the wake of the exhibition of his decade long project “Urban Songlines” on view at the Tate I sat down with Dutch artist Allard van Hoorn for a conversation about art and its relationship to public space By designing collaborative participatory choreographies for architecture Allard van Hoorn encourages us to find novel ways of connecting experiencing and listening to public spaces David Van Leeuwen: How did you come to be interested in public space and the relationship of art to public space Allard van Hoorn: I guess I wasn’t always an artist in my youth I followed a path dictated by my environment at one point I felt like I wanted to develop a personal relationship to the world I felt that the point to start is public space because that is where I had to learn I had to step out of my door and feel and see and look at the world in order to be able to relate to it my idea was to investigate public space by doing some interventions in them.  I remember the very first thing I brought from Holland was a roll of duct tape and that coincided with the date I moved to Barcelona There was a free market in the streets of Amsterdam and kids and grownups tape a few square feet in the sidewalk before Queen’s day and they occupied that space with the tape so I found one in an old factory and the studios were only divided by the beams in the ceiling I was in the space for a while I was thinking about how to relate to the studio but at that point I had no idea what to do with the studio I eventually decided to reproduce what I saw last I taped the circumference of that square space that was mine I would put the cross in it as people in Amsterdam would do for Queens’ day I put the words on the wall that people put in it saying ‘occupied’ I left for a week and came back to the space the neighbor on the right-hand side had pushed an empty shopping cart containing a blank white canvas into the space and it was sitting squarely in the space a starting point.  The neighbor on the other side had drawn on the wall with an arrow pointing at my space ‘sin trabajo’ which translates in Spanish into ‘without work,’ the literal translation is jobless “hey if you don’t do anything with this space you may end up unemployed.” I thought it was amazing that just me doing a contemplative passive exercise with this space for myself as a thinking exercise led to two strong reactions from my neighbors I started to do interventions in the streets I wanted to see what possibilities would open up if we listened to spaces like buildings and bridges instead of just inhabiting them It is baked into our memories and it largely guides our attention How is sound important to your work and to public space I got very interested in the idea of how to work with public space and sound after reading “Songlines” a book by Bruce Chatwin in 2008 I was struck by the technique of aboriginals to translate their public space by singing the shape in language and in tone it was a threefold method in relating to the land it is a sonic map that you get to learn when you are born when the aboriginal mother is pregnant and she feels the first kick she marks the ground and the tribal elders assign a stretch of land from that point on to the horizon that is the songline that they learn to sing – the songline passes throughout the entire continent The tonal structure of the sung description of this landscape allows them to envision how the landscape will look like without knowing the language they relate to the land in a spiritual way And the third is a sense of management and taking care of the land to maintain balance I learned that I was super interested in sound being a representation of public space More than half of the people I know live in cities but sorts I have always liked the idea of the artist putting up their mirror to the world I learned that it was previously an active fishing and mining town and that there was a close-knit community that was more and more broken up by recent tourism These cute houses are rented out as holiday homes to tourists whose presence has gentrified the area pushing older communities into the suburbs helping each other out seem to have been lost to a certain extent the idea of supersizing the silver ball felt like a tool for self-reflection in relation to the sense of community gentrification; the slow-moving very reflective ball became a rolling mirror through public space that allowed participants to see each other together in the landscape What kind of choreographies would they design How would they reimagine the function of the silver ball if they had control over it We organized workshops with the different communities and any other community that wanted to collaborate We invited them to design their own workshops together with me and the Tate team and to see how they wanted to use that reflection that mirror that rolls through public space as a way to reconnect and discuss the idea of community sense in their town Guy Debord speaks of the re-humanizing effects of participation How do you view the value of participation by the audience and collaborators in your works generally participation and collaboration is something that develops while you are doing the project and its kind of a feedback system Buckminster Fuller had this design science implementation model where feedback is essential Continuous feedback loop into the original system and the more we get to the day we all perform together and activate this thing in public space something changes because somebody gave this very insightful input into the system I think it is essential that the spirit of the project has a life after the artist came and activated it in a certain community I try to create projects where the local community has as much ownership as possible if not entirely Their participation is the activation of the artwork Upon watching the video of the performance it is clear that there was a large turnout from the St What was it about the project that resonated with them The feedback I got was that people had a lot of ownership over the project They came up with the choreography for their own parts they have a ritual where they beat the grounds with a stick to wake up the apple trees that takes place in the community orchard in February the event is meant to stimulate a good harvest It was their idea to beat the silver ball as a community tool to wake it up and wake up its senses As the apple-orchard group handed over the ball to the skate park community meeting each other talking with each other in that landscape in the reflection of the ball that allowed a wider conversation Are there artists or thinkers that have influenced your ideas about art and its relation to public space or the value of public spaces in general I already mentioned Richard Buckminster Fuller I see his creativity and his discovery and research into the relationship of man in the special and mechanical world was an exercise that ended up in useable and tangible objects such as architecture and vehicles What I learned from this practical approach to art is that we can create tools that function in communities to spark conversation or incite change that have done projects that don’t seem like art performances because they are placed out of that context yet again in the public spaces where you don’t expect them to be art For artists working with interventions in public space this also goes for Christo and Jeanne Claude is that the audience extends beyond an art audience When you are in a white cube or in a museum When you encounter something in public space and you do not have to know anything about art and when you can interact with it differently I am interested in art and artists who allow meaning to be created by their audience or users; the openness that void where you have the chance to put in your own meaning A renowned architect once said that what would define the greatest architects How do your ideas of space relate an architect’s idea of space Architecture has to be more than the function alone The form must invite certain other aspects that are common to our lives and that is why I often introduce dance in buildings because I see dance as somewhat of a polar opposite to architecture which is regarded as one of the highest physical technological achievements of society Where architecture is static and rigid in its power If the body moves in public space it choreographs the space self-controlled piece of architecture in public space Then these two are opposites on the spectrum To create architecture came from the body and walking “Walkscapes” by Francesco Careri the very first architectural interventions in public This places him further forward and higher over the bottom bracket which should help him to get the power out when riding in an aggressive position The Cube Litening C:68X TE (the TE stands for ‘Team Edition’) is one of the best aero road bikes available, and it’s no surprise to see van der Hoorn sticking with this model, rather than switching to the new lightweight Cube Litening we spotted at the Giro d’Italia Though it nominally only has clearance for 28c tyres in practice it can take a bit more than that so it arguably makes it an ideal bike for a flat (We're yet to confirm what size tyres van der Hoorn had fitted for the Tour's cobbles but we'd have expected most teams to run 30c rubber – ed.) And it certainly looks as if van der Hoorn’s Cube has seen its fair share of cobbles tidy and well maintained when we photographed it ahead of the Grand Départ in Copenhagen it also had its fair share of battle scars On top of the aerodynamic riding position and bike van der Hoorn has also optimised his drivetrain on the day before the opening stage of this year’s race it had an enormous set of 56/44t chainrings on it Those are chainrings we’d have previously seen almost exclusively on time trial bikes optimised for flat courses van der Hoorn had an 11-34t cassette – a wide range option which likely helps him stay in that whopping 56t chainring more of the time Given the mostly flat parcours of the Tour to date we’d be surprised if van der Hoorn has strayed from that setup since The explanation for this is likely to be as simple as a limited availability of new parts – even former Tour winner Geraint Thomas is still using the same setup (albeit with less extreme gearing) Leaving no stone unturned, van der Hoorn also has the CeramicSpeed OSPW Aero system – a €739 derailleur cage assembly which is claimed to reduce drivetrain friction and improve a bike’s aerodynamics When we last checked in with van der Hoorn, he was running Continental GP5000TL tubeless tyres on Newmen carbon wheels Since then, though, he and the rest of his team have upgraded to the latest Continental GP5000 S TR tyres It’s a tyre which has already secured back-to-back victories at Paris-Roubaix, a win at the world time trial championships, and, perhaps most importantly of all, a coveted spot at the top of our best road bike tyres round-up The Conti tyres were mounted on Newmen Advanced R.50 Streem carbon wheels Notably, Intermarché Wanty Gobert’s head mechanic, Mikey van Kruinigen, told us the team isn’t using tubeless tyre inserts at this year’s Tour Though they offer a run-flat solution in the event of a puncture, van Kruinigen told us the foam inserts have a tendency to soak up tubeless sealant meaning they needed to add more than would otherwise be required that wasn’t acceptable to the team’s weight-conscious riders One nice detail we spotted was a sticker on each rim which noted how much sealant had been added to the system it’s crucial to check its state periodically in order to maintain its puncture-sealing performance The wheels also had a well-worn set of of their parts While van der Hoorn was narrowly pipped to the line after a gruelling stage over the Roubaix cobbles the former Giro stage winner clearly has good legs and a bike setup that works for him so we’re looking forward to seeing what else is to come over the next two weeks of the Tour Metrics details Adapted plant pathogens from various microbial kingdoms produce hundreds of unrelated small secreted proteins (SSPs) with elusive roles we used AlphaFold-Multimer (AFM) to screen 1879 SSPs of seven tomato pathogens for interacting with six defence-related hydrolases of tomato This screen of 11,274 protein pairs identified 15 non-annotated SSPs that are predicted to obstruct the active site of chitinases and proteases with an intrinsic fold Four SSPs were experimentally verified to be inhibitors of pathogenesis-related subtilase P69B including extracellular protein-36 (Ecp36) and secreted-into-xylem-15 (Six15) of the fungal pathogens Cladosporium fulvum and Fusarium oxysporum Together with a P69B inhibitor from the bacterial pathogen Xanthomonas perforans and Kazal-like inhibitors of the oomycete pathogen Phytophthora infestans P69B emerges as an effector hub targeted by different microbial kingdoms consistent with a diversification of P69B orthologs and paralogs This study demonstrates the power of artificial intelligence to predict cross-kingdom interactions at the plant-pathogen interface In response to apoplast colonization by bacterial the host plant secretes a broad diversity of metabolites and proteins that are presumably toxic and harmful to extracellular microbes have learned to live in this challenging environment but molecular mechanisms that these pathogens use to avoid or suppress extracellular immunity are largely unknown we demonstrate that AFM can also be used for cross-kingdom discovery screens for protein–protein interactions at the plant pathogen interface illustrated with the discovery of four pathogen-secreted inhibitors targeting a tomato-secreted immune protease P69B facilitating statistical analysis that demonstrates that AFM scores are statistically different between compatible and incompatible complexes (p = 2.1e–09 and 1.8e–9 To analyse the structures of the best models for each of these 376 protein pairs we established a custom script in Python to present the surface of the hydrolase structure in gray with the active site in red and the putative inhibitor as cartoon and lines colored using a rainbow scheme based on the plDDT scores This presentation facilitated a quick classification of how the SSP binds to the hydrolase a The 376 candidate complexes were manually screened for complexes where the SSP blocks the active site of the hydrolase with an intrinsic fold b The remaining 36 candidate complexes included 13 complexes between Kazal-like inhibitors and P69B and cystatin-like inhibitors with Pip1 and 23 candidate complexes with other candidate inhibitors transcriptome analysis of infected plants showed that genes encoding eight SSPs are not expressed at the tested conditions whereas genes encoding 11 SSPs are expressed during infection No transcriptomic datasets were available for Xp (4 SSPs) d Distribution of the final 15 candidate hydrolase-inhibitor complexes over the tested tomato pathogens and target hydrolases The 36 complexes included eight complexes of Kazal-like proteins from Pi bound to P69B, and five complexes of cystatin-like proteins from Pi bound to Pip1 (Fig. 3b) The selection of these inhibitors validated our manual screening method since these interactions could also be predicted by sequence homology No transcripts were detected for eight candidate inhibitors Although the expression of these candidates might have been missed by chosen conditions and materials these eight candidates were not analyzed further There was no expression data available for Xp infections but these four candidates were all retained No novel inhibitors were identified from pathogens Ps and Pi but some inhibitors may not have been included in our SSP selection or have been missed by AFM as false negatives putative chitinase inhibitors are also produced by bacterial pathogens is not in a complex with proteins that have structural similarity to our tomato hydrolase models our 15 SSP-hydrolase complexes uncover candidate targets of these SSPs These data confirm that all four tested candidate inhibitors indeed inhibit P69B so it is likely that XpSsp1 homologs play an active role during Xanthomonas infections although these proteins are highly conserved the residue predicted to occupy the S1 pocket is highly variant (K despite the high structural diversity of the five P69B inhibitors most inhibitors seem to occupy the S4 and S2 pockets with similar residues but the predicted residues occupying the S1 pocket can be strikingly diverse and include both basic (Lys) and acidic (Asp) residues We successfully used AFM as a discovery tool to identify cross-kingdom interactions at the plant-pathogen interface We used AFM to predict complexes between 1879 SSPs with six extracellular hydrolases and from 376 complexes with high scores we manually selected 15 putative inhibitors that block the active site with an intrinsic fold and are likely expressed during infection Four of the candidates were produced and confirmed to be P69B inhibitors This work demonstrates that the use of artificial intelligence to predict cross-kingdom protein complexes can make instrumental contributions to predicting protein functions in host-microbe interactions but these are not in complex with proteins that have structural similarity to the tomato hydrolases these AFM models correctly predicted that four of these SSPs are indeed P69B inhibitors although further assays are required for validation of the predicted structures we successfully used AFM to identify functions of four unrelated We found that the vast majority of the SSPs in AFM-predicted complexes with high scores are probably not hydrolase inhibitors rather be substrates or allosteric regulators which remains to be explored in the future we were successful with identifying inhibitor candidates because we used a stringent selection by manually screening the structures for SSPs that block the active site and have an intrinsic structure This stringent selection resulted in a high hit rate because all four tested candidates were confirmed to be P69B inhibitors In addition to previously described Kazal-like inhibitors of Phytophthora infestans we discovered four P69B inhibitors from three additional tomato pathogens: XpSsp1 from Xanthomonas perforans; CfEcp36 from Cladosporium fulvum and FoTIL and FoSix15 from Fusarium oxysporum These pathogens secrete P69B inhibitors because they are exposed to very high levels of P69B during apoplast colonization This suggests that other tomato pathogens probably also secrete P69B inhibitors that remain to be identified We may have missed some putative P69B inhibitors produced by other pathogens because they were too large (>35 kDa) were not detected in the used transcriptomic dataset It seems likely that other proclaimed chitinases may have antibacterial activities and that this is why they are targeted by bacterial inhibitors It seems likely that P69B has many additional substrates in tomato and its apoplastic pathogens our AFM screen identified 17 pathogen-produced SSPs that interact with the substrate binding groove of P69B but lack an intrinsic structure and might therefore be substrates that can be studied further These ‘ring-of-fire’ positions will likely cause differential sensitivity of the paralogs for the different pathogen-derived inhibitors This variation indicates that the P69B paralogs evolved from parallel arms races with pathogen-secreted inhibitors resulting in gene duplication and diversification in the ancestral Solanum species these observations indicate a fascinating arms race at the plant-pathogen interface These low scores indicate that AFM can produce false negatives Some of the low scores might be due to low mean non-gap MSA depth for some of the SSPs which is below the desired 100 MSA for 45% of the tested SSPs This implies that new interactions might be discovered when additional SSP sequences are added to the database screens for hydrolase inhibitors can be automated using a script that searches for residues of candidate inhibitors that are in close proximity to the active site We propose artificial intelligence to predict plant-pathogen interactions will be a revolutionary approach in future research Mean non-gap amino acid depth for chains of each protein were calculated using the features.pkl output file generated by AFM (Supplementary Data 1 outfox the peloton on a sprint stage at a Grand Tour so it was fantastic to see Taco Van Der Hoorn (what a great name by the way) of Team Intermarché - Wanty - Gobert Matériaux take a win on stage three of the 2021 Giro d’Italia To understand why events like this are so rare How can one rider overcome the collective strength of the teams working together to bring everything back together in time for a sprint finish and Van Der Hoorn expertly exploited the brinkmanship taking place between the top sprint teams It also looked very much like the peloton underestimated the Dutch rider’s strength giving him too much leeway without realising he had the firepower to make the move stick It was also an exciting victory for us tech nerds because Van Der Hoorn was using a highly optimised clothing and bike setup With a final gap of just four seconds over the chasing peloton I’d argue those optimisations were absolutely crucial to his victory that’s not to take anything away from his physical performance But racing is about combining brains and brawn and Van Der Hoorn clearly has both in spades The rider is the biggest cause of aerodynamic drag in the rider plus bike system, accounting for around 80 per cent of the total Anything you can do to improve the way the air flows over your body can make a significant difference to how much power is required to ride at a given speed you’ll be able to ride faster for the same effort Van Der Hoorn has clearly done his research, because his position of riding in a tucked down position, with his hands on the hoods and forearms parallel to the ground, is one of the fastest positions available on a road bike When we tested this position in a velodrome we found it saved a whopping 94 watts when riding at 45kph versus riding with straight arms on the hoods #AeroGainz don’t come much bigger than that is also something we’re increasingly seeing throughout pro cycling You can see how much more narrow Van Der Hoorn’s arms are compared to his breakaway companions and narrow equals fast as it reduces your frontal aera Part two of optimising the aerodynamics of your body is aero clothing and kit Van Der Hoorn has been meticulous in his clothing choices with both halves of the garment sewn together to reduce drag It also uses fabrics designed to be skin tight and aerodynamically efficient at race speeds On top of this, Van Der Hoorn was also using aero overshoes like those we would normally see used for time trials he was wearing arm warmers made out of an aerodynamic fabric too When it comes to saving energy for that big attack every little helps and it starts right from kilometre 0 On top of all this, Van Der Hoorn was wearing an aero road helmet and he’d even taken the time to tape over the vents on the front While an aero road helmet is a decent drag-reducing tactic (the difference between a Specialized Prevail and a Specialized Evade is apparently around 11 watts at 45kph) the latter detail may be a bit more hit and miss Continuing on the ‘aero is everything’ theme, Van Der Hoorn was also riding a full-on aero road bike, the Cube Litening C:68X, which is said to reduce aerodynamic drag by 30 per cent compared to the previous, non-aero-optimised Cube Litening fully hidden cables and a slippery integrated handlebar and stem Though the body is the largest contributor to aerodynamic drag the bike accounts for around 20 per cent of total drag which apparently equates to about 3 seconds per kilometre product manager for bicycle tyres at Continental Van Der Hoorn's team performed its own testing and found a 12 watt difference at 45kph between the GP5000 TL tubeless tyres and the Continental Competition tubulars they had been using previously Departments The DCS cohort is an on-going three others but disappeared around 8:30 p.m The other three were rescued from the water Another person is missing in the Markermeer after a boat capsized between Hoorn and Edam The emergency services responded to the Zuidelijk Havenhoofd with helicopters and boats after the report of the swimmers in trouble. They called off the search for the last missing swimmer when it got too dark to see, said the local security office, Haaglanden Veilig An eyewitness saw rescue workers pull two “boys” out of the water. “They both had legs covered in blood. There was also a mother there who was panicking and screaming,” the witness told Omroep West. According to them, the boys tried to save the missing swimmer but couldn’t get close. “The water was so wild, really impassable. Those boys barely survived.” The rescued swimmers were taken to the hospital. Their current condition is unknown. Another person is missing in the Markermeer after a boat capsized between Hoorn and Edam at around 5:15 p.m. on Sunday, a spokesperson for the police confirmed to ANP. The boat had four people on board, three of whom ended up in the water. One managed to get back on board under their own power, and the rescue society KNRM rescued the second. The coast guard launched a large search for the missing person but had to call it off without success when it became too dark to see. De Hoorn was where the first Stella Artois was brewed in 1926. There will be parties on Monday and Tuesday evening and guided factory tours for 1,600 people on Wednesday and Thursday. On Thursday evening, 100 residents of Leuven will be served a "beer pairing" dinner in the old brewery. The week will culminate in a party for 800 people on Friday. To celebrate this milestone, parent company AB InBev is launching special editions of Stella Artois cans, and the drink will be servied in vintage glasses in more than 1,000 bars and cafés throughout Belgium. "We wanted to pay tribute to the packaging of yesteryear, including the original logo from 1964 and the typical Stella Artois colours: red, white and gold,” says Olivier Maillet, director of the
