This article is also available in Italian / Questo articolo è disponibile anche in italiano the European Commission unveiled its Vision for Agriculture and Food a document outlining a roadmap for the future of Europe’s agri-food sector and supply chain The strategy aims to bolster the sector’s competitiveness and resilience by prioritising simplification and digitalisation to enhance efficiency across the entire production chain the document appears to present internal contradictions and lacks clarity on the role of farmers in this transformation there is no clear vision on how these principles will be put into practice We discussed these topics with Cristina Guarda who is deeply engaged in food system policies What is your overall opinion of the EU Vision on European food systems presented by the Committee on Agriculture The Vision (here you can find the document editor’s note) presents a sharply focused analysis of the current state of agriculture—a sector characterised by an ageing workforce and by uncertain incomes which are significantly below both the European average and those of other economic sectors A field in which there is a lack of services in rural areas together with a pronounced gender gap that represents an additional challenge through the analysis of environmental and climate impact on agriculture how much production capacity depends on these factors it presents a highly lucid picture of the sector’s current problems Rather than addressing these challenges in line with the Farm to Fork strategy—which aimed to strengthen farmers’ roles within the food supply chain through progressive measures—we are witnessing a shift in direction Agriculture Commissioner Christophe Hansen appears to be steering policy back towards a more conservative approach focused solely on competitiveness and attractiveness of the sector accompanied by popular terms like simplification and innovation Could you explain this last statement in more detail The document makes no concrete reference to what is actually meant by simplification It provides no practical examples of which bureaucratic aspects Europe intends to remove through this political strategy Nor is it clear whether it is about deregulation or real it is evident that this would do nothing to ease the administrative burden on farmers Another issue is that much of this bureaucracy is not imposed by Europe but stems from national strategic plans There seems to be an almost ideological stance as if technological advancement alone can provide all the answers It paints a future where technology will miraculously solve every problem—advanced systems to capture emissions assisted evolution technologies (AET) to create ultra-drought-resistant seeds these practices are not yet a viable option no one is investing in research areas that could make a difference: improving seeds for organic farming developing agroecological practices to reduce water consumption and generating ecosystem services essential for food production The Vision also fails to acknowledge those exploring new economic distribution models based on short supply chains High-performing research and innovation networks already exist built on democratic research and the direct involvement of local communities in experimentation Yet there is no mention of any of this in the document the risk remains the same: promoting costly technological solutions which inevitably lead to small and medium-sized farmers taking on debt as they often don't have the resources to afford them What are the main proposals that the Greens/EFA group is promoting we have outlined key priorities for our vision of the future CAP It is clear to us that payments should be tied to job creation rather than simply distributed based on cultivated land That is why we propose capping payments at €60,000 per farming business and redistributing the remaining CAP funds to those who need them most—new and young farmers and funding supporting the transition to climate- and biodiversity-friendly agriculture Every farmer should have the right to a fair wage we want to make economic eco-schemes both more attractive and more rewarding We know very well that farmers who adopt agroecological practices require more time and space for production they provide an ecosystem service that benefits the entire community and all economic sectors The principle we propose is simple: those who produce ecosystem services must be paid for their work Farm businesses must be assessed as a whole with financial recognition for all measures taken to protect the environment This would mean creating a new and stable income stream What are the other elements of your proposal We have a dependency problem from other countries: we import chemical fertilisers The Commission’s Vision suggests addressing this by boosting domestic production promoting more extensive livestock farming and tackling livestock emissions through the previously mentioned “innovations” You cannot talk about extensive farming without reducing the number of animals reared A more sustainable rearing strategy must go hand in hand with a structural review of the sector This is why we advocate for a genuine strategy on protein diversification Encouraging more sustainable livestock systems is not enough—we must also ensure equal support and promotion for plant-based and alternative protein sources But this transition cannot happen without engaging citizens and policymakers cannot afford to ignore it Yet these two aspects are entirely absent from the current Vision despite being well addressed in the Farm to Fork strategy We propose clear and transparent labelling that provides information on environmental impact and animal welfare standards—ensuring consumers can make informed choices The underlying problem in this Vision is that it speaks of resilient production systems while simultaneously focusing strongly on exports That is why we ask that the focus must first be European we oppose the Mercosur agreement because we support a model that valorises regional markets and strengthens short supply chains rather than one built on global competition We advocate for alternative partnerships that are more sustainable and resilient per- and polyfluoroalkyl substances: how important is it today to fully understand the real distribution of PFAS and to start remediation interventions to deal with a situation that is now critical in some areas There are two key issues that must be tackled we need to stop the production and release of PFAS which requires investment in research to develop viable alternatives we must find ways to eliminate the PFAS already present in the environment Halting production alone will not solve the problem—we also need to understand how to destroy them effectively and safely We already know how to capture PFAS using activated carbon But the real challenge lies in destruction Current methods—including microwaves and high-temperature thermal processing—are extremely expensive and often generate even smaller Serious investment in research is essential to develop effective PFAS destruction methods without creating new risks We must also work to prevent companies from continuing to release them into the environment—for instance by promoting closed-loop systems that stop contaminated wastewater from reaching rivers and aquifers farmers in affected areas also need support in adopting cultivation techniques that prevent plants from absorbing PFAS from the soil This is not just a water pollution issue—the contamination of soil has major implications for agricultural production Another crucial step is banning PFAS in industries where safer alternatives already exist there is no justification—replacement solutions for PFAS are available and manufacturers must be pushed to adopt them without further delay The path forward is clear: stop production and release This leave to chance; if you lose access to these “private keys,” you may never get your cryptocurrency back The first question when choosing a crypto wallet is whether you want a hot wallet or a cold wallet The main difference between a hot wallet and a cold wallet is that hot wallets are connected to the internet while cold wallets are not Other questions to ask when comparing wallets include: How many types of digital assets are supported, how easy is it to move crypto offline, into a cold wallet, and are there resources for in-app staking or rewards programs offering add-on services such as trading or staking in exchange for fees A hot wallet makes it relatively easy to carry out transactions using crypto but it may be more vulnerable to hackers who could theoretically reach your crypto over the internet Here are NerdWallet’s top picks for hot crypto wallets Zengo has emerged as a next-generation wallet that claims to build on the technology of previous wallets Zengo uses a form of cryptography known as multi-party computation Zengo claims this is the first instance of a consumer wallet (as opposed to an institution) using MPC Seed phrases and private keys have been at the heart of self-custody in crypto since the beginning but Zengo aims to enhance security while doing away with the issues that come from managing your own keys and seed phrases no Zengo wallets have been hacked or stolen We look at more than 15 factors to evaluate crypto wallets Crypto.com Onchain (formerly Crypto.com DeFi Wallet) is a wallet created by a company mostly known for its crypto exchanges. It’s specifically tailored for use in what is known as decentralized finance (DeFi) which is a sector of the crypto world focused on lending saving and other financial products that don’t rely on a central broker Users of Crypto.com Onchain can use their crypto holdings to interact with DeFi products both on their mobile app and in a browser extension It also has a desktop app that integrates with Ledger hardware wallets Crypto.com Onchain emerged as a winner here thanks to the wallet's connection to the main Crypto.com app This makes it extremely simple to use Crypto.com's well-reviewed centralized trading app then transfer assets to your own self-custody wallet there is a 0.0008 BTC withdrawal minimum (a little more than $60 at the time of last update) So you'd need more than $60 worth of BTC to instantly transfer it to your Onchain wallet all-purpose crypto wallet whose users can access their crypto via mobile Users of Guarda can move crypto into cold storage through an integration with the hardware wallet Ledger Guarda also has staking programs available Exodus offers a solid set of software tools trade or stake cryptocurrency directly from their wallets and it has an integration with the Trezor cold wallet intended to help people easily move crypto from hot to cold storage also has a solid library of explanatory content for people who are learning about crypto Assets supported: More than 10 million digital assets and over 100 blockchains Trust Wallet also does not provide a full list of supported cryptocurrencies on its website it's likely the wallet will be able to support just about any cryptocurrency you come across The company operates the largest U.S.-based crypto exchange But Coinbase Wallet is a separate product that allows users to store cryptocurrency themselves rather than keeping it in the custody of Coinbase Because the Coinbase Wallet was created by Coinbase it integrates easily with its company’s exchange and may be a good introduction for users who haven’t used a non-custodial wallet before The free application has some other strengths such as mobile and browser-based connections to decentralized applications It also lacks some functionality offered by competitors though its browser extension connects to the Ledger hardware wallet with the option to manually add unsupported ERC-20 assets If you’re a regular user of applications on the Ethereum blockchain MetaMask can store any digital asset that’s built on Ethereum MetaMask also integrates with many of the “Web3” applications that exist on the Internet and require crypto transactions to work MetaMask has mobile and browser-based wallets It also offers staking and DeFi apps through its MetaMask Portfolio feature and integrations with multiple hardware wallets including Ledger and Trezor Just remember that if you’re not a big user of Ethereum and its related tokens There is a large selection of hot wallets on the market, and most of them can support hundreds or even thousands of cryptocurrencies. They also generally can hold at least some types of NFTs, or non-fungible tokens and many connect directly to exchanges where you can buy or sell crypto Cold wallets are usually gadgets that are disconnected from the internet that help you store sensitive crypto information because you have to buy a piece of actual hardware that’s set up to store your crypto Because they are not connected to the internet they may be harder for other users to reach hardware wallets come preloaded with software and other usability and security features that make the process smoother you may want to consider details such as price and security measures you should check to make sure the hardware you’re considering will work with your software wallet Here are NerdWallet’s top picks for cold crypto wallets Ledger is one of the best-known names in crypto wallets and Ledger can integrate with many popular software wallets such as Crypto.com and Guarda plus a dedicated desktop app and a browser extension Ledger also has two-factor authentication available and a library of educational content for users » You can buy Ledger wallets directly from the Ledger shop here Trezor offers some of the highest-end hardware available in crypto storage While its entry-level model (the Tezor Model One) costs around $49 it also has a more expensive model (price at last update was $169) with a touch screen Trezor has integrations with other crypto firms such as Exodus though it also provides built-in services such as staking and crypto purchases through its software products as well as a mobile app (Trezor Suite Lite) for Apple and Android One slight inconvenience: Trezor wallets do not have Bluetooth capability although the company notes that this is a deliberate decision meant to reduce the wallet's vulnerability to cyberattacks » You can shop all of Trezor's products in the company's official store here Non-custodial crypto wallets are the type of storage option preferred by many crypto enthusiasts because they place you in control of your own private data. Unlike when you keep assets on a cryptocurrency exchange you don’t have to trust a third party to secure your private keys That’s not to say non-custodial wallets are without risk They are less forgiving of errors such as lost passwords and you have to trust that your wallet’s hardware and software will work as intended And moving assets between wallets regularly can become costly because of transaction fees imposed by underlying blockchain networks For these reasons some crypto users have multiple types of wallets: some for long-term safekeeping and others for active trading One way to choose a wallet is by looking at the website of the cryptocurrency you’re hoping to store. There are many single-purpose solutions built for individual cryptocurrencies. For example, MetaMask is often considered a top Ethereum wallet because it focuses solely on ERC-20 tokens Electrum is among the top Bitcoin wallets because it's built specifically for Bitcoin power users But if you’re planning to protect multiple kinds of cryptocurrencies in one place » Not ready for a wallet? Here are the best crypto exchanges and platforms You can also keep your crypto on a cryptocurrency exchange but then you're trusting a third party to secure your assets allow you to buy crypto through accounts with them as well Bitcoin ETFs and Ethereum ETFs are also an option for 401(k) investors or others who can't invest directly in crypto — but these involve trusting someone else to safeguard your crypto investment If you want total control of your crypto assets Hot crypto wallets generally offer an account recovery process if you forget your username or password although the terms of that recovery process may vary between providers If you physically lose a cold crypto wallet, or it breaks, don't despair — the crypto isn't actually gone; cold wallets just store private key information needed to access or move your crypto. The cold wallets we review offer users the ability to regenerate their wallets using a seed recovery phrase in the event of loss That depends on the broker. Some, like Robinhood, do work with external wallets, while others do not. firsts for Green MEP Cristina Guarda Not only is this the Italian lawmaker’s first term in the European Parliament but she also welcomed her first baby at the end of September.  But her own welcome into the world of EU politics has been less than warm.  the MEP has worked remotely in the last weeks of her pregnancy Home-office was working well for Guarda — that is until she requested to join the upcoming commissioner hearings remotely.  The hearings give MEPs the chance to grill and ultimately vote on the commissioner-designates the only Green who sits on both the agriculture and regional committees next week’s hearing gives her the chance to directly quiz hard-right candidate Raffaele Fitto who will oversee both agrifood and rural affairs in his role as executive vice president of cohesion and reforms “I’m not even asking to vote — I’m just asking to speak to put my question to him in my own words,” the MEP told EUobserver Her request to speak during next Tuesday’s hearing was denied in late October by a conservative majority in the governing body of the European Parliament – the Conference of Presidents (CoP) The coalition consisted of four men from the European People’s Party (EPP) European Conservatives and Reformists (ECR) Patriots for Europe (PFE) and the Europe of Sovereign Nations (ESN) four women from progressive forces backed Guarda’s demand an EPP spokesperson group said allowing such a move could open the door to “all kinds of requests to intervene online” from MEPs in situations such as sick leave or travelling and for other meetings or even during plenary sessions a spokesperson from the European Conservatives and Reformists (ECR)  group agreed on the need to address the issue long-term given the “time constraints” the group encouraged Guarda to put her question in writing and have it read out by a colleague from her group which is the currently proposed compromise agreement.  But this is not an acceptable solution for the Green MEP “It's frustrating to see someone else do your job and [ask] your question without your background and your knowledge,” she said But Guarda sees the move as an attempt from conservative men to keep women 'in their place' “My message is that a woman should decide for herself how much time she spends with the baby and how much time she spends working,” she said adding that it “helps her mental health” to continue being able to continue participating in meetings.  The situation is familiar to parliament president Robert Metsola was also unable to vote on key decisions after having her fourth child back in 2016 This included voting on the EPP group’s election of Antonio Tajani as nominee for the parliament president or any of the December 2016 plenary votes Attributing the slow progress on parental rights to “technical and political” reasons an official in Metsola’s office said she “remains committed” to finding a way forward that is “legally sound and which does not put unnecessary pressure on women who are on maternity leave” Her office said she has in recent years approached various parliamentary technical groups and relevant services “multiple times” to explore options — but that experts warned this raises a “number of technical Her team also said that Metsola is committed to keeping this issue high on the political agenda sees “no legal or technical argument” to block Guarda’s participation pointing out that all the necessary technical infrastructure was put in place both for meetings and votes during the Covid pandemic Reintke — who put Guarda’s request to the CoP meeting —  described the parliament’s decision as proof of as “lack of political will” This is “striking” given that the groups that opposed the move position themselves as 'pro-family and pro-children' yet “chose to deny a young mother’s chance to participate,” she added.  who chairs the Parliament’s FEMM committee “Being a mother or father should not prevent MEPs from fulfilling our obligations and represent the citizens who elected us,” Galvez added.  Together with group leaders from the Socialists have written a statement in an effort to reverse the parliament's decision “We believe that this refusal impedes the rights of an MEP and is a discriminatory limitation to the work and political mandate of an elected member not being able to travel after having had a baby but who wants to undertake her work voluntarily,” the statement reads.  the move should also be a “wake-up call” for the parliament to re-think its approach to parents and carers. “This is not the 1980s anymore – we have the digital means to make this possible and we should do all we can to make the parliament accessible,” she also said Natasha Foote is a Brussels-based freelance journalist 0 decision taken in the Conference of Presidents last 24 October 2024 to refuse the request of our fellow MEP Cristina Guarda to participate remotely in the Hearing of Commissioner-designate We believe that this refusal impedes the rights of a Member of the European Parliament and is a discriminatory limitation to the work and political mandate of an elected Member not being able to travel after having had a baby but who wants to undertake her work voluntarily We would like to stress that the request of Cristina Guarda only refers to her intention to pose an oral question to a Commissioner-designate Given the specific circumstances of the case we believe there should be no technical nor political barrier for Cristina Guarda to speak remotely for one minute and represent herself in a crucial political meeting as a Hearing of a Commissioner We also wish to stress that such derogatory decisions are taken on a regular basis at Coordinators level for Committees without undermining the right to maternity leave We believe that allowing her participation to this meeting is essential to ensure that constituents are not unrepresented while their Member is on leave or pregnant if she is able and wants to continue performing her role Exercising one's mandate and representative role is not only the right of the elected person but it is also essential to guarantee the effective participation of citizens in the EU’s democratic life call on the President to find a way to allow Ms Cristina Guarda exercise her right and mandate and be able to participate remotely and formulate a question to Commissioner-designate The European Parliament in its Gender Action Plan adopted in 2020 commits as a gender sensitive parliament to provide equal access and participation for all genders and to work for improved work-life balance conditions and better accommodate family commitments We ask therefore on the President to live up to this commitment We expect a close attention to this urgent matter and we look forward to any workable solution Guarda do Embaú is home to a world class wave that breaks consistently year-round and is known as one of the best surf spots in Il concerto è stato registrato all’iconico Roxy Theatre sul Sunset Strip in occasione del release show del loro ultimo album “Dark Parade“ Quella sera i Cirith Ungol hanno eseguito il disco per intero più alcuni classici come ‘Join The Legion’ ‘Black Machine’ e ‘Atom Smasher’ Di seguito la prima anteprima del DVD: il video di ‘Down Below’ One of the world’s most iconic music venues – the Sunset Strip’s Roxy Theatre – hosted a legendary 2024 gig by one of the world’s most iconic metal bands The one-night-only performance was an album release show for the band’s 2023 Dark Parade LP It captured the lineup at the height of its considerable powers along with a set of classic songs including “Join The Legion,” “Frost And Fire,” “Black Machine,” and “Atom Smasher.” The end result is a stellar live album and DVD There was no choice other than the Roxy for CIRITH UNGOL to record this momentous live collection “It’s owned by a Hollywood music legend Lou Adler,” notes cofounding drummer Rob Garven “and is next door to the famous Rainbow Bar and Grill which includes the Whisky and The Starwood [now gone] so every time we played one of these historic venues It was great to see many of our friends and fans from all over the world at the show.” there has been a live bootleg record famous in the underground from their last appearance there in1984 titled Cirith Ungol I felt it fitting for the band to return there 40 years later to officially document a live performance in a way the guys would be proud of and Goldenvoice all came together in support to actually make it happen.” CIRITH UNGOL‘s Live At The Roxy holds its own against other legendary live records “The Who made an extremely powerful album with Live At Leeds had several pretty spectacular live albums and 1993’s Uriah Heep Live is another outstanding live album It’s hard to accurately capture all the sound and feeling of a live concert and I think our Live At The Roxy does also.” Live At The Roxy will be released on April 25th via Metal Blade Records today the band presents a taste of the monumental event with video footage of “Down Below.” “This song strung together by the haunting acoustic guitar is a gift to all of you on this Doomed Planet the culmination of all the pain and suffering absorbed into our being from the pandemic It features some of my best drumming and one of [guitarist] Jimmy [Barraza]’s best solos As the song rises to the ultimate crescendo all this pent-up emotion is unleashed on the listener with our final gasping breath Truly a churning maelstrom of metal chaos descending!“ Live At The Roxy will be available digitally and as a 2xCD/DVD set or 2xLP/DVD set in the following color variants: “The band’s strong late-period form is now also documented on an impressive live album with a whopping 100 minutes of playing time includes the complete release show for the 2023 album The Dark Parade Recorded at the legendary Roxy Club on L.A.’s Sunset Strip CIRITH UNGOL practically offer a complete showcase of their work culminating in the frenetically acclaimed ‘Join The Legion’ The audience sound is also beautifully captured so that Live At The Roxy can be recommended to every CIRITH UNGOL cultist to heart.” – Deaf Forever (DE) FRISCO – FC Dallas has waived midfielders Bryan Leyva and Bruno Guarda as well as forwards Ruben Luna and Matias Jara Portugal was again shown at Sunday’s stage four which exploded quite far from the finish in Guarda A combined attack from W52 – Porto and Efapel put race leader Alejandro Marque’s Atum / Tavira squad in the end by the Spaniard who brought back by himself half of the two-minute advantage Figueiredo and Amaro Antunes (W52) held over the penultimate climb to Guarda (Cat-3) Abner was the third strongest within those in the peloton and sits almost at the same overall place he had a day prior: he’s now in sixth 1’10” from Marque and 45″ away from the podium The ‘Grandíssima’ will enjoy its (feat ORM)” accompagnato da un video diretto da Moshmallows: BAEST commented about the “Colossal” album and the band’s current activities as follows: “Paying homage to the heavy metal roots of our music This album showcases our passion for melody and a new direction for our death metal Keeping the energy of former releases and adding new flavours to the inferno ‘Colossal’ stands out as an honest Recording it over the course of 2 years at Antfarm Studio with Tue Madsen has been a different but exciting process leaving a lot of space in between material released off the album with the two first singles ‘Colossus’ and ‘Imp Of The Perverse’ being almost a year old by now We found that giving songs more time to grow felt right and we also had to reinvent our own style and sound in that process which leads to our satisfaction with ‘Colossal’ – it’s a broad album sound wise Playing some of the new tracks live on our recent tour with Danish rock legends D-A-D was helpful in cementing the appeal of the new tracks and helped us finish the album recordings between those shows we embark on our first ever US/North America run supporting Arch Enemy and we are following this tour up with a ton of European festivals This will be the perfect platform to show the new material and get feedback from the fans – see you on the road!” BAEST also checked in with the following comment about the song: “‘Misfortunate Son’ is the newest single release from our upcoming 4th album ‘Colossal’ Recorded at Antfarm Studio with Tue Madsen (The Haunted it brings forth a blend of melody and brutality The song showcases our passion for classic heavy metal riffing while incorporating the signature catchiness we always strive for with BAEST We brought our Danish heroes ORM to add treatment is based on hydroxyurea (HU) which decreases leukocyte counts and increases fetal hemoglobin synthesis Different cell types are thought to contribute to vaso-occlusion the role of monocytes subsets remains unclear We investigated frequencies of monocytes subsets in blood and their response to HU therapy testing their ability to express pro-inflammatory molecules and tissue factor (TF) We identified major changes in monocyte subsets with classical monocytes (CD14++CD16−) appearing highly frequent in who were not taking HU whereas those with patrolling phenotype (CD14dimCD16+) were enriched in individuals undergoing therapy IL-8 as well as TF by the LPS-activated monocytes frequency of TF-expressing monocytes is increased in patients with previous vaso-occlusion activated monocytes expressing TF produced several pro-inflammatory cytokines simultaneously Such polyfunctional capacity was dramatically dampened by HU therapy The frequency of classical monocytes subset was positively correlated with percentage cytokine producing cells upon LPS stimulation These findings suggest that classical monocytes are the subset responsible for multiple pro-inflammatory cytokine production and possibly drive inflammation and vaso-occlusion in SCA which is damped by HU Understanding the mechanisms driving susceptibility to VOE is critical to develop optimization of clinical management and development of new therapeutic approaches of SCA patients The exact mechanisms by which TF-expressing monocytes may drive VOE and/or cardiovascular complications of SCA patients are not completely described Considering the intricate mechanisms related to SCA pathogenesis we aimed to investigate in detail the effect of HU therapy on circulating monocytes subsets and on their ability to express TF as well as pro-inflammatory cytokines upon activation in SCA patients we tested association between monocyte activation phenotypes and occurrence of VOE Our findings indicate that HU therapy induces substantial changes in frequency of monocyte subsets as well as in their capacity to promote inflammation and coagulation which was associated to occurrence of VOE in SCA our data suggest that HU treatment modulate the inflammatory response driven by the monocytes The two patients underoing HU therapy experienced one episode of VOE six months prior to blood drawn and referred HU use for the last 6 years (C) Frequencies of indicated monocyte subsets PBMC between the study groups were compared using the Mann-Whitney U test Hydroxyurea therapy negatively impacts production of pro-inflammatory cytokines of monocytes in response to LPS PBMC from sickle cell anemia patients were incubated with 100 ng/mL LPS in vitro and intracellular cytokine staining assay was performed to test whether hydroxyurea treatment in vivo induces changes in the capacity of monocytes to respond to LPS by producing TNF-α the study groups were compared using the Mann-Whitney U test Sickle cell anemia-associated tissue factor production by monocytes in response to LPS is diminished by hydroxyurea treatment in vivo (A) PBMC from sickle cell anemia patients were incubated with 100 ng/mL LPS in vitro and intracellular cytokine staining assay was performed to test whether hydroxyurea treatment in vivo induces changes in the capacity of monocytes to respond to LPS by producing tissue factor (TF) HU group n = 17 and no HU group n = 20 At each experimental condition (B) Mean Fluorescence Intensity (MFI) of TF expression by monocytes at indicated experimental conditions is shown No statistically significant differences were observed (C) Frequency of TF-expressing monocytes upon LPS stimulation was compared between SCA patients presenting or not previous occurrence of vaso-occlusive events (VOE) The study groups were compared using the Mann-Whitney U test (D) Receiver Operator Characteristics (ROC) curve analyses was employed to test whether frequency of TF-expressing monocytes after LPS stimulation could discriminate patients with previous occurrence of VOE from those who had not as a way to measure strength of association (E) Frequencies of TF-expressing monocyte subsets was compared were compared between the indicated groups using the Mann-Whitney U test These results uncover differential ability to induce TF expression among the distinct subsets of monocytes in SCA patients Hydroxyurea therapy reduces the capacity of activated monocytes to produce multiple pro-inflammatory cytokines Polyfunctional analysis of TF− (A) and TF+ (B) monocytes upon LPS stimulation was performed in PBMC from SCA patients undertaking or not hydroxyurea Data were compared using the Mann-Whitney U test (C) The overall cytokine expression profiles of activated TF− or TF+ monocytes from SCA patients treated or not with hydroxyurea were compared using the chi-square test (D) Frequencies of monocytes producing more than 1 cytokine in vitro were compared between SCA patients undertaking (n = 17) or not (n = 20) hydroxyurea using the Mann-Whitney U test Ex vivo frequency of CD14++CD16− monocytes in peripheral blood directly correlates with the capacity of activated monocytes to produce pro-inflammatory cytokines (A) Frequencies of classical monocytes in peripheral blood from SCA patients undertaking or not hydroxyurea were tested for correlations with frequencies of monocytes expressing indicated markers after LPS stimulation in vitro Data were compared using the Spearman correlation rank test (B) Spearman correlations between frequency of monocytes subsets and frequency of monocytes expressing IL-1 β Bars represent the strength of correlation (r values) Red bar indicates statistically significant correlation (p < 0.05 after adjustment for multiple comparisons) while grey bars were nonsignificant We hypothesize that HU may directly induce differentiation of classical monocytes into non-classical/patrolling phenotype by increasing CD16 expression Future studies are warranted to answer this question Considering that heme is released during hemolysis these findings argue that intravascular hemolysis may play a pivotal role in monocyte activation in SCA these data indicate that HU affects not only monocyte subsets but also the ability of these cells to produce pro-inflammatory cytokines In sterile inflammatory conditions such as SCA the polyfunctionality still remains to be evaluated we investigated the cytokine profile production of both TF+ and TF− monocytes and also tested the effect of HU in production of multiple pro-inflammatory cytokines Our data provide evidence that patients who were not under HU therapy have increased frequency of monocytes simultaneously producing TF HU substantially dampened such production without affecting cell death This result suggests that the inflammatory response promoted by activated monocytes relies on the production of multiple pro-inflammatory cytokines and is directly affected by HU therapy this is the first study to determine ex vivo characterization of monocytes subsets and to identify their polyfunctionality in SCA the association between TF-expressing monocytes and occurrence of VOE also highlights the importance of these cells in vascular complications linked to SCA our data corroborate with previous studies that show beneficial effects of HU therapy in SCA We show that HU is associated with the improvement of laboratory parameters to decreased frequency and activation of the classical inflammatory monocytes HU therapy directly dampened the polyfunctional capacity of monocytes suggesting an overall anti-inflammatory property which the molecular mechanism still requires elucidation Considerations regarding monocytes subsets activation profile and cytokine production are useful to suggest novel therapeutic targets and may help to understand the inflammatory mechanism underlying SCA Thirty-seven pediatric SCA patients (HbSS genotype) were enrolled in the present study all seen at the Bahia Hemotherapy and Hematology Foundation from August 2017 to December 2017 The patients had an average age of 14.16 ± 3.08 years and a median age of 14 years (interquartile range [IQR]: 12–17 years) All patients were in steady state of sickle cell anemia characterized as the absence of acute crisis in the past three months prior to blood collection procedures Three patients have had stroke and were under blood transfusion therapy; one patient had received transfusion 10 months before study enrollment and the other two had received 30 days before enrollment Twenty patients were not under HU therapy while 17 patients were taking HU for at least 5 months Prior to enrollment in the study informed consent was obtained from all individual participants Legal guardians agreed to allow the biological sample collection procedures and signed terms of informed consent of all individuals under 18 years while individuals older than 18 years have signed the assent form This study received approval from the Institutional Research Board of São Rafael Hospital (protocol number: 1400535) and is in compliance with the ethical principles of the revised Declaration of Helsinki clinical data regarding the occurrence of previous clinical manifestations (e.g VOE) were collected using a standardized questionnaire (self-reported or reported by the parents) and confirmed by the medical records Patients or their legal guardians were asked whether they ever had or not Hospital admissions were defined as hospitalization for more than three days and VOE were defined as acute pain affecting any body part lasting several hours in association with swelling especially in the joints and soft tissues requiring medication Patients with previous history of VOE presented at least one episode of VOE (ranging from 1 to 5 events) in the past six months Hematological parameters were obtained using a Beckman Coulter LH 780 Hematology Analyzer (Beckman Coulter USA) and hemoglobin patterns were confirmed by high-performance liquid chromatography employing an HPLC/Variant-II hemoglobin testing system How can we collaborate to empower women who fight for the future of their work? – Following this thought  Entre Redes was born” says Puga. Seamlessly incorporating the “redeiras” of A Guarda in the design process of a collection of objects – she gracefully fuses the individuality of their artisan techniques with the possibility of innovation.  The resulting products, Bolina and Saia, are deeply inspired by marine objects and tools, handmade with recycled nets from the sea and woven with traditional fishing knots utilized by the netmakers throughout history.  At first, Bolina skillfully incorporates unused fishing rope pieces from the process of net making.  Handwoven by the local women netmakers of A Guarda – this one-of-a-kind chair is entirely joined by traditional fishing knots, leading to an innovative reinterpretation of the technique, yet maintaining the identity of the craft. Cava is taking concrete steps toward solving the myriad problems the industry faces today This much has been apparent since April 2021 Cava announced a series of sweeping regulation changes concerning the production of Cava de Guarda Superior this elite category includes the specific designations of Cava Reserva (with a minimum of 18 months of bottle aging) and Cavas de Paraje Calificado (Cavas sourced from specific single-vineyard plots and aged a minimum of 36 months) which the denomination described as “the most exacting standards in the world for quality D.O sparkling wines made using the strict traditional method,” all grapes destined for the Guarda Superior designation must be hand-harvested from vineyards with a minimum of 10 years of age In addition to strictly limiting yields (to a maximum of 4.9 tons per acre) and requiring vintage and disgorgement dates for all Guarda Superior wines Cava also requires estates to comply with strict traceability audits from harvest to bottle It would save its most radical update for last declaring a commitment to convert all Cava de Guarda Superior to 100 percent organic by 2025 To the region’s faithful legions of growers and producers this watershed decision aligns with a wider effort to strengthen Cava’s qualitative foundations while ensuring a more sustainable future for the territory where world-class traditional method wines have been crafted for more than a century estates both large and small are busy putting plans in place to adapt to this ambitious new mandate Such a massive paradigm shift cannot help but invite questions and bring a certain set of challenges With the 2025 deadline quickly approaching what are the implications for the style and quality of Cava as well as its long-term growth and distribution And how will overcoming the short-term hurdles involved in reaching this milestone help to ensure the enduring success of Spain’s most famous and historically celebrated sparkling wine As extraordinary as D.O. Cava’s decision might seem, in many ways it represents the natural extension—and official formalization—of an evolution that had already been brewing for quite some time. Over recent years, the denomination has staked its claim as a global pioneer of organic viticulture. According to its 2022 Global Report the annual number of bottles of Cava labeled as organic has grown by more than 40 percent compared to 2021 this transformation will only accelerate as Cava continues to lead the push toward more resilient forms of agriculture “The number of bottles of organic Cava produced has now reached 13,780,711 units 34.09 percent of which corresponds to premium organic Cava.” Cava’s considerable effort in promoting organic production Pagés stresses that “both the consumer and the market are demanding it.” Ultimately he views this evolution as being driven by deeper currents than market forces alone; he believes it amounts to an act of conservation for the territory itself “This care and preservation of the territory is something with which we identify closely.” “The region’s relatively low disease pressure and sufficient sunlight support the growth of healthy vines with fewer chemical interventions.”  Despite these natural advantages, the exacting standards the D.O. established to track compliance, requiring estates to trace every step of the production process from vineyard to bottle, still presents a challenge for some winemakers. “The process is extremely difficult to manage if you’re not used to it,” says Marta Casas, a winemaker and co-owner at Parés Baltà the pioneering organic estate that first gained certification in 2003 before converting to biodynamic in 2013 “Even for us it has been a significant challenge and we’ve been farming organically since 2003.” the end results more than compensate for all the hard work their efforts will preserve the land for future generations their work takes a critical step toward defining Cava both as a world-class sparkling wine and a unique expression of place “It’s a big change in terms of the mentality of the producers but in the end it will present us with an equally big reward,” says Casas These changes will allow us to demonstrate to the consumer that our Cava comes from a very specific part of the world and that each step of the process is controlled and verified from the very beginning.” it’s still too early to predict how these major shifts at the level of production will impact the sales and distribution of Cava’s top wines in the global market Buyers and consumers alike can expect what Aura calls “the introduction of a dynamic dimension to production and distribution.”  “pricing in the market might reflect the additional effort and cost associated with organic farming.” Any such realignment would simply represent a fair-market adjustment helping to “position Cava de Guarda Superior as a premium product that aligns with consumer preferences for sustainable and high-quality wines.” To Maite Esteves, a partner and the manager of Vins El Cep another pioneer of organic sparkling wine in Comtats de Barcelona the revolutionary changes taking place within Cava’s top tier ultimately promise to reinvigorate the category as a whole “The most important thing is that all this means a change for the entire sector and that it serves to improve the positioning of Cava,” she says “The future of Cava depends on increasing its value so that the entire chain is strong and committed to quality.”  While it’s impossible to see into the future one thing’s for certain: We don’t need to wait until 2025 to taste the results Armed with a new story to tell about itself and the passionate artisans who make it the denomination’s highly anticipated transformation is already underway Don’t miss the latest drinks industry news and insights—delivered to your inbox every week winemaker Nicole Marchesi has helped usher Far Niente into the future while honoring the legendary winery’s roots PDO Santorini’s distinctive volcanic terroir the exceptional qualities of its Assyrtiko grapes and the unique vine-growing methods employed by its winemakers all contribute to the unparalleled character of its wines Italian icon Florio has both pioneered and perfected the famous fortified wine in the town of Marsala Far Niente Wine Estates has built a portfolio of emblematic Napa Valley Cabernet Sauvignons at every price point Learn more about how Provi simplifies the complex process of ordering and promoting wholesale alcohol between buyers SevenFifty Daily is an award-winning online magazine about the business and culture of the beverage alcohol industry covering all three tiers of the alcohol industry Joe: You're a child He's the moon when I'm lost in darkness and warmth when I shiver in cold His heart overflows with the kindness of which this world is not worth of Nicky: You're an incurable romantic.
