With a newly added volume including the Council Chamber, Reset architecture has reorganized the town hall of Geldrop- Mierlo (Dutch town next to Eindhoven). This transparent meeting centre communicates the public nature of the building and engages the citizens with the municipality. Internally, it is the core of the building complex where visitors, the council and civil servants come together.

With this rearrangement a new spatial clarity arises in which the villa and the newly added volume together form the public entity, centrally placed between the two office wings. This unity is reflected in architecture.

The entire centre has an open and spacious appearance with meeting rooms on two floors and a lower, more intimate part, with the Council Chamber.
© Stijn PoelstraThe cohesion of the elegant space is enhanced by a continuous rhythm of columns in the four facades and all- round light through the glass facades. This carré of columns subtly positions the new space between the existing office wings outer walls. Spatially they appear to be load-bearing but are only a curtain wall construction.
© Stijn PoelstraThe lightweight superstructure is built upon existing columns and foundation. Reset architecture has proposed a T-shape remontable steel structure with a wooden floor and a steel roof. This clear construction scheme contributes to the logical embedding of the program in between the columns grid. The routing is located outside the columns and surrounded by the columns of the façades.
SectionOn the longitudinal side, a façade strip of oak slats has been installed to hide the connection with the office wings gutter. Maybe the oak provides a mental connection with the woods outside but surely it contributes to a warm atmosphere experience of the space, which furthermore consists of calm gray tones in its finishes.
The town hall is heated by low-temperature heating connected to air-to-water heat pumps. An additional VRF system further extends the ability to heat or cool the building. The air handling units for the ventilation of the building are fitted with heat recovery elements. Adaptive LED lighting has been used to further reduce the energy usage of the building which is to a large extent generated by PV panels.
© Stijn PoelstraSustainability is more than saving energy. The new construction is carried out in a light, mountable structure in line with a circular way of thinking. During construction, (demolition) parts are reused immediately or made available for reuse. The project makes a small, contribution to the biodiversity in the built environment. Bat and swallow boxes have been built into the north facade of the renewed gable ends.

The universal appearance of a curtain wall, as an addition to the building complex, has been gratefully used to clearly communicate the public function and inviting open character. Nevertheless, the curtain wall detailing is not standard, the system has been refined to ensure that the universal also matches its context. The design of the new building shows a far-reaching abstraction of the vertical articulation and corner pillars in the eclectic villa.

Distinct micro-environmental properties have been reported to be essential for maintenance of neural precursor cells (NPCs) within the adult brain
Due to high complexity and technical limitations
the natural niche can barely be studied systematically in vivo
By reconstituting selected environmental properties (adhesiveness
we show that NPCs can be maintained stably at high density over an extended period of time (up to 8 days)
they would expand and (in the case of neurospheres) differentiate rapidly

Next, the specificity of the interactions between the NPCs and the engineered hydrogel matrices was tested. Figure 4B graphically summarizes cellular responses upon addition of an integrin-β1 blocker (Echistatin) and a MMP-inhibitor (GM6001)
inhibition of RGD-mediated signaling and MMP-degradation
significantly decreased object density in +RGD/+DL hydrogels measured on day 6 (OD+RGD/+DL = 623.1 ± 64.3 µl−1 against OD+RGD/+DL+Echistatin = 363.3 ± 50.5 µl−1 and OD+RGD/+DL+GM6001 = 317.1 ± 23.8 µl−1; both p < 0.001; two-way ANOVA F(10,126) = 6.94) but not in −RGD/−DL geldrop cultures
the NPCs need to be able to (i) receive integrin signals and to (ii) modulate the architecture of their surrounding ECM
The cell capability of the cells to invade the surrounding hydrogel is largely dependent on the activity of MMPs
at day 6 the highest cell density was found in the stiffest γ1.5 hydrogel (OD+RGD/+DL = 824.7 ± 67.7 µl−1)
the two gel matrices with medium and stiff mechanical properties had equally high cell densities (for γ1.0: OD+RGD/+MMP = 813.5 ± 85.5 µl−1
for γ1.5: OD+RGD/+MMP = 824.6 ± 90.0 µl−1)
Since gels with a higher degree of crosslinking contained a greater number of degradable linkers
the proliferation-enhancing effect could either be caused by increased stiffness or by increased matrix degradability

Our geldrop culture experiments show that adding extracellular cues profoundly alters how NPCs act in culture
the novel culture environment allows to maintain them in a stem-like state with low rates of baseline expansion in which the cells
other than commonly observed under neurosphere conditions
do not show signs of instantaneous differentiation
Monolayer cultures do not show spontaneous differentiation as long as they are kept in the presence of growth factors
resembling the picture in monolayer cultures
but are often aligned in parallel and actual migration will likely be limited due to the mechanical properties of the drop and the dense cell mass
an adhesive 2D substrate appears to be a factor that both promotes and limits proliferation
These conditions set an upper limit in terms of final cell density as well as for the possible time in culture without passaging
neurospheres do not rely on the availability of adhesive substrates and are consequently not restricted in this respect
the naturally resulting spherical shape of the cell clusters is not beneficial for the continued supply by growth factors by diffusion
doubling the diameter of the sphere increases the surface by a factor of 4
whereas the volume increased by a factor of 8
growth factor supply through the surface becomes increasingly insufficient with greater spheres
the proliferation of the NPCs eventually reaches an upper limit of cell numbers that can be maintained
arrangement of NPCs in elongated clusters as in the geldrop cultures presumably assured a beneficial surface-to-volume ratio
The comparison is based on two volumetric measures
which in the case of the nuclear EdU-signal is an unusual approach because the volume does not directly translate into a count
which is highly error-prone for densely packed cells
But under the assumption that the volume of a nucleus is constant
the total amount of EdU-volume is a linearly-dependent measure for proliferating cells without the error through mis-segmentation
The fit of a linear model allows the conclusion that measuring GFP-signal is a valid proxy for proliferation
A disadvantage of the gel drop cultures is that immunocytochemistry is often impossible
presumably due to antibody scavenging and penetration issues
This limits the use of markers that can be applied
so that our conclusions about the nature of cytoskeletal changes
also in response to ROCK inhibition remains limited
sustained proliferation of cells no longer leads to rapid (in monolayer) or subtle (in neurospheres) collapse of the NPC population
Instead it is possible to steer cell cluster growth towards a state that allows sufficient presentation of adhesion sites and diffusible growth factors
this proteolysis seemed robust towards changes in the sequence of the substrate
maintenance of NPCs in +RGD/+DL hydrogels relied on a functional cell-microenvironment interaction
We observed a bi-directionality of this interaction: perturbation on both the matrix side (by scrambled linker peptides) and on the cell side (by molecular inhibitors) equally led to a loss of the maintenance effect
This cellular ‘mechanical memory’ influenced cell fate decision at a later time point
In the context of cultured NPCs there is thus a need for additional studies targeting the proteomic and transcriptomic level to further clarify
if and how the cells are intrinsically affected by their culture microenvironment
results from secondary cultures highlight the potential of the geldrop culture to strongly instruct cells during the period of culture while in parallel maintaining their cardinal features – a fundamental requirement for the in vitro maintenance of NPCs
In summary we have demonstrated the applicability of geldrop cultures to maintain NPCs at high density in vitro
The specific advantage of that approach over monolayer and neurosphere cultures originates from an in vivo-like representation of the microenvironment
cell-cell and cell-ECM interactions can act in concert rather than excluding each other (as seen in the two extreme conventional platforms)
It was possible to systematically vary matrix-dependent signals and thereby optimize the microenvironment for NPCs
An optimized hydrogel matrix then allowed cell cultivation under high cell densities in a chemically defined 3D environment without losing the multipotency and self-renewal potential
While the incompatibility of the hydrogel with immunohistochemistry poses a serious practical limitation
the fact that precursor cells can become stabilized in a reductionistic artificial microenvironment is an important step towards building synthetic stem cell niches
The experiments were conducted in accordance with the applicable European and National regulations (Tierschutzgesetz) and approved by the responsible authority (Regierungspräsidium Dresden)
For the following discussion “RGD” refers to the adhesion sequence and “DL” refers to the degradable
Presence of functional groups within a respective hydrogel is indicated by +/− prefix in the text
For inhibition of Rho-associated protein kinase (ROCK) the inhibitor Y-27632 (Sigma-Aldrich) was added at a concentration of 10 µM
Collagenase A was dissolved at 1 mg/ml in complete Neurobasal medium
Then gels possessing a MMP-cleavable peptide sequence were digested by incubation with Collagenase solution for 5 min at 37 °C
Remaining cell aggregates were minced by repetitive up-and-down pipetting

Upon fixing with 4% PFA for 20 min at room temperature samples were washed once with PBS
a commercial Click-iT EdU imaging kit was used (Cat-#: C10337
According to the kit’s protocol the samples were incubated for 30 min with the Click-iT reaction cocktail
After one washing step the samples were additionally counterstained with Hoechst 33342 (1:4000) for 15 min and washed 3 times
Raw images were pre-processed using ‘Background Subtraction’ followed by ‘Gamma Correction’
images were segmented using automated OTSU thresholding
The resulting binary images were than quantified for object counts using the plug-in ‘3D Object Counter’ with a lower size cut-off of 100 voxel (voxel size: dx = 0.645 µm
the sum of all object counts (derived from multiple z-stacks per geldrop) was divided by the total scanned gel volume that had been investigated by spinning disk microscopy in that experiment
The scanned gel volume can be calculated as area of field-of-view times the height of a z-stack
By normalizing object counts on a per-volume-base
the metric of object density is calculated

the possibilities for renting out a room were broadened
an online platform was launched to connect students and landladies
the municipality wants to do something about the shortage of student housing
The tunnel for cyclists and pedestrians at the Geldrop train station is being refurbished
The underpass will have a more fresh look and new lighting will be installed
The municipality has agreed this with ProRail and the NS
The modifications should improve the appearance of the tunnel
Safety for cyclists and pedestrians must also be strengthened with this
the entrance on the side of the Zesgehuchten will be finished in the same way as the entrance on the centre side of Geldrop

the municipality must accommodate 231 asylum seekers
Geldrop-Mierlo currently only meets the quota of 54 minor refugees
the municipality has the task of housing 232 Ukrainian refugees
There are plans to place a few temporary units for the remaining twenty
The special youth evening in Geldrop during the annual KommuS Kasteelfestival (castle festival) in July will not take place
The reason is that the organisation no longer receives money from the municipality

The plan was to organise an evening for young people
the organisation received €20,000 to organise this evening
but this year the municipality has decided not to do so anymore

The town also created the world’s densest Tolkien-themed street grid: an estate covering about a quarter of a square mile
avenues and paths carrying names taken from Tolkien’s various epics
The area was built up in the late 20th century in Genoenhuis
a district of Geldrop bordered by the E34 motorway in the south and the Eindhoven to Weert railway in the east
part of the estate looks ring-shaped – which gave one town planner the idea to link the area to Tolkien

Jack says he still managed to get through the corona years reasonably well
but now that he is once again facing understaffing and also sees purchasing and energy prices rising

“I still have a top business and now there are people who want to take over from me”

Our fries are made at the factory and delivered here frozen twice a week
The quality is beyond question as far as I’m concerned
although we do use the slogan here: ‘There’s no arguing about taste’ “
The end of an era is approaching for Corrie and Leo Hendriks
they ran the Primera together in the Coevering shopping centre in Geldrop
“I normally work about 70 hours a week
Going out spontaneously isn’t easy for him
Corrie will continue to work for the new owner
“I want to give myself at least a six-month break
Now I want to invest my time in my hobbies”
She likes to play the clarinet and has been with the local orchestra for years
