This change comes just days after the Army resumed training flights in the area following January’s deadly midair collision between a Black Hawk and American Eagle CRJ-700 On Thursday, two separate commercial flights—operated by Republic Airways and Delta Air Lines—were forced to go-around while on approach to Runway 19 at Ronald Reagan Washington National Airport (KDCA) An air traffic controller instructed the two aircraft to go around in order to avoid a Black Hawk helicopter The Black Hawk was operating under the Priority Air Transport (PAT) call sign the same one involved in January’s accident According to CBS News the Army had a four-week plan to restart helicopter training in the region after the collision Now, flights will be halted again into the Pentagon, Politico reported An Army spokesperson told the outlet that this pause is pending an internal inquiry The suspension applies to operations in the Army’s 12th Aviation Battalion the same unit involved in January’s accident This pause only pertains to flights into the Pentagon which is located less than 2 miles from Reagan National The National Transportation Safety Board (NTSB) is probing Thursday’s incident. This event is essential for raising community awareness and connecting people to services beyond today.” Among the array of offerings were dental services, screenings for breast cancer and colorectal issues, trauma prevention strategies, CPR training, and heart health promotion. People also received practical items, like toothbrushes and bracelets, along with American Heart Association's portable mannequins to practice CPR at home. “Even in rural areas, where it might take time for EMS to arrive, this training can exponentially increase the chances of saving a life,” Ewing said. : : FLI1 fusion oncoproteins; thus any approach aimed at blocking or inactivating EWSR1 : : FLI1 is expected to interfere with tumor growth Another possible strategy to interfere with EWSR1 : : FLI1 function is the use of siRNAs to knock down EWSR1 : : FLI1 expression This strategy has been extraordinarily successful for the generation of cell models of EWSR1 : : FLI1 knockdown which have been very helpful for understanding the biology of Ewing sarcoma potential therapies based on the use of siRNAs designed to knockdown EWSR1 : : FLI1 mRNAs have the same drawbacks as those described above concerning the use of small molecules designed to block EWSR1 : : FLI1 function to achieve the best EWSR1 : : FLI1 inactivation genetic inactivation of the EWSR1 : : FLI1 gene exquisite control of Cas9 expression must be achieved to rule out any unwanted effects on normal cells we demonstrate that this precise and exquisite control could be achieved by using an Ewing-specific promoter formed by concatenated repeats of the tetranucleotide GGAA (GGAAprom) the use of a consensus TATA box downstream of the GGAA repeats increased the activity of the promoter by 2–3 times compared to that of the GGAA repeats only The high specificity of this DNA sequence for Ewing sarcoma cells was demonstrated both in vitro and in vivo GGAAprom was active in Ewing sarcoma cells expressing EWSR1 : : FLI1 proteins (A673 which encompass three different EWSR1 : : FLI1 isoforms) but not in non-Ewing sarcoma cells such as the fibrosarcoma cell line HT1080 or the osteosarcoma cell lines U2-OS and Saos-2 The absence of GGAAprom activation in HT1080 cells is particularly relevant because these cells express high levels of native FLI1 indicating that GGAAprom is only active in Ewing sarcoma cells expressing EWSR1 : : FLI1 This important finding was soundly confirmed in in vivo experiments when athymic nude mice were injected intravenously with adenovirus harboring a luciferase gene under the control of GGAAprom no luciferase expression in any of the 12 tissues analyzed was detected This means that a gene therapy in which the therapeutic gene is under the control of the Ewing-specific promoter will be active only in Ewing sarcoma cells expressing EWSR1 : : FLI1 This opens the possibility of using this type of approach with a multitude of therapeutic genes that may include not only nucleases such as the one proposed in this work to inactivate EWSR1 : : FLI1 but also genes capable of inducing cell death (for example suicide genes) or immunomodulatory genes capable of activating an antitumor immune response still one or two orders of magnitude higher than that used in our preclinical animal studies This indicates that there is still room to use higher doses that would have yielded even more remarkable results further experiments with higher doses were not possible due to the limitation in the amount of adenovirus produced the authors use a complex strategy for in vivo delivery based on the use of lentiviral vectors and an adapter antibody that can make more difficult its preclinical development Although in vivo gene editing certainly presents numerous risks that must be carefully considered the development of highly specific expression strategies such as the one presented in this work opens new opportunities to permanently inactivate the EWSR1 : : FLI1 fusion gene characteristic of Ewing sarcoma we reported a gene therapy approach to permanently inactivate EWSR1 : : FLI1 in Ewing sarcoma cells in a highly specific way no other approach has been shown to determined to become a successful leader once he graduates from Missouri S&T. Through academic excellence and leading cadets in S&T’s Army ROTC Stonehenge Battalion “I’m involved in basically every opportunity that you can be involved in,” Ewing says “I just have this philosophy: I always want to be better tomorrow than I am today He will graduate twice this year: earning a bachelor’s degree in May and a master’s degree in December.   Both of Ewing’s grandfathers were military officers (Army and Air Force) Ewing says they passed away before seeing him choose the military Ewing’s path to the military has certainly been a busy one.  Ewing’s first college year began with glowing success. Along with getting involved in the ROTC color group, the cannon crew and various campus clubs, Ewing excelled in the Ranger Challenge a military skills competition for the nation’s military academies and ROTC programs.   The final competition, the Sandhurst Military Skills Competition comprises the top 16 ROTC teams in the nation plus 15 international teams and military academies The task force and brigade competitions determine which 16 ROTC teams advance to Sandhurst Ewing’s team won the task force competition and placed second in the brigade competition held at the United States Military Academy at West Point “We became the first school in Missouri to make it to Sandhurst,” Ewing says “It was the first time in our school’s history that we ever made it to an international competition of that caliber.”  Ewing’s summer following his first year of college was just as busy. Ewing completed an internship with the United States Army Corps of Engineers, where he worked at Fort Leonard Wood on a hospital replacement project.   “I worked as a quality control and quality assurance engineer with Major Steven Lanni and saw how the Army Corps of Engineers operates in the engineering regiment itself,” Ewing says.   Sophomore year: Developing solutions  In addition to serving as the chair of a national scholarship committee Ewing became more involved during his sophomore year as a team leader within S&T’s Stonehenge Battalion By earning the highest combat fitness test score at S&T he achieved the role of master fitness trainer in which he developed personal training plans and curriculum for S&T cadets.   He became involved in the recruiting committee and even developed a software program that helped recruit more cadets.  “That software I helped develop — that’s some of the knowledge I got from my degree that really helped our program,” Ewing says “It basically automated our process that we already did but just streamlined it and made it more efficient so we didn’t have to do it the old way with a pencil.”  In recognition of his service to the campus and community named for a professor of engineering management at Missouri S&T for over 30 years.   he completed a course at the University of North Carolina through Project Global Officer to learn Mandarin Chinese in just two months.  He learned how to safely jump out of an airplane wearing a parachute.  “I did jump school because one of my biggest fears is heights but how I’ve defined myself is I want to do the things that I’m the most scared of That was my way of conquering what I was most afraid of.”   he became captain of the Ranger Challenge team and a platoon sergeant within the battalion.   Year three is primarily preparing students to get ready for cadet summer training Ewing joined Missouri S&T’s eSports club and became an officer in the American Society for Engineering Management (ASEM) Myers Scholarship to help determine the next recipient.  Ewing attended cadet summer training in Fort Knox He was evaluated on individual military skills and his ability to lead a platoon of 40 cadets through a rigorous field exercise 1 out of 640 cadets that participated in his regiment This exceptional performance contributed to his ranking as No 13 out of 5,140 cadets in his graduation year group from across the nation.  From there Ewing flew to Fort Lewis in Washington to complete another internship in the 46th Aviation Support Battalion He worked with the Multi-Domain Effects Battalion “I was learning how to be an officer in a real-life environment,” Ewing says learning signal operations such as how we secure our communication networks across the Pacific Ocean.”  Ewing became the cadet battalion commander the highest officer position available for a cadet — and leads 56 cadets.  “Being a battalion commander basically means that you assume all responsibility,” he says “Any training that we conduct is 100% on me to make that decision.”  Ewing explained that he thinks of big ideas for the battalion then ensures those ideas happen by bringing them to his staff who then push it all the way down to each leader.  “The major reason why I like leading is developing others and seeing other people succeed,” Ewing says “That’s been my primary motivator for ROTC and being a leader within the organization.”  Ewing said his senior year has been especially validating — this is the year they get back all of their results everything a student does in their first three years is tabbed and put into an order of merit score then an order of merit list where they get stacked against all 6,000 cadets across America.  11 in the entire country out of all 6,000 cadets that competed in my year group,” Ewing says.   he had the benefit of choosing where his future path goes he was officially awarded Cyber (the US Army Cyber Command) as his branch “It’s a cool honor to have the hard work over the last four years awarded,” Ewing says “I would enjoy going to work at the National Security Agency or to work for a multi-domain task force protecting national security for the country the spearhead of the future of the military is where I want to put myself because while they are one of the most rigorous and time-consuming jobs; the Daytona Tortugas this past week It was a great week for the pitching staff who allowed more than three runs in just two of the six games and four steals in just five games against Daytona Kevin Villavicencio also had an excellent week though just one (a double) went for extra bases and he now has a .394 average and .945 OPS in nine games with St Nate Dohm made two excellent starts in the series and he also allowed six hits and three walks Edgar Moreta held the Tortugas scoreless in his start going five innings and allowing just two hits while striking out three Moreta has struggled and owns a 4.97 ERA through four appearances Perhaps the most interesting start in St. Lucie this past week came from Raimon Gomez who is not just one of the hardest throwers in the Mets’ system but in all of professional baseball but the most notable stat from his start was that he reached a ridiculous 104.5 miles-per-hour with his fastball in the first inning AVG – Marco Vargas (STL)/Kevin Villavicencio (STL/BRK) – .409 SO – Blade Tidwell (SYR) – 37 giving the Mets no choice but to promote him Ewing will likely hit an adjustment period heading to Brooklyn but his development has been a
