Recent research into the role of the caspase-2 enzyme, co-authored by Associate Professor Marcin Poręba, PhD, Dsc, Eng from the Faculty of Chemistry, showed that inhibition of its enzymatic activity leads to preventing the growth of cancer cells and, consequently, their death. The results have just been published in the journal “Science Advances“
Prof. Marcin Poręba, who works at the Department of Biological Chemistry and Bioimaging of the Faculty of Chemistry and also holds the position of Vice-Dean for General Affairs at the Faculty of Medicine, is researching the role of caspase-2 in acute myeloid leukaemia in collaboration with the team headed by Lisa Bouchier-Hayes, PhD from the Children's Hospital in Houston, Texas (USA)
Their joint work resulted in the publication entitled “Caspase-2 is essential for proliferation and self- renewal of nucleophosmin-mutated acute myeloid leukemia”
The authors examined the role of caspase-2 in the development of acute myeloid leukaemia (AML) connected with nucleophosmin (NPM1) mutations
“The NPM1 mutation is a protein connected with the nucleus of the cell
and its mutations lead to the protein being displaced into the cytoplasm (NPM1c+)
Marcin Poręba and adds that although these are the most common mutations driving the development of AML
the mechanisms behind their action are not fully understood
Poręba and Dr Bouchier-Hayes has shed new light on the role of caspase-2 in NPM1-mutated myeloid leukaemia
“This enzyme is crucial not only for apoptotic processes but also for the regulation of the cell cycle and the proliferation of cancer cells,” says the scientist from Wrocław University of Science and Technology
“We’ve shown that inhibition of the enzymatic activity of caspase-2 leads to inhibition of cancerous cells’ growth and subsequent death
Our researcher’s role in the project was to develop new caspase-2 inhibitors that
may help develop new therapies to enhance the treatment of this cancer
The first steps in this direction were already taken in July 2024
at which time the Polish-American team submitted a joint grant R01 “Regulation of nucleophosmin mutated AML self-renewal by caspase-2” to the largest US research agency
The task of the group working at Wrocław University of Science and Technology
will be to develop caspase-2 inhibitors that can be used in preclinical studies in mice with acute myeloid leukaemia
“Caspases are a group of enzymes that play a key role in the process of programmed cell death,” explains Prof
“They are activated in response to signals indicating cell damage or stress
The first includes apoptotic caspases that initiate and carry out the process of apoptosis
which is immunologically silent and doesn’t cause an inflammatory response
resulting in leakage of cell contents to the outside and inflammation
“The role of caspases in cell death is well known
few people know that caspases are also involved in cell proliferation

CHARLOTTE, NC (Monday, December 19, 2022) – Charlotte FC today announced the Club has signed midfielder David Poreba to an MLS NEXT Pro contract on loan from Polish top flight club Stal Mielec. The Club retains an option for a permanent transfer following the 2023 season.
The American and Polish dual national becomes the first signing for Charlotte FC’s MLS NEXT Pro team which begins play in 2023.
Poreba joined Stal Mielec from Chicago FC in 2021 and has appeared in two matches in Poland’s top professional league, the Ekstraklasa.
“David is a talented player who has shown his potential at the professional level throughout the Stal Mielec system. He is the exact type of player we are passionate about developing and we’re delighted to sign the Club’s first-ever player,” said Charlotte FC’s MLS NEXT Pro President Darrius Barnes. “2023 is a groundbreaking year for player development and player progression at Charlotte FC and we look forward to continuing to build out our MLS NEXT Pro roster this winter.”
During his time with Stal Mielec, Poreba made two additional professional appearances in the Fortuna Polish Cup.

CHARLOTTE, NC (Wednesday, May 17, 2023) – Charlotte FC today announced the Club has signed Crown Legacy FC midfielder and captain David Poreba to a Short-Term Loan Agreement ahead of tonight’s MLS match versus Chicago Fire FC. Poreba will be available for the match against Chicago.
Poreba has seven starts in eight appearances for undefeated Crown Legacy FC in MLS NEXT Pro. The Polish-American midfielder has been a key piece for Head Coach Jose Tavares’ side, chipping in two goals and three assists so far this season.
Per MLS rules, a club may sign a player from its MLS NEXT Pro team to a maximum of four Short-Term Loan Agreements each season. An individual player may be included on up to four MLS league season match rosters, but they may appear in no more than two MLS matches. An individual player may appear in any number of non-league matches during the terms of his four Short-Term Loan Agreements.
This is the first Short-Term Loan Agreement of 2023 that Poreba has signed with Charlotte FC.

Midfielder Lukasz Poreba speaks to HSV.de about his second year with the club, his style of play, important moments in his career and his past as a prolific winger.
HSV.de: Lukasz, it’s now day three of pre-season. How does it feel to be back out there?
Lukasz Poreba: I’m pleased to be back. We had a long break and we’re now all motivated to get back to work. We’ve got quite a few training sessions planned. It’s tiring stuff, but that’s absolutely fine. We need all this training to prepare well for the new season.
HSV signed you on a permanent basis from RC Lens following the conclusion of last season. How happy are you to be staying here in Hamburg after your loan spell?
I’m really happy about it. I tried my best to get this contract last season. It wasn’t an easy year for me, particularly in the first half when I had a few injury problems and didn’t play much. Things went a lot better after the winter break, though I still wasn’t able to fully show what I’m capable of. That’s my aim now for the new campaign.
You joined on loan really late on in pre-season last year. How different does your situation feel now?
It’s a lot easier for me because I know the coach, my teammates and the staff a lot better. They also know what I’m like as well. As a result of that, I’m a lot more comfortable. It’s generally tough to settle into a club and a team when you’ve been at several clubs in a short period of time, as was the case with me.
The language can also play an important role. How is your German?
I understand lot, maybe around 50-60% of what people say. I’m also becoming more confident when speaking it too. I’ve set myself the aim of putting more energy into learning the language this year. It’s an important thing when it comes to integrating, otherwise there will always be a barrier there.
Your game speaks for itself on the pitch. You always look to be in control and play with real clarity. Despite your age, you come across as a calm and confident player. Where has this style come from?
I changed my style of play a little bit when I started playing adult football. I was a bit hectic as a youngster and I’ve grown up in this regard. I try to keep things simple, only take two touches and keep my position in midfield. I can control my emotions during games, I’m always fully focused. That’s my job. Whether I score a goal, make a tackle or misplace a pass, I simply try to keep my emotions in check. This is the mindset I always have.
What would have to happen for you to get angry?
I’m a calm guy on the pitch, but I’m completely different off it. (laughs) Everyone that knows me is familiar with how crazy I can be, and that I can have some silly ideas. The language barrier is obviously something that often stops me showing this side of myself. I’m full of energy!
During competitive things. I’m somebody that always wants to win. My friends, my little brother and my girlfriend could back that up. It doesn’t matter what I’m playing, I always take it very seriously and want to win.
You are only 24 years old, yet you’ve played in three different countries and leagues already. If you take a look back at your still short career, what were the key moments that helped you become the player you are today?
What have you learnt about HSV after a year at the club?
To be honest, I found out that HSV is a really big club in Germany when I was young, but the size and stature of the club still surprised me somewhat. The standards here are really high in every regard. When you play for HSV, you come up against opponents that give 200% against you, which is a really useful experience and definitely helps you. You obviously have to learn how to deal with pressure too, but regularly playing in front of 50,000 of your own fans is a special feeling.
You also scored a few goals in the second half of last season. Even though you’re a natural defensive midfielder, you were deployed slightly higher up the pitch during this period. What are your thoughts on this role?
I’ve mainly played as a number 6, alongside another one, or as a deeper number 8 in recent years. The coach used me in a more attacking role and I showed that I can score goals and create chances from this position. Playing as a number 8 is a bit more challenging for me, but I’m more confident in the role the more I train and play there. I ultimately just want to play as much football as possible and help us to achieve our main goal.
Final question: have you always been a defensive midfielder or did you play in a different position when you were younger?
I was actually a right winger as a kid. I used to love dribbling on the wing and I scored lots of goals too. “Dribble and score” was my motto in the 5v5 games back then. I moved into midfield in my youth and then at the start of my professional career, and I’ve played in all positions since in the heart of the midfield. I don’t dribble like I used to! (laughs)
Charges of second-degree murder in the death of Russell Hitchon were suddenly withdrawn last week against Rorey Grant Tyler Hill just weeks after he had been ordered to stand trial in the man’s death.
The surprising move by assistant Crown attorney Sean Bradley, was handled over a Zoom link in Superior Court where Hill appeared on screen from Maplehurst Correctional Centre.
Hill, 40, and Jessica Elizabeth Poreba, 43, had each been facing charges of first-degree murder in connection with the death of Hitchon, who was found dead in his Colborne Street apartment on July 22, 2022.
Last month, after a preliminary hearing, Justice Colette Good discharged Hill of first-degree murder but ordered that he stand trial for second-degree murder.
The judge said there wasn’t enough evidence presented during the hearing to commit Poreba to trial and said she was free to go.
In a long statement, explaining why he was requesting Hill’s charges also be withdrawn, Bradley said on June 20, there wasn’t enough evidence to proceed.
“While we continue to believe Mr. Hill committed this crime, the Crown can no longer say it can be proved beyond a reasonable doubt,” said Bradley.
“With no reasonable prospect of conviction, we are therefore duty-bound to withdraw this charge.”
During a preliminary hearing, evidence was presented that Hitchon, Poreba and Hill were roommates, drug users and dealers.
Hitchon died after suffering multiple blunt force injuries to his head, face and neck, was stomped on and possibly strangled.
His lungs were pierced and his body was lacking about a third of his blood.
A friend finally found Hitchon, bloated and cold, lying on his bed, with footprints on his body.
In his comments to the judge, Bradley said the Crown’s office has to constantly assess a case to see if there is a reasonable prospect of a conviction at the end of it.
He said he “digested” Good’s ruling and her withdrawal of the charges against Poreba after the lengthy preliminary hearing.
As well, he reviewed transcripts and all the evidence available, and assessed the credibility of witnesses before making a decision.
Bradley said there were “inconsistent statements” at the hearing and the case wasn’t likely to get any stronger.
“An objective assessment leads to the conclusion we would not be successful (in getting a conviction.)”
Bradley noted that Hill, who was arrested in August 2022, chose not to apply for bail.
In fact, when the matter was scheduled to be dealt with a week earlier so that Hill would be released, he asked that the matter be delayed for a week.

A 40-year-old man has been ordered to stand trial for second-degree murder in the death of a 68-year-old Brantford man in 2022.
Rorey Grant Tyler Hill and Jessica Elizabeth Poreba, 43, had been facing charges of first-degree murder.
During a judgment on committal in Brantford’s Ontario Court of Justice on Tuesday, Justice Colette Good discharged Hill of first-degree murder but ordered him to stand trial for second-degree murder in the death of Russell Hitchon, who was found dead in his Colborne Street apartment on July 22, 2022.
In her lengthy judgment, Good said there wasn’t enough evidence presented in a preliminary hearing to have Hill’s co-accused, Poreba, stand trial and told her she was free to go.
Hill, Poreba and Hitchon knew each other.
Hill will appear in assignment court on May 30 before his trial. No date has yet been set for the trial.

Another MEP's undeclared Azerbaijan trip is helping to build pressure for reform in the EU Parliament

So said Tomasz Poręba from Poland's ruling Law and Justice party on Azeri state media in Baku last July
Europe and Azerbaijan in an interview entitled 'Poland needs gas'
which Azerbaijan wants to sell to the EU in larger quantities
Poręba was there as an MEP and as a member of the EU Parliament's transport committee (Tran)
according to an official report by Azerbaijan's parliament
and multiple Azeri-government media stories at the time
He also visited the war-torn Nagorno-Karabakh region
where senior EU diplomats fear to tread in case it inflames tensions with Armenia
But if he was representing the European Parliament
Poręba didn't bother to declare the visit in his EU Parliament register
And the Tran committee has no record of him going there on its behalf
in what amounts to deja vu on three other MEPs
When Swedish investigative news site Blankspot contacted Poręba
his Belgian law firm replied that Poręba wasn't in Azerbaijan in his capacity as an MEP
Poręba was accompanied by New Direction's executive director Witold de Chevilly
And if Azeri media claimed Poręba had represented anyone other than New Direction that was simply the journalists' mistake
But even if New Direction did pay for everything
the MEP should still have declared this as a gift worth over €150 in his EU Parliament registry — which he didn't
And if Poręba's trip was nothing to do with his MEP status
then Azerbaijan's EU embassy told a contradictory story
Poręba and the three other MEPs who visited Azerbaijan last year (Latvian socialist Anders Ameriks
and German liberal Engin Eroglu) were all invited by the \"EU-Azerbaijan Parliamentary Cooperation Committee in the Parliament of Azerbaijan\"
helped organise their trips as part of his normal duties
some of the MEPs later admitted Azerbaijan had paid for their hotels in Baku and for excursions to the mountainous and remote Nagorno-Karabakh region
nor other members of the European Parliament received 'any free hospitality'
If you are referring to small gestures such as offering tea
hotels are covered by the visiting guests themselves,\" it said
There is no suggestion any of the four MEPs broke laws in their dealings with the petro-dictatorship
But Azerbaijan has a track record of EU disinformation as well as so-called 'caviar diplomacy' — lavish hospitality for foreign VIPs
The way Poręba and others rode roughshod over EU Parliament transparency rules also makes Brussels look bad
in the wake of the Qatargate bribery scandal
One fellow MEP, German Green Niklas Nienaß, resigned from a parliament cross-party group
following earlier revelations of their freewheeling Azeri diplomacy
And this investigation sheds light on how foreign regimes make friends and influence people in the EU power-bubble
the 49-year old Pole didn't start out as Azerbaijan's friend
Baku declared him persona non grata when he travelled to Nagorno-Karabakh in 2010 — long before Azerbaijan reconquered parts of the region from Karabakh-Armenian forces in the Second Nagorno-Karabakh War in 2020
Poręba went there as an EU Parliament rapporteur on Armenia
he signed a letter declaring full respect for Azerbaijan's territorial integrity and his visa-ban was lifted
His trip back to Azerbaijan in July 2022 was his first one since then
He didn't give a reason for the U-turn in his sympathies
Poręba has worked hard to tie Europe and Poland closer to Azerbaijan
He submitted a proposal to link European transport corridors to Azerbaijan via Turkey last November
He convened a special meeting with Azerbaijan's transport minister about the corridors in December
And he discussed the same topic with a think-tank belonging to Tural Ganyaliyev
have even advocated for the opening of the Zangezur Corridor — a highly sensitive route
whose status is still being negotiated by Armenia and Azerbaijan and which remains closed for now
And Azeri media continue to report that Poręba hosts his meetings as a Tran committee member
even as Tran says it has no record of him representing them at such events
Poręba's engagement goes way beyond any EU mandate he might have
When Azerbaijan attacked Armenia in September 2022
Poręba took to the airwaves to denounce Armenia as a close ally of Russia
he also gave a public statement on the quality of an Azerbaijani football club
while adding that he had helped two Polish coaches to get an internship with the team

After approaching the Walton County Board of Commissioners in June 2023 with her vision
Poreba went about making her dream a reality with the assistance of some other dedicated animal advocates in the community

“We are putting the finishing touches on the shelter facility now and working through the process to receive our shelter license from the Georgia Department of Agriculture
We expect to be able to start taking in animals early November and plan to be open to the public mid-November,” she said
“I feel a mix of excitement and gratitude as we near the realization of this dream

the facility will have 20 indoor kennels for dogs
eight outdoor kennels for dogs and two large play yards.
the dogs will be taken in rotation to the outdoor kennels to spend some time in the fresh air and others will be taken to the play yards to be socialized and trained to help encourage their adoption
there also will be walking trails and volunteers will be able to take the dogs in the facility for walks around the property
Poreba said there also are plans to expand on the current building
“We will have some basic medications and vaccines on site
but sterilization surgery and more complex medical needs will be handled at our partner vets off-site,” Poreba said
“We will be serving only Walton County animals at this facility.”
Poreba said she has learned that cats do much better when they are not in cages so they will be kept in the large room where they plan to have runners along the wall for the cats to climb on
There also will be a window perch for them to be able to get a chance to look outside
recently commented on the situation regarding cats and how difficult it is for the shelter to contend with the number of cats that end up there
He said that the county is overrun with feral and stray cats and consequently many of them end up at the shelter
“We respond to calls and trap feral and stray cats
Many feral cats are not socialized to live with people
We take in 100 to 250 cats per month,” he said
Poreba is hoping to be able to really make a difference with the Walton County Humane Society.
“We won’t have any specific time limits for animals awaiting adoptions,” Poreba said
“We will keep each animal until they are adopted or in rare circumstances
until they become too behaviorally or medically unsound for adoption. There are a lot of very involved rescue organizations and individuals in the community
We don’t anticipate the need to send animals from our facility to other rescue organizations
but we do hope to be able to support and build up relationships with other organizations that are also supporting the Walton County Animal Shelter.”

“We currently have a full time executive director and are hiring a full time shelter manager
We are currently evaluating additional staffing needs but anticipate hav- ing a robust volunteer program,” Poreba said
They have already been contacted by Eagle Scout programs with offers to do projects at the shelter to earn merit badges
And we would like volunteers to manage off-site events
We welcome volunteers with all levels of experience working in shelters and with animals
“While our mission is about supporting the animals in the community
we really look forward to enhancing the human-animal bond and supporting people in the com- munity as well by making adoption and spay/neuter easy and accessible to all residents.”
