established
BD patients exhibit impaired decision making in the IGT
Using post-reward/punishment decision-making measurements developed from animal studies

crisscrosses a nation so small and flat that it’s easy to use bicycles to transport people

where the teenager hid during World War II and wrote her famous diary; the city’s lovely canals; and massive Dam Square
including the comprehensive Van Gogh Museum
the Rijksmuseum and the Rembrandt House Museum
all of which are taking part in Rembrandt 400
and Amsterdam is celebrating his life in grand style throughout this year with special museum exhibits and events
The museum lines weren’t as long as I expected
salon and other rooms and found it particularly interesting that he bought the expensive home in 1639
about the same time he received a large commission to paint “Night Watch,” probably his greatest work
depicting a group of militiamen preparing to march
Amsterdam was once the European capital of culture and commerce

Tourists visit it today for its cultural treasures
its excellent cafes and its charming canals — and sometimes to taste the forbidden fruits of its red-light district or the coffee shops authorized to sell cannabis
Bikes are everywhere in the city; thousands are stowed near the Central Train Station
and every post and pole in the city seems to have one bike or several chained tightly to it
“Amsterdam was made for horses and boats originally,” said Els Wamsteeker of the city’s tourism bureau
an inviting town on the West Frisian peninsula that still has a flotilla of colorful wooden windjammers
we could easily have gone by train; its rail station is a terminus on the line to Amsterdam
which has a scenic old town and central square
where we abandoned the car for rented bicycles
It’s a storybook town of cobblestone streets
tidy little shops and rustic canals and bridges
We rode through the village and countryside
then topped a dike along Lake IJsselmeer’s edge
and I could see sailboats straining again the wind on the lake and feel my face turning bright red in the chill

Though now known as something of a shopping destination
Arnhem has a rich World War II history to explore
Be cautious of vehicular and bicycle traffic when you're on foot -- pedestrians do not have the right of way
A stop in Arnhem makes for a lovely day of wandering through historic sights
within walking distance of the city's main shopping district
as well as the Battle of Arnhem Information Center at the foot of the John Frost Bridge
There are a handful of restaurants in the immediate vicinity and on a sunny day their terraces are a nice place to sip a drink and look out over the water
you'll need to walk about 10 minutes further into the city
it is imperative that you watch out for bicyclists and cars when walking around
Pedestrians do not have the right of way in the Netherlands
This is especially true where bicyclists are concerned
They do not slow down and they can come from any direction unexpectedly
so be sure to look every which way before stepping onto a bike lane
cars and trucks will drive on many of the narrow streets -- and even in squares -- so always keep an eye and ear open for oncoming traffic
Most of Arnhem's main attractions are too far afield to walk; an organized tour is your best bet as most itineraries do not spend a full day in the city and so your time is tight
A short walking tour through the city will take you past the Historic Cellars and St
If your riverboat doesn't have bikes and you're interested in renting one for a few hours
head 10 to 15 minutes into the city to the central rail station and look for the "Fietsenstalling" sign where you can rent a bike for the day
The Netherlands is part of the European community
While credit cards are accepted in many stores
in almost all cases you need a card with a chip and pin
including two ING banks (just look for the bright orange lions on the sign) with ATMs; the larger of the two is on Willemsplein about two minutes away from the information center
Dutch is the official language of Arnhem but almost everyone speaks English
Just say hello first and you'll be greeted in English
Only riverboats can visit the small city of Arnhem
with all boats mooring along the Lower Rhine (or Nederrijn) between the Nelson Mandela Brug (Bridge) and the John Frost Brug (what was once the "Bridge Too Far")
There are no port facilities and no parking lot
a small street dotted with Turkish and Indian restaurants right along the water
From here you can make your way by foot further into city center
There are no buses in the immediate vicinity

lightning and hail began to hit the Netherlands on Monday afternoon
At least 180 flights at Schiphol Airport were canceled during the afternoon and evening hours due to the expected gusts
The wind also led to the cancellation of trains running between the central stations in Amsterdam and Rotterdam on the high-speed line
The flight cancellations at Schiphol primarily concern 90 roundtrip flight pairings
Some 207 departures were facing delays throughout the day
and another 186 arrivals were also expected to land later than scheduled
no trains will run between Amsterdam Centraal Station and Rotterdam Centraal Station on the high-speed line on Monday
The passenger train operator said it expects that the direct high-speed connection between the two cities will not restart before Tuesday afternoon at around 3 p.m
The strong wind was also causing a disruption on the high-speed route between Rotterdam and Breda
Fewer trains will run on that route on Monday until at least around 8 p.m
A weather warning in place in Noord-Holland

The KNMI also warned of thunderstorms with heavy gusts of winds between 75 and 90 kilometers per hour everywhere else in the Netherlands, which should taper off into the evening hours on Monday. A code yellow weather warning applies to the entire country

but the western coastal provinces will remain at risk of heavy gusts until the night into Tuesday

A group of determined pensioners who credit a cycling group for 'changing their lives' has returned from a cycling trip all the way to Enkhuizen in Holland
Headed up by 72-year-old Lawrence O’Halleron - who received an MBE for his dedicated work to improve the lives of residents in Gateshead - the cycling club
take on hundreds of miles of terrain every week and are by no means restricted to this country
Lawrence's passion for fostering a sense of community and improving mental and social wellbeing has led him to lead three rides each week and organize various events throughout the year
The group comprises more than 50 members with many of the members being in their 60s
70s and 80s - some people have taken early retirement and joined the group
Lawrence said: "Teams Wheelers offers a lot of people a way to be active after they retire
they might not want to cycle huge distances or to new places alone - and even if they do
Without the support of the wider community and people turning up to cycle and chat week in
the trip to Holland wouldn't be possible."
As chair of Big Local Gateshead for the past eight years
Lawrence has seen the area's community grow stronger and stronger
The area was given £1m of funding from National Lottery funds 10 years ago to spend on bettering the community
supported by the work of charity organisation Local Trust
Many of the members of Teams Wheelers got involved not just because they enjoyed the sport
but because they wanted to make new connections after retiring from work
Lorraine Jacques is one of the newest members of Teams Wheelers
She said: "I am nearly blind in my right eye so I didn't have the confidence to cycle alone
Lawrence has led a group of people that are caring
Everyone made the effort to make me feel welcome and supported me throughout the rides."
Peter Irvine seconds Lorraine that joining Teams Wheelers was the 'best
life-changing decision' he's made and said: "Teams Wheelers are always encouraging new riders to join
which involves supporting them and teaching them the ways of the roads
The huge effect on people's health and wellbeing helps the NHS and local community greatly and it gets people fit and out of the house."

there is a void that needs filling - not just with the activity
Joining Teams Wheelers and cycling with a group of people of a similar age has helped massively with the transition into retirement."

This morning it was announced that the Dutch national broadcaster
would announce the Dutch representative of Eurovision 2025 in Basel (Switzerland)
and it turns out that Claude is going to represent the Netherlands
full name Claude Kiambe (September 16th 2003)
is a Dutch singer that is from Congolese and Kinshasan decent
In 2013 Claude fled Congo when he was just 9 years of age together with his mother
They ended up in a Dutch refugee camp in the city of Alkmaar
This is where Claude’s love for music started
In 2017 he won his first local talent show in Enkhuizen en in the Spring of 2019 he took part in the eight season of “The Voice Kids” in the Netherlands
where he sang “Papaoutai” of world-famous Belgian singer
this single became number 1 in the Netherlands
but also got a lot of recognition in Belgium
This song was heavily influenced by Claude’s great example
In 2023 Claude was announced as the pre-show of his idol Stromae
but due to health reasons Stromae experienced at the time all the concerts were cancelled
In March and April of 2024 Claude had his first own club tour with which he toured the Netherlands
On September 21st Claude was featured in the Dutch hit show
In 2024 Claude hinted that he was ready to do Eurovision if all the stars would align
and today he was announced as the Dutch representative for Eurovision 2025
The Eurovision Song Contest 2025 will take place in Basel (Switzerland)
