The Department of Defense is suspending helicopter training flights in the Washington, D.C., region, specifically near the Pentagon. This change comes just days after the Army resumed training flights in the area following January’s deadly midair collision between a Black Hawk and American Eagle CRJ-700
On Thursday, two separate commercial flights—operated by Republic Airways and Delta Air Lines—were forced to go-around while on approach to Runway 19 at Ronald Reagan Washington National Airport (KDCA)
An air traffic controller instructed the two aircraft to go around in order to avoid a Black Hawk helicopter
The Black Hawk was operating under the Priority Air Transport (PAT) call sign
the same one involved in January’s accident
According to CBS News
the Army had a four-week plan to restart helicopter training in the region after the collision
Now, flights will be halted again into the Pentagon, Politico reported
An Army spokesperson told the outlet that this pause is pending an internal inquiry
The suspension applies to operations in the Army’s 12th Aviation Battalion
the same unit involved in January’s accident
This pause only pertains to flights into the Pentagon
which is located less than 2 miles from Reagan National
The National Transportation Safety Board (NTSB) is probing Thursday’s incident. The FAA stated that the Black Hawk crew did not follow published procedures, instead opting for a “scenic route.”

This 'freak athlete' Mets prospect is creating a buzzApril 18th
NEW YORK -- Earlier this week, Mets director of player development Andrew Christie was chatting with JT Schwartz, a longtime Mets farmhand who was rehabbing at the team’s Spring Training complex in Port St. Lucie, Fla. While in Florida, Schwartz had a firsthand look at A.J. Ewing
the buzziest Mets prospect of the early Minor League season
“And he really is,” Christie later said in a telephone interview
Selected with the 2023 compensatory pick the Mets received for losing Jacob deGrom in free agency, at 134th overall, Ewing entered the year as Pipeline’s No. 28 Mets prospect
finishing a homer shy of the cycle to raise his batting average to .459 -- tops of anyone at that level
he ranked Top 5 in batting average in the entire Minor Leagues
“It just took him a little bit to get acclimated to pro ball,” Christie said
He’s got a really good idea of the strike zone
One scout who’s seen Ewing play this spring defined him as a likely bench player
But Ewing has been hitting the ball in the air with regularity and recently bashed a 109 mph home run -- indications that he can grow as a hitter
Although Ewing has far more swing-and-miss to his game than Morabito
he’s walked more than he’s struck out so far this season
when Christie told him he’d be returning to Single-A St
Lucie (the Mets’ lowest full-season affiliate) to start the year
Ewing responded: “I’m going to make your life very difficult to keep me there.”
Said Christie: “He is delivering thus far.”
Triple-A SyracuseNo. 11 prospect Drew Gilbert -- one of the pieces the Mets received from the Astros for Justin Verlander in 2023 -- finally made it back to Syracuse this week after a delayed start to his season
The Mets have progressed slowly with Gilbert
who strained his hamstring early last April and didn’t return until late July
Team officials felt that an elongated ramp-up could only help Gilbert
who’s been working not just on healing his hamstrings
but on strengthening them to avoid future problems
Gilbert set a new personal best sprint speed on the field
Double-A BinghamtonRyan Clifford, the other player the Mets received for Verlander, is heating up at the plate. After going hitless in his first three games, Clifford had two multi-hit efforts in his next six. Three of his half-dozen hits over that stretch went for extra bases, and Clifford walked as often has he struck out during those games. The Mets’ No. 4 prospect
Clifford spent his final 98 games of last season at Binghamton
That makes him ripe for an early-summer promotion
High-A BrooklynThe hottest hitter at Brooklyn, without question, is No. 22 prospect Chris Suero
The catcher hit five home runs in his first nine games
with a .353/.405/.941 slash line over that stretch
which is why the Mets have given him reps at first base and left field in addition to behind the plate
He could even eventually see time in center
meaning his hot start may soon result in a bump up the ladder ..
but he’ll probably need to cut down on his hefty strikeout rate before that happens

NORFOLK, Va. — Sentara Norfolk General Hospital on Saturday hosted a community health event aimed at promoting education and access to care for residents of Hampton Roads.
Attendees found more than 50 tables filled with resources and services tailored to meet various healthcare needs.
Sentara Associate Chief Nursing Officer Misa Ewing emphasized the importance of such events.
“Today’s objective is to provide education, offer access to care, and create collaboration opportunities among our community partners," she told Norfolk Neighborhood reporter Jay Greene. This event is essential for raising community awareness and connecting people to services beyond today.”
Among the array of offerings were dental services, screenings for breast cancer and colorectal issues, trauma prevention strategies, CPR training, and heart health promotion. People also received practical items, like toothbrushes and bracelets, along with American Heart Association's portable mannequins to practice CPR at home.
“Even in rural areas, where it might take time for EMS to arrive, this training can exponentially increase the chances of saving a life,” Ewing said.
The event not only focused on health but also included a talent acquisition department providing job opportunities. "We holistically provide services beyond health and education," Ewing noted. "We also have job offerings today, allowing us to serve the community in multiple dimensions."
Ewing shared anecdotes about attendees discovering health issues they weren’t aware of prior, which empowered them to seek necessary care. “Through our ACC clinic, we connect individuals with the full gamut of care services they may need,” she said.
While the event was filled with valuable health services, the ambiance was enjoyable and engaging, featuring music, food trucks, a petting zoo, and bubbles to ensure families felt welcome.
For those who couldn't attend, Ewing recommended visiting Sentara's website for more information and resources.

An announcement to “build out a brand-new air traffic control infrastructure system” is coming soon
Transportation Secretary Sean Duffy said during a press briefing on Thursday
President Donald Trump and Duffy have been roadshowing a plan to completely overhaul the United States’ aging air traffic control system
which Trump called “very obsolete.” He continued to say that January’s deadly midair collision at Reagan Washington National Airport (KDCA) near Washington

Duffy added that an announcement on this overhaul will be coming in a matter of days
we’re going to talk about the infrastructure on the airspace,” he said
On Thursday, the FAA and the National Air Traffic Controllers Association (NATCA) detailed a plan to give cash bonuses to air traffic controllers (ATCs) for agreeing to work in hard-to-staff or high-cost-of-living regions

“And we can’t snap our fingers to make up the numbers
Trump tossed around potential private contractors to overhaul the ATC network
But it would be ideal to have a single contractor
“We’re going to have a broader press conference next week on what that plan looks like,” Duffy said Thursday
30 years behind the curve on building a brand-new system.”
Duffy shared that he’s already laid out this plan to Trump: “I’ve had a conversation with the president
I’ve laid out our ideas for a brand-new air traffic control system.”
“I’ve heard him for years talk about the problems within the American [ATC] system and how he wants to fix it and

US political commentator Jennifer Ewing has highlighted a peculiar detail about Donald Trump's decision to rename VE Day as Victory Day for World War II
The former president announced the change on his Truth Social platform
Ewing pointed out to GB News that Americans don't actually celebrate VE Day in the first place
making the entire announcement rather perplexing
Trump declared on Truth Social: "I am hereby renaming May 8th as Victory Day for World War II and November 11th as Victory Day for World War I."
The former president claimed America had done "more than any other country" in achieving victory in World War II
Ewing told GB News: "I can completely see how it’s disrespectful to other countries
what’s confusing me and confusing a lot of Americans is that we don’t celebrate VE Day in America
this announcement didn’t come with an executive order
Congress would need to approve any name change for a holiday
I go back to the point that we don’t celebrate this day in America
I would say that if we did celebrate this day and he was going to change it
I could see how it would be disrespectful to others who
participated with us and led us in that war for sure."
nobody was close to us in terms of strength
but we never celebrate anything," Trump wrote
He attributed this lack of celebration to the fact that "we don't have leaders anymore
Trump concluded his announcement by declaring: "We are going to start celebrating our victories again!"
Trump's claims about America's role in the wars have sparked controversy
particularly his assertion that "nobody was close to us" in terms of contribution
Critics have pointed out that his statement might offend allies like Britain and Russia
The Russian Victory Day on 9 May commemorates the 27 million Soviet citizens who died during the conflict
approximately 418,500 Americans were killed in both European and Pacific theatres

Back in a 1999 column, my old boss, Bill Simmons, coined the Ewing Theory. A friend of his "was convinced that Patrick Ewing's teams (both at Georgetown and with New York) inexplicably played better when Ewing was either injured or missing extended stretches because of foul trouble," he later wrote for ESPN
Ewing was a star center for the New York Knicks
his career directly coincided with a guy named Michael Jordan
so Ewing's Knicks won lots and lots of games
Ewing had just gone down with an Achilles injury during the 1999 Eastern Conference finals
Everyone assumed the Packers would walk to the NBA finals
and the Knicks immediately won three straight games to clinch the series
not been back to the Eastern Conference finals in the two decades since Ewing left the team
Simmons only landed on Ewing for the theory because a buddy of his was obsessed with that particular example, but the idea was more important than the individual. As he wrote in a 2013 update:
That's the thing about the Ewing Theory ..
or mistakenly believed he was more valuable than he was
an injury or departure can lead to more minutes for players who fit that team's style and framework better
it might take a simple injury for everyone to realize
everything falls into place and you're a team
Let's dig into why Mbappé made Real Madrid worse
and what it all says about Mbappé as a player
Madrid's defense was already getting by on individual brilliance -- great goalkeeping
the athletic range of their young midfielders and elite center-back play
who is genuinely one of the worst and least active out-of-possession forwards in the world
was going to put even more stress on a defense that wasn't protected by a sound underlying structure
These are their adjusted goals allowed (a blend of 70% expected goals and 30% goals) over the past two seasons
it's not even clear that Mbappé's arrival has made the defense worse at all
we'd expect Madrid's defensive performance to decline a little bit
That should've been a massive win for Madrid
I can't stress this enough: The defending Champions League winners added arguably the best attacker in the world ..
Here are those same adjusted goal numbers for the other end of the field:
These players all have multiple elite-level skills so
we might see Raphinha drive the ball forward and slip a through ball to Yamal
who plays a cut back for Lewandowski to score the goal
Lewandowski will dart across to the near post to take more defenders with him
and Raphinha will crash the far post to finish off a curled ball in from Yamal
you might have attacks where the players have more static skills -- each one is really good at one particular thing -- but it usually works because each of those skills complement each other
Their two best attackers pretty much do the exact same thing: peel off to the left wing and try to run into the penalty area
Just take a look at Mbappé's heat map of touches for the season:
Perhaps this could work if there was a third attacker who was constantly crashing into the center of the box
or maybe a ball-dominant creative type on the right who was springing Mbappé and Vinícius into these spaces on the left side
But among wingers in Europe's Big Five top leagues over the past 365 days
Rodrygo ranks in the 13th percentile for expected goals assisted
The common thread among Madrid's three attackers is that none of them are getting on the ball in the middle of the field
And here's a little industry secret about the middle of the field: it's where the goal is
On top of all this, Madrid also lost midfielder Toni Kroos to retirement over the offseason
so the team never really had any kind of reliable
systematic way of getting the ball up to these forwards
Perhaps a more consistent pattern of ball progression would have brought the ball up to the forwards in a way where even their lopsided movements would've been able to rip apart unsettled or under-manned defenses
Instead, more often than not, it's looked like what we saw against Arsenal:
Needing to win by at least three goals at home
this collection of uber-expensive and super-talented attackers only created one decent chance off of a defensive error and otherwise were left shooting from tight angles and long distances -- far away from the center of the goal
I still think we're better in attack and defense without Mbappé."
That's PSG manager Luis Enrique
right after his club knocked Liverpool out of the Champions League
By those same numbers we looked at for Madrid, here's how PSG have progressed defensively in Ligue 1:
and allowing significantly fewer touches inside their own penalty area
they're attempting almost four extra shots per game
and getting into the opposition penalty area more often
The only thing that's gotten "worse" is the quality of shots they're allowing: 0.09 xG per shot last season
is a good thing because it's indicative of a team that's able to press much more aggressively and much higher than last year
the best is that you'll allow better chances but also fewer chances
and it'll all add up to a decline in goals conceded.)
there's just no way PSG would've been able to press Liverpool off the field like they did this season
The attack also benefits from a press that keeps the ball in the attacking third for most of every match
and from the opportunities created by winning the ball back right as the opposition is transitioning from a defensive to an attacking structure
Without Mbappé, who doesn't touch the ball a ton, is most effective in space, and likes to peel off left, the attack is also much more fluid and unpredictable. With the arrival of Khvicha Kvaratskhelia from Napoli in January
PSG now have four attackers who are devastating with the ball at their feet
but also comfortable running into space and slipping passes through the opposition backline
The effectiveness of the press, too, allows both of their fullbacks, Nuno Mendes and Achraf Hakimi
to get forward aggressively and contribute to the attack
While the various pieces of Madrid's lineup almost seem to actively negate each other
PSG's players all seem to amplify each other's strengths
While I tend to believe that talent always trumps tactics -- and while almost every great manager is on the record in agreement with me -- that's the remaining elephant in the room
When Bill Simmons outlined the Ewing Theory
he made it clear he wasn't saying that Patrick Ewing was necessarily worse than everyone thought
he was saying that specific circumstances sometimes create a situation where a team plays better without its best player
and the coaching circumstances at PSG and Real Madrid couldn't be any different
Luis Enrique is one of the most tactically demanding managers in the world -- he has a clear way that he wants his teams to play
And he's done a brilliant job at PSG so far this season
It's not a given that a bunch of ball-dominant dribble-happy wingers should work so well in an attack, but Luis Enrique has figured it out, largely by freeing up Ousmane Dembélé to play centrally and do what he does best: everything, with either foot
Meanwhile, Carlo Ancelotti said this at the beginning of last season: "I think the mistake new generation coaches make is that they give too much information about the system on the ball
I think old school coaches like me prefer not to give too much information and allow freedom for creativity."
That approach has worked brilliantly at Madrid
a club that's able to attract the best creative players in the world
Vinicius and Mbappé's instincts always take them to the same area of the field
Mbappé is still having a fantastic individual season
which values everything a player does on the ball
has Mbappé adding 10.57 goals compared to the average player at his position
That's the highest number of any player in Europe's Big Five leagues
and Liverpool's Salah (10.12) is the only other player with a number north of nine
But Madrid are simply a significantly worse team since he arrived
And the difference between Mbappé and everyone else on Madrid is that everyone else had already been a significant contributor to this team when it was the best team in the world
We've seen them do it -- we still haven't seen him do it
Mbappé is just a really hard player to fit into an elite team
he limits the approaches you can take without the ball
he's not the kind of high-touch player you can just ping the ball out to over and over again while he creates a ton of chances for himself and his teammates
You need a very specific blend of players both offensively and defensively to get the most out of Mbappé
until further notice -- until we see Mbappé actually be the best player on the best team in the world -- we're officially designating Mbappé as soccer's face of the Ewing Theory
what would be the greatest triumph for the Ewing Theory?" At the time
he suggested the 2001 Seattle Mariners winning their first-ever World Series after eventual Hall-of-Famers Randy Johnson
The Mariners won an MLB-record 116 games in 2001
but lost in the American League Championship Series to the New York Yankees
Well, PSG lost Neymar and Lionel Messi two seasons ago
and they're just three games away from winning their first-ever Champions League
Ewing argues that schools have been used to harm Black and Native people
Ewing is a self-proclaimed “school person,” a declaration she makes often in discussing her new book
Original Sins: The (Mis)education of Black and Native Children and the Construction of American Racism
and University of Chicago professor contends that the American education system has been deeply shaped by systemic prejudice and has historically favored white students and belittled their Black and Indigenous peers
she challenges readers to confront this uncomfortable truth so they can reimagine what schools could be
You have described this book as subject matter you’d felt was missing from the curriculum
Why was it so important for you to write it
I became very aware of race at a young age
You don’t have to try very hard as a kid to notice how things feel unfair or illogical — and how often those feelings fall along racial lines
I learned more about the history of schooling in the United States and read documents about the education of Indigenous and formerly enslaved people
What shocked me was the explicitness of it — to see people say very expressly in print things like “If we want to make war against Native people
it’s actually less expensive to take their kids and wage war in the classroom.” But then you also have W.E.B
Du Bois basically saying that Black people invented the idea of universal public education in the South during Reconstruction
So you have these two things in tandem: schools being integral to the freedom dream of a people
and schools used very intentionally as a source of harm and social control
I’d cobble together readings for my students to explore these ideas
Where’s the book that ties this all together
“Schools are not bystanders in the project of racial capitalism and genocide in the United States; they have been laboratories where those ideas were honed
You’ve written and spoken a lot about how schools are important places of nurturing and community building
you make a strong case that the broader education system has done irrecoverable damage
One example is how in the mid-19th century
Boston ruled to uphold segregated public schools due to the “mental and moral” differences between races
which is page after page of damning evidence about the harm schools have caused
I still consider myself a “school person.” I believe in the promise of schools because
there are also stories of incredible creativity
Schools can be sites of celebration and community strength building
How do we reimagine what these spaces could be
The book argues that there is a kind of smog in the historical record that has obscured the mistreatment of Black and Native people in schools
who celebrated Black education but also wrote that Black children “could not compare” with white ones
Why is it important to correct that record now
Schools are not bystanders in the project of racial capitalism and genocide in the United States; they have been laboratories where those ideas were honed
Correcting the record isn’t just about understanding the past
It’s important to have those facts to understand what it is that we’re actually fighting
this book is about providing a firm evidentiary basis for those who are committed to justice and want to think more critically about how schools perpetuate harm and what social transformation might look like
we’ve seen an intense political battle emerge over how American history is taught
Original Sins dives into some uncomfortable territory
like how government leaders sought to “civilize” Native children and saw schools as a means to assimilate them
How are you preparing for the inevitable backlash
I think about what Toni Morrison taught us: The function of that kind of reaction is distraction
Every moment I spend fighting those battles is a moment I’m not writing
I love real critique and discussion from people who read the book and have a shared desire to love and celebrate children
That’s a discussion I welcome and I’m excited about
Everything else I can’t really be bothered with
you note that the book is not intended to be a how-to guide for fixing schools
you hope that it starts a broader conversation
what does a “good school” look like to you
One issue is that we’ve become good at fiddling around the edges without tackling the root questions: What is a school
a good school is a place that serves as a home for imagination
and philosophical — in a way that nourishes not only students but the broader community
It could be schooling that is integrated with the natural environment or one where there are people of all different ages
It could be a place where the line between who’s a teacher and who’s a learner is a lot blurrier
It would look different for different communities
I want people to develop a sense of curiosity about the spaces they’re building together and to make those points of connection for themselves — and to question the habits we have picked up as an institution that we don’t even think about
How do we see connections between those practices and histories
Where can we be more imaginative in doing things differently
This isn’t about a step-by-step toolkit; it’s about asking deep
open-ended questions and fostering collective work toward justice and joy
What excites me is knowing that educators and community members are always doing that kind of reinvention work
Du Bois famously posed this haunting question in 1903
in his classic work The Souls of Black Folk
especially whenever I encounter one particular line from Thomas Jefferson’s Notes on the State of Virginia: “Among the blacks is misery enough
sending me spinning through my own vignettes of the Black poets who set a course for my thriving
my class took a three-day trip to Washington
Parts of Jefferson’s legacy were omnipresent in my experience of schooling

I honestly didn't know Patrick Ewing shoes were a thing
I'm telling you that if I'm ever just casually standing next to a basketball goal and hear someone say
Kelsey?" I hope it's Patrick Ewing wearing these shoes
and I hope that he immediately dunks over me and breaks the rim before looking back at me to say: "Whoops."
My bad. That fever dream is actually just that amazing old Snickers commercial that (apparently) lives rent-free in my head for the rest of my life.
Regardless, NBA legend Patrick Ewing has been selling re-released versions of the iconic shoes he wore back in the 1990s, as well as other stylish retro sneakers, since relaunching his Ewing Athletics brand in 2012
Ewing Athletics continues to thrive as the first independent basketball brand
catering to a dedicated fanbase and honoring its rich heritage," says the company's website
Featured below are two of the limited edition shoes currently on sale on the site in select sizes. Both are modified versions of Ewing Brand's 33 HI Retro sneakers.
the 33 HI is an exact retro of the 1990 original
and features a classic reversible ankle strap that can be worn on the front or back of the shoe
and a full-length PU midsole for cushioning," says the website
the Big Pun x Ewing 33 Hi uses the color scheme from Pun’s debut album Capital Punishment
it was the only album released during Big Pun’s lifetime (he tragically passed away at age 28) and was the first solo Latin hip hop record to go Platinum
"This special edition collaboration was originally released in 2018 and sold out instantly
we have brought it back for a limited time
special branding and packaging from the original 2018 edition
Everything from the leather to logos to the box has been upgraded to a premium level to complete the collector's edition feel."
47 out of 48 of the reviews for these shoes are five out of five stars
and classic design," says another.
"To honor the legacy of Hip Hop pioneers Kid 'N Play
we worked on a special 33 HI inspired by their debut album
The black-and-white sneakers feature a black wool upper accented with white leather
directly tying into the outfits Kid 'N Play rocked on the album cover
Design details include Kid 'N Play’s logo on the tongue
the number “2” replacing the signature “33” on the strap
and the word “hype” added to the heel as a nod to 2 Hype
we included colorful gradient laces for anyone in a house party mood
and they come in a commemorative box to complete the tribute to their debut album."
All 12 reviews for these sneakers are also five stars out of five
and I love the throwback to my childhood," says another.

We have recently demonstrated that genetic inactivation of EWSR1 : : FLI1 by CRISPR/Cas9
successfully blocks cell proliferation in a cell model of Ewing sarcoma
CRISPR/Cas9-mediated gene editing can exhibit off-target effects
precise regulation of Cas9 expression in target cells is essential to develop gene-editing strategies to inactivate EWSR1 : : FLI1 in Ewing sarcoma cells
we demonstrate that Cas9 can be specifically expressed in Ewing sarcoma cells when located downstream a promoter consisting of GGAA repeats and a consensus TATA box (GGAAprom)
Cas9 is selectively expressed in Ewing sarcoma cells that express EWSR1 : : FLI1 oncoproteins
but not in cells expressing wild-type FLI1
Ewing sarcoma cells infected with GGAAprom>Cas9 and a specific gRNA designed to inactivate EWSR1 : : FLI1
showed successful EWSR1 : : FLI1 inactivation and the subsequent blockade of cell proliferation
GGAAprom>Cas9 can be efficiently delivered to Ewing sarcoma cells via adenoviral vectors both in vitro and in vivo
highlighting the potential of this approach for Ewing sarcoma treatment
Our results demonstrate that the CRISPR/Cas9 machinery is safe and specific for Ewing sarcoma cells when driven under a GGAAprom
paving the way for the development of cancer gene therapies based on the selective expression of genes with therapeutic potential
One way to improve specificity could include designing a molecular strategy in which the Cas9 nuclease is expressed exclusively and specifically in Ewing sarcoma cells
This characteristic is exclusive to EWSR1 : : FLI1 and to our knowledge
no other transcription factors have been shown to have similar characteristics
which shares the same DNA-binding domain as EWSR1 : : FLI1
is not able to transactivate these GGAA repeat sequences
we show that a minimal GGAA-based promoter
which contains concatemerized GGAA repeats and a TATA-box (GGAAprom)
is able to drive high Ewing-specific expression of a gene of interest
Using Cas9 and specific sgRNAs designed to inactivate EWSR1 : : FLI1 as a therapeutic approach
we demonstrated that this strategy is effective in vitro when lentiviral and adenoviral vectors are used as delivery strategies
the use of adenovirus in vivo revealed that this strategy allows the expression of luciferase only in tumor cells but not in normal tissues
we demonstrated that adenovirus harboring Cas9 under the regulation of the GGAA promoter were able to reduce tumor growth in vivo
Our results justify additional preclinical research to advance in the development of this new gene therapy for Ewing sarcoma

the animals were first ordered from largest to smallest tumor volume
and then randomly distributed to one group or the other
Animals were treated with three intratumoral injections at 48–72 h intervals
Two independent experiments using two different doses of adenovirus were performed (1 × 109 VP/dose and 5 × 1010 VP/dose)

EWSR1 : : FLI1 induced specifically the expression of the reporter gene luciferase (mean ± SD of one experiment done by triplicate)
B Schematic representation of the different promoters used for lentiviral infections
indicating the presence or absence of GGAA repeat region and TATA-box
C A673 and MHH-ES1 Ewing sarcoma cells and HT1080 fibrosarcoma cells were infected at two different MOI with lentivirus harboring the luciferase reporter gene under the control of the promoters described in B)
Luciferase activity in stably infected cells was quantified and normalized to cells infected with an empty vector
High luciferase activity was observed in Ewing sarcoma cells infected with GGAAprom including the GGAA repeats and the TATA-box
Luciferase expression was not observed in HT1080 infected with any construct (mean ± SD of three experiments)
luciferase activity was slightly lower in A673 cells infected at an MOI of 0.5 than in the same cells infected at an MOI of 5 with GGAAprom
luciferase activity was even greater in MHH-ES1 cells than in A673 cells infected with GGAAprom
which contains a GGAA-rich region and a linked
can be efficiently activated by EWSR1 : : FLI1 in A673 and MHH-ES1 Ewing sarcoma cells but not in HT1080 cells
which express high levels of the native FLI1 protein
This characteristic makes GGAAprom particularly suitable as a specific promoter for gene therapy in Ewing sarcoma because of its high specificity for EWSR1 : : FLI1
A Cas9 expression in A673 and MHH-ES1 Ewing sarcoma cells and HT1080 fibrosarcoma cells stably transduced with GGAAprom>Cas9
GGAAless>Cas9 and EFS>Cas9 lentivirus
High levels of Cas9 expression were specifically achieved in Ewing sarcoma cells infected with GGAAprom>Cas9
B Effect of gRNA FLI-EX2 and FLI1-EX9 on cell proliferation
Stably transduced cells were additionally infected with lentivirus harboring a gRNA against FLI1 exon 2 (negative control) or a gRNA against FLI1 exon 9
which causes the genetic inactivation of EWSR1 : : FLI1
The effect on cell proliferation was monitored for 7 days
Cas9 expression combined with gRNA FLI1-EX9 blocked cell proliferation only in Ewing sarcoma cells (mean ± SD of a representative experiment from a total of three; ****P < 0.0001
sgRNA directed against FLI1 exon 2 can be used as a control sgRNA in Ewing sarcoma cells because this FLI1 exon is not included in the EWSR1 : : FLI1 fusion
and Ewing sarcoma cells do not express native FLI1; thus
editing at this exon is not deleterious for Ewing sarcoma cells
an RNA guide targeting FLI1 exon 9 affects the DNA-binding domain of FLI1
which is an essential region for EWSR1 : : FLI1 function
no effects on cell proliferation were observed when GGAAprom>Cas9
or EFS>Cas9 HT1080 cells were infected with FLI1 exon 2 or exon 9 gRNAs
These results demonstrate that the combination of Cas9 under the control of an Ewing-specific promoter (GGAAprom) and a gRNA targeting exon 9 of FLI1 is sufficiently efficient to block cell proliferation in Ewing sarcoma cells
while lentiviral vectors are excellent proof-of-concept tools
the use of a lentiviral-based approach to deliver gene therapy to Ewing sarcoma cells is not suitable for clinical development
we decided to explore the use of adenovirus as a suitable alternative to deliver gene therapy to Ewing sarcoma cells
Although adenoviruses have several drawbacks
they remain one of the most commonly used viral vectors in cancer gene therapy
with higher transduction efficiencies observed at higher MOI
A Luciferase activity in cells infected with adenovirus (MOI = 100) harboring a constitutive promoter (EF1A > LUC2) or the Ewing specific promoter (GGAAprom>LUC2)
GGAAprom was only active in Ewing sarcoma cells (red bars) (mean ± SD of a representative experiment from a total of three)
Ad EF1A > LUC2 (n = 4) or Ad GGAAprom>LUC2
and luciferase activity quantified by IVIS imaging 72 hours post-injection
No luciferase activity was observed at all in mice injected i.v
C Quantification of luciferase activity in tissue extracts
Twelve different tissues were extracted from each animal and the corresponding protein lysates were analyzed for luciferase activity
Each point represents the luciferase activity obtained in one animal (mean ± SEM)
D Luciferase expression in tumors injected i.t
Luciferase activity was detected in Ewing sarcoma tumors injected with Ad-EF1A > LUC2 (constitutive expression) and Ad-GGAAprom>LUC2 (Ewing-specific expression)
Luciferase activity in tumor tissue lysates is also shown
Each point represents the luciferase activity obtained in one tumor (mean ± SEM)
These findings strongly support that gene therapy based on the expression of a therapeutic gene under the control of the GGAAprom could represent a therapeutic strategy with remarkable specificity for Ewing sarcoma cells
without impacting normal cells of the organism
tumors injected with either Ad-EF1A > LUC2 or Ad-GGAAprom>LUC2 exhibited high levels of luciferase activity
These results indicate that Ad-GGAAprom>LUC2 is able to infect Ewing sarcoma cells in vivo and efficiently induce the expression of a gene of interest upon reaching the target tissue
A Effect of Ad-GGAAprom>Cas9/gRNA EX2 and EX9 on cell proliferation
SK-N-MC and HT-1080 cells were infected with the mentioned adenovirus (MOI = 25)
and cell proliferation was monitored by cell counting
Cell proliferation was severely impaired in A673 and SK-N-MC Ewing sarcoma cells infected with Ad-GGAAprom>Cas9/gRNA EX9 (mean ± SD of a representative experiment from a total of three
and EWSR1 : : FLI1 target genes CD44 and NR0B1 following adenovirus infection (MOI = 100)
Treatment with Ad-GGAAprom>Cas9/gRNA EX9 dramatically reduced EWSR1 : : FLI1 expression
concomitantly affecting the expression of EWSR1 : : FLI1 target genes CD44 and NR0B1
C Effect of adenovirus infection on cell morphology
with A673 cells and split into two groups on day 19
The group treated with adenovirus received intratumoral injections of Ad-GGAA>Cas9/gRNA EX9 at days 19
Control group received intratumoral PBS (n = 5)
Tumor volume and tumor volume normalized to day 19 (start of treatment) for each animal are showed
adenovirus treatment (5 × 1010 VP/dose) was started on day 9 (n = 8) and administered at days 10
Control group received intratumoral PBS as above (n = 7)
Tumor volume and tumor volume normalized to day 9 (start of treatment) for each animal are showed
A significant reduction in tumor growth was observed in the treated group (*P < 0.05; two-way ANOVA)
and gene editing was quantified by ultra-deep next generation sequencing
Significant gene editing was observed in animals treated with 5 × 1010 VP/dose (mean ± SEM; ****P < 0.0001
A Tumor sections were stained with an anti-Ki67 antibody to assess cell proliferation
Graph shows percentage of Ki67+ cells in control animals (PBS
n = 5) and animals treated with 5 × 1010 VP/dose of Ad-GGAAprom>Cas9/gRNA EX9 (Ad-Cas9-EX9
B Representative tumors for each group stained with Ki67 antibody (bar = 100 µm)
Ewing sarcoma is a devastating rare tumor affecting children and young adults
for which the development of new therapeutic strategies is imperative
Ewing sarcoma is characterized by tumor-specific chromosomal translocations that produce tumor-specific fusion proteins
such as the oncogenic chimeric transcription factor EWSR1 : : FLI1
Ewing sarcoma cells are addicted to these EWSR1 : : FLI1 fusion oncoproteins; thus
any approach aimed at blocking or inactivating EWSR1 : : FLI1 is expected to interfere with tumor growth
Another possible strategy to interfere with EWSR1 : : FLI1 function is the use of siRNAs to knock down EWSR1 : : FLI1 expression
This strategy has been extraordinarily successful for the generation of cell models of EWSR1 : : FLI1 knockdown
which have been very helpful for understanding the biology of Ewing sarcoma
potential therapies based on the use of siRNAs designed to knockdown EWSR1 : : FLI1 mRNAs have the same drawbacks as those described above concerning the use of small molecules designed to block EWSR1 : : FLI1 function
to achieve the best EWSR1 : : FLI1 inactivation
genetic inactivation of the EWSR1 : : FLI1 gene
exquisite control of Cas9 expression must be achieved to rule out any unwanted effects on normal cells
we demonstrate that this precise and exquisite control could be achieved by using an Ewing-specific promoter formed by concatenated repeats of the tetranucleotide GGAA (GGAAprom)
the use of a consensus TATA box downstream of the GGAA repeats increased the activity of the promoter by 2–3 times compared to that of the GGAA repeats only
The high specificity of this DNA sequence for Ewing sarcoma cells was demonstrated both in vitro and in vivo
GGAAprom was active in Ewing sarcoma cells expressing EWSR1 : : FLI1 proteins (A673
which encompass three different EWSR1 : : FLI1 isoforms) but not in non-Ewing sarcoma cells such as the fibrosarcoma cell line HT1080 or the osteosarcoma cell lines U2-OS and Saos-2
The absence of GGAAprom activation in HT1080 cells is particularly relevant because these cells express high levels of native FLI1
indicating that GGAAprom is only active in Ewing sarcoma cells expressing EWSR1 : : FLI1
This important finding was soundly confirmed in in vivo experiments
when athymic nude mice were injected intravenously with adenovirus harboring a luciferase gene under the control of GGAAprom
no luciferase expression in any of the 12 tissues analyzed was detected
This means that a gene therapy in which the therapeutic gene is under the control of the Ewing-specific promoter will be active only in Ewing sarcoma cells expressing EWSR1 : : FLI1
This opens the possibility of using this type of approach with a multitude of therapeutic genes that may include not only nucleases such as the one proposed in this work to inactivate EWSR1 : : FLI1 but also genes capable of inducing cell death (for example
suicide genes) or immunomodulatory genes capable of activating an antitumor immune response
still one or two orders of magnitude higher than that used in our preclinical animal studies
This indicates that there is still room to use higher doses that would have yielded even more remarkable results
further experiments with higher doses were not possible due to the limitation in the amount of adenovirus produced
the authors use a complex strategy for in vivo delivery based on the use of lentiviral vectors and an adapter antibody that
can make more difficult its preclinical development
Although in vivo gene editing certainly presents numerous risks that must be carefully considered
the development of highly specific expression strategies such as the one presented in this work opens new opportunities to permanently inactivate the EWSR1 : : FLI1 fusion gene characteristic of Ewing sarcoma
we reported a gene therapy approach to permanently inactivate EWSR1 : : FLI1 in Ewing sarcoma cells in a highly specific way
no other approach has been shown to inactivate so effectively
We believe the results shown here support the use of this strategy to treat Ewing sarcoma and justify continuing its preclinical development

Jacob Ewing is a senior in engineering management with an emphasis and minor in explosives engineering and leads the cadets in S&T’s Army ROTC Stonehenge Battalion
Jacob Ewing is determined to become a successful leader once he graduates from Missouri S&T. Through academic excellence and leading cadets in S&T’s Army ROTC Stonehenge Battalion
“I’m involved in basically every opportunity that you can be involved in,” Ewing says
“I just have this philosophy: I always want to be better tomorrow than I am today
He will graduate twice this year: earning a bachelor’s degree in May and a master’s degree in December.
Both of Ewing’s grandfathers were military officers (Army and Air Force)
Ewing says they passed away before seeing him choose the military
Ewing’s path to the military has certainly been a busy one.
Ewing’s first college year began with glowing success. Along with getting involved in the ROTC color group, the cannon crew and various campus clubs, Ewing excelled in the Ranger Challenge
a military skills competition for the nation’s military academies and ROTC programs.
The final competition, the Sandhurst Military Skills Competition comprises the top 16 ROTC teams in the nation
plus 15 international teams and military academies
The task force and brigade competitions determine which 16 ROTC teams advance to Sandhurst
Ewing’s team won the task force competition and placed second in the brigade competition
held at the United States Military Academy at West Point
“We became the first school in Missouri to make it to Sandhurst,” Ewing says
“It was the first time in our school’s history that we ever made it to an international competition of that caliber.”
Ewing’s summer following his first year of college was just as busy. Ewing completed an internship with the United States Army Corps of Engineers, where he worked at Fort Leonard Wood on a hospital replacement project.
“I worked as a quality control and quality assurance engineer with Major Steven Lanni and saw how the Army Corps of Engineers operates in the engineering regiment itself,” Ewing says.
Sophomore year: Developing solutions
In addition to serving as the chair of a national scholarship committee
Ewing became more involved during his sophomore year as a team leader within S&T’s Stonehenge Battalion
By earning the highest combat fitness test score at S&T
he achieved the role of master fitness trainer
in which he developed personal training plans and curriculum for S&T cadets.
He became involved in the recruiting committee
and even developed a software program that helped recruit more cadets.
“That software I helped develop — that’s some of the knowledge I got from my degree that really helped our program,” Ewing says
“It basically automated our process that we already did but just streamlined it and made it more efficient
so we didn’t have to do it the old way with a pencil.”
In recognition of his service to the campus and community
named for a professor of engineering management at Missouri S&T for over 30 years.
he completed a course at the University of North Carolina through Project Global Officer to learn Mandarin Chinese in just two months.
He learned how to safely jump out of an airplane wearing a parachute.
“I did jump school because one of my biggest fears is heights
but how I’ve defined myself is I want to do the things that I’m the most scared of
That was my way of conquering what I was most afraid of.”
he became captain of the Ranger Challenge team and a platoon sergeant within the battalion.
Year three is primarily preparing students to get ready for cadet summer training
Ewing joined Missouri S&T’s eSports club and became an officer in the American Society for Engineering Management (ASEM)
Myers Scholarship to help determine the next recipient.
Ewing attended cadet summer training in Fort Knox
He was evaluated on individual military skills and his ability to lead a platoon of 40 cadets through a rigorous field exercise
1 out of 640 cadets that participated in his regiment
This exceptional performance contributed to his ranking as No
13 out of 5,140 cadets in his graduation year group from across the nation.
From there Ewing flew to Fort Lewis in Washington to complete another internship in the 46th Aviation Support Battalion
He worked with the Multi-Domain Effects Battalion
“I was learning how to be an officer in a real-life environment,” Ewing says
learning signal operations such as how we secure our communication networks across the Pacific Ocean.”
Ewing became the cadet battalion commander
the highest officer position available for a cadet — and leads 56 cadets.
“Being a battalion commander basically means that you assume all responsibility,” he says
“Any training that we conduct is 100% on me to make that decision.”
Ewing explained that he thinks of big ideas for the battalion
then ensures those ideas happen by bringing them to his staff
who then push it all the way down to each leader.
“The major reason why I like leading is developing others and seeing other people succeed,” Ewing says
“That’s been my primary motivator for ROTC and being a leader within the organization.”
Ewing said his senior year has been especially validating — this is the year they get back all of their results
everything a student does in their first three years is tabbed and put into an order of merit score
then an order of merit list where they get stacked against all 6,000 cadets across America.
11 in the entire country out of all 6,000 cadets that competed in my year group,” Ewing says.
he had the benefit of choosing where his future path goes
he was officially awarded Cyber (the US Army Cyber Command) as his branch
“It’s a cool honor to have the hard work over the last four years awarded,” Ewing says
“I would enjoy going to work at the National Security Agency
or to work for a multi-domain task force protecting national security for the country
the spearhead of the future of the military
is where I want to put myself because while they are one of the most rigorous and time-consuming jobs; they are the future.”
Ewing said not only did S&T’s ROTC program help lead him toward his future in leadership
but his academic programs also played a major part
ROTC was the thing that glued all of it together and connected all the dots
I’ll take that experience that I learned about being technical
and take it into the military as a leader.”
teaching professor and associate chair for undergraduate studies in engineering management and systems engineering
was a pivotal part of Ewing’s success as his advisor
She noticed his leadership potential early on.
regardless of whether he stays in the Army or whether he transfers into the civilian world
because that’s just who he is,” Schuman says
“He’s not one of those leaders that comes in and tells everybody what to do
Schuman says problem-solving and critical thinking are the top skills the program focuses on.
“Our program is a little different from other engineering programs,” Schuman says
“We try to look at the whole picture and how everything fits together
I think that we’ve provided that for him
and I think the Army ROTC has also provided a different piece of that.”
says he’s proud of how much Ewing has grown as a leader in the past few years.
“I think one of the coolest things about him is he’s not just really good at academics
and he does it all effortlessly,” Weir says
“But the biggest thing for me is that he does it with empathy
and he cares for his soldiers and his people
I think that truly will help him rise to the top.”
former professor of military science at S&T
he gave Ewing a very meaningful review.
“The best cadet I have observed in five years in the U.S
and his extra effort is demonstrated by his top accomplishment marks…(he’s) an exceptional young leader who will make a difference in our Army.”
“The part where he said I was the best cadet he had seen in five years in the cadet command stands out as the greatest remark anyone has ever said about me,” Ewing says
“He had worked in ROTC for five years and saw thousands of cadets
which makes that comment very touching … It really showed me that all my hard work paid off and motivated me to continue doing the best I could.”
Ewing plans to become an officer in the Army’s Cyber branch and hopes that his influence as a leader will help develop others.
“I’ll never define my success as an Army officer based on my personal achievements
but on the achievements of those I developed and led,” Ewing says
then I will define my career as having been successful.”

Beverly Daniel Tatum is a New York Times bestselling author and award-winning psychologist
DECATUR, Ill. – Noted psychologist and New York Times bestselling author Dr. Beverly Daniel Tatum took the stage at Millikin University’s Albert Taylor Theatre on Wednesday, February 26, for the Spring 2025 Thomas W. Ewing Lecture
sharing her message about the importance of embracing discussions about race and identifying the struggles that many have in the United States to engage in that conversation.
and Other Conversations in an Era of School Resegregation.” Her presentation for the Ewing Lecture was titled “Closing the Gap: Embracing Cross-Racial Dialogue."
Tatum shared that many white people listen to discussions on racial topics but are often uncomfortable sharing their thoughts and experiences
asking if they had experienced a racially centered event as a child and if they were able to speak about it with an adult afterward
A portion of the audience hadn’t had a follow-up conversation
and Tatum argued that not having those conversations strengthens the idea that some racial topics are not for some people.
“I've had many conversations about race; I do it professionally
If I'm conversing with someone who's never done it
It may feel like it's not a peer-to-peer conversation
but it might feel more like a teacher and a learner,” Tatum said
“I have a friend who has an expression that I love: There's a teacher and a learner in every student
Even if I'm talking to someone who hasn't had as much practice as me
I still have something to learn from that person
white people in cross-racial dialogues think they don't have anything to share
and they enter into that conversation only wanting to listen
From the point of view of the students of color
if we're talking about a topic that is important to me as a student of color
and the white students in my class are not saying anything
that makes me feel like they don't care and aren't interested
Tatum served as President of Spelman College from 2002 to 2015
and her upcoming book to be released September 2
“Peril and Promise: College Leadership in Turbulent Times,” delves into the challenges that higher education faces while also acknowledging that colleges also provide the solutions to the divisions that disrupt our society.
many people say that our society feels highly polarized
you might have voted for somebody different than me
but I'm willing to spend the next 15 weeks meeting (in a class) with you and others to listen and share,” Tatum said
You are learning something you didn't know before
you will begin to get sick of this subject
“That's what I'm calling the "Argument of Discomfort." If you weren't required to attend
You get to the point where you're able to talk about it in the company of people you've come to trust because you've been sitting with them for several weeks
and visiting lecturers are selected based on their prominence in those fields and ability to inspire others to serve

respectively) and will now head to High-A Brooklyn
1 BB in eight innings) have been promoted from High-A Brooklyn to Double-A Binghamton
against the Worcester Red Sox this past week
but they rallied to win the final three games
including the series finale in which they won by an astounding score of 20-12
Singleton batted .360 for the series and had an OPS of 1.067
Billy McKinney also had a solid series
The former major league Met has not had a strong start to the season but could look to build off the momentum of this last series
Baty naturally made an impact in his first game
The best start from a starting pitcher in this series came from Brandon Waddell
It wasn’t a particularly strong start
as he allowed two runs (one earned) in just four innings
Waddell owns a 1.54 ERA through five starts
Kevin Herget made an impression out of the bullpen
tossing four scoreless innings over two appearances
The veteran owns a 3.72 ERA through nine appearances
Photo by Bronson Harris of Binghamton Rumble Ponies
the Binghamton Rumble Ponies also had to play seven games this past week
They had more success than their Triple-A friends
including the first four in the series against the New Hampshire Fisher Cats
The series win has the Rumble Ponies at 12-8 on the season
Ryan Clifford had an up-and-down week
but three of his four hits went for extra bases (two home runs and a double)
Clifford has a low average and has struck out 25 times in 62 at-bats this season
but he’s raised his OPS to .786 as he looks to get going
Nick Morabito had a strong week
and a home run with three RBIs and two stolen bases
The speedster had been off to a slow start in his first taste of Double-A
Jonah Tong had an excellent week on the mound
displaying the swing-and-miss stuff that made him a breakout prospect a year ago
Tong made two starts and allowed two runs (one earned) in 8 1/3 innings while striking out a ridiculous 19 batters
Tong lowered his ERA this year to 3.63 and now has 30 strikeouts in 17 1/3 innings
Zach Thornton and Ryan Lambert will join this productive pitching staff following their promotions
The Brooklyn Cyclones played just five games this week
The Cyclones won the series against the Jersey Shore BlueClaws three games to two
dropping the series opener and finale but winning the middle three games
Carson Benge and Eli Serrano III
two of the Mets’ highly touted 2024 draft picks
Benge went 6-for-19 with a home run and four RBIs
Serrano has fared better than Benge in the early going
owning an OPS over .100 points higher (.826 to .725)
Estarling Mercado also continued his fast start to the year
He had just a .665 OPS between Low-A and High-A ball in 2024
R.J. Gordon
struck out nine and allowed just one run in five innings during his start against Jersey Shore
who made his first pro ball appearance this year
has a 2.70 ERA through four appearances with Brooklyn
Díaz allowed just two hits and struck out five
while Hall allowed seven hits and two walks with four strikeouts
Both pitchers have been fantastic this year
with Díaz not allowing a run through his first 15 2/3 innings
while Hall owns a 1.45 ERA through four starts
and Chandler Marsh will get their first taste of baseball above the Low-A level
Lucie Mets took four of six games from the Daytona Tortugas this past week
It was a great week for the pitching staff
who allowed more than three runs in just two of the six games
and four steals in just five games against Daytona
Kevin Villavicencio also had an excellent week
though just one (a double) went for extra bases
and he now has a .394 average and .945 OPS in nine games with St
Nate Dohm made two excellent starts in the series
and he also allowed six hits and three walks
Edgar Moreta held the Tortugas scoreless in his start
going five innings and allowing just two hits while striking out three
Moreta has struggled and owns a 4.97 ERA through four appearances
Perhaps the most interesting start in St. Lucie this past week came from Raimon Gomez
who is not just one of the hardest throwers in the Mets’ system but in all of professional baseball
but the most notable stat from his start was that he reached a ridiculous 104.5 miles-per-hour with his fastball in the first inning
AVG – Marco Vargas (STL)/Kevin Villavicencio (STL/BRK) – .409
SO – Blade Tidwell (SYR) – 37
giving the Mets no choice but to promote him
Ewing will likely hit an adjustment period heading to Brooklyn
but his development has been a pleasant one so far for the Mets
He hadn’t been off to the best start to the year
but his two starts in this last series cemented why the Mets are so excited about his potential
Tong is still just 21 years old and will look to continue to establish himself as one of the best pitchers on the Mets’ farm in 2025
