Amyloid fibrils have emerged as innovative tools to enhance the transduction efficiency of retroviral vectors in gene therapy strategies
we used cryo-electron microscopy to analyze the structure of a biotechnologically engineered peptide fibril that enhances retroviral infectivity
Our findings show that the peptide undergoes a time-dependent morphological maturation into polymorphic amyloid fibril structures
The fibrils consist of mated cross-β sheets that interact by the hydrophobic residues of the amphipathic fibril-forming peptide
The now available structural data help to explain the mechanism of retroviral infectivity enhancement
provide insights into the molecular plasticity of amyloid structures and illuminate the thermodynamic basis of their morphological maturation
In the absence of an experimentally determined structure of a virus-binding fibril
it is difficult to estimate how these peptide properties manifest themselves at the quaternary structural level of the assembled amyloid fibril
Our findings show that the structure is based on a fundamental building principle
Each protofilament (PF) is built by two paired cross-β sheets that interact by the hydrophobic side of the amphipathic peptide
while the polar residues are pointing outwards
These structural features provide insights into the molecular basis of amyloid polymorphism and virus binding
and improves our understanding of the molecular basis of peptide nanofibrils as retroviral transduction enhancers
a Negative Stain TEM images of in vitro formed PNF-18 fibrils at different times of incubation
b Relative abundance of fibril widths of in vitro formed PNF-18 fibrils at different times of incubation
a Cryo-EM images of fibril morphologies I − III and others
b Crossover distance and fibril width values of fibril morphologies I − III and others (n = 20)
c Relative abundance of fibril morphologies I − III and others in samples after one and 14 days of incubation (n = 500
We conclude that the specific morphology of PNF-18 fibrils is not crucial for their virus enhancing infectivity
a 4.8 Å tick slice of one molecular layer of the fibril (top)
Below are 1.2 Å thick slices through one layer of the reconstructed density of Morphology II (bottom)
b Cross-sectional view of one molecular layer of the reconstructed density (gray)
c Side view of a stack of four layers of the molecular model showing the staggering of the peptide stack towards each other
a Depiction of the amphiphilic nature of the peptide
showing the polar (green) and apolar (yellow) sites in the molecule
b Hydrophobic contact mode between two type α peptide stacks (c) Hydrophilic contact mode between a α and a β peptide stack
a Cross-sectional view of the molecular model of Morphology II showing the polar (green) and apolar (yellow) sites in the molecule
b Coulomb surface potential of the cross-sectional view of Morphology II and (c) side view of the fibril
Morphology I seems to contain four peptides in each molecular layer and Morphology III eight
The general structural similarity of the two fibrils to Morphology II implies the existence of common hierarchical structural principles underlying the formation of differently structured PNF-18 amyloid fibrils and that the three fibril morphologies differ mainly in the number of their constituting PFs
suggesting that the altered tilt helps to compensate the higher torsional strain at outer radial positions
Biochemical calculation example of the benefit of the conversion of six fibrils of Morphology I (left) into four fibrils of Morphology II (right)
The gain is the formation of two additional polar contact modes
Chemically synthetic PNF-18 peptide was obtained from the group of Prof
Tanja Weil (Max Planck Institute for Polymer Research
The peptide was incubated at 0.3 mg/mL concentration in 50 mM HEPES buffer
for a period of up to 60 days at room temperature
After different time points aliquots were withdrawn from the reaction and analyzed as indicated in each experiment
4 µL of the sample solution were placed onto a glow-discharged carbon coated
The sample was incubated on the grid for 1 min at room temperature
Surplus solution was removed with filter paper (Whatman)
The grid was washed three times with 10 µL water and stained three times with 10 µL 2% (w/v) uranyl acetate in water
The dried grids were examined in a JEM-1400 TEM (JEOL) equipped with a F216 camera (TVIPS) that was operated at 120 kV
Electron Microscopy Sciences) were glow-discharged for 40 s at 20 mA using a PELCO easiGlow glow discharge cleaning system (TED PELLA)
4 µL of sample were then applied on the grid and incubated for 30 s
followed by both side blotting for 8 s (temperature 21°
humidity >90%) and plunging into liquid ethane using a Vitrobot Mark 3 (Thermo Fisher Scientific)
Grids were screened using a JEM-2100 transmission electron microscope (Jeol) at 200 kV
High-resolution data sets of the fibrils were recorded with a Titan Krios (Thermo Fisher Scientific) microscope that was equipped with a K2-Summit detector (Gatan) and operated at an acceleration voltage of 300 kV
The width and crossover distance of morphologies were determined from cryo-TEM images using ImageJ software
Together with the imposing of a C2 symmetry during reconstruction yielded clearly separated β-strands
3D classification with local optimization of helical twist and rise and a C2 symmetry were used
The best 3D class was finally reconstructed with local optimization of the helical parameters using 3D auto-refinement
All 3D classification and auto-refine processes were carried out by using a central part of 10% of the intermediate asymmetrical reconstruction
The final reconstruction was post-processed with a soft-edge mask and B-factor sharpened
The estimate of the resolution of the reconstruction was carried out based on the FSC at 0.143 between two independently refined half-maps
The error bars shown in this manuscript report the standard deviation

Race One: Noel Zanocz was just too good in the first moto – he wasn’t top three at the start but it didn’t take him long to get to the front passing his Fantic team mate
Simone Mancini – he even had time to wave to his friends in the crowd after making the move
Mancini was riding well considering the conditions but then Garcia passed him on the last lap for second
Hindersson rode well for fifth ahead of Doensen who will be looking for much more – he should be at the front
Team GB’s Jamie Keith rode superb for sixth
Cole McCullough got an awful start with only a couple of riders behind him but charged through the field – he got to twelfth with plenty of riders just ahead of him before a costly crash
Race Two: Mancini got another holeshot and was looking good at the front but Zanocz was buried down the order outside the top fifteen but he went into beast mode and was passing riders everywhere around a brutal track
The Hungarian got up to second which was enough for the overall with Mancini taking the win – he is getting stronger in the sand
Garcia ended up on the podium with a third overall
McCullough had another crash but battled through to tenth
Race One: Moritz Ernecker got the holeshot and was looking superb at the front
He did have Levi Townley in hot pursuit as the top five stayed relatively close for the first five minutes
Ernecker had a crash which allowed Townley to led and he then started to pull away at the front – he ended up with the win
Sleny Goyer from France is good in the sand and finished runner up with Finnish talent

learned the owners of Northern Paws Pet Lodge
they decided they wanted to take over the business
before the Heerdes bought the business Dec
“It takes a lot to make it go ’round,” said Ashley
The previous owners have also continued to work at the lodge
and keep the same care pets and their families have come to expect
She says they have run the pet lodge much the same as the previous owners and really only made one major update
because it was a really old tub and stuff,” said Ashley
Ashley says they currently do not offer grooming services
but do give a complimentary bath to dogs who stay for six or more days at a time
The business has 31 dog rooms altogether – 24 standard rooms in the East and West Pines wings
and seven suites for well-behaved and house-trained dogs
The suites also have TVs in the room for the pets and a raised bed
though Ashley says families are always welcome to bring bedding that smells like home
The building also has in-floor heat to make the space more comfortable for the pets
There is also an 11-acre outdoor play area for dogs
so they have some time to use the bathroom and play with a staff member for a bit
Dogs are not intermingled with others from outside their household
so that makes it nice,” said Ashley of the play area
adding there is a smaller fenced in yard for smaller dogs
Northern Paws also offers daycare for dogs to hang out for the day
There are six cat condos and they also get time to explore
with an indoor playroom for the felines to take turns visiting
where Ashley says they try to share pictures of the pets who stay with them
“We really try to treat them like our own,” said Ashley

The Uluru Statement
Almost as soon as one school district's bond election ended
The Harrisburg school board is asking voters to pass a $40 million school bond next week to expand the district's high school and build a new elementary school by August 2020.
And they're doing it without a tax increase to residents
but residents can vote absentee or early between now and Monday.
It's the first bond election for the district since 2014, when voters passed a $25 million bond to build Horizon Elementary
school board vice chairwoman Linda Heerde said Tuesday
And it comes on the coattails of voters passing the largest school bond in the state's history for the Sioux Falls School District about a month ago
we're either building or preparing to build," Heerde said
I almost feel like we've developed a little bit of an art or science to this because we do it so often that we've gotten kind of good at building buildings
The district's population has nearly doubled in a decade
making it the fastest growing district in the state
More: Flexibility key for fast-growing Harrisburg district
With no chance of growth slowing down
Heerde said the current high school will be over capacity by fall 2020
Liberty and Endeavor Elementary schools are also expected to be beyond capacity by the same time
with Freedom Elementary expected to be overflowing by 2021.
Expanding the high school by 450 additional students and adding another elementary school would alleviate the pressure
school board member Jay Hutton said.
"It's kind of known we're not behind the eight-ball," Hutton said
"We're really building for where we see growth in the numbers of students."
The idea to expand came more than a year ago after focus group meetings and community input
do we want to build on to elementary schools that are reaching their max capacity?" Heerde said
"Was it OK with everybody to bump up and build on to making these elementary schools bigger or was it more important for us to keep that small town
More: Harrisburg proposes building 'freshman academy,' field house to ease strain on high school
That became the central debate before the board called for the bond
And the focus groups easily showed the thing that makes Harrisburg unique was making sure students felt apart of the community
"Making big buildings was not going to accomplish that for us," she said.
The new high school addition comes with a nearly $22 million price tag for construction, including classrooms, music rooms, an auxiliary gym, a physical education facility, a building construction shop and a possible bus barn, according to the district's website
The price tag also includes the cost of the architecture
landscaping and furnishing technology and equipment
The additions will be about 98,500 square feet
the new elementary will cost about $17 million and be located east of Bakker Crossing Golf Course
That includes the same work for the addition to the high school
building a playground and parking
The bond also includes insurance fees and interest payment amounts totaling about $1.8 million
"Everybody wants to remain one Tiger nation nation as long as we can," Heerde said.
Harrisburg School District hasn't raised taxes in 12 years because it operates under a self-imposed tax levy of $12 per $1,000 on owner-occupied properties.
With more houses and businesses being built in the area
the district has seen an increase in taxable valuation
That allows the district to build more schools without having to change the tax rate
the district has built seven schools and several auxiliary buildings
Where can you vote? The district is also changing up its voting locations this year
"The thing people get confused about for voting is city of Sioux Falls residents in the Harrisburg School District do get a vote in this election," Heerde said
and that's one thing people get confused about the most."
Any registered voter in the district's boundaries can vote by mail or during regular office hours at these nine different locations in Sioux Falls and Harrisburg between now and Monday:
residents will only have two locations to cast their ballots at between 7 a.m
Residents residing in the the city of Sioux Falls and Delapre township can vote at North Middle School next Tuesday
Lynn and Perry townships can vote at the community room in Liberty Elementary School

Initiatives to protect homeless young people during the pandemic were effective
but we need a long term strategy that delivers better support and helps prevent homelessness
By Dr Jess Heerde, Professor George Patton, Associate Professor Rohan Borschmann, Professor Stuart Kinner and Dr Jesse Young
In 2008 Australia had a national strategic agenda to halve homelessness by 2020
with a strategy of early intervention and improved service provision to finally break the homelessness cycle
This strategic agenda has long since been discarded
Instead we have been left with rising demand for homelessness services and a national homelessness support system struggling to keep up with this demand
The raw daily experience of what it’s like to be homeless
and the damage it does to young people’s health
wellbeing and life chances are largely hidden from view
We can just walk past the problem on our streets
without seeing it and without understanding it
But homelessness and the threat of homelessness puts relentless pressure on young people and their families
The young have had the greatest recent increases in homelessness. Rates among those aged 25-34 years have steadily increased over the past three Censuses
to 18,411 in 2011 and 24,224 in 2016 –a 32 per cent increase between 2011 and 2016
According to the 2019-20 Australian Institute of Health and Welfare specialist homelessness services annual report
290,500 Australians accessed homelessness services over this period
and of these 40 per cent were aged under 25 years
COVID-19 has reminded us that it doesn’t need to be this way
The pandemic has highlighted the extreme vulnerabilities of the homeless, especially the young homeless. It triggered unprecedented intervention by Australian governments through health
which successfully reduced the incidence of the virus among the homeless
The success of these COVID-19-driven housing and social strategies in protecting young Australians from homelessness could and should be the catalyst for strategic and enduring solutions
real and enduring reform has not yet occurred
Investing in social housing during a pandemic
We are facing the risk of a new wave of youth homelessness
Lockdowns and associated economic downturns have brought an increased demand for homelessness services, demand that is not likely to subside any time soon. The temporary supports put in place at the height of the pandemic are receding, funding for emergency and temporary accommodation is being wound back
This is compounded by high rates of youth unemployment
These factors now threaten to fuel a new and larger wave of homelessness in the young
it doesn’t provide funding to increase capacity for services to respond to the growing homelessness crisis
This is also missed in the Oppositions’s pledge of 20,000 social housing properties
government policy has relied on intervening only when young people are at serious risk of falling into homelessness or are already homeless
The danger is that we will keep relying on short-term
crisis-driven approaches such as those precipitated by the COVID-19 pandemic
Ultimately, these approaches do little to address the underlying drivers of homelessness among the young. Nor do they address the adverse health and social consequences of homelessness
Population-wide and targeted prevention strategies are rarely discussed or funded. We need to be focusing on the needs of young people prior to their becoming at risk of homelessness. It is possible to identify the drivers of homelessness from early in adolescence
and the interrelationships among them that operate at individual
Targeting these drivers should be central in driving policy reform to reduce homelessness and support young Australians to make a successful transition into stable housing
In addition to targeting the drivers of homelessness, it is also necessary to focus on the health needs of the young homeless
we must track the health needs and outcomes of these young people at a national level to drive whole of-government policy reform
Victorian kids’ mental health hit hardest during 2020
and make a strong case for the minimisation of co-payments required for health services
Although feasible
currently this absence of knowledge is a public health opportunity missed
To reduce the number of homeless young Australians our approach needs to improve
crisis-driven approaches will not prevent a new wave of homelessness or better respond to the health needs of homeless young people
It is time for an inter-sectoral alliance of researchers
and young people with lived experience of homelessness to come together and be included in planning processes

Professor van Heerde moved to New Zealand in 2006 and joined Massey in January 2013
“It’s nice to be able to represent Massey like this and be part of a culture where success is celebrated,” he says
Professor van Heerde has a PhD in economics and his research focuses on measuring the quantitative effect of marketing
including the impact of advertising on sales and the interaction between corporate communications
He was also instrumental in developing Massey's Master of Analytics qualification
which aims to fill the “big data” skills shortage
and he holds the MSA Charitable Trust Chair in Marketing
The quality and impact of Professor van Heerde’s research has received plenty of recognition recently
he received both the Jan-Benedict EM Steenkamp Award and the INFORMS Society for Marketing Science Long Term Impact Award
Professor van Heerde’s work on building brands

Richard van Heerde dominated this past weekend’s (April 19-20) INEX Legends second round at Zwartkops Raceway to emerge as joint points leader with Jayden Goosen
was also favoured by fortune after drawing pole position for Saturday’s first race
with Van Heerde joining her on the front row
Debutant Gavin Botha shared the second row with Goosen
while Neels Jooste and Schaun Kleynhans completed the top six
Berlin Robinson and Rico Raker were the next to line up
Tyler Robinson’s advantage increased her advantage over the rest of the field in Race 1
Goosen made up places early to slot into an early second place
The sizeable crowd’s attention soon shifted to the battle for third place
Roos made up a host of places to trail Botha and Van Heerde
It did not take Roos long to pass van Heerde
he moved into the final podium spot by passing Botha
Van Heerde dispatched Botha to fifth place
Padayachee kept a watchful eye on the battle en route to sixth place
Repsold held off Jacobs Jnr for seventh place
while Raker and Jooste completed the list of drivers who went the full race distance
Berlin Robinson moved to ninth place before retiring
while Kleynhans retired just after the half-distance mark
Both the latter drivers failed to complete the minimum distance to earn a result classification
Padayachee drew pole position for the second race
and Raker completed the rest of the line-up
the race soon got neutralised following incidents that sidelined Tyler Robinson
and a subsequent Safety Car followed by a red flag
the final classification was based on the aggregate times for both parts of the race
This saw Jacobs Jnr winning his maiden race
Padayachee took third after trading positions with Roos
Repsold and Berlin Robinson completed the top six as they finished
Tyler Robinson’s DNF in Race 2 came to her aid for the Race 3 starting grid
which was determined by inverting the points sheet for the day
with the rest of the grid comprising Repsold
Robinson took the early lead en route to her fifth win of the season
Goosen hounded her to the flag but without success
Jacobs Jnr occupied third place in the early stages before Van Heerde traded places with him
while Berlin Robinson completed the race in a lonely sixth place
the last of the drivers to go the full race distance
Van Heerde’s consistency throughout paid off
with him taking the Round 2 victory by 20 points from Goosen
Jacobs Jnr completed the podium for the event
Van Heerde also took the Masters Class win from Repsold and Jooste
Goosen won the Professional Class ahead of Tyler Robinson and Botha
Padayachee beat Raker to the Semi-Professional Class win
while Jacobs Jnr took the Young Lions honours from Roos and Berlin Robison
Van Heerde and Goosen are now tied on 1020 points at the top of the overall points table

Professor Harald Van Heerde from UNSW Business School achieves highest ever placing in prestigious Marketing Research Productivity Lists
Ibrar KhanUNSW Media & Content+61 2 9385 0853ibrar.khan@unsw.edu.au
A UNSW Business School academic has been ranked 7th in the world for his contribution to the field of marketing
having been in the top 10 for a while now – I’m very happy to be representing this part of the world and UNSW at that level,” Professor Harald Van Heerde said
The Marketing Research Productivity Lists are based on the number of articles written in the top four journals over the past 10 years
UNSW Sydney is the only Australian university to feature an academic in the top 20 positions of the list which ranks Author Productivity in the Premier Marketing Journals (2009-2018)
“Most people in the rankings are US based academics or have been trained in the US
I’ve worked in New Zealand and Australia having gained my PhD in the Netherlands
so it’s special to be a bit of an outlier in that field of mostly American colleagues,” Van Heerde said
Professor Van Heerde earned his PhD at the University of Groningen in the Netherlands and has attracted significant research grants for projects in the Netherlands
He also serves as the only non-US based Co-editor of the Journal of Marketing
Traditionally it’s been about consumers and firms but it’s now beginning to open up the scope
so it’s now becoming more about social marketing
using marketing to create a better world,” Prof Van Heerde said
“A lot of marketing articles are about privacy concerns
technological driven products such as mobile marketing
Uber) – these are very interesting times to be a marketeer.”
Professor Van Heerde was appointed by the Business School under the SHARP program
or Strategic Hires and Retention Pathways recruitment - a UNSW 2025 program to attract the world's best scholars to UNSW Sydney to position it as 'Australia's Global University'
Dean UNSW Business School said: “He is an outstanding addition to our group of globally recognised marketing scholars."
The 2024 FIM Junior Motocross World Champions and World Cup Winner have been crowned today at the conclusion of racing on the Circuit Kamperweg Heerde in the heart of the Netherlands
Champions came from as far afield as Hungary and New Zealand
but the home crowd also had one of their own claim a winner’s trophy on the podium
Noel Zanocz continued his fantastic year with the World Junior 125cc title in his pocket after a stunning display of riding in the sand
The Hungarian also leads the EMX125 Championship at the time of writing
New Zealander Levi Townley justified the long journey with a stunning overall win to add another FIM Gold Medal to the family collection as father Ben watched on proudly
the home crowd cheered the success of young charger Kash van Hamond
who used his skills in the sand to claim the 65cc World Cup
It was also joy for the local supporters in the Nations standings
as Team Netherlands claimed the overall team victory
ahead of Team France in second and Team Great Britain in third
with the top 10 of this year’s EMX125 Championship present alongside top racers from all the other continents
but it was Sweden’s Sandor Sols who claimed the first race holeshot ahead of local ace Dean Gregoire
it was Simone Mancini who dispelled the rumours that young Italians can’t ride sand by taking the lead on the opening lap
son of 1990s star Marcel and no relation to WMX star Lotte
but Zanocz was on a mission and charged through to take second by the end of lap three
he set the fastest time of the whole race on lap four
and dived past Mancini for the lead on lap six
Sols dropped back with a bike issue to finish 14th
but Spaniard Francisco Garcia put in a solid ride to take third on lap six
then chased down Mancini with a pass on the very last lap of the race
with current EMX125 Championship challenger Doensen ending his topsy-turvy race in fifth
Zanocz cruised to a 14-second victory to leave everyone else scratching their heads
That super Swede Sols grabbed the holeshot again in race two
Mancini grabbed the lead on lap two this time
but Zanocz was outside the top 15 and giving his opponents some hope
Gregoire and Van Drunen were again giving their home fans something to cheer for
but it was Doensen who climbed to 2nd at the end of lap five
it was that Hungarian menace Zanocz who had powered up to second place past his series rival
With Doensen staying solid in third ahead of Garcia
who also fought through from behind at the start
it meant that Zanocz was in the driving seat for the overall
but he still put everything into catching his teammate
with a small mistake just interrupting his rhythm enough to give it to the Italian
Local ace Dani Heitink claimed fifth ahead of Hindersson
but it was Zanocz who claimed the World Junior Championship with his 1-2 scoreline ahead of Mancini’s 3-1
Garcia completed the podium ahead of Doensen and Hindersson
with Brit Jamie Keith taking a good sixth overall with consistent 6-8 finishes
The 85cc class had taken the 40 starters from 108 entrants on Saturday
so it was always going to be a full and frantic circuit for the buzzing machines
Austrian Moritz Ernecker took the holeshot and held the early lead from Levi Townley and French flyer Sleny Goyer
before Finland’s Viktor Leppälä moved into contention
remounting in 16th and recovering to finish an eventual 8th
This gave the elder of the two Townley brothers
as Goyer moved up to claim second with three laps to go
leaving Leppälä to hold off a charge from Estonian Lucas Leok
The USA’s Wyatt Duff claimed fifth ahead of another Estonian
who put a last lap pass on France’s Kenzo Ferez
and he simply would not be caught as he made an early gap and claimed a solid 12-second victory by the flag
The battle raged behind him as the Dane Casey Karstrom put his Yamaha into second initially before local ace Dex van den Broek completed his charge from outside the top five to claim second in the race
but Levi Townley battled past Brit Finley Pickering
enough to keep Leok back in the points and claim the first ever World Junior Championship for New Zealand
Van den Broek claimed third overall ahead of Goyer and Leppälä
with American Braxtyn Mes taking sixth overall by virtue of a stunning charge right from the back in race two
who has also been racing in the AMA recently
who claimed the first race holeshot in the 65cc class and was never overtaken to win by nearly ten seconds
Van Hamond fought from outside the top five to claim second in the race
ahead of Estonian Theo Kolts and a stunning charge from Great Britain’s Cohen Jagielski
Belgium’s Xen Temmerman took fifth ahead of fast-starting Frenchman Timoteï Cez
Verhoeven again took the holeshot in race two
but Van Hamond had charged from outside the top ten to claim second by lap three
then dived past the New Zealander to take the lead
Despite his efforts to fight for the World Cup overall victory
Nico lost positions to Germany’s Luca Nierychlo
enough only for second overall behind popular local ace Van Hamond
Verhoeven’s AMA rival Jaydin Smart came through from an awful start to claim fifth
with his countryman Joseph Vicari claiming sixth in race two
it was Van Hamond who claimed the first 65cc title for a Dutch rider since Ivano van Erp in 2017
with Jagielski recovering from a second race crash to finish 12th
with two overall podium finishers and a fourth place
were easily the overall winners in the team Championship
and France just edged out Team Great Britain for second overall
with Estonia fourth and a fighting USA in fifth
Team Italy was declared the winner of the Ride Green Cup for demonstrating once again their commitment to environmental protection
it was a fantastic event at the Dutch woodland circuit that many will take some amazing racing memories away from
