he wasn't in a position to run to become the city's first mayor in 2017 he's running on a platform of empowering Framingham.  “I want to be the mayor for the people He waited to pursue dual he has been flying in the area for over 20 years Alexandre Germani 1st on podium of Brazilian Serial Class  in Real Minas Open 2002 in Governador Valadares it is one of the most known flying spots here in Brazil with a high takeoff and delicious flying routes and its traditional championships that are very fun to attend One of my first ever championships participation was in Governador Valadares The most evident difference is not in the place itself because 20 years ago it already had a great structure and all but what I recall the most and it was painful is that in championships we used to go up to the take-off riding some “pau de arara” trucks that were flatbed trucks with some wooden boards to sit on it was a tuff 1h ride that you arrive in the take-off already tired and with back pain Flying in GV in 2002 (source Alexandre Germani)          "Pau de arara" truck in Brazil      (source Wikimedia) Governador Valadares always has a major significance in the Brazilian paragliding scene we have there one stage of the Brazilian Nationals almost every year and already had some major international comps too For example Paragliding Worlds in 2005 and PWC Superfinal in 2014 and 2017 There have been a lot of very cool tasks there and the ones that I liked the most were the ones that we went out and return to land in the “Feira da Paz” landing But it is not allowed anymore due to security reasons The most common route is the south route heading to Caratinga but Governador Valadares is known for its light winds so in competitions is usual to make triangles and more complex tasks The classic condition is usually March to April so we expect to have great conditions in the Pan-Am Governador Valadares is a well-known place for a lot of pilots and the Brazilian nationals took place there 2 weeks before Pan-Am so I think the Brazilians will arrive only right at the competition Governador Valadares has usually light winds at take off and sometimes it varies between the 2 directions (N and S) so take off early when the condition is good use a good sweat-resistant sunscreen it’s a very hot region and you will sweat a lot and take some extra water with you Take care with the rock wall in the side of the take-off (it’s sometimes very turbulent) take care with power cables in the landing (there is a lot of power cables crossing the valleys) and take care of the puddles at landing (when you see a perfect flat grass field for landing beware that is probably a quagmire) I’ve learned that our pre-conceived information on the place the regions that work best and this kind of stuff is not always a good thing and the fact that a large group of good pilots flying together is more important Governador Valadares usually has strong and consistent thermals so it is a place where you can risk getting low and out front Thank you Alexandre for the insights and we wish you a successful competition FAI - Follow her on Twitter @zanerazz Metrics details Leishmaniasis is a highly diverse group of diseases caused by kinetoplastid of the genus Leishmania with human pathogenic species separated into two subgenera according to their development site inside the alimentary tract of the sand fly insect vector The disease encompasses a variable spectrum of clinical manifestations with tegumentary or visceral symptoms Leishmania (Leishmania) amazonensis is an important etiological agent of human cutaneous leishmaniasis that accounts for more than 8% of all cases in endemic regions (L.) amazonensis is generally found in the north and northeast regions of Brazil (L.) amazonensis from dogs with clinical manifestations of visceral leishmaniasis in Governador Valadares an endemic focus in the southeastern Brazilian State of Minas Gerais where L These isolates were characterized in terms of SNPs chromosome and gene copy number variations confirming that they are closely related to a previously sequenced isolate obtained in 1973 from the typical Northern range of this species The results presented in this article will increase our knowledge of L (L.) amazonensis-specific adaptations to infection parasite survival and the transmission of this Amazonian species in a new endemic area of Brazil although hindgut development appears to be ancestral These distinct disease features have been classified into tegumentary (TL; also known as cutaneous leishmaniasis or CL) and visceral leishmaniasis (VL) The municipality of Governador Valadares in the southeastern Brazilian state of Minas Gerais is an endemic area for TL and a focus of intense transmission of VL identification of Leishmania species using molecular or biochemical tools has not been conducted in this area Our study has found important differences in terms of SNPs chromosome and gene copy number variation within L (L.) amazonensis and with the closely related species L (L.) mexicana that have not been explored previously As well as expanding our knowledge of the diversity and epidemiology of L this information will ultimately contribute to the understanding of some of the mechanisms of L (L.) amazonensis infection and survival as well as provide conclusive evidence of the presence of this species in VL-endemic urban areas Molecular genotyping indicates that isolates S3 and S6 belonged to the Leishmania mexicana complex due to the matched restriction profile with L. (L.) amazonensis/L. (L.) mexicana (Supplementary Fig. S3) whereas the remaining 34 isolates were genotyped as L (L.) infantum isolates were sequenced on the Illumina HiSeq 2000 v3 platform at The Wellcome Trust Sanger Institute Raw sequence data was deposited in the European Nucleotide Archive with the accession number ERP016755 base and mapping quality as described in the methods This approach identified 23,921 and 17,624 SNPs between JPCM5 and isolates S3 and S6 13,474 variants were shared between both isolates This result contrasts with the lower number of variants called in the other three L (L.) infantum isolates against the JPCM5 (2,342; 2,318 and 2,149 SNPs in S1 This finding is consistent with PCA result for these isolates that shows a cluster of three isolates with the JPCM5 reference strain, while samples S3 and S6 were markedly different (Fig. 1). This result confirmed that isolates S3 and S6 were somehow unrelated to the rest of the isolates and therefore required further analysis. PCA shows that the S3 and S6 isolates to do not group with L Competitive mapping against the L. (L.) mexicana U1103 and the L. (L.) amazonensis M2269 reference genomes resulted in more than 78% of the reads from both the S3 and S6 isolates mapping specifically to L. heterozygous sites on disomic chromosomes should have frequencies close to 0.5 while those on trisomic chromosomes will show frequencies of 1/3 or 2/3 and those on tetrasomic chromosomes can show peaks at 1/4 The blue lines represent normalized counts of the proportions at heterozygous positions for all chromosomes The results support that most chromosomes are disomic in both isolates (L.) amazonensis is also likely to act during the transition to infecting metacyclic promastigotes These tandem gene arrays include surface antigen protein 2 (PSA2) may be particularly important for the more aggressive disease phenotype associated with L (L.) amazonensis that can range from DCL to VL infection The genomic analysis performed in this study of the two L (L.) amazonensis isolates allowed us to explore some unique features in terms of chromosome and gene copy number variations that are unique to this species (L.) amazonensis could be the result of different selection pressures to other Leishmania species Gene copy number variations in relation to L (L.) mexicana show that species-specific expansions exist despite the high similarity especially in expanded genes and tandem arrays in proteins potentially involved in cell differentiation cellular trafficking and parasite host interaction this mechanism could be crucial for invasion and survival inside host macrophages playing an important role for PSA2 and EF-1α and it may also be partially responsible for the broad clinical phenotype associated with different isolates including TL It is important to emphasize that because we are analyzing only two L it is difficult to obtain robust information about determinants of the disease This question should be addressed in a larger study with an increased number of L amazonensis isolates and oriented towards a comparative perspective against a VL species this article presents the first report of L (L.) amazonensis in Governador Valadares and shows a potential risk for current control efforts in the area that have been designed considering only the presence of L (L.) amazonensis presented all clinical symptoms shown by dogs infected with L This information provides evidence of the severity of L (L) amazonensis infection and might indicate potential involvement of L (L.) amazonensis in canine VL that should be further investigated We also find evidence that the presence of L (L.) amazonensis in southern Brazilian regions correspond to an ancient dispersion event rather than a recent introduction of this species as our two southern isolates are estimated to have a common ancestor almost two thousand years ago Sequence data from additional isolates from both foci would help to shed further lights on the epidemiology of L and exclude an alternative explanation of multiple introductions of this species to Southern Brazil Our results indicate that the EIE-LVC kit and possibly other diagnostic kits based on similar antigens can cross-react with L This cross-reactivity may have resulted in an underestimation of the distribution and prevalence of L This important drawback underscores the need to develop better diagnostic methods capable of discriminating between L (L.) infantum and to encourage the use of genomics into epidemiological studies Given the different clinical presentations of human infections with L surveillance efforts and control activities in this region should consider the presence of L (L.) amazonensis and address putative vectors for this species and the risks of co-infections in human subjects The upper right inset shows the location of the Minas Gerais State in Brazil the city of Governador Valadares (black circle) and the isolation site of the M2269 L (L.) amazonensis reference in the State of Para (green circle) we show the neighborhoods where isolates S3 and S6 were collected in Governador Valadares (Vila Mariana and Santa Helena Parasites were cultivated in Schneider culture medium supplemented with 10% fetal bovine serum and 1% penicillin and streptomycin for up to three passages Genomic DNA was extracted from ≈109 promastigotes using the DNeasy Blood and Tissue Kit (Qiagen) using the manufacturers protocol Sera from dogs were tested in triplicate by ELISA using the EIE-LVC kit supplied by Biomanguinhos following the manufacturer’s standard protocol This kit consists of soluble antigens of L (L.) major and has been widely used in public health laboratories in Brazil for the diagnosis and surveillance of canine visceral leishmaniasis Based on results from molecular typing we sequenced the genomes of five selected isolates Genomic DNA was sheared into 400–600-base pair fragments by focused ultrasonication (Covaris Adaptive Focused Acoustics technology (AFA Inc. Joni ValadaresHelena CavellPublished26 August 2016Updated 8 May 2017There are many awareness days throughout the year but the one on Saturday 10 September is particularly poignant It's a subject people find hard to contemplate - let alone talk about - but one spokesperson is doing his best to discuss the subject and more importantly help those who are suffering with suicidal thoughts Mr Gay England is a down-to-earth lad from Gateshead At the Mr Gay Europe pageant in August he was first runner up to the eventual winner he's proud to have represented his country and so he should be Although most pageants could be considered a bit…erm – in this case that couldn’t be further from the truth Although a pageant sounds fun (or terrifying) the thing that drew Joni to the competition was the chance to be an ambassador for the LGBT community “They ask you in your interview what you would want to focus on with your title how can you help improve the LGBT community suicide in the LGBT youth is very very high…” That’s an understatement; according to Young Minds 44% of young (16-24 year old) LGBT people have considered suicide Having lost his father to suicide when he was seven years old Joni is passionate about doing all he can to help prevent it “My goal will always be suicide prevention because I lost my father.” But he understandably didn’t feel that way when he was younger: “I was so against suicide I used to find it so selfish…I wouldn't even go to the funeral of a friend who committed it It was a subject that I had to face and conquer but it wasn’t until this year that finally I began to understand and wanted to work with charities to help prevent suicide I wasted so many years hating but in the end I turned my passion against it to preventing it!” The BBC is not responsible for the content of external sites.And that’s where pageantry comes in Joni was given a stage to raise awareness for LGBT suicide prevention he’s working with various charities to keep the ball rolling “A lot of suicide charities don’t actually have LGBT sectors at the moment so we’re going to start setting that up – help specifically dedicated towards LGBT youth charity knowledge and my position in the LGBT community I want to tell the youth everywhere that its OK to be gay.. Mr Gay Europe gave him the perfect platform to broach these issues; for Joni entering seemed like a no brainer But what does it actually take to enter MGE Can anyone with a worthwhile cause and a photogenic face sign up No swimsuit contest here; Mr Gay Europe is actually really intense How would you cope with a two month long presidential election-style campaign Plus a sports challenge that involved running up and down a Norwegian mountain As well as numerous social media challenges including having to present yourself to the world on YouTube (no thanks Not to mention the written exams which test your knowledge of LGBT rights It’s not just walking up and down the catwalk looking pretty… I’m not in any way insulting other pageants because I’m sure what they do is very hard but I don’t see them running up and down a hill four times round in the middle of the Norwegian hills!” And Joni had little support outside his friends and family “I even contacted [here he names and shames a local newspaper] and I couldn’t even get a story in there local lad from Gateshead is in the final for Mr Gay Europe… when they phoned me up they said they’d heard I was in Mr Europe The reaction was more positive in other countries though; Mr Belgium I mean their Prime Minister streams the final live in their country…they are so behind the LGBT cause in Belgium so I think the right winner was chosen because he’ll be able to achieve so much more than I could Joni ValadaresThat seems to be the idea behind pageants like these: garner support Hopefully the MGE pageant itself will indirectly help suicide prevention through paving the way for broad inclusion and understanding around the LGBT community it’s that for the LGBT community acceptance is essential It certainly is for Joni: “we were taken into the octagon in the Cathedral in Trondheim which is where Kings and Queens were married We were the first gay men to be allowed in there It was a huge step forward for the LGBT community” Through organising events like this MGE is sending out a clear and loud message to the LGBT community: you are not alone But between Joni’s suicide prevention campaigns and the pageant’s efforts to encourage LGBT acceptance So you can stop thinking of pageants as an old-fashioned beauty content Mr Gay Europe is redefining pageantry and doing wonders for the LGBT community along the way If you're affected by the issues discussed in this piece, there are organisations that offer information and support.
